Objective: In this paper we compared the two methods of cell sorting (magnetic cell sorting and flow cytometry sorting) for the isolation and function analysis of mouse CD4+ CD25+ regulatory T (Treg) cells, in order to inform further studies in Treg cell function. Methods: We separately used magnetic cell sorting and flow cytometry sorting to identify CD4+ CD25+ Treg cells. After magnetic cell separation, we further used flow cytometry to analyze the purity of CD4+ CD25+ Treg cells, trypan blue staining to detect cell viability, and propidium iodide (PI) staining to assess the cell viability. We detected the immune inhibition of CD4+ CD25+ Treg cells in the in vitro proliferation experiments. Results: The results showed that compared to flow cytometry sorting, magnetic cell sorting took more time and effort, but fewer live cells were obtained than with flow cytometry sorting. The CD4+ CD25+ Treg cells, however, obtained with both methods have similar immunosuppressive capacities. Conclusion: The result suggests that both methods can be used in isolating CD4+ CD25+ Treg cells, and one can select the best method according to specific needs and availability of the methodologies.

Genistein, a potent antioxidant compound, protects dopaminergic neurons in a mouse model of Parkinson＇s disease. However, the mecha- nism underlying this action remains unknown. This study investigated human SH-SYSY cells overexpressing the A53T mutant of α-synuclein. Four groups of cells were assayed： a control group （without any treatment）, a genistein group （incubated with 20 μM genistein）, a rote- none group （treated with 50 μM rotenone）, and a rotenone ＋ genistein group （incubated with 20 μM genistein and then treated with 50μM rotenone）. A lactate dehydrogenase release test confirmed the protective effect of genistein, and genistein remarkably reversed mitochondrial oxidative injury caused by rotenone. Western blot assays showed that BCL-2 and Beclin ! levels were markedly higher in the genistein group than in the rotenone group. Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling revealed that genistein inhibited rotenone-induced apoptosis in SH-SYSY cells. Compared with the control group, the expression of NFE2L2 and HMOX1 was significantly increased in the genistein ＋ rotenone group. However, after treatment with estrogen receptor and NFE2L2 channel blockers （ICI-182780 and ML385, respectively）, genistein could not elevate NFE2L2 and HMOX1 expression. ICI-182780 effectively prevented genistein-mediated phosphorylation of NFE2L2 and remarkably suppressed phosphorylation of AKT, a protein downstream of the estrogen receptor. These findings confirm that genistein has neuroprotective effects in a cell model of Parkinson＇s dis- ease. Genistein can reduce oxidative stress damage and cell apoptosis by activating estrogen receptors and NFE2L2 channels.

Let m, n ∈ N, and V be an m-dimensional vector space over a field F of characteristic 0. Let U = F ＋ V and Rn be the rook monoid. In this paper, we construct a certain quasi-idempotent in the annihilator of U^×n in FRn, which comes from some one-dimensional two-sided ideal of rook monoid algebra. We show that the two-sided ideal generated by this element is indeed the whole annihilator of U^×n in FR^n.

BACKGROUND Preoperative radiochemotherapy is widely used in locally advanced rectal *** can improve local control of rectal cancer. However, some researchers believe it increases the incidence of surgical complications. They doubt its safety. Patients with locally advanced rectal cancer receive three different treatments in our hospital, including long-course radiochemotherapy, short-course radiotherapy,and surgery directly. We can compare their differences in postoperative *** To investigate surgical complications caused by different preoperative radiotherapy *** We retrospectively analyzed 1197 patients admitted between 2008 and 2010 with locally advanced rectal cancer. Three hundred and forty-six patients were treated with preoperative long-course radiochemotherapy(25 × 2 Gy) followed by total mesorectal excision(TME) 6–8 wk later, and 259 patients received short-course radiotherapy(10 × 3 Gy) and subsequently TME 7–10 d later. The remaining 592 patients underwent TME alone without neoadjuvant therapy. According to Clavien–Dindo classification, surgical complications were evaluated for up to 30 d after discharge from *** There were no deaths in 30 d in all groups after treatment. The majorcomplications were anastomotic leakage and perineal wound complications. The results suggested that both long-course [odds ratio(OR) = 3.624, 95% confidence interval(CI): 1.689–7.775, P = 0.001] and short-course(OR = 5.150, 95%CI:1.828–14.515, P = 0.002) radiotherapy were associated with anastomotic *** ileostomy was a protective factor for anastomotic leakage(OR =6.211, 95%CI: 2.525–15.385, P < 0.001). BACKGROUND At present,the existing internal medicine drug treatment can alleviate the high glucose toxicity of patients to a certain extent,to explore the efficacy of laparoscopic jejunoileal side to side anastomosis in the treatment of type 2 diabetes,the report is as *** To investigate the effect of jejunoileal side-to-side anastomosis on metabolic parameters in patients with type 2 diabetes mellitus(T2DM).METHODS We retrospectively analyzed the clinical data of 78 patients with T2DM who were treated via jejunoileal lateral *** indicators were collected preoperatively,as well as at 3 and 6 months *** metabolic indicators analyzed included body mass index(BMI),systolic blood pressure(SBP),diastolic blood pressure(DBP),fasting blood glucose(FBG),2-hour blood glucose(PBG),glycated hemoglobin(HbA1c),fasting C-peptide,2-hour C-peptide(PCP),fasting insulin(Fins),2-hour insulin(Pins),insulin resistance index(HOMA-IR),βCellular function index(HOMA-β),alanine aminotransferase,aspartate aminotransferase,serum total cholesterol(TC),low-density lipoprotein cholesterol(L DL-C),triglycerides(TG),high-density lipoprotein,and uric acid(UA)*** SBP,DBP,PBG,HbA1c,LDL-C,and TG were all significantly lower 3 months postoperative vs preoperative values;body weight,BMI,SBP,DBP,FBG,PBG,HbA1c,TC,TG,UA,and HOMA-IR values were all significantly lower 6 months postoperative vs at 3 months;and PCP,Fins,Pins,and HOMA-βwere all significantly higher 6 months postoperative vs at 3 months(all P<0.05).CONCLUSION Side-to-side anastomosis of the jejunum and ileum can effectively treat T2DM and improve the metabolic index levels associated with it.

An Estimation of the Initial Period of Pulsars

The initial period of a pulsar is an important factor in our understanding of the formation of neutron stars and of the nature of the equation of state of neutron star matter. Up to now this quantity can only be obtained for a few pulsars for which accurate age and braking index are known. Based on the theory of the off-center dipole emission, in which pulsars obtain their high velocities depending on the initial periods, we calculate the initial period using the proper motion data. Because the orbital velocity of the progenitor and asymmetric kick in the supernova explosion may also contribute to the observed velocity of the pulsar, the derived values of initial periods are lower limits. For normal pulsars, the initial periods are in the range of 0.6 - 2.6ms. For the millisecond pulsars, the initial periods are comparable to their current periods, and the ratio between the initial period and the current period increases with the decrease of the current period. For PSR B1937+21 with the shortest period of 1.56ms, the ratio is 0.77.
