Near-infrared light(NIR)triggered transdermal drug delivery systems are of great interest due to their on-demand drug release,which enable to enhance drug treatment efficiency as well as reduce side ***,a NIR-triggered microneedle(MN)patch array has been fabricated through depositing the photothermal conversion agent and anti-diabetic drug-loaded polymer vesicles with upper critical solution temperature(UCST)into dissolvable polymer *** UCST-type polymer has a clearing point temperature of 41℃ and the drug-loaded polymer vesicles present excellent NIR-triggered and temperature responsive drug release behavior in vitro due to the disassociation of polymer vesicles upon NIR *** applying MNs to diabetic rats,significant hypoglycemic effect is achieved upon interval NIR irradiation and the blood glucose concentration can decrease to normal state for several hours,which enables to achieve the goal of on-demand drug *** work suggests that the NIR-triggered MN drug release device has a potential application in the treatment of diabetes,especially for those requiring an active drug release manner.

Polyacrylonitrile beads（PAN） cysteine（CS） was synthesized from polyacrylonitrile beads（PAN） and cysteine（CS）.The content of the functional group and the percentage conversion of the functional group of PAN-CS prepared under the optimum condition using response surface methodology（RSM） for the first attempt were 3.22 mmol/g and 35.78%.The structure was characterized by ET-IR and elemental *** adsorption properties of the resin for Cu（Ⅱ） were investigated by batch and column *** adsorption results suggested that PAN-CS had higher adsorption capability for Cu（Ⅱ）than other metal ions and maximum saturated adsorption capacity was 184.7 mg/*** resin and its metal complexes were studied by ***,the resin can be eluted easily using 1 mol/L ***-CS can provide a potential application for selective removal of copper from waste solution.

The effect of addition temperature of MgO particles(MgOp)on their dispersion behavior and the efficiency of grain refinement in AZ31 Mg alloy was *** addition,the grain refinement mechanism was systematically studied by microstructure characterization,thermodynamic calculation,and analysis of solidification *** results show that the grain size of AZ31 Mg alloy initially decreases and then increases as the MgOp addition temperature is increased from 720 to 810℃,exhibiting a minimum value of 136μm at 780℃.The improved grain refinement efficiency with increasing MgOp addition temperature can be attributed to the reduced Mg melt viscosity and enhanced wettability between MgOp and Mg ***,a corresponding physical model describing the solidification behavior and grain refinement mechanism was proposed.

Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

This study aimed to develop a pan-genotypic and multifunctional small interfering RNA(siRNA)against hepatitis B virus(HBV)with an efficient delivery system for treating chronic hepatitis B(CHB),and explore combined RNA interference(RNAi)and immune modulatory modalities for better viral *** synthetic siRNAs targeting consensus motifs distributed across the whole HBV genome were designed and *** lipid nanoparticle(LNP)formulation was optimized by adopting HO-PEG_(2000)-DMG lipid and modifying the molar ratio of traditional polyethylene glycol(PEG)lipid in LNP *** efficacy and safety of this formulation in delivering siHBV(tLNP/siHBV)along with the mouse IL-2(mIL-2)mRNA(tLNP/siHBVIL2)were evaluated in the rAAVHBV1.3 mouse model.A siRNA combination(terms“siHBV”)with a genotypic coverage of 98.55%was selected,chemically modified,and encapsulated within an optimized LNP(tLNP)of high efficacy and security to fabricate a therapeutic formulation for *** results revealed that tLNP/siHBV significantly reduced the expression of viral antigens and DNA(up to 3log_(10)reduction;vs PBS)in dose-and time-dependent manners at single-dose or multi-dose frequencies,with satisfactory safety *** studies showed that tLNP/siHBVIL2 enables additive antigenic and immune control of the virus,via introducing potent HBsAg clearance through RNAi and triggering strong HBV-specific CD4^(+)and CD8^(+)T cell responses by expressed mIL-2 *** adopting tLNP as nucleic acid nanocarriers,the co-delivery of siHBV and mIL-2 mRNA enables synergistic antigenic and immune control of HBV,thus offering a promising translational therapeutic strategy for treating CHB.

BACKGROUND Glycosylation,a commonly occurring post-translational modification,is highly expressed in several tumors,specifically in those of the digestive system,and plays a role in various cellular pathophysiological *** the importance and detection methods of glycosylation in digestive system tumors have garnered increasing attention in recent years,bibliometric analysis of this field remains *** present study aims to identify the developmental trends and research hotspots of glycosylation in digestive system *** To find and identify the developmental trends and research hotspots of glycosylation in digestive system *** We obtained relevant literature from the Web of Science Core Collection and employed VOSviewer 1.6.19 and CiteSpace(version 6.1.R6)to perform bibliometric *** A total of 2042 documents spanning from 1978 to the present were analyzed,with the research process divided into three phases:the period of obscurity(1978-1990),continuous development period(1991-2006),and the rapid outbreak period(2007-2023).These documents were authored by researchers from 66 countries or regions,with the United States and China leading in terms of publication *** Celso A had the highest number of publications,while Pinho SS was the most cited ***-occurrence analysis revealed the most popular keywords in this field are glycosylation,expression,cancer,colorectal cancer,and pancreatic ***,the Journal of Proteome Research was the most prolific journal in terms of publications,while the Journal of Biological Chemistry had the most *** The bibliometric analysis shows current research focus is primarily on basic research in this ***,future research should aim to utilize glycosylation as a target for treating tumor patients.

