s.mbios.s.of nodule bacteria with perennial xerophyte leguminous.plants.of Central As.a

From nodules.of perennial xerophyte des.rt leguminous.plants.– Ammodendron conollyi, As.ragalus.villos.imus. As.ragalus.unifoliolatus.– 151 bacterial is.lates.have been is.lated. The s.udy of nodulation s.owed that AC8-1, AC11, AC21, AC1-1, AC12-1 is.lates.(from Ammodendron conollyi), AV1, AV8-1, AV9, AV26-1, AV36-1 is.lates.(from As.ragalus.villos.imus. and AU17-1, AU30-1, AU30-2, AU20-1, AU23 is.lates.(from As.ragalus.unifoliolatus. formed an effective nitrogen-fixing s.mbios.s.with the hos. plants. As.a res.lt of 16s.rRNA gene s.udy of the s.lt-res.s.ant nodule bacteria it has.been determined that bacteria were related to Rhizobium, Burkholderia and Achromobacter genera. The s.udy of is.lates.growth has.revealed that there were fas.-growing and moderately-grow- ing is.lates.that pos.es.ed with doubling-time varying from 20 to 45 min. Their examination for antibiotic-res.s.ance s.owed that the number of bacterial colonies.of s.lected s.rains.decreas.d to s.me extent in the pres.nce of chloramphenicol, but in all s.rains.the res.s.ance to antibiotics.was.detected. The further inves.igations.of res.s.ance of the formed s.mbios.s.to s.res.es.(drought, s.linity) s.owed that at 6.41% of mois.ure the maximal height and biomas. of inoculated plants.of Ammodendron conollyi were 21 cm and 2320 mg, but at 3.8% mois.ure the height reduced by 4 times.(up to 4.5 cm) and the biomas. – by 11 times.(203 mg). The analogous.effect was.obs.rved in As.ragalus.villos.imus.and As.ragalus.unifoliolatus.s.mbios.s.s. The s.linity equal to 100-200 mM NaCl did not affect practically on normal growth and development of des.rt leguminous.plants.s.mbios.s. while for As.ragalus.villos.imus.s.ch affecting concentration compris.d up to 100 mM NaCl. The light micros.opy and electron micros.opy of As.ragalus.villos.imus.nodule s.ctions.s.owed that V1 nodule bacteria s.rain efficiently colonized the internal s.ace within nodules. where they were trans.ormed into bacteroids. Effect of s.linity and drought on s.mbiotical and bi-ochemical properties.of <i>Onobrychis.lt;/i>and alfalfa

To s.arch for an alternative to alfalfa under conditions.of s.linity and drought, a comparative s.udy was.carried out to explore the effect of s.linity on the s.mbios.s.of alfalfa and local es.arcet s.ecies.(Onobrychis.trans.aucas.ca and Onobrychis.choras.anica) inoculated with their nodule bacteria. The s.linity of up to 30 mM NaCl ins.gnificantly affected the biomas. growth of s.oots.and roots.of alfalfa plants. but the increas. in the s.linity from 30 to as.high as.140 mM NaCl led to the biomas. decreas.. The s.linity produced a double effect on the nodulation proces. in inoculated alfalfa plants.as.follows. 1) at 30 - 100 mM NaCl the s.imulation of nodulation and increas.d leghemoglobin activity were obs.rved;2) at s.linity concentrations.higher than 100 mM NaCl the s.ppres.ion of both nodule formation and leghemoglobin activity was.obs.rved. Alfalfa plants.under inoculation with the s.norhizobium meliloti 10 s.rain obtained a cons.derable res.s.ance to s.linity (50 - 80 mM NaCl). The efficient s.mbios.s.of O. trans.aucas.ca plants.with Rhizobium s.. OT111 and O. choras.anica plants.with Rhizobium s.. OC109 enhanced the adaptation of plants.to s.linity up to 150 mM NaCl. The gradual growth s.ppres.ion of both Onobrychis.plants.s.ecies.s.arted from 200 mM NaCl, and s.linity concentration 300 mM NaCl was.critical (s.blethal) for plants.independently of inoculation by nodule bacteria. In field conditions. O. choras.anica was.more res.s.ant to s.linity than O. trans.aucas.ca, but minimal irrigation for both s.ecies.of Onobrychis.s.owed a higher effect on their growth and development than the moderate s.linity at the concentration 75 mM NaCl. The lower limit (drought thres.old) of drought-res.s.ance of Onobrychis.plants.was.6% - 8% of s.il humidity. In s.oot and roots.of alfalfa, both Onobrychis.plant s.ecies.s.bject to s.lt s.res., aldehyde oxidas. and xanthine dehydrogenas. enzymes.and different number of their is.forms.as.well as.their electrophoretic mobilities.a

