Benefit-Risk Assessment of Chinese Medicine Injections for Primary Liver Cancer Based on Multi-criteria Decision Analysis

ObjectiveTo evaluate the benefit-risk of 3 commonly used Chinese medicine injections, Aidi Injection (ADI), Cinobufagin Injection (CINI) and Compound Kushen Injection (CKI), in the treatment of primary liver cancer (PLC), so as to provide a reference for clinical decision-making.MethodsRandomized controlled trials (RCTs) of ADI, CINI and CKI in the treatment of PLC published in the databases of China National Knowledge Infrastructure, Wanfang, China Science and Technology Journal Database, SinoMed, PubMed, Cochrane Library, and Web of Science were retrieved from January 2020 to October 2022. The data of benefit and risk indicators were combined to obtain the effect value. The multi-criteria decision analysis (MCDA) model was applied to build the decision tree. The benefit value, risk value and benefit risk value of the 3 injections in PLC treatment were calculated. Monte Carlo simulation was carried out to calculate the 95% confidence interval and probability of differences among the 3 injections, so as to optimize the evaluation results.ResultsA total of 71 RCTs were included. The benefit values of ADI, CINI and CKI combined with transcatheter arterial chemoembolization (TACE) were 42, 38 and 36, respectively. The risk values were 42, 25 and 37, respectively. The benefit risk values were 42, 31 and 37, respectively. The benefit risk differences of ADI vs. CINI, ADI vs. CKI, and CKI vs. CINI were 11 (−0.86, 17.75), 5 (−5.01, 11.09), and 6 (−1.87, 12.63), respectively. The probability that ADI superior to CINI, ADI superior to CKI, and CKI superior to CINI was 96.26%, 77.27%, and 92.62%, respectively.ConclusionBased on the results of MCDA model, CINI combined with TACE has the greatest risk in the treatment of the PLC. Considering the efficacy and safety, the possible priority of the 3 Chinese medicine injections combined with TACE in the treatment of PLC is ADI, CKI and CINI.

Two cardenolide glycosides,corotoxigenin 3-0-[β-D-glucopyranosyl-(l→4)-6-deoxy-β-D-glucopyranoside](1)and coroglaucigenin 3-0-[β-D-glucopyranosyl-(l→4)-6-deoxy-β-D-glucopyranoside](2),were isolated from the seed fairs of Asclepias curassavica.The structures of 1-2 were determined based on the combination of the analysis of their MS,NMR spectroscopic data and acid hydrolysis.The inhibitory effects of compounds 1 and 2 on human colorectal carcinoma cells(HCT116),non-small cell lung carcinoma cells(A549)and hepatic cancer cells(SMMC-7721)were evaluated.The results showed that both compounds 1 and 2 significantly inhibited the viability,proliferation,and migration of A549,HCT116 and SMMC-7721 cells,suggesting that compounds 1 and 2 can be applied in the treatment of lung,colon and liver cancers in clinical practice.This study may not only provide a scientific basis for clarifying the active ingredients in A.curassavica,but also help to understand its antitumor activity,which can promote the application of A.curassavica in clinical treatment of various cancers.

目的探讨多能干细胞(iPSC)分泌的外泌体(Exos)促进角质细胞的增殖、侵袭与迁移,从而促进伤口愈合的作用。方法提取iPSC-Exos并通过透射电子显微镜、纳米颗粒追踪分析技术和蛋白质印迹法进行鉴定。将带有PKH26标记纯化的iPSC-Exos加到角质细胞(HaCaT)里进行角质细胞摄取外泌体的测定;用细胞计数试剂盒(CCK-8)检测外泌体最佳工作浓度。HaCaT细胞分为对照组(细胞划痕)和实验组(细胞划痕之后加入最佳工作浓度的外泌体)。分别用CCK-8、5-乙炔基-2′脱氧尿嘧啶核苷(EdU)、划痕实验及Transwell实验检测各组细胞的增殖、迁移以及侵袭能力。结果iPSC-Exos呈现圆形或椭圆形的膜囊泡结构,直径为(120.00±25.00)nm。实验组iPSC-Exos特征性表面标志物CD9、CD63、CD81的表达均呈阳性,对照组均呈阴性。HaCaT细胞能够摄取iPSC-Exos。干预24 h后,对照组和实验组的划痕愈合率分别为(25.70±1.07)%和(71.60±12.76)%,Transwell侵袭细胞数分别为(86.33±10.79)和(166.33±24.13)个,EdU阳性比例分别为(45.30±3.17)%和(78.10±6.29)%,实验组的上述指标与对照组比较,在统计学上差异均有统计学意义(均P<0.05)。结论多能干细胞分泌的外泌体能够促进角质细胞的增殖、迁移及侵袭,从而间接促进伤口愈合。

