A genetic variant study of bortezomib-induced peripheral neuropathy in Chinese multiple myeloma patients

Background:Bortezomib results in peripheral neuropathy(PN)in approximately 50%of patients,during multiple myeloma(MM)treatment,a complication known as Bortezomib-induced peripheral neuropathy(BIPN).The drug response varies among individuals.Genetic factor may play an important role in BIPN.Methods:A nextgeneration sequencing(NGS)panel containing 1659 targets from 233 genes was used to identify risk variants for developing BIPN in 204 MM patients who received bortezomib therapy.mRNA expression of MTHFR and ALDH1A1 in 62 peripheral blood samples was detected by real-time quantitative PCR(RT-qPCR).Serum homocysteine(Hcy)levels were detected in 40 samples by chemiluminescent microparticle immunoassay(CMIA).Results:Compared with the non-BIPN group(n=89),a total of 8 significantly associated single nucleotide polymorphisms(SNPs)were identified in the BIPN group(n=115):MTHFR(rs1801131,rs1801133,rs17421511),EPHX1(rs1051740),MME(rs2016848),ALDH1A1(rs6151031),HTR7(rs1935349)and CYP2A6(rs8192720).The mRNA expression level of MTHFR in newly diagnosed patients with peripheral neuritis after treatment(NP group)was lower than that of newly diagnosed patients without peripheral neuritis after treatment(NnP group)(1.70±0.77 vs.2.81±0.97,p=0.009).Serum Hcy levels were significantly higher in BIPN group than in non-BIPN group(11.66±1.79μmol/L vs.8.52±3.29μmol/L,p=0.016)and healthy controls(11.66±1.79μmol/L vs.8.55±2.13μmol/L,p≤0.001).Conclusion:CYP2A6,EPHX1,MTHFR,ALDH1A1,HTR7,MME and BIPN are linked in Chinese MM patients.BIPN is more likely to occur in patients with lower MTHFR mRNA expression,which might result in higher serum Hcy levels.

Prediction of the thermal conductivity of Mg–Al–La alloys by CALPHAD method

Mg-Al alloys have excellent strength and ductility but relatively low thermal conductivity due to Al addition.The accurate prediction of thermal conductivity is a prerequisite for designing Mg-Al alloys with high thermal conductivity.Thus,databases for predicting temperature-and composition-dependent thermal conductivities must be established.In this study,Mg-Al-La alloys with different contents of Al2La,Al3La,and Al11La3phases and solid solubility of Al in the α-Mg phase were designed.The influence of the second phase(s) and Al solid solubility on thermal conductivity was investigated.Experimental results revealed a second phase transformation from Al_(2)La to Al_(3)La and further to Al_(11)La_(3)with the increasing Al content at a constant La amount.The degree of the negative effect of the second phase(s) on thermal diffusivity followed the sequence of Al2La>Al3La>Al_(11)La_(3).Compared with the second phase,an increase in the solid solubility of Al in α-Mg remarkably reduced the thermal conductivity.On the basis of the experimental data,a database of the reciprocal thermal diffusivity of the Mg-Al-La system was established by calculation of the phase diagram (CALPHAD)method.With a standard error of±1.2 W/(m·K),the predicted results were in good agreement with the experimental data.The established database can be used to design Mg-Al alloys with high thermal conductivity and provide valuable guidance for expanding their application prospects.

Effect of Blood Circulation in Veins on Resonance Suppression of the Dragonfly Wing Constructed by Numerical Method

To reveal the resonance suppression mechanism of the blood circulation in dragonfly wings,a numerical modeling method of dragonfly wings based on Voronoi diagrams is proposed,and the changes in mass,aerodynamic damping,and natural frequencies caused by blood circulation in veins are investigated.The equivalent mass of blood,boundary conditions,and aerodynamic damping are calculated theoretically.Modal analysis and harmonic response analysis of wing models with different blood circulation paths are performed using the finite-element method(FEM).The vibration reduction ratioδis introduced to compare the damping efficiency of different mass regions.Finally,a natural frequency testing device is constructed to measure the natural frequencies of dragonfly wings.The results indicate that the shape,mass,and natural frequencies of the dragonfly wing model constructed by numerical method agree well with reality.The mass distribution on the wing can be altered by blood circulation,thereby adjusting the natural frequencies and achieving resonance suppression.The highestδof 1.013 is observed in the C region when blood circulates solely in secondary veins,but it is still lower than theδof 1.017 when blood circulates in complete veins.The aerodynamic damping ratio(1.19–1.79%)should not be neglected in the vibration analysis of the beating wing.