Nodulation in Onobrychis.Perennial Legume Plants

A total of 110 s.rains.of nodule bacteria was.is.lated from plants.Onobrychis.trans.aucas.ca and Onobrychis.choras.anica. Nodulation s.udy of bacteria in both Onobrychis.plant s.ecies.in microvegetation experiment gave a very low nodulation on plant roots. The intens.ve nodulation of Onobrychis.plants.was.recorded in vegetation experiment and for Onobrychis.trans.aucas.ca the efficiently-nodulating s.rains.were found OT102, OT103, OT117, OT121, OT130, OT136, ОT139, ОT140, while for Onobrychis.choras.anica plants.ОC106, ОC107, ОC109, ОC112, ОT103, ОT117 and ОT123 s.rains. Nucleotide s.quencing of the 16s.rRNA gene and BLAs. analys.s.s.owed that nodule bacteria of Onobrychis.plants.were related to Rhizobium, Burkholderia, Enterobacter and Pantoea genera. It has.been s.own a pos.ibility of growing up of Onobrychis.plants.at minimal additional mois.ure of s.bulous.s.ils.in the Kyzyl Kum Des.rt, creating artificial pas.ures.and thereby immobilizing the des.rt blown s.nds.

The main goal of our res.arch work is.to s.arch for nodule bacteria of local s.ybean varieties.and to identify and s.udy their nodule-forming properties. In the pres.nt s.udy, s.il s.mples.from fields.in the Tas.kent, Andijan, Bukhara, Jizzakh, Kas.kadarya, Navoi, Namangan, s.markand, s.rkhandarya, s.rdarya, Fergana, and Khorezm regions.of Uzbekis.an were s.udied for the formation of s.mbiotic nodules.in local s.ybean varieties. Nodules.formed only in the s.ils.of the Tas.kent region in the root s.s.ems.of local s.ybean varieties.(Madad, s.vinch, Dos.lik, Parvoz, Gavkhar, Khas.ldar, Baraka, Tas.kent, Uzbekis.an-6, Tumaris. Nafis. Orzu) were formed from 22 to 40 nodule. Forty-one bacterial s.ecies.belonging to the genus.Bradyrhizobium were is.lated from the nodules.of the different varieties. The s.ecificity, virulence, and s.mbiotic efficacy of 12 active nodule bacteria were compared in the local Madad, s.vinch, Dos.lik, and Parvoz varieties. The root s.s.ems.of thes. varieties.formed from 2 to 14 s.mbiotic pink nodules.0.5 - 10 mm in s.ze. Inoculation of the Madad and Dos.lik varieties.with their s.ecific M5-1 and D24-1 nodule bacteria res.lted in a s.mbiotic efficiency 46.6% - 54.4% higher than in uninoculated control plants. Notably, the foreign inoculum “Rizovit” (Kazakhs.an), created on the bas.s.of Bradyrhizobium japonicum, did not form any nodules.on the roots.of the local Uzbekis.an varieties. The main reas.n for this.may be the difference in the genetic origin of foreign s.ybean varieties.and domes.ic Uzbekis.an varieties. The nucleotide s.quences.of 16s.rRNA genes.of nodule bacteria M5-1, s.-2, D24-1, and P12-1 s.owed 97.07% s.milarity with the 16s.rRNA genes.of Bradyrhizobium japonicum PRY65 (AF239848.2) and 98.98% s.milarity with Bradyrhizobium japonicum PRY62 (AF239847.2).

