Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

CuO nanoparticles were successfully synthesized via a two-jet electrospun method,and then screen-printed on silver-carbon electrodes,forming CuO-modified Ag-C(CuO/Ag-C)disposable strip *** natural environment condition for glucose detection,the obtained CuO/Ag-C electrodes show a high sensitivity of 540 nA·mM^(-1)·cm^(-2),and a low limit of detection(0.68 mM)in a wide linear response range of 0.68 mM and 3 mM(signal/noise=3),*** addition,the CuO/Ag-C electrodes also exhibit excellent anti-interference,air stability and *** a result,the fabrication of CuO nanoparticles via an electrospun process and the technique of screen-printed electrodes are of great significance for glucose detection.

Nitrogen addition promotes soil carbon accumulation globally

Soil is the largest carbon(C) reservoir in terrestrial ecosystems and plays a crucial role in regulating the global C cycle and climate *** nitrogen(N) deposition has been widely considered as a critical factor affecting soil organic carbon(SOC) storage, but its effect on SOC components with different stability remains unclear. Here, we analyzed extensive empirical data from 304 sites worldwide to investigate how SOC and its components respond to N addition. Our analysis showed that N addition led to a significant increase in bulk SOC(6.7%), with greater increases in croplands(10.6%) and forests(6.0%) compared to grasslands(2.1%). Regarding SOC components, N addition promoted the accumulation of plant-derived C(9.7%–28.5%) over microbial-derived C(0.2%), as well as labile(5.7%) over recalcitrant components(–1.2%), resulting in a shift towards increased accumulation of plant-derived labile C. Consistently, N addition led to a greater increase in particulate organic C(11.9%) than mineral-associated organic C(3.6%), suggesting that N addition promotes C accumulation across all pools, with more increase in unstable than stable pools. The responses of SOC and its components were best predicted by the N addition rate and net primary productivity. Overall, our findings suggest that N enrichment could promote the accumulation of plant-derived and non-mineral associated C and a subsequent decrease in the overall stability of soil C pool, which underscores the importance of considering the effects of N enrichment on SOC components for a better understanding of C dynamics in soils.

Ambient air temperature effects on the temperature of sewage sludge composting process

Using data obtained with a full-scale sewage sludge composting facility, this paper studied the effects of ambient air temperature on the composting temperature with varying volume ratios of sewage sludge and recycled compost to bulking agent. Two volume ratios were examined experimentally, 1 ： 0 ： 1 and 3 ： 1 ： 2. The results show that composting temperature was influenced by ambient air temperature and the influence was more significant when composting was in the temperature rising process： composting temperature changed 2.4-6.5℃ when ambient air temperature changed 13℃. On the other hand, the influence was not significant when composting was in the high-temperature and/or temperature falling process： composting temperature changed 0.75-1.3℃ when ambient air temperature changed 8-15℃. Hysteresis effect was observed in composting temperature＇s responses to ambient air temperature. When the ventilation capability of pile was excellent（at a volume ratio of 1：0：1）, the hysteresis time was short and ranging 1.1-1.2 h. On the contrary, when the proportion of added bulking agent was low, therefore less porosity in the substrate（at a volume ratio of 3：1：2）, the hysteresis time was IonQ and ranging 1.9-3.1 h.

Due to the lack of accurate data and complex parameterization,the prediction of groundwater depth is a chal-lenge for numerical *** learning can effectively solve this issue and has been proven useful in the prediction of groundwater depth in many *** this study,two new models are applied to the prediction of groundwater depth in the Ningxia area,*** two models combine the improved dung beetle optimizer(DBO)algorithm with two deep learning models:The Multi-head Attention-Convolution Neural Network-Long Short Term Memory networks(MH-CNN-LSTM)and the Multi-head Attention-Convolution Neural Network-Gated Recurrent Unit(MH-CNN-GRU).The models with DBO show better prediction performance,with larger R(correlation coefficient),RPD(residual prediction deviation),and lower RMSE(root-mean-square error).Com-pared with the models with the original DBO,the R and RPD of models with the improved DBO increase by over 1.5%,and the RMSE decreases by over 1.8%,indicating better prediction *** addition,compared with the multiple linear regression model,a traditional statistical model,deep learning models have better prediction performance.

