Our prior investigations have established that Inonotus obliquus(Chaga)possesses hypoglycemic *** hyperglycemia is known to precipitate renal function *** functionality of the kidneys is intricately linked to the levels of cyclic guanosine-3',5'-monophosphate(cGMP),which are influenced by the activities of nitric oxide synthase(NOS)and phosphodiesterase(PDE).Enhanced cGMP levels can be achieved either through the upregulation of NOS activity or the downregulation of PDE *** objective of the current study is to elucidate the effects of Chaga on disorders of glucolipid metabolism and renal abnormalities in rats with type 2 diabetes mellitus(T2DM),while concurrently examining the NOS-cGMP-PDE5 signaling pathway.A model of T2DM was developed in rats using a high-fat diet(HFD)combined with streptozotocin(STZ)administration,followed by treatment with Chaga extracts at doses of 50 and 100 mg·kg^(−1)for eight *** findings revealed that Chaga not only mitigated metabolic dysfunctions,evidenced by improvements in fasting blood glucose,total cholesterol,triglycerides,and insulin resistance,but also ameliorated renal function markers,including serum creatinine,urine creatinine(UCr),blood urea nitrogen,24-h urinary protein,and estimated creatinine ***,enhancements in glomerular volume,GBM thickness,podocyte foot process width(FPW),and the mRNA and protein expressions of podocyte markers,such as nephrin and wilms tumor-1,were *** was found to elevate cGMP levels in both serum and kidney tissues by increasing mRNA and protein expressions of renal endothelial NOS and neural NOS,while simultaneously reducing the expressions of renal inducible NOS and *** summary,Chaga counteracts HFD/STZ-induced glucolipid metabolism and renal function disturbances by modulating the NOS-cGMP-PDE5 signaling *** research supports the potential application of Chaga in the clinical prevention and treatment of T2DM and

Comprehensive SummaryThe detection of critical endogenous species, such as bacteria in microenvironments in the body, requires better imaging tools for visualization and monitoring of biological events. Bioluminescence imaging is the most popular strategy for obtaining real-time in living cells and organisms. Herein, we introduced a nitroaryl group on the C-3 position and a hydroxy group at the C-6 phenyl ring on furimazine to report the first bioluminescent probe (7) based on NanoLuc-furimazine bioluminescent pair for the detection of nitroreductase in bacteria. The probe, which possessed up to 560-fold intensity increase with a low detection limit of 16 ng/mL of nitroreductase, has the most efficient uncage efficiency in comparison with other bioluminescent congeners, thus enabling highly selective and sensitive visualization of NTR activity in a panel of clinical priority pathogens. Additionally, imaging of the recombinant strain as well as the NTR from mouse feces indicated the potential of this probe in the application of different mouse disease models.

目的探讨中医药治疗单纯性肥胖的研究热点与趋势,为相关研究提供参考。方法检索中国知识资源总库、中文科技期刊数据库(VIP)、中国学术期刊数据库(万方数据)建库至2023年8月30日发表的中医药治疗单纯性肥胖相关文献,采用Excel2020软件分析纳入文献发文时间、来源期刊,采用CiteSpace6.1.R3软件进行核心作者共现、机构共现、关键词聚类、突现分析,采用VOSviewer1.6.18软件进行作者合作关系共现、高频关键词共现分析。结果共纳入文献1 376篇,发文量总体呈上升趋势。纳入文献涉及2 648位作者,发文较多的有刘志诚、周仲瑜、王佳捷等。976篇期刊文献来源于297种期刊,《针灸临床杂志》为该领域发文量最多的期刊。发文量前5位机构依次是广州中医药大学、南京中医药大学、黑龙江中医药大学、湖北中医药大学及山东中医药大学。高频关键词有单纯性肥胖、针刺、电针、瘦素、临床研究等。结论中医药治疗单纯性肥胖发展前景良好,主要治疗手段为电针、针刺、针灸和耳穴贴压等,研究热点集中在针刺、针灸、拔罐、穴位埋线等方面,穴位埋线研究可能是未来研究趋势。