Cloning and Expression of Actinobacillus pleuropneumoniae Gene Coding for TbpA and Development of an Indirect TbpA-ELISA

This study presents the cloning and expression of gene encoding transferrin-binding protein A from Actinobacillus pleuropneurnoniae in Escherichia coli expression system and the development of an indirect TbpA-ELISA. The gene coding TbpA was amplified from the A. pleuropneumoniae serotype 2 genome using polymerase chain reaction and cloned to pET-28b expression vector under the control of strong, inducible T7 promoter. The recombinant plasmid was expressed in E. coli BL21 （DE3）. The expressed fusion protein was analyzed using SDS-PAGE and Western blotting. The diagnostic potential of recombinant TbpA （rTbpA） was evaluated through an antibody-detection indirect ELISA based on the purified rTbpA. The TbpA antibodies were detectable in mice on day 7 after vaccination with purified rTbpA protein or infection with A. pleuropneumoniae serotype 10 with the TbpA-based ELISA. In addition, the TbpA-ELISA was able to detect 12 serotyping rabbit antisera postinoculation （PI） with A. pleuropneumoniae 12 serotypes experimentally. The comparable result was obtained by detecting the 117 clinical serum samples using, respectively, the TbpA-ELISA and indirect hemagglutination test （IHA） based on multiplex antigen. The result indicates that the TbpA-ELISA was the more sensitive method compared with the Mix-IHA method because of its consistent presence in A. pleuropneumoniae serotypes. In conclusion, the conserved TbpA of A. pleuropneumoniae can be used for the development of a cross-serotype diagnostic method for the detection of antibodies against A. pleuropneurnoniae.

A passive sampler for synchronously measuring inorganic and organic pollutants in sediment porewater:Configuration and field application

In situ measurement of multiple pollutants coexisting in sediment porewater is an essential step in comprehensively assessing the bioavailability and risk of pollutants,but to date,this needs to be better developed.In this study,a passive sampler,consisting of an“I-shaped”supporting frame and inorganic/organic sampling units,incorporating equilibrium dialysis theory and kinetic/equilibrium sorption principle,was developed for the synchronous measurement of inorganic(e.g.,phosphorus and metal(loid)s)and organic pollutants(e.g.,parent and substituted PAHs).The equilibrium time and sampling rates were explored in laboratory tests to support in situ application.Profiles of pollutants in porewater within a vertical resolution of centimeters,i.e.,1 cm and 2 cm for inorganic and organic pollutants,respectively,were obtained by field deployment of the sampler for further estimation of diffusive fluxes across the sediment-water interface.The results suggested that the role of sediments for a specific pollutant may change(e.g.,from“sink”to“source”)during the sampling time.This study demonstrated the feasibility of synchronous measurement of inorganic and organic pollutants in sediment porewater by the passive sampler.In addition,it provided new insight for further investigation into the combined pollution effects of various pollutants in sediments.