BACKGROUND Enhancing awareness and use of pre-exposure prophylaxis(PrEP)and postexposure prophylaxis(PEP)is vital to curb human immunodeficiency virus(HIV)***-risk behaviors prevalent among sexually transmitted infection clinic outpatients underscore the need for increased PrEP/PEP education in this *** To investigate the effects of both onsite and online health education on the knowledge of,and willingness to use,PrEP and PEP among individuals receiving PEP *** Participants were drawn from a cohort study on PEP service intervention at an STD/AIDS outpatient clinic in designated HIV/AIDS hospitals in Beijing,conducted from January 1 to June 30,*** education was provided both onsite and online during *** assessing knowledge of,and willingness to use,PrEP/PEP were administered at baseline and again at 24 wk *** A total of 112 participants were enrolled in the study;105 completed the follow-up at week *** percentage of participants with adequate knowledge of,and willingness to use,PrEP significantly increased from 65.2%and 69.6%at baseline to 83.8%and 82.9%at the end of the intervention(both P<0.05).Similarly,those with adequate knowledge of,and willingness to use,PEP increased from 74.1%and 77.7%at baseline to 92.4%and 89.5%at week 24(P<0.05).Being between 31 years and 40 years of age,having a postgraduate degree or higher,and reporting a monthly expenditure of RMB 5000 or more were found to be significantly associated with knowledge of PrEP and PEP(both P<0.05).CONCLUSION The findings show that both onsite and online health education significantly improved the knowledge of,and increased willingness to use,PrEP and PEP in individuals utilizing PEP services.

紫楠天然种群表型变异特征

[目的]揭示紫楠天然种群的表型变异程度及规律。[方法]以其分布区内20个种群为研究对象,运用巢式方差分析等方法,对紫楠11个表型性状及其与地理生态因子的相关性进行系统研究。[结果]紫楠11个表型性状在种群间和种群内均存在极显著差异,其变异系数为8.54%~43.75%,均值为20.83%,且6个叶片性状的平均变异系数(26.27%)高于5个种子性状(14.31%),即种子比叶片具有更大的遗传稳定性;紫楠种群间的变异(50.44%)高于种群内的变异(23.42%),平均表型分化系数为66.46%,种群间变异是紫楠表型变异的主要来源。在调查分布范围内,叶片随海拔升高而变小,呈由西向东逐渐变宽的趋势,而种子大小则表现为由东北向西南逐渐变大的趋势。依据叶片和种子性状可将20个种群聚为三大类群,基本可代表紫楠在不同分布区域的表型特征。[结论]紫楠11个表型性状在种群间和种群内均存在极显著差异,以种群间变异为主。叶片的平均变异系数高于种子,呈由西向东逐渐变宽趋势,种子则呈由东北向西南逐渐变大的趋势。聚类分析将20个紫楠天然种群依地理位置分为三大类。

Cloning and sequence analysis of nine novel MYB genes in Taxodiaceae plants

Myeloblastosis （MYB） is one of the largest transcribed factor families in plants. To gain an overall picture of the evolution of MYB genes in relict plants, we cloned nine novel MYB genes in Taxodiaceae plants （ Taxodium distichum, Taxodium ascendens, Cryptomeria japonica var. Sinensis, Cryptomeria japonica cv. Araucarioides, Cryptomer Ja- ponica, Metasequoia glyptostroboides, Cunninghamia lanceolata, Tai- wania cryptomerioides and Glyptostrobus pensilis）. The deduced amino acid sequences for MYBs showed that the nine MYB proteins contained two DNA binding domains. The first domain is from amino acid position 29 to 78, wherein three tryptophanes at 33, 53 and 73 were separated by 19 amino acids, respectively. The second domain is from amino acid position 82 to 127, wherein three tryptophanes at 86, 105 and 124 were separated by 18 amino acids, respectively, whereas the first tryptophane at amino acid position 86 is replaced by a phenylalanine. The characteri- zation of these conserved domains at nine MYBs indicated that they all belong to the R2R3-MYB group. The secondary structure analysis showed that a-helix and 13-turn are the major motifs of the predicted secondary structure of MYBs. The three dimensional model of each MYB protein showed that the structure is like clip, making it more flexi- ble and mobile. The similarities between the nine MYB proteins in Taxodiaceae were calculated. The highest identical value of 99% is be- tween CjsMYB, CjMYB and CjaMYB, whereas the lowest value of 82% is between TaMYB and C1MYB. According to the phylogenetic tree, the distances between different genera were relatively large whereas those within genera were relatively small. As expected, accessions of the same genus formed a subgroup before being grouped with other genera.

Amplified consensus genetic marker （ACGM） is a PCR-based marker technique that uses primers designed within conserved regions of coding sequences. After a comparison of Cryptomeria japonica and Arabidopsis ESTs to search for conserved sequences, 237 single e-PCR products were obtained. We randomly selected 110 candidate ACGM markers to test. Of the 110 candidate ACGM markers tested, 106 yielded stable and clear PCR products in C. japonica. We then tested the utility of these 106 primer pairs in 10 species, representing 7 genera of Taxodi- aceae. The number of specific amplification primer pairs among those 10 species varied from 49 to 103 （or 46.2±97.2%）. The 106 primer pairs （ACGM loci） were high transferable to Cryptomeria fortunei Hooibrenk （97.2%） but were low in Metasequoia glyptostroboides （46.2%）. The number of PCR bands per primer pair ranged from 1.06 to 1.15, which means that most of the ACGM primers can obtain a single band within these 10 Taxodiaceae species. In summary, our study shows that ACGM is a technique applicable for marker development even in species with limited sequence data.
