T-type calcium channels(Cav3.1,Cav3.2 and Cav3.3) make one of the families of the voltage-gated calcium channels(VGCC).T-type channels are distinct from other VGCC types in that they are activated at very negative voltages(activation threshold near-60 mV) Thus,in excitable cells T-type channel activity contributes to the control of excitability by setting up an action potential firing *** evidence indicates that the T-type calcium channels are expressed in the receptive fields of peripheral sensory neurons,suggesting that they may play an important role in *** until now the mechanism of the regulation of the T-type calcium channels are still largely *** investigated regulation of T-type Ca channels by inflammatory *** application of bradykinin(200 nM) did not significantly affected the amplitude of currents though Cav3.2 heterologousely erexpressed in HEK293 cells,but induced a leftward shift the channel voltage *** inflammatory mediator,substance P(10 uM) acutely inhibited CaV3.2 current in HEK293 cells and the native T-type Ca current in cultured DRG neurons by 30.2%±6.8%(n = 11,P<0.05) and by 25.9%±5.3%(n = 11,P<0.05) *** inhibition was not reversible upon washout but was completely reversed by the application of reducing agent DTT(1mM) suggesting that SP effect on T-type channels is mediated by the oxidative *** support of this conclusion substitution of redox sensitive histidine at the position 191 in Cav3.2 by glutamine completely abohshed SP-induced *** further experiments we found that pretreatment of cultured rat DRG neurons with a cocktail of inflammatory mediators(bradykinin,50 nM;ATP,2 uM;prostaglandin E2,0.5 μM;norepinephrine,0.5 μM) overnight strongly increased the T-type Ca current density(from 64.1±7.4 pA to 105.6 ± 16.5 pA,n=28 and n = 37,P<0.05)and a proportion of neurons which express T-type Ca current(from 28/63 to 37/63).Our results put forward ne

Ca -activate chloride current(CACC) plays important physiological roles in different tissues including neuronal *** is not clear whether there is specificity in CACC activation by Ca from different sources,namely Ca influx through voltage-/ligand-gated Ca channels in the plasma membrane,vs Ca release from intracellular *** this study we investigated the activation of CACC by intracellular Ca induced by the membrane depolarization in rat DRG *** whole-cell patch recordings were performed on DRG neurons from 7-day-old SD *** the condition of regular external(high[NaCl]) and internal(high[KC1]) solution,brief depolarization evoked a large inward tail current at -80 *** depolarization-induced tail current selectively appeared in a subset of medium-diameter DRG neurons and could be partially blocked by chloride current inhibiters DIDS and NFA,indicating a possibly component of *** tail current could also be blocked by Ca free external solution,pretreatment with BAPTA-AM,and NiCl,indicating Ca influx is essential for the activation of *** fura-2AM-loaded neurons,brief depolarization range from -30 to +90 mV induced a depolarization-dependent increase of intracellular Ca. However it seems that the depolarization-induced CACC activation and intracellular Ca increase is not merely a result of opening of voltage-dependent Ca *** Na and internal K also affected the depolarization-induced CACC activation and intracellular Ca *** the membrane depolarization-induced activation of CACC may use intracellular Cathrough Ca channels as well as transporters.

Membrane phosphatidylinostol 4,5-bisphosphates(PtdIns(4,5)P2,PIP2) is fundamentally important for maintaining function of a large number of ion channels and *** showed in our previous study that membrane depolarization induced elevation of membrane phosphatidylinostol 4,5-bisphosphates(Ptdlns(4,5) P2,PIP2) and subsequently increased KCNQ2/Q3 currents expressed in Xenopus oocytes through increased activity of PI4 *** underlying mechanism for this depolarization-induced enhancement of PIP2 synthesis was further *** present results indicated that activation of protein kinase C(PKC),specifically activation of PKC isozymesβⅡwas responsible for the enhanced PIP *** found phorbol-12-myristate,13-acetate(PMA),an activator of PKC,mimicked effects of the membrane depolarization by increasing KCNQ2/Q3 activity,elevating membrane PIP level and increasing activity of PI4 *** the other hand,the membrane depolarization enhanced PKC *** effects of both depolarization and PMA were blocked by PKC inhibitor or PI4K RNA *** results demonstrated that the depolarization selectively activated PKCβⅡisoform and enhanced the PKC interaction with PI4 *** results reveal that depolarization-induced elevation of membrane PIP2 is through activation of PKC and subsequently increased activity of PI4 kinase.

Charge-exchange(CX) recombination spectroscopy is a powerful tool monitoring ion temperature and plasma rotation with good temporal and spatial resolutions. A compact, new design for a high-throughput, tri-band high spectral resolution spectrometer has been developed for the charge-exchange recombination spectroscopy measurement on the hl-2A tokamak. The simultaneous measurements of He II(468.57 nm), C VI(529.1 nm), and Dα(656.1 nm accompanied by beam emission spectra) with an acquisition frequency up to 400 Hz are achieved by vertically binning the spectrum from each fiber in experiments. Initial results indicate that the system can provide radial profiles of not only ion temperature and rotation velocity,but also concentration of carbon. For the case of helium, the measurements for the ion temperature and rotation velocity are straightforward but the apparent concentration associated with the observed CX intensity is obviously too high. Modeling of the active He II CX feature including plume contributions needs to be carried out to extract the true helium *** spectrometer could become a prototype for the ITER charge-exchange recombination spectroscopy diagnostic and the pilot experiments, as presented here, demonstrate the possibility of impurity concentrations measurements based on the combined measurement of local beam emission and charge-exchange recombination spectroscopy spectra.

Fast neutron flux measurements with high count rates and high time resolution have important applications in equipment such as *** this study,real-time neutron and gamma discrimination was implemented on a self-developed 500-Msps,12-bit digitizer,and the neutron and gamma spectra were calculated directly on an FPGA.A fast neutron flux measurement system with BC-501A and EJ-309 liquid scintillator detectors was developed and a fast neutron measurement experiment was successfully performed on the hl-2 M tokamak at the Southwestern Institute of Physics,*** experimental results demonstrated that the system obtained the neutron and gamma spectra with a time accuracy of 1 *** count rates of up to 1 Mcps,the figure of merit was greater than 1.05 for energies between 50 keV and 2.8 MeV.

在hl-3装置1MA电流等离子体欧姆放电实验期间,米尔诺夫(Mirnov)探针测量到两种高频磁流体(MHD)不稳定性。在等离子体启动阶段,电流爬升受到抑制,碘化钠探测器测量到逃逸电子增加。同时Mirnov探针测量到多分支频率f为20~400k Hz的高频MHD不稳定性。结果表明,这种不稳定性的出现与逃逸电子增加密切相关,并抑制了等离子体电流的爬升,损失了大约7%伏秒数。等离子体电流和密度爬升时,Mirnov探针测量到频率f为110~160k Hz的高频MHD不稳定性。这种不稳定性频率与芯部阿尔芬频率成正比,处于环向阿尔芬本征模频率范围。随着这种不稳定性的演化,等离子体发生了破裂。这些高频MHD不稳定性的研究对于hl-3后续放电及未来ITER的初始放电有重要的参考意义。
