复方卡比多巴和多巴丝肼的药代动力学和药效学比较研究

目的研究复方卡比多巴(左旋多巴/卡比多巴为250 mg/25 mg)和多巴丝肼(左旋多巴/苄丝肼为200 mg/50 mg)在帕金森病(PD)患者中的药代动力学及药效学特点。方法本试验用左旋多巴冲击实验,随机入组和交叉试验设计。20例PD患者第1周分别空腹口服复方卡比多巴275 mg或多巴丝肼250 mg,第2周接受另一种药物治疗。用高效液相色谱-串联质谱法检测左旋多巴血药浓度,用国际运动障碍协会帕金森病统一评分量表Ⅲ评估运动症状。结果有17例复方卡比多巴组和18例多巴丝肼组的患者数据纳入分析。服药后,复方卡比多巴组和多巴丝肼组的Cmax分别为(3563.76±1003.06)和(3642.44±1192.70)ng·mL^(-1),t_(max)分别为(1.10±0.44)和(1.03±0.55)h,t_(1/2)分别为(1.52±0.15)和(1.68±0.27)h,AUC_(0-t)分别为(7625.19±1706.85)和(5846.07±1191.16)ng·mL^(-1)·h,平均驻留时间(MRT)分别为(2.39±0.36)和(2.14±0.37)h。2组患者的C_(max)、t_(max)和t1/2比较,在统计学上差异均无统计学意义(均P>0.05);与多巴丝肼组相比,复方卡比多巴组中左旋多巴的AUC明显增加,MRT明显延长(均P<0.05)。2组各个时间点的运动症状改善和左旋多巴血药浓度呈现出较为一致的变化趋势,复方卡比多巴组在服药后第6和8小时的运动功能改善均显著优于多巴丝肼组[(-10.82±8.91)vs(-5.17±6.78)分,(-7.88±10.05)vs(-2.11±4.84)分;均P<0.05]。结论复方卡比多巴的药代动力学和药效学特点与多巴丝肼类似。

With the continuous appearance and expansion of high-strength hydrogels in emerging fields such as industry,medicine,and green development,the synthesis and application of high-strength hydrogels have developed dramatically and achieved remarkable results from the aspects of raw materials,preparation methods,and reinforcement ***,there is still a lack of systematic reviews on high-strength ***,we first discuss the advantages of natural and synthetic materials,and the characteristics of high-strength hydrogels prepared from different raw materials;we then expound on the influence mechanism of physical interactions or chemical bonds on the strength of the hydrogel from three aspects:physical cross-linking,chemical cross-linking,and dynamic chemical cross-linking;at last,we systematically expound the strengthening strategies,including double network/multi-network,nanocomposite,topology,supramolecular polymerization,and characteristics and strengthening mechanisms of such high-strength *** addition,based on the development status of high-strength hydrogels,we combined the application requirements and the existing drawbacks of high-strength hydrogels to propose their possible development directions in the future.

A fiber-optic liquid-level sensor is proposed and experimentally *** is a Mach–Zehnder interferometer(MZI)composed of a thin-core single-mode fiber(TCSMF)without coating sandwiched between two single-mode fibers(SMFs).The transmission spectrum properties of the MZI modified by liquid around the TCSMF are used for detecting the liquid *** is found that the sensor exhibits an excellent linear relationship between the level and the shift of interference dip wavelength and its achieved sensitivities are 0.160 nm/mm and 0.288 nm/mm for liquid with an RI of 1.3330 and 1.3696,*** the spectrum differential integration(SDI)method to analyze transmission spectra can increase the detecting resolution of the liquid *** to its advantages of an extremely easy fabrication process,high sensitivity and a large sensing range,the sensor is an ideal candidate for continuous liquid level sensing.

