Small extracellular vesicles derived from human induced pluripotent stem cell-differentiated neural progenitor cells mitigate retinal ganglion cell degeneration in a mouse model of optic nerve injury

Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

Srgap2 suppression ameliorates retinal ganglion cell degeneration in mice

Slit-Robo GTPase-activating protein 2(SRGAP2) plays important roles in axon guidance, neuronal migration, synapse formation, and nerve regeneration. However, the role of SRGAP2 in neuroretinal degenerative disease remains unclear. In this study, we found that SRGAP2 protein was first expressed in the retina of normal mice at the embryonic stage and was mainly located in the mature retinal ganglion cell layer and the inner nuclear layer. SRGAP2 protein in the retina and optic nerve increased after optic nerve crush. Then, we established a heterozygous knockout(Srgap2+/–) mouse model of optic nerve crush and found that Srgap2 suppression increased retinal ganglion cell survival, lowered intraocular pressure, inhibited glial cell activation, and partially restored retinal function. In vitro experiments showed that Srgap2 suppression activated the mammalian target of rapamycin signaling pathway. RNA sequencing results showed that the expression of small heat shock protein genes(Cryaa, Cryba4, and Crygs) related to optic nerve injury were upregulated in the retina of Srgap2+/– mice. These results suggest that Srgap2 suppression reduced the robust activation of glial cells, activated the mammalian target of rapamycin signaling pathway related to nerve protein, increased the expression of small heat shock protein genes, inhibited the degeneration of retinal ganglion cells, and partially restored optic nerve function.

Elucidation of Regional Mechanical Properties of Brain Tissues Based on Cell Density

Research on the mechanical properties of brain tissue has received extensive ***,most of the current studies have been conducted at the phenomenological *** this study,the indentation method was used to explore the difference in local mechanical properties among different regions of the porcine cerebral ***,hematoxylin-eosin and immunofluorescence staining methods were used to determine the correlation between the cellular density at different test points and mechanical properties of the porcine cerebral *** frontal lobe exhibited the strongest *** temporal lobe displayed the lowest sensitivity to changes in the indentation speed,and the occipital lobe exhibited the highest shear ***,the shear modulus of different areas of the cerebral cortex was negatively correlated with the total number of local cells per unit area and positively correlated with the number of neuronal cell bodies per unit *** of the mechanical properties of the local brain tissue can provide basic data for the establishment of a finite element model of the brain and mechanical referential information for the implantation position of brain chips.

Prognostic factors for acute central retinal artery occlusion treated with hyperbaric oxygen:The Hong Kong study report number five

Pancreatic stent improves the success rate of needle-knife papillotomy in patients with difficult biliary cannulation

BACKGROUND Needle-knife precut papillotomy(NKP)is typically performed ***,it remains unclear whether pancreatic stent(PS)placement can improve the outcomes of *** To explore whether PS placement improves the success rate of NKP in patients with difficult biliary *** This single-center retrospective study included 190 patients who underwent NKP between January 2017 and December 2021 after failed conventional biliary *** cases with incidental pancreatic duct cannulation during conventional biliary cannulation,the decision for pre-NKP PS placement was made at the endoscopist's *** primary outcome was the difference in the NKP success rate between patients with and without PS placement;the secondary outcome was the adverse event *** Among the 190 participants,82 received pre-NKP PS(PS-NKP group)whereas 108 did not[freehand or freehand NKP(FH-NKP)group].Post-NKP selective biliary cannulation was successful in 167(87.9%)patients,and the PS-NKP had a significantly higher success rate than the FH-NKP group(93.9%vs 83.3%,P=0.027).The overall adverse event rates were 7.3%and 11.1%in the PS-NKP and FH-NKP groups,respectively(P=0.493).A periampullary diverticulum(PAD)and significant intraoperative bleeding during NKP were independently associated with NKP failure;however,a pre-NKP PS was the only predictor of NKP *** the 44 participants with PADs,the PS-NKP group had a non-significantly higher NKP success rate than the FH-NKP group(87.5%and 65%,respectively;P=0.076).CONCLUSION PS significantly improved the success rate of NKP in patients with difficult biliary cannulation.