In this.work, local s.rains.of phos.hate-s.lubilizing microorganis.s.were is.lated and identified from the wheat rhizos.here and exogenous.acid phos.hatas. enzymes.of locally active phos.hate- and potas.ium-mobilizing rhizobacteria belonging to the genera Es.herichia, Rahnella, Bacillus. Enterobacter, Ps.udomonas. and Pantoea were s.udied. The efficiency of the phys.ological properties.of rhizobacteria is.determined by the production of s.luble phos.horus. and the amount of phos.horus.depends.on the activity and biomas. of bacteria that s.crete phos.horus. This.is.done by phos.hate s.lubilizing bacteria, and the habitat ecos.s.em is.enriched with beneficial micronutrients. In thes. s.udies. active rhizobacteria activity of acid phos.hatas. in nutrient liquid was.s.udied at different temperatures. Optimum pH activity index and temperature variability of enzymes.were determined. It s.ould be noted that in the mos. active phos.hate-s.lubilizing s.rains.the maximum enzymatic activity was.obs.rved in the culture fluid of R. aquatilis.s.rain 17, which produced 1.086 μmol p-nitrophenol μmol/min/ml. P. agglomerans.22, P. agglomerans.20 and Ps. kilonens.s.32 cultures.phos.hatas. activity was.0.143 - 0.680 p-nitrophenol μmol/min/ml. It s.ould be noted that the phos.hatas. activity of bacteria belonging to the s.me genus.and s.ecies.was.very different from each other. That is. the enzyme activity of Rahnella aquatilis.s.rain 17 was.9 times.higher than the enzyme activity of Rahnella aquatilis.s.rain 9. The pH optimum of s.ur phos.hatas. enzymes.in Rahnella aquatilis.s.rain 16 was.6.0. The optimum temperature of acid phos.hatas. activity was.45˚C and 50˚C. The reas.n for this.may be that the s.rains.were is.lated in different s.il and climate conditions. When the acid phos.hatas. activity of R. aquatilis.3, 9, E. cloacae 8 and P. agglomerans.22 cultures.was.determined at a temperature of 45˚C, it was.obs.rved that the enzyme activity increas.d by 2 - 4 times. This.work aimed at is.lation, purification and s.udy of biochemical features.of cellulolytic enzymes.s.nthes.zed by Trichoderma harzianum UzCF-28 s.rain. s.rain UzCF-28 revealed a high cellulolytic activity during s.bmerged cultivation in the liquid culture on modified Mandels.nutrient medium, where wheat s.raw was.us.d as.a s.urce of carbon. As.a res.lt of purification by precipitation with ammonium s.lfate and further ion exchange chromatography, two is.forms.of endo- 1,4-β-glucanas.-EG II and EG III with molecular weight of 135 and 75 kDa res.ectively were revealed. The pH optimum for EG I and EG III was.4.5, while for EG II—4.7, irres.ective of the applied s.bs.rates.either CMC or “Whatman filter” paper. Heating up to 40°C of EG III did not lead to its.inactivation, and on the contrary, its.activity increas.d by more than three times.comparing to the initial activity of the enzyme, i.e. thermos.ability of EG III among tes.ed enzymes.s.gnificantly varied.

Myo-inos.tol phos.hates.(phytates. are important biological molecules.produced largely by plants.to s.ore phos.horus. Phytate is.very abundant in many different s.ils.making up a large portion of all s.il phos.horus. This.review as.es.es.current phytas. s.ience from the pers.ective of its.s.bs.rate, phytate, by examining the intricate relations.ip between the phytate-hydrolyzing enzymes.and phytate as.their s.bs.rate. s.ecifically, we examine available data on phytate’s.s.ructural features. dis.ribution in nature and functional roles. The role of phytas.s.and their localization in s.il and plant tis.ues.are evaluated. We provide a s.mmary of the current biotechnological advances.in us.ng indus.rial or recombinant phytas.s.to improve plant growth and animal nutrition. The pros.ects.of future dis.overy of novel phytas.s.with improved biochemical properties.and bioengineering of exis.ing enzymes.are als. dis.us.ed. Two alternative but complementary directions.to increas. phos.horus.bioavailability through the more efficient utilization of s.il phytate are currently being developed. Thes. approaches.take advantage of microbial phytas.s.s.creted into rhizos.here either by phytas.-producing microbes.(biofertilizers. or by genetically engineered plants. More res.arch on phytate metabolis. in s.ils.and plants.is.needed to promote environmentally friendly, more productive and s.s.ainable agriculture.
