Hepatocellular carcinoma (HCC) is one of the most common tumor types and remains a major clinical challenge. Increasing evidence has revealed that mitophagy inhibitors can enhance the effect of chemotherapy on HCC. However, few mitophagy inhibitors have been approved for clinical use in humans. Pyrimethamine (Pyr) is used to treat infections caused by protozoan parasites. Recent studies have reported that Pyr may be beneficial in the treatment of various tumors. However, its mechanism of action is still not clearly defined. Here, we found that blocking mitophagy sensitized cells to Pyr-induced apoptosis. Mechanistically, Pyr potently induced the accumulation of autophagosomes by inhibiting autophagosome-lysosome fusion in human HCC cells. In vitro and in vivo studies revealed that Pyr blocked autophagosome-lysosome fusion by upregulating BNIP3 to inhibit synaptosomal-associated protein 29 (SNAP29)-vesicle-associated membrane protein 8 (VAMP8) interaction. Moreover, Pyr acted synergistically with sorafenib (Sora) to induce apoptosis and inhibit HCC proliferation in vitro and in vivo. Pyr enhances the sensitivity of HCC cells to Sora, a common chemotherapeutic, by inhibiting mitophagy. Thus, these results provide new insights into the mechanism of action of Pyr and imply that Pyr could potentially be further developed as a novel mitophagy inhibitor. Notably, Pyr and Sora combination therapy could be a promising treatment for malignant HCC.

Ozone(O_(3))pollution is usually linked to warm weather and strong solar radiation,making it uncommon in cold ***,an unusual occurrence of four high O_(3)episode days(with maximum hourly concentrations exceeding 100 ppbv and peaking at 121 ppbv)was recorded in January 2018 in Lanzhou city,*** these episodes,the average daytime concentration of total non-methane volatile organic compounds(TVOCs)reached 153.4±19.0 ppbv,with alkenes—largely emitted from the local petrochemical industry—comprising 82.3±13.1 *** we show a photochemical box model coupled with a Master Chemical Mechanism to elucidate the mechanisms behind this unusual wintertime O_(3)*** find that the typically low temperatures(−1.7±1.3℃)and weak solar radiation(263.6±60.7 W m^(-2))of those winter episode days had a minimal effect on the reactivity of VOCs with OH ***,the ozonolysis of alkenes generated Criegee intermediates,which rapidly decomposed into substantial ROx radicals(OH,HO_(2),and RO_(2))without *** radical production led to the oxidation of VOCs,with alkene ozonolysis ultimately contributing to 89.6±8.7%of the O_(3)formation during these *** mechanism did not activate at night due to the depletion of O_(3)by the NO titration ***,the findings indicate that a reduction of alkenes by 28.6%or NO_(x)by 27.7%in the early afternoon could significantly mitigate wintertime O_(3)***,this study unravels the unique mechanism of alkene-induced winter O_(3)pollution and offers a reference for winter O_(3)reduction strategies in the petrochemical industrial regions.

High precision magnetometer(HPM)is a magnetic field detection payload onboard China Seismo-Electromagnetic Satellite(CSES),including two fluxgate magnetometers(FGM)and a coupled dark state magnetometer(CDSM).Observations show that FGM appears to be influenced when tri-band beacon(TBB)is powered on and emits electromagnetic *** interference phenomenon is further validated based on both in-orbit observation analysis and electromagnetic compatibility(EMC)tests on the ground.A joint correction algorithm based on the least square fitting and first-order difference method according to scalar magnetometer data is proposed to eliminate the *** algorithm significantly improves the consistency of HPM *** correction,the average scalar deviation error could be reduced from 9.0 nT to around 0.7 nT.

设计了一款轻量化和紧凑型的二自由度操纵手柄,并基于该操纵手柄提出了一种无力传感器的自适应变阻抗控制策略。根据操纵员操纵力大小对阻抗参数进行相应的调整以适应操纵员不同的操纵习惯;除了保证精确的位置控制外,加入了操纵手柄末端的速度控制,增强了操纵员的操纵感受。另外,在变阻抗柔顺控制的基础上加入了操纵力估计控制策略,从而避免了由于加装力传感器的额外接线导致系统复杂度增加以及由此带来的测量噪声。证明了所提控制策略在Lyapunov意义下的稳定性,说明了控制误差是收敛的,整个闭环系统的信号响应是一致极限有界的。最后,通过仿真实验以及实际台架实验验证了上述策略的有效性和稳定性。