A simple, economical, and sensitive capillary electrophoresis (CE) method integrated with capacitively coupled contactless conductivity detection was developed for the determination of metal ions such as K+, Na+, Mg2+, Sr2+, Ca2+ in drinking water. 18-Crown-6 ether and Hexadecyltrimethylammonium Bromide (CTAB) were employed as complexing reagents. The effects of electrolyte additives, citric acid buffer solution, and other separation conditions of CE were comprehensively investigated and carefully optimized. The best results were obtained in a running buffer solution composed of citric acid (12 mM), 18-crown-6 ether (0.2 mM), and CTAB (0.015 mM) at pH 3.5. Under these conditions, a complete separation of five metal ions was successfully achieved in less than 12 min. The limits of detection for the optimal procedure were determined to be in the range of 0.02 - 0.2 mg·L-1. The repeatability with respect to migration times and peak areas, expressed as relative standard deviations, was better than 2.3% and 5.1%, respectively. Evaluation of the efficiency of the methodology indicated that it was reliable for the determination of metal ions in six different brands of drinking water samples.

Permanganate/sulfite(Mn(VII)/S(IV))process is a promising pre-oxidation technology for sequestering the emerging organic contaminants in drinking water treatment ***(IPM),a representative of iodinated X-ray contrast media,has been widely detected in water sources and has the risk of forming iodinated disinfection byproducts(I-DBPs)in water treatment *** this study,we investigated the evolution of iodine species during the IPM degradation by the Mn(VII)/S(IV)process and its effect on the subsequent formation of I-DBPs during chlorination at pH 7.0 and *** could be effectively degraded in the Mn(VII)/S(IV)process at environmentally relevant pH(pH 7.0 and 8.0).The results of quenching and competitive oxidation kinetic experiments revealed that SO^(·-)_(4)was the major reactive oxidizing species contributing to the degradation of IPM whereas the contributions of HO·and reactive manganese species were negligible in the Mn(VII)/S(IV)process.I–and IO–3were generated while no HOI was detected during the degradation of IPM in the Mn(VII)/S(IV)*** effects of IPM oxidation by Mn(VII)/S(IV)on the subsequent formation of chlorinated disinfection by-products(Cl-DBPs)during chlorination were related to the category of *** pre-oxidation of IPM by Mn(VII)/S(IV)resulted in the generation of I-DBPs during the disinfection process although no I-DBPs were detected if no pre-oxidation was *** finding of this study suggested that attention should be paid to the toxicity of DBPs when water containing iodinated organic contaminants is treated by Mn(VII)/S(IV)process or other pre-oxidation technologies.

This study aimed to investigate the effects of phosphorylated-ovalbumin(P-OVA)at different concentrations(0.5%and 1.0%)on the gel properties of pork myofibrillar protein(MP).The results showed that the textural properties such as gel hardness,cohesiveness,springiness and chewiness were improved with P-OVA addition at 0.5%.The water holding capacity(up to 75.89%)and gel strength(up to 168.56 g·mm)of MP gel were markedly increased after P-OVA *** absolute value of zeta potential reached 13.85 mV and maximum hydrophobicity(15.2μg)resulted from the addition of 0.5%*** storage modulus(G’)and loss modulus(G’’)of MP gel were significantly increased from 50°C,and the G’and G’’significantly increased after 1.0%P-OVA addition,evidencing that the cross-linking effect of MP protein gel was *** addition,the P-OVA addition improved the structure of MP gel protein by reducing theα-helix,while increasing theβ-sheet and the r-value(the ratio between two ultraviolet second-derivative peak-to-trough distances),which further promoted the uniform and compact gel network *** work demonstrated that P-OVA is a highly effective modifier that significantly improves the quality of MP *** a view of practice,0.5%P-OVA is the optimal addition amount.