Age-stage, two-sex life tables of Bactrocera cucurbitae （Coquillett）（Diptera： Tephritidae） with a discussion on the problem of applying female age-specific life tables to insect populations

Age-stage, two-sex life tables of the melon fly, Bactrocera cucurbitae （Coquil- let-t）（Diptera： Tephritidae）, reared on cucumber （Cucumis sativus L.）, sponge gourd （Luffa cylindrica Roem） and a carrot medium （mashed Daucus carota L. mixed with sucrose and yeast hydrolysate） were constructed under laboratory conditions at 25 4- loc, 65% 4- 0.5% relative humidity, and a photoperiod 12 ： 12h （L ： D）. The intrinsic rates of increase of B. cucurbitae were 0.144 6, 0.141 2 and 0.068 8 days on cucumber, sponge gourd, and carrot medium, respectively. The highest net reproduction rate was 172 offspring per fly reared on sponge gourd. The mean generation times ofB. cucurbitae ranged from 34 days reared on cucumber to 56 days reared on carrot medium. The life history raw data was analyzed using the traditional female age-specific life table and compared to results obtained using the age-stage, two-sex life table. When the age-specific female life table is applied to an age-stage-structured two-sex population, survival and fecundity curves will be improperly manipulated due to an inability to include variation in preadult development time. We discussed different interpretations of the relationship between the net reproductive rate and the intrinsic rate of increase to clarify possible misunderstanding in the literature.

Effects of microstructure on uniaxial strength asymmetry of open-cell foams

Based on the elongated Kelvin model, the effect of microstructure on the uniaxial strength asymmetry of open-cell foams is investigated. The results indicate that this asymmetry depends on the relative density, the solid material, the cell morphology, and the strut geometry of open-cell foams. Even though the solid material has the same tensile and compressive strength, the tensile and compressive strength of open-cell foams with asymmetrical sectional struts are still different. In addition, with the increasing degree of anisotropy, the uniaxial strength as well as the strength asymmetry increases in the rise direction but reduces in the transverse direction. Moreover, the plastic collapse ratio between two directions is verified to depend mainly on the cell morphology. The predicted results are compared with Gibson and Ashby＇s theoretical results as well as the experimental data reported in the literature, which validates that the elongated Kelvin model is accurate in explaining the strength asymmetry presented in realistic open-cell foams.

Automatic context induction for tone model integration in mandarin speech recognition

Tone model （TM） integration is an important task for mandarin speech recognition. It has been proved to be effective to use discriminatively trained scaling factors when integrating TM scores into multi-pass speech recognition. Moreover, context-dependent （CD） scaling can be applied for better interpolation between the models. One limitation of this approach is a large number of parameters will be introduced, which makes the technique prone to overtraining. In this paper, we propose to induce context-dependent model weights by using automatically derived phonetic decision trees. Question at each tree node is chosen to minimize the expected recognition error on the training data. First order approximation of the minimum phone error （MPE） objective function is used for question pruning to make tree building efficient. Experimental results on continuous mandarin speech recognition show the method is capable of inducing the most crucial phonetic contexts and obtains significant error reduction with far fewer parameters, compared with that obtained by using manually designed context-dependent scaling parameters.

Application of split Hopkinson tension bar technique to the study of dynamic fracture properties of materials

A novel approach is proposed in determining dy- namic fracture toughness （DFT） of high strength steel, using the split Hopkinson tension bar （SHTB） apparatus, com- bined with a hybrid experimental-numerical method. The center-cracked tension specimen is connected between the bars with a specially designed fixture device. The fracture initiation time is measured by the strain gage method, and dynamic stress intensity factors （DSIF） are obtained with the aid of 3D finite element analysis （FEA）. In this approach, the dimensions of the specimen are not restricted by the connec- tion strength or the stress-state equilibrium conditions, and hence plane strain state can be attained conveniently at the crack tip. Through comparison between the obtained results and those in open publication, it is concluded that the ex- perimental data are valid, and the method proposed here is reliable. The validity of the obtained DFT is checked with the ASTM criteria, and fracture surfaces are examined at the end of paper.