Urban lake ecosystems are significant for social development,but currently we know little about the geographical distribution of algal community in urban lakes at a *** this study,we investigated the algal community structure in different areas of urban lakes in China and evaluated the influence of water quality parameters and geographical location on the algal *** results showed that obvious differences in water quality and algal communities were observed among urban lakes in different geographical *** was the dominant phylum,followed by cyanobacteria in all *** network analysis indicated that algal community composition in urban lakes of the western and southern area showed more variations than the eastern and northern areas,*** analysis and structural equation model revealed that nutrients and p H were dominant environmental factors that affected the algal community,and they showed higher influence than that of iron,manganese and COD Mn ***,algal community and density exhibited longitude and latitude *** general,these results provided an ecological insight into large-scale geographical distributions of algal community in urban lakes,thereby having potential applications for management of the lakes.

本研究采用超声萃取-液相色谱-三重四极杆质谱法,对土壤中12种苯胺类化合物的前处理方法和标准曲线配制方式进行优化研究,确定了超声提取、离心、浓缩等前处理条件,并通过性能指标和实际样品的测定,考察了实验方法的适用性。实验证明:采用优化后的方法进行实际样品前处理和配制校准曲线,除2,4-二甲基苯胺和4-硝基苯胺相关系数>0.995外,其余化合物相关系数均在0.999以上。12种苯胺类化合物方法检出限为1~4μg/kg,低、中、高空白加标样品相对标准偏差为1.5%~7.3%,加标回收率为76.2%~110%。该方法条件明确,操作简便,在检测土壤样品时,可提高样品检测的准确性和工作效率。

The magnetic field is one of the most important parameters in solar physics,and a polarimeter is the key device to measure the solar magnetic *** crystals based Stokes polarimeter is a novel technology,and will be applied for magnetic field measurement in the first space-based solar observatory satellite developed by China,Advanced Space-based Solar ***,the liquid crystals based Stokes polarimeter in space is not a mature ***,it is of great scientific significance to study the control method and characteristics of the *** retardation produced by a liquid crystal variable retarder is sensitive to the temperature,and the retardation changes 0.09°per 0.10℃.The error in polarization measurement caused by this change is 0.016,which affects the accuracy of magnetic field *** order to ensure the stability of its performance,this paper proposes a high-precision temperature control system for liquid crystals based Stokes polarimeter in *** order to optimize the structure design and temperature control system,the temperature field of liquid crystals based Stokes polarimeter is analyzed by the finite element method,and the influence of light on the temperature field of the liquid crystal variable retarder is analyzed *** analyzing the principle of highprecision temperature measurement in space,a high-precision temperature measurement circuit based on integrated operational amplifier,programmable amplifier and 12 bit A/D is designed,and a high-precision space temperature control system is developed by applying the integral separation PI temperature control algorithm and PWM driving heating *** experimental results show that the effect of temperature control is accurate and stable,whenever the liquid crystals based Stokes polarimeter is either in the air or *** temperature stability is within±0.0150℃,which demonstrates greatly improved stability for the liquid crystals based Stokes polarim

