Objective:Peritoneal fibrosis(PF)is the main cause of declining efficiency and ultrafiltration failure of the peritoneum,which restricts the long-term application of peritoneal dialysis(PD).This study aimed to investigate the therapeutic effects and mechanisms of bone marrow mesenchymal stem cells-derived exosomes(BMSC-Exos)on PF in response to ***:Small RNA sequencing analysis of BMSC-Exos was performed by second-generation sequencing.C57BL/6J mice were infused with 4.2.%glucose-based peritoneal dialysis fluid(PDF)for 6 consecutive weeks to establish a PF model.A total of 36 mice were randomly divided into 6 groups:control group,1.5%PDF group,2.5%PDF group,4.2.%PDF group,BMSC-Exos treatment group,and BMSC-Exos+TP53 treatment *** transcription quantitative polymerase chain reaction(RT-qPCR)was performed to measure the expression level of miR-2.a-3p in BMSC-Exos and peritoneum of mice treated with different concentrations of *** and Masson staining were performed to evaluate the extent of *** therapeutic potential of BMSC-Exos for PF was examined through pathological examination,RT-qPCR,Western blotting,and peritoneal function ***-mesenchymal transition(EMT)of HMrSV5 was induced with 4.2.%*** were divided into control group,4.2.%PDF group,BMSC-Exos treatment group,and BMSC-Exos+TP53 treatment *** Counting Kit-8 assay was used to measure cell viability,and transwell migration assay was used to verify the capacity of BMSC-Exos to inhibit EMT in HMrSV5 ***:Small RNA sequencing analysis showed that miR-2.a-3p was highly expressed in BMSC-derived exosomes compared to *** RT-qPCR results showed that the expression of miR-2.a-3p was upregulated in BMSC-Exos,but decreased in PD *** found that PF was glucose concentration-dependently enhanced in the peritoneum of the PD *** with the control mice,the PD mice showed high solute transport and decreased ultrafiltration volume as well as an obvious f

BACKGROUND Wnt/FZD-mediated signaling pathways are activated in more than 90%of hepatocellular carcinoma(HCC)cell *** a well-known secretory glycoprotein,Wnt3 can interact with FZD receptors on the cell surface,thereby activating the Wnt/β-catenin signaling ***,the N-glycosylation modification site of Wnt3 and the effect of this modification on the biological function of the protein are still *** To investigate the effect of Wnt3 N-glycosylation on the biological function of HCC *** Site-directed mutagenesis was used to verify the Wnt3 N-glycosylation sites,actinomycin D treatment was used to detect the stability of Wnt3 after site-directed mutation,the binding of the N-glycosylation site-directed mutant Wnt3 to FZD7 was observed by laser confocal microscopy,and the effects of the N-glycosylation site-directed mutation of Wnt3 on the Wnt/β-catenin signaling pathway and the progression of HCC cells were detected by western blot and cell function *** Wnt3 has two N-glycosylation-modified sites(Asn90 and Asn301);when a single site at amino acid 301 is mutated,the stability of Wnt3 is weakened;the binding ability of Wnt3 to FZD7 decreases when both sites are mutated simultaneously;and the level of proteins related to the Wnt/β-catenin signaling pathway is *** proliferation,migration and invasion are also weakened in the case of single 301 site and double-site *** These results indicate that by inhibiting the N-glycosylation of Wnt3,the proliferation,migration,invasion and colony formation abilities of liver cancer cells can be weakened,which might provide new therapeutic strategies for clinical liver cancer in the future.

Testosterone is closely associated with lipid metabolism and known to affect body fat composition and muscle mass in ***,the mechanisms by which testosterone acts on lipid metabolism are not yet fully understood,especially in *** this study,cyp17a1-/-zebrafish(Danio rerio)exhibited excessive visceral adipose tissue(VAT),lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis(DNL)*** assay for transposase accessible chromatinwithsequencing(ATAC-seq)results demonstrated that chromatin accessibility of DNL genes was increased in cyp17a1-/-fish compared to cyp17a1+/+male fish,including stearoyl-CoA desaturase(scd)and fatty acid synthase(fasn).Androgen response element(ARE)motifs in the androgen signaling pathway were significantly enriched in cyp17a1+/+male fish but not in cyp17a1-/-*** androgen receptor(ar)-/-and wildtype(WT)zebrafish administered with Ar antagonist flutamide displayed excessive visceral adipose tissue,lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis *** Ar agonist BMS-56492. reduced the content of VAT and lipid content,and down-regulated acetyl-CoA carboxylase a(acaca),fasn,and scd ***,the rescue effect of testosterone on cyp17a1-/-fish in terms of phenotypes was abolished when ar was additionally ***,these findings reveal that testosterone inhibits lipid deposition by down-regulating DNL genes via Ar in zebrafish,thus expanding our understanding of the relationship between testosterone and lipid metabolism in teleosts.

The convergence of Internet of Things(IoT),5G,and cloud collaboration offers tailored solutions to the rigorous demands of multi-flow integrated energy aggregation dispatch data *** generative adversarial networks(GANs)are instrumental in resource scheduling,their application in this domain is impeded by challenges such as convergence speed,inferior optimality searching capability,and the inability to learn from failed decision making ***,a cloud-edge collaborative federated GAN-based communication and computing resource scheduling algorithm with long-term constraint violation sensitiveness is proposed to address these *** proposed algorithm facilitates real-time,energy-efficient data processing by optimizing transmission power control,data migration,and computing resource *** employs federated learning for global parameter aggregation to enhance GAN parameter updating and dynamically adjusts GAN learning rates and global aggregation weights based on energy consumption constraint *** results indicate that the proposed algorithm effectively reduces data processing latency,energy consumption,and convergence time.

The facile construction of nanoscale NbH_(x)supported by Ti_(3)C_(2.(NbH_(x)@Ti_(3)C_(2.)was achieved using facile ball milling strategy to improve the hydrogen desorption kinetics and reversibility of magnesium borohydride(Mg(BH_(4))_(2.).The doping of 30 wt%NbH_(x)@Ti_(3)C_(2.catalyst reduced the onset dehydrogenation temperature of Mg(BH_(4))_(2.to 71.2..Additionally,the Mg(BH_(4))_(2.+30NbH_(x)@Ti_(3)C_(2.composite achieved remarkable hydrogen release of over 9.2.wt%at a temperature as low as 2.0℃,indicating unexpected dehydrogenation ***,the reversibility of NbH_(x)@Ti_(3)C_(2.doped Mg(BH_(4))_(2.was retained to more than 4.2.wt%after four cycles,which was increased by 68%compared to that of undoped Mg(BH_(4))_(2..The strong catalysis of NbH_(x)@Ti_(3)C_(2.catalyst could be attributed to the synergistic effect of NbH_(x)and Ti_(3)C_(2.in hydrogen spillover and ***_(x)acted as a"hydrogen pump"for effective hydrogen spillover,while Ti_(3)C_(2.created numerous diffusion channels for hydrogen dissolution and provided the active catalytic site to facilitate the re/dehydrogenation of Mg(BH_(4))_(2..
