The formation of Ca^(2+) gradients at the cleavage furrows during cytokinesis of Zebrafish embryos

In dividing embryos,a localized elevation in intracellular Ca^(2+)([Ca^(2+)]i)at the cleavage furrow has been shown to be essential for ***,the underlying mechanisms for generating and maintaining these[Ca^(2+)]_(i) gradients throughout cytokinesis are not fully *** the present study,we analyzed the role of inositol 1,4,5-trisphosphate receptors(IP3Rs)and endoplasmic reticulum(ER)distribution in determining the intracellular Ca^(2+) gradients in early zebrafish *** of the injected Ca^(2+) indicator,Indo-1,showed that during the first cell division a standing Ca^(2+) gradient was formed~35 min after fertilization,with the[Ca^(2+)]_(i) spatially decaying from 500–600 nmol/L at the cleavage furrow to 100–200 nmol/L around the *** the IP3R immunohistochemical fluorescence was relatively concentrated in the peri-furrow region,ER labeling was relatively enriched in both peri-furrow and peri-nuclear *** simulation suggested that a divergence in the spatial distribution of IP3R and the locations of Ca^(2+) uptake within the ER was essential for the formation of a standing Ca^(2+) gradient,and the Ca^(2+) gradient could only be well-established under an optimal stoichiometry of Ca^(2+) uptake and ***,while inhibition of IP3R Ca^(2+) release blocked the generation of the Ca^(2+)gradient at a lower[Ca^(2+)]_(i) level,both Ca^(2+) release stimulation by inositol 1,4,5-trisphosphate(IP3)injection and ER Ca^(2+) pump inhibition by cyclopiazonic acid also eliminated the Ca^(2+) gradients at higher[Ca^(2+)]_(i) *** results suggest a dynamic relationship between ER-mediated Ca^(2+) release and uptake that underlies the maintenance of the perifurrow Ca^(2+) gradient and is essential for cytokinesis of zebrafish embryos.

Improving the Seasonal Forecast of Summer Precipitation in Southeastern China Using a CycleGAN-based Deep Learning Bias Correction Method

Accurate seasonal precipitation forecasts,especially for extreme events,are crucial to preventing meteorological hazards and their potential impacts on national development,social activity,and ***,the intensity of summer precipitation is often largely underestimated in many current dynamic *** study uses a deep learning method called Cycle-Consistent Generative Adversarial Networks(CycleGAN)to improve the seasonal forecasts for June-JulyAugust precipitation in southeastern China by the Nanjing University of Information Science and Technology Climate Forecast System(NUIST-CFS 1.0).The results suggest that the CycleGAN-based model significantly improves the accuracy in predicting the spatiotemporal distribution of summer precipitation compared to the traditional quantile mapping(QM)*** the unpaired bias-correction model,we can also obtain advanced forecasts of the frequency,intensity,and duration of extreme precipitation events over the dynamic model *** study expands the potential applications of deep learning models toward improving seasonal precipitation forecasts.

Disordered hyperuniformity and thermal transport in monolayer amorphous carbon

Disordered hyperuniformity(DHU)is a recently discovered novel state of amorphous systems characterized by strongly suppressed density fluctuations at large length scales as in crystalline materials,which offers great potential for achieving unusual mechanical,electronic,and photonic ***,despite the fundamental and technological importance of thermal transport in amorphous solids,the effect of DHU remains largely ***,we theoretically study thermal transport in a class of two-dimensional DHU materials—monolayer amorphous carbon(MAC).Beginning with a perfect graphene lattice,we continuously apply Stone-Wales transformations to generate a series of MAC models with varied degrees of disorder and defects,which are quantified through comprehensive structural analysis including the so-called hyperuniformity index(H),where a smaller H indicates a higher degree of ***,we conduct molecular dynamics simulations to obtain the thermal conductivity(κ).A significant correlation between the thermal transport behavior and degree of hyperuniformity is observed,with the room-temperatureκdecreasing from 26.3 to 5.3 W m^(-1)K^(-1)while H is tuned from 0.0004 to ***,two distinct transport regimes are identified,including a nearly-DHU regime at small H(<0.005)whereκdrops sharply and a non-DHU region at larger H whereκbecomes relatively ***-resolved analysis reveals longer lifetime and higher participation ratio for the heat carriers in nearly-DHU MAC,implying that the hidden long-range correlations could support extended modes that enhance *** work highlights the impact of DHU on the thermal properties of amorphous materials and represents a conceptual advancement that is worthy of future exploration.

