E-DNA scaffold sensors and the reagentless, single- step, measurement of HIV-diagnostic antibodies in human serum

The multiplexed,point-of-care measurement of specific antibodies could improve the speed with which diseases are diagnosed and their treatment *** this end,we are developing E-DNA scaffold sensors,which consist of a rigid,nucleic acid“scaffold”attached on one end to an electrode and presenting both a redox reporter and an epitope on the *** the absence of antibody,the reporter efficiently transfers electrons when interrogated ***-induced steric hindrance limits movement,reducing electron transfer in a manner that is both easily measured and quantitatively related to target *** we have used monoclonal antibodies to explore the analytical performance of E-DNA sensors,showing that they support the rapid,single-step,quantitative detection of multiple antibodies in small volume ***,in contrast,we employ authentic human samples to better explore the platform’s clinical ***,we developed E-DNA sensors targeting three HIV-specific antibodies and then compared the analytical and clinical performance of these against those of gold standard serological *** so we find that,although the multistep amplification of an ELISA leads to a lower detection limits,the clinical sensitivity of ELISAs,E-DNA sensors and lateral-flow dipsticks are indistinguishable across our test *** thus appears that,by merging the quantitation and multiplexing of ELISAs with the convenience and speed of dipsticks,E-DNA scaffold sensors could significantly improve on current serological practice.

Background Alzheimer’s disease(AD)is a progressive multifaceted neurodegenerative disorder for which no disease-modifying treatment *** is central to the pathology progression,with evidence suggesting that microglia-released galectin-3(gal3)plays a pivotal role by amplifying neuroinflammation in ***,the possible involvement of gal3 in the disruption of neuronal network oscillations typical of AD remains *** Here,we investigated the functional implications of gal3 signaling on experimentally induced gamma oscillations ex vivo(20-80 Hz)by performing electrophysiological recordings in the hippocampal CA3 area of wild-type(WT)mice and of the 5×FAD mouse model of *** addition,the recorded slices from WT mice under acute gal3 application were analyzed with RT-qPCR to detect expression of some neuroinflammation-related genes,and amyloid-β(Aβ)plaque load was quantified by immunostaining in the CA3 area of 6-month-old 5×FAD mice with or without Gal3 knockout(KO).Results Gal3 application decreased gamma oscillation power and rhythmicity in an activity-dependent manner,which was accompanied by impairment of cellular dynamics in fast-spiking interneurons(FSNs)and pyramidal *** found that the gal3-induced disruption was mediated by the gal3 carbohydrate-recognition domain and prevented by the gal3 inhibitor TD139,which also prevented Aβ42-induced degradation of gamma ***-more,the 5×FAD mice lacking gal3(5×FAD-Gal3KO)exhibited WT-like gamma network dynamics and decreased Aβplaque *** We report for the first time that gal3 impairs neuronal network dynamics by spike-phase uncoupling of FSNs,inducing a network performance ***,our findings suggest gal3 inhibition as a potential therapeutic strategy to counteract the neuronal network instability typical of AD and other neurological disorders encompassing neuroinflammation and cognitive decline.

Background:Cities differ from non-urban environments by the intensity,scale,and extent of anthropogenic pres-sures,which can drive the occurrence,physiology,and behavior of the organisms thriving in these *** as green cover often predict the occurrence patterns of bird species in urban ***,anthropogenic noise and artificial light at night(ALAN)could also limit the presence and disrupt the behavior of ***,there is still a dearth of knowledge about the influence of urbanization through noise and light pollution on nocturnal bird species *** this study,we assessed the role of green cover,noise,and light pollution on the occurrence and vocal activity of the Mottled Owl(Ciccaba virgata)in the city of Xalapa(Mexico).Methods:We obtained soundscape recordings in 61 independent sites scattered across the city of Xalapa using autonomous recording *** performed a semi-automated acoustic analysis of the recordings,corroborating all Mottled Owl *** calculated two measures of anthropogenic noise at each study site:daily noise(during 24 h)and masking noise(mean noise amplitude at night per site that could mask the owl’s vocalizations).We further performed generalized linear models to relate green cover,ALAN,daily noise,and masking noise in relation to the owl’s occurrence(i.e.,detected,undetected).We also ran linear models to assess relationships among the beginning and ending of vocal activity with ALAN,and with the anthropogenic and masking noise levels at the moment of which vocalizations were ***,we explored variations of the vocal activity of the Mottled Owl measured as vocalization rate across ***:The presence of Mottled Owls increased with the size of green cover and decreased with increases in both artificial light at night and noise *** the temporal scale,green cover was positively related with the ending of the owl’s vocal activity,while daily noise and ALAN levels were not related to the tim

We present,to the best of our knowledge,the first implementation of full-field quantum optical coherence tomography(FF-QOCT).In our system,we are able to obtain full three-dimensional(3D)information about the internal structure of a sample under study by relying on transversely resolved Hong–Ou–Mandel(HOM)interferometry with the help of an intensified CCD(ICCD)*** system requires a single axial scan,obtaining full-field transverse single-photon intensity in coincidence with the detection of the sibling photon for each value of the signal-idler temporal *** believe that this capability constitutes a significant step forward toward the implementation of QOCT as a practical biomedical imaging technique.