随着山羊(Capra hircus)养殖业的发展，交通运输引起的山羊健康问题已不容忽视。本研究旨在探讨运输应激对赣西山羊胃壁组织细胞凋亡及凋亡相关基因B细胞淋巴瘤-2(B-cell lymphoma-2, Bcl-2)和Bcl-2相关X蛋白(Bcl-2 associated X protein, Bax)表达的影响。将12只1周岁的健康赣西公山羊随机均分为：对照组(n=4)、2 h运输组(n=4)、6 h运输组(n=4)。运输过程中禁食、禁水，待运输结束后收集山羊各胃壁的组织并进行相关的组织学和定量分析。末端脱氧核苷酸转移酶介导的dUTP生物素缺口末端标记法(terminal dexynucleotidyl transferase-mediated dUTP-biotin nick end labeling, TUNEL)结果显示，与对照组相比，2 h运输组和6 h运输组山羊胃壁组织的细胞凋亡率发生了显著升高(P<0.05)，并且除瓣胃外其余胃壁组织6 h运输组细胞凋亡率显著高于2 h运输组(P<0.05)；免疫组织化学结果显示，Bax和Bcl-2主要定位于前胃(瘤胃,网胃,瓣胃)的黏膜上皮和皱胃的固有层，与对照组相比，两运输组中Bax在各胃壁上述部位的表达增强，而Bcl-2的表达减弱(除瓣胃外)；qPCR结果显示，两运输组各胃壁组织中Bax的mRNA表达水平除瓣胃外均发生了显著上调(P<0.05)，而Bcl-2的mRNA表达水平均发生了显著下调(P<0.05),Bax/Bcl-2的比率发生了显著上调(P<0.05);Western blot结果也显示，Bax和Bcl-2的蛋白表达量也均发生了与mRNA相似的变化趋势(P<0.05)。结果表明，运输应激可通过调控Bax和Bcl-2的表达而促进山羊胃壁组织细胞发生凋亡。本研究可为改善运输应激对山羊器官组织损伤的方法提供理论依据。

AIM To evaluate the association of 12 tag single nucleotide polymorphisms(tag SNPs) in three onco-long non-coding RNA(lnc RNA) genes(HOTTIP,CCAT2,MALAT1) with the risk and prognosis of hepatocellular cancer(HCC). METHODS Twelve tag SNPs covering the three onco-lnc RNAs were genotyped by the KASP method in a total of 1338 samples,including 521 HCC patients and frequencymatched 817 controls. The samples were obtained from an unrelated Chinese population at the First Hospital ofChina Medical University from 2012-2015. The expression quantitative trait loci(e QTL) analyses were conducted to explore further the potential function of the promising SNPs. RESULTS Three SNPs in HOTTIP,one promoter SNP in MALAT1,and one haplotype of HOTTIP were associated with HCC risk. The HOTTIP rs17501292,rs2067087,and rs17427960 SNPs were increased to 1.55-,1.20-,and 1.18-fold HCC risk under allelic models(P = 0.012,0.017 and 0.049,respectively). MALAT1 rs4102217 SNP was increased to a 1.32-fold HCC risk under dominant models(P = 0.028). In addition,the two-way interaction of HOTTIP rs17501292-MALAT1 rs619586 polymorphisms showed a decreased effect on HCC risk(P interaction = 0.028,OR = 0.30) and epistasis with each other. HOTTIP rs3807598 variant genotype showed significantly longer survival time in HBV negative subgroup(P = 0.049,HR = 0.12),and MALAT1 rs591291 showed significantly better prognosis in female and HBV negative subgroups(P = 0.022,HR = 0.37; P = 0.042,HR = 0.25,respectively). In the study,no significant effect was observed in e QTL analysis. CONCLUSION Specific lnc RNA(HOTTIP and MALAT1) SNPs have potential to be biomarkers for HCC risk and prognosis.

正交胶合木(CLT)是一种适合于建筑工业化的人工绿色材料,具有可预制、可再生、自重轻但强度和刚度较高等特点,有较好的环境效应。如果能够在正交胶合木基础上加上预应力,则该组合结构受力性能将会得到进一步提升。主要对不同初始张拉力下预应力-正交胶合木组合结构进行试验研究:考虑预应力钢绞线的2种不同初始张拉力,设计了2个CLT剪力墙构件进行拟静力往复加载试验,得到了2个构件的骨架曲线、刚度退化曲线、等效粘滞阻尼系数、变形模式等内容;同时,通过将试验结果与国内相关研究成果进行对比分析,发现施加预应力对CLT结构整体刚度、耗能能力等有较大提升;相比于预应力-木组合结构组装于钢底梁上,预应力-木组合结构组装在混凝土底梁上在一定程度上减少了钢节点的使用,缩小了两者之间的刚度差,使得混凝土底梁情况下构件屈服荷载、屈服位移以及最大承载力方面有一定的提升。