U-Net Models for Representing Wind Stress Anomalies over the Tropical Pacific and Their Integrations with an Intermediate Coupled Model for ENSO Studies

El Niño-Southern Oscillation(ENSO)is the strongest interannual climate mode influencing the coupled ocean-atmosphere system in the tropical Pacific,and numerous dynamical and statistical models have been developed to simulate and predict it.In some simplified coupled ocean-atmosphere models,the relationship between sea surface temperature(SST)anomalies and wind stress(τ)anomalies can be constructed by statistical methods,such as singular value decomposition(SVD).In recent years,the applications of artificial intelligence(AI)to climate modeling have shown promising prospects,and the integrations of AI-based models with dynamical models are active areas of research.This study constructs U-Net models for representing the relationship between SSTAs andτanomalies in the tropical Pacific;the UNet-derivedτmodel,denoted asτUNet,is then used to replace the original SVD-basedτmodel of an intermediate coupled model(ICM),forming a newly AI-integrated ICM,referred to as ICM-UNet.The simulation results obtained from ICM-UNet demonstrate their ability to represent the spatiotemporal variability of oceanic and atmospheric anomaly fields in the equatorial Pacific.In the ocean-only case study,theτUNet-derived wind stress anomaly fields are used to force the ocean component of the ICM,the results of which also indicate reasonable simulations of typical ENSO events.These results demonstrate the feasibility of integrating an AI-derived model with a physics-based dynamical model for ENSO modeling studies.Furthermore,the successful integration of the dynamical ocean models with the AI-based atmospheric wind model provides a novel approach to ocean-atmosphere interaction modeling studies.

Testing Linearity in Functional Partially Linear Models

For the functional partially linear models including flexible nonparametric part and functional linear part,the estimators of the nonlinear function and the slope function have been studied in existing literature.How to test the correlation between response and explanatory variables,however,still seems to be missing.Therefore,a test procedure for testing the linearity in the functional partially linear models will be proposed in this paper.A test statistic is constructed based on the existing estimators of the nonlinear and the slope functions.Further,we prove that the approximately asymptotic distribution of the proposed statistic is a chi-squared distribution under some regularity conditions.Finally,some simulation studies and a real data application are presented to demonstrate the performance of the proposed test statistic.

Self-Triggered Set Stabilization of Boolean Control Networks and Its Applications

Set stabilization is one of the essential problems in engineering systems, and self-triggered control(STC) can save the storage space for interactive information, and can be successfully applied in networked control systems with limited communication resources. In this study, the set stabilization problem and STC design of Boolean control networks are investigated via the semi-tensor product technique. On the one hand, the largest control invariant subset is calculated in terms of the strongly connected components of the state transition graph, by which a graph-theoretical condition for set stabilization is derived. On the other hand, a characteristic function is exploited to determine the triggering mechanism and feasible controls. Based on this, the minimum-time and minimum-triggering open-loop, state-feedback and output-feedback STCs for set stabilization are designed,respectively. As classic applications of self-triggered set stabilization, self-triggered synchronization, self-triggered output tracking and self-triggered output regulation are discussed as well. Additionally, several practical examples are given to illustrate the effectiveness of theoretical results.

Residual levels and health risk assessment of trace metals in Chinese resident diet

Large-scale metal contamination across the food web is an intractable problem due to increasing pollutant emissions,atmospheric transport,and dry and wet deposition of elements.The present study focus on several trace metals that are rarely studied but have special toxicity,including tin(Sn),antimony(Sb),gold(Au),hafnium(Hf),palladium(Pd),platinum(Pt),ruthenium(Ru),tellurium(Te)and iridium(Ir).We investigated trace metals residues and distribution characteristics,and further evaluated the potential health risks from major daily food intakes in 33 cities in China.Sn,Sb,Ir,Hf,and Au were frequently detected in food samples with the concentrations ranged from ND(not detected)to 24.78μg/kg ww(wet weight).Eggs exhibited the highest residual level of all detected metals(13.70±14.70μg/kg ww in sum),while the lowest concentrations were observed in vegetables(0.53±0.17μg/kg ww in sum).Sn accounting for more than 50%of the total trace metals concentration in both terrestrial and aquatic animal origin foods.In terrestrial plant origin foods,Sn and Ir were the most abundant elements.Hf and Au were the most abundant elements in egg samples.In addition,Sb and Ir showed a clear trophic dilution effect in terrestrial environments,while in aquatic ecosystems,Sn,Hf,and Au exhibited obvious trophic amplification effects.The calculated average estimated daily intake(EDI)via food consumption in five regions of China was 0.09μg/(kg·day),implying the health risk of aforementioned elements was acceptable.