Objective:To investigate the protective effects of stir-fried Semen Armeniacae Amarum(SAA)against aristolochic acidⅠ(AAⅠ)-induced nephrotoxicity and DNA adducts and elucidate the underlying mechanism involved for ensuring the safe use of Asari Radix et ***:In vitro,HEK293T cells overexpressing Flag-tagged multidrug resistance-associated protein 3(MRP3)were constructed by Lentiviral transduction,and inhibitory effect of top 10 common pairs of medicinal herbs with Asari Radix et Rhizoma in clinic on MRP3 activity was verified using a self-constructed fluorescence screening *** m RNA,protein expressions,and enzyme activity levels of NAD(P)H quinone dehydrogenase 1(NQO1)and cytochrome P4501A2(CYP1A2)were measured in differentiated Hepa RG *** toxicity after inhibition of AAⅠmetabolite transport was detected using cell counting kit-8 *** vivo,C57BL/6 mice were randomly divided into 5 groups according to a random number table,including:control(1%sodium bicarbonate),AAⅠ(10 mg/kg),stir-fried SAA(1.75 g/kg)and AAⅠ+stir-fried SAA(1.75 and 8.75 g/kg)groups,6 mice in each *** 7 days of continuous gavage administration,liver and kidney damages were assessed,and the protein expressions and enzyme activity of liver metabolic enzymes NQO1 and CYP1A2 were determined ***:In vivo,combination of 1.75 g/kg SAA and 10 mg/kg AAⅠsuppressed AAⅠ-induced nephrotoxicity and reduced d A-ALI formation by 26.7%,and these detoxification effects in a dose-dependent manner(P<0.01).Mechanistically,SAA inhibited MRP3 transport in vitro,downregulated NQO1 expression in vivo,increased CYP1A2 expression and enzymatic activity in vitro and in vivo,respectively(P<0.05 or P<0.01).Notably,SAA also reduced AAⅠ-induced hepatotoxicity throughout the detoxification process,as indicated by a 41.3%reduction in the number of liver adducts(P<0.01).Conclusions:Stirfried SAA is a novel drug candidate for the suppression of AAⅠ-induced liver and kidney dam

An improved GCN−TCN−AR model for PM_(2.5) predictions in the arid areas of Xinjiang,China

Specific spatiotemporal control of intracellular calcium concentration([Ca]) is crucial in regulating cell functions in eukaryotic cells.A local calcium elevation specifically localized to the cleavage furrow before and during furrow formation is essential for cell division in the early zebrafish embryo. The dynamics and function of this furrow-specific calcium signal are not fully understood. Here,we showed that the localized[Ca] change was normally in the range of 100 nM to 400 nM,and could be blocked by heparin, a IP3R blocker that inhibited calcium release from endoplasmic reticulum,the major intracellular calcium *** calcium elevation was spatially confined to the peri-furrow region. Immunostaining of IP3Rs and live-cell tracking of endoplasmic reticulum showed both IP3Rs and the ER calcium store were slightly enriched at the peri-furrow region. Blocking of calcium uptake of ER with cyclopiazonic acid（CPA） produced a homogenous calcium elevation in the animal pole of the embryo. This expanded calcium elevation did not prevent the initiation of the furrow,but lead to its regressionsoon after its *** results characterized the dynamics and function of the furrow-specific calcium elevation during cytokinesis of zebrafish *** spatial confinement of this calcium elevation was partially due to the spatial enrichment of internal calcium store and calcium release ***,our data suggested that the global calcium gradient produced by the local calcium signal was essential to maintain the furrow after its formation,but was not necessary for the process of furrow initiating.

The transformation of nor-seco-cucurbit[10]uril to cucurbit[5]uril and cucurbit[8]uril controlled by its own concentration

Nor-seco-cucurbit[10]uril(ns-CB[10]) is a kinetic product with unique structure. The single bridged methylene in its structure makes the molecular cavity of ns-CB[10] more deformable when compared to ordinary cucurbit[n]uril, reducing its structural stability. Repeated experiments showed that ns-CB[10] gradually cracks in an acidic solution and changes the specificity of cucurbit[5]uril(CB[5]) and cucurbit[8]uril(CB[8]) under more robust acidic solutions and when heated. A series of experiments were designed to study the transformation behavior of ns-CB[10]. It was found that the concentration of ns-CB[10] was correlated with the content distribution of CB[5] and CB[8]. This study explores the influencing factors and mechanisms of the transformation of ns-CB[10] to CB[5] and CB[8]. The results are of great significance for the application of ns-CB[10], understanding the formation mechanism of cucurbit[n]urils. Furthermore,it provides a new pathway for synthesizing new cucurbit[n]urils.

Gas-Solid Flow,Combustion Characteristics,and Gas Emissions in a 75 t/h CFB Boiler Based on the CPFD Method

A 3D simulation using Computational Particle Fluid Dynamics(CPFD)methods was used to calculate coal combustion in a 75 t/h industrial-scale circulating fluidized bed(CFB)*** characteristics,gas-solid flow characteristics,and gaseous pollutant emissions of CFB boilers from combustion ignition to stable operation were systematically evaluated in this *** show that the temperature distribution is relatively uniform throughout the *** the combustion process unfolds within the boiler,the gas composition curve strikingly portrays the inverse correlation between CO_(2)and O_(2)*** the combustion reaction progresses,it becomes evident that the concentration of CO_(2)progressively increases,while the concentration of O_(2)concurrently *** inverse relationship underscores the fundamental combustion reaction,where carbon-based fuels react with oxygen to produce carbon dioxide and release ***,a comprehensive analysis has revealed that,from ignition to stable combustion,both nitric oxide(NO)and sulfur dioxide(SO_(2))emissions exhibit a declining *** reduction in pollutant generation is attributed to the improvement in combustion *** complete combustion leads to lower levels of unburned hydrocarbons,which are prone to NO ***,the sulfur content in the fuel is more efficiently oxidized to sulfur trioxide(SO_(3))or bound in sulfates,reducing SO_(2)*** steady state in the simulation,the SO_(2)mass flow rate varies significantly with the furnace height,gradually increasing from 0.07 kg·s^(-1)at 4 m at the bottom of the furnace to a peak of 0.078 kg·s^(-1)at 8 m in the center,and then decreasing to 0.06 kg·s^(-1)at 20m at the top of the furnace.