The CSES(China Seismo-Electromagnetic Satellite)is the electromagnetism satellite of China’s zhangheng mission which is planned to launch a series of microsatellites within next 10 years in order to monitor the electromagnetic environment,gravitational *** CSES 01 probe(also called ZH-1)was launched successfully on 2 February 2018,from the Jiuquan Satellite Launch Centre(China)and is expected to operate for 5 years in *** second probe CSES 02 is going to be launched in *** scientific objectives of CSES are to detect the electromagnetic field and waves,plasma and particles,for studying the seismic-associated *** meet the requirements of scientific objective,the satellite is designed to be in a sun-synchronous orbit with a high inclination of 97.4°at an altitude around 507 *** carries nine scientific payloads including Search-coil magnetometer,Electric Field Detector,High precision Magnetometer,GNSS occultation Receiver,Plasma Analyzer,Langmuir Probe,two Energetic Particle Detectors(including an Italian one),and Tri-Band *** to now,CSES has been operating in orbit for 2 years with stable and reliable *** using all kinds of data acquired by CSES,we have undertaken a series of scientific researches in the field of global geomagnetic field re-building,the ionospheric variation environment,waves,and particle precipitations under disturbed space weather and earthquake activities,the Lithosphere-Atmosphere-Ionosphere coupling mechanism research and so on.

混合交通流作用下大跨径桥梁车辆荷载效应分析

卡车列队是一种通过自动控制、无线通讯等技术实现卡车成队近距离跟驰的交通模式,小间距行驶的车队直接导致桥面重车密度增加,且对于大跨径桥梁其可以容纳的卡车队数多,结构的整体和区段性能均可能受到影响。为研究卡车列队的荷载效应水平及其与普通车辆形成的混合交通流对桥梁结构的影响,评估现行规范的适用性,调研了国内外关于卡车列队的实际测试和理论研究,基于在役大跨径桥梁的实测交通流数据,提出了卡车列队荷载模型和混合交通流模拟方法。以一座大跨径悬索桥为主要背景工程,考虑实际的桥梁运行情况,实现了混合交通流模拟,研究了代表性混合交通场景中结构的车辆荷载效应水平和安全性,分析了卡车列队模式以及混合交通流对于大跨径悬索桥汽车荷载效应的影响。分析结果表明,混合交通流作用下的荷载效应水平较普通车辆车流有明显提高,但仍低于规范加载值。不同加载场景下的效应值约为规范值的35%~80%,现行规范对于卡车列队参与的混合交通流作用下结构的整体效应和区段效应均可提供较好的安全裕度,能够保证桥梁在未来相应运营状态下的安全服役。

With the increasing penetration rate of distributed energy,the low inertia level of microgrid becomes increasingly *** low inertia of the system leads to the deterioration of the frequency and voltage fluctuations,which makes the system difficult to deal with the instantaneous power ***,researchers developed a virtual synchronous generator(VSG) and connected it to the microgrid,which significantly alleviated the ***,previous VSG control algorithms have limitations due to their focus on parameter adjustment and cannot cope with large power imbalance *** overcome these challenges,this paper proposes a virtual synchronous generator control strategy based on adaptive model predictive *** control strategy first establishes an adaptive prediction model of the system,optimizes the control parameters of the VSG and updates the prediction model through an adaptive *** required power change is calculated from the prediction model and the quadratic programming(QP) objective function,and finally the power reference value of the virtual synchronous machine is *** results show that the proposed control strategy provides inertial support under microgrid load switching conditions and effectively suppresses frequency fluctuations in power imbalance scenarios,verifying the superiority and great autonomous performance of the proposed control strategy.

Objective: To explore the main chemical compounds in Xiaoer Qixing Cha Formulae (XQCF), and investigate its mechanisms for the treatment of infantile functional dyspepsia (IFD). Methods: The chemical components were identified by UPLC-QTOF/MS analytic technique. Targets of the compounds were screened from TCMSP and SWISS database, and disease targets were screened from OMIM and TTD online database. Candidate targets of compounds were mapped to the disease targets as predict therapeutic targets for XQCF. Several networks were constructed and analyzed by Cytoscape ver. 3.2.1. Meanwhile, prescription compatibility in XQCF was interpreted from the network perspective based on distribution of the number of targets. Furthermore, Gene Ontology (GO) enrichment analysis and KEGG pathway analysis were operated via Clue Go to illustrate complex relationships between the potential targets and pharmacological mechanisms. Results: A total of fifty-three compounds were recognized or tentatively characterized belonging to XQCF based on MS data and online chemical database. Sixty-three therapeutic targets were screened. AKT1, FOS, SLC6A4, COMT and 5-HT receptors were focused as therapeutic targets of XQCF. Pathways including carbohydrate digestion and absorption, serotonergic synapse, calcium signaling pathway and cAMP signaling pathway were predicted as significant regulatory pathways. The results indicated that the predicted targets and pathways related in brain-gut axis to a great extent, which could be potential pharmacological mechanism of XQCF for the treatment of IFD. Conclusions: The findings in this study provided the experimental and theoretical basis for further research for XQCF. Those also illustrated a reasonable method worth intensive study on pharmacodynamic mechanisms of TCM Formulae.