A novel porous and crystalline two-dimensional(2D)electrochemically active covalent organic framework(COF)based on ortho-quinone units has been prepared as an innovative approach towards the development of organic cathode materials with multiple redox sites as an efficient electrocatalyst for the oxygen reduction reaction(ORR).In contrast with most of the previously reported COFs as electrocatalysts for the ORR,the electrocatalytic application of this material towards ORR has been investigated without adding any metal or conductive supporting material and avoiding any additional carbonization ***,the electrochemical properties of the COF material have been compared with two analogue amorphous frameworks with similar chemical composition,which points out the important role of the enhanced crystallinity and porosity of the COF network in its superior performance as an electrocatalyst towards ORR.

Medical schools that received the PROMED (program for the encouragement of curricular changes in medical courses)—Preconditions and change process

Since the Curriculum Guidelines (CG) which were created in 2001 and PROMED (Program for the Encouragement of Curricular Changes in Medical Courses), several schools have applied for this incentive. Medical Schools (MS) have faced important changes in their curriculum throughout the years. The purpose is to verify if medical schools that received PROMED already had a historical of curricular changes. Several interviews, which were carried out with PROMED coordinators, were recorded, transcribed and analyzed according to the Bardin’s content analysis technique. Later, these interviews were later categorized into nine themes. This paper analyses the Medical School History category which encompasses three subcategories: existence of a former changing history;need to overcome the model and changing period. It is highlighted that medical schools that received PROMED had a previous changing historical which was sometimes located in anti-hegemonic niches. It is noticeable, however, that such changes were not enough to affect the comprehension of healthillness process and its consequent results. PROMED was, effectively, the reference for medical schools implementing the C.G. which represents, now, a benchmarking for all new and old schools in the country.

The Brazilian public healthcare system and its participation in medical training

Introduction: In Brazil, in a joint initiative of the Ministries of Health and of Education and Culture, the Program for the Promotion of Changes in Medical School Curricula (PROMED) was created, in order to give financial support to medical schools for the development and implementation of curricular changes to the medicine courses. Objective: To ascertain the influence of PROMED on the interaction between the Brazilian public healthcare system (SUS) and the universities. Method: Using the qualitative method by way of content analysis of the Bardin, interviews with 19 coordinators of the medicine courses which received financial support from PROMED were carried out and analyzed. Results: Subcategories which express the current relation between SUS and the universities were found: the need to train SUS professionals and the teaching staff involved, the lines of research directed toward SUS and partnership relations as part of the student learning process, the strengthening of SUS and the internal problems. Conclusions: Observing the needs of the relationship between SUS and the curricular changes it was noted that the public health services are of extreme importance as part of the training process of the medical student. Besides favoring practice, it establishes student activities within the health services, the evaluation of the policies, planning and management of the health services in activities of training and social communication in healthcare, linked to community organizations or diverse social entities. Thus, problems associated with the healthcare service can be identified, as well as the health conditions and life style of the population.

FPGA芯片的应用越趋普及，因为这种芯片可以提高系统设计的灵活性，例如可以轻易为系统增添功能或利用代码修改设计，所以许多系统设计工程师都喜欢采用FPGA。但要为FPGA提供足够的供电便要面对一些挑战。首先要解决的是多条供电干线的问题。一般来说，供电系统必须为FPGA芯片提供最少两个供电，一个是内核的供电，此外，输入／输出群组也必须另有供电（可能超过一个）。但这只是最基本的要求，内置FPGA的系统可能还要另外加设更多供电干线，以便为DDR存储器、收发器、以太网物理层芯片（PHY）、模拟／数字转换器或小型微控制器等提供供电。除此之外，这些供电干线也必须符合一些特别的要求，例如输出不可超过1．25V、单调升压、供电排序及上升时间必须受控等。

Effects of Silicon Nanoparticles on the Transient Liquid Phase Bonding of 304 Stainless Steel

Transient liquid phase （TLP） bonding of 304 stainless steel with nickel based filler metal, BNi-9, was performed to study the influence of silicon nanoparticles （NPs） on the mechanical and structural properties of the bonding area. It was found that silicon NPs act as a melting point depressant in the brazing process; the formation of silicon TLP induces the dissolution of elements of the metal filler and promotes a uniform distribution in the bonding area. Silicon NPs induce the development of smaller eutectic structures in the melting zone; it has been related to microhardness measurements, which are lower when the silicon NPs are used in the brazing process.