Sequential pyrolysis for understanding specific influence of cellulose- and lignin-derived volatiles on properties of counterpart char

Interactions of cellulose-and lignin-derived intermediates have been well documented during pyrolysis of lignocellulosic biomass.The reaction network for the interactions is rather complex,as cellulose-derived volatiles could interact/react with not only lignin-derived volatiles but also lignin-derived char and vice versa.To probe specifically the impacts of cellulose-derived volatiles on the lignin-derived char or the opposite,herein the sequential pyrolysis was performed by arranging cellulose in the upper bed with lignin in the lower bed or lignin above with cellulose below at 350 and 650℃,respectively.The results indicated that the sequential pyrolysis of cellulose→lignin or lignin→cellulose at 350℃induced increased char yield from formation of carbonaceous deposits via volatiles-char interactions.Compared with the lignin-derived volatiles,the cellulose-derived volatiles,especially aldehyde fractions,were more reactive towards the lignin-derived char at 350℃,forming oxygen-rich lignin-derived char with a higher yield,an abundance of aliphatic structures and consequently lower thermal stability.In sequential pyrolysis of lignin→cellulose,more aromatics-rich species were deposited on cellulose-derived char,but the lignin-derived volatiles were less reactive for enhancing the char yield.At 650℃,instead of polymerisation of the volatiles on the char,either the cellulose-or lignin-derived char catalyzed the cracking of the counterpart volatiles to remove the aliphatic functionalities,which made the char more aromatic and thermally more stable.

Matching Algorithms of Minimum Input Selection for Structural Controllability Based on Semi-Tensor Product of Matrices

In 2011,Liu,et al.investigated the structural controllability of directed networks.They proved that the minimum number of input signals,driver nodes,can be determined by seeking a maximum matching in the directed network.Thus,the algorithm for seeking a maximum matching is the key to solving the structural controllability problem of directed networks.In this study,the authors provide algebraic expressions for matchings and maximum matchings proposed by Liu,et al.(2011)via a new matrix product called semi-tensor product,based on which the corresponding algorithms are established to seek matchings and maximum matchings in digraphs,which make determining the number of driver nodes tractable in computer.In addition,according to the proposed algorithm,the authors also construct an algorithm to distinguish critical arcs,redundant arcs and ordinary arcs of the directed network,which plays an important role in studying the robust control problem.An example of a small network from Liu’s paper is used for algorithm verification.

Identification and fine mapping of qSW2 for leaf slow wilting in soybean

Drought is one of the abiotic stresses limiting the production of soybean(Glycine max).Elucidation of the genetic and molecular basis of the slow-wilting(SW)trait of this crop offers the prospect of its genetic improvement.A panel of 188 accessions and a set of recombinant inbred lines produced from a cross between cultivars Liaodou 14 and Liaodou 21 were used to identify quantitative-trait loci(QTL)associated with SW.Plants were genotyped by Specific-locus amplified fragment sequencing and seedling leaf wilting was assessed under three water-stress treatments.A genome-wide association study identified 26 SW-associated single-nucleotide polymorphisms(SNPs),including three located in a 248-kb linkage-disequilibrium(LD)block on chromosome 2.Linkage mapping revealed a major-effect QTL,qSW2,associated with all three treatments and adjacent to the LD block.Fine mapping in a BC_(2)F_(3) population derived from a backcross between Liaodou 21 and R26 confined qSW2 to a 60-kb interval.Gene expression and sequence variation analysis identified the gene Glyma.02 g218100,encoding an auxin transcription factor,as a candidate gene for qSW2.Our results will contribute significantly to improving drought-resistant soybean cultivars by providing genetic information and resources.

The cross-dimensional dynamical systems have received increasing research attention in recent years. This paper characterizes the structure features of the cross-dimensional vector space.Specifically, it is proved that the completion of cross-dimensional vector space is an infinite-dimensional separable Hilbert space. Hence, it means that one can isometrically and linearly embed the crossdimensional vector space into the ?2, which is known as the space of square summable sequences.This result will be helpful in the modeling and analyzing the dynamics of cross-dimensional dynamical systems.

Physical Vapor Deposited(PVD)TiAlN coatings are extensively utilized as protective layers for cutting tools,renowned for their excellent comprehensive performance.To optimize quality control of TiAlN coatings for cutting tools,a multi-scale simulation approach is proposed that encompasses the microstructure evolution of coatings considering the entire preparation and service lifecycle of PVD TiAlN coatings.This scheme employs phase-field simulation to capture the essential microstructure of the PVD-prepared TiAlN coatings.Moreover,cutting simulation is used to determine the service temperature experienced during cutting processes at varying rates.Cahn-Hilliard modeling is finally utilized to consume the microstructure and service condition data to acquaint the microstructure evolution of TiAlN coatings throughout the cutting processes.This methodology effectively establishes a correlation between service temperature and its impact on the microstructure evolution of TiAlN coatings.It is expected that the present multi-scale numerical simulation approach will provide innovative strategies for assisting property design and lifespan prediction of TiAlN coatings.