Microstructure and Properties of Hot Working Die Steel H13MOD

Compared with H13 steel, the influences of different heat treatment process on the microstructure and properties of the new type of hot working die steel H13MOD were studied. The results show that the complete aus tenitizing temperature of H13MOD is around 1030 ℃ and the quenching hardness achieves the maximum value at this temperature. While for H13, the complete austenitizing temperature is above 1100 ℃ and the quenching hardness rise constantly with the quenching temperature increasing. In quenching process, the undissolved MC carbides can prevent the coarsening of grain in both steels. With the rise of quenching temperature, when MC carbides dissolve completely, the grain grows quickly. The hardness and strength of H13MOD at higher tempering temperature （above 570 ℃） are nearly the same as those of H13, but its toughness is higher than that of H13. Mo2C carbide is the main strengthening phase in H13MOD, which is attributed to the higher content of Mo. The quantity of VC eutectic car- bides is reduced because of lower content of V in H13MOD, which plays an important role in enhancing the impact toughness of H13MOD. Under a certain strength condition, H13MOD steel can be used in the environment that higher toughness is required and the service life of die casting mold can be improved.

An innovative wood-chip-framework substrate used as slow-release carbon source to treat high-strength nitrogen wastewater

Removal of nitrogen in wastewater before discharge into receiving water courses is an important consideration in treatment ***,nitrogen removal efficiency is usually limited due to the low carbon/nitrogen（C/N） ratio.A common solution is to add external carbon sources,but amount of liquid is difficult to ***,a combined wood-chip-framework substrate（with wood,slag and gravel） as a slow-release carbon source was constructed in baffled subsurface-flow constructed wetlands to overcome the *** show that the removal rate of ammonia nitrogen（NH_4~＋-N）,total nitrogen（TN） and chemical oxygen demand（COD） could reach 37.5%-85%,57.4%-86%,32.4%-78%,respectively,indicating the combined substrate could diffuse sufficient oxygen for the nitrification process（slag and gravel zone） and provide carbon source for denitrification process（wood-chip zone）.The nitrification and denitrification were determined according to the location of slag/gravel and wood-chip,*** removal was efficient at the steady phase before a shock loading using slag-wood-gravel combined substrate because of nitrification-denitrification process,while nitrogen removal was efficient under a shock loading with wood-slag-gravel combined substrate because of ANAMMOX *** study provides a new idea for wetland treatment of high-strength nitrogen wastewater.

Production of 5-Hydroxymethylfurfural and Furfural from Lignocellulosic Biomass in Water-Tetrahydrofuran Media with Sodium Bisulfate

5-Hydroxymethylfurfural (HMF) and furfural (FF), two bio-based platform chemicals, were produced from various raw lignocellulosic materials (corncob, corn stover, wheat straw, rice straw and sugarcane bagasse) in a water-tetrahydrofuran media by using NaHSO4 as catalyst. The influences of reaction temperature (160–200 °C), reaction time (30–120 min), solvent volume ratio, feedstock concentration (2.4wt%–11.1wt%) and catalyst dosage were studied. The highest HMF and FF yields obtained from corncob were 47mol% and 56mol% under condition of 190 °C, 90 min, 10/1 of THF/H2O. Besides, the lignin in the raw biomass was also depolymerized into organosolv lignin.

Vacuum Ultraviolet Excited Photoluminescence Properties of Y_2O_2S:Eu^(3+),Bi^(3+) Phosphor

As an Hg-free lamp using phosphor, the Bi^3＋ and EH^3＋ co-doped Y2O2S phosphors were prepared and their luminescence properties under vacuum ultraviolet（VUV） excitation were investigated. The VUV photoluminescent intensity of Y2O2S：Eu^3＋ was weak, however, considerably stronger red emission at 626 nm with good color purity was observed in Y2O2S：Eu^3＋,Bi^3＋ systems. Investigation on the photoluminescence reveals that the strong VUV luminescence of Y2O2S：Eu^3＋,Bi^3＋ at 147 nm is mainly because the Bi^3＋ acts as a medium and effectively performs the energy transfer process： Y^3＋-O^2-→Bi^3＋→Eu^3＋, while the intense emission band at 172 nm is attributed to the absorption of the characteristic ^1So-^1P1 transition of Bi^3＋ and the direct energy transfer from Bi^3＋ to Eu^3＋. The Y2O2S：Eu^3＋,Bi^3＋ shows excellent VUV optical properties compared with the commercial （Y,Gd）BO3：Eu^3＋. Thus, the Y2O2S：Eu^3＋,Bi^3＋ can be a potential red VUV-excited candidate applied in Hg-free lamps for backlight of liquid crystal display.
