Black radish root extract alleviates sodium valproate-induced liver damage via inhibiting mitochondrial membrane potential collapse and oxidative stress in mice

Objective:To explore the effect of black radish(Raphanus sativus *** niger)root extract on liver enzymes,oxidative stress,and histopathological alterations in mice with sodium valproate-induced ***:Thirty-two mice were divided into four groups:the control group received drinking water by gavage,the second group was administered with 100 mg/kg of sodium valproate,the third group received 300 mg/kg of black radish root extract,and the fourth group was given both sodium valproate(100 mg/kg)and black radish root extract(300 mg/kg).After 28 days,the mice were euthanized,and serum levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),and alkaline phosphatase(ALP),along with liver malondialdehyde(MDA),reactive oxygen species(ROS),mitochondrial parameters,tumor necrosis factor-alpha(TNF-α)gene expression,and histopathological changes were ***:Sodium valproate caused hepatic damage in mice,characterized by elevated serum levels of liver enzymes,increased MDA and ROS levels and TNF-αgene expression,as well as histopathological *** black radish root extract significantly alleviated sodium valproate-caused hepatic injury by decreasing the serum levels of ALT and AST,MDA,ROS,TNF-αgene expression,as well as mitochondrial impairment,but did not have a significant effect on sodium valproate-induced histopathological ***:The black radish root extract demonstrates protective effects against sodium valproate-induced liver injury,possibly through mitigating oxidative stress,mitochondrial impairment,and inflammatory mediator expression.

Study on the Interaction of Vitamin B12 with DNA by Spectroscopy and Electrochemical Methods

The Interaction between vitamin B12 （VBI2） and fish sperm DNA was investigated in physiological buffer （pH 7.4） using the methylene blue （MB） dye as a spectral probe by spetcrophotometery, viscosity measurements and cyclic voltammetry. The apparent binding constant of vitamin B12 with DNA was found to be 3.2 × 10^5 mol^-1·L. The voltammetric behavior of vitamin B12 has been investigated at glassy carbon electrode using cyclic voltammetry. Thermodynamic parameters including △H^0, △S^0 and △G^0 for the interaction between VB12 and DNA have determined as -2.3 × 10^4, 27.54 and -3.1 × 10^4j·mol^-l·K^-1 respectively. One indication of DNA binding mode with VBI2 was the change in viscosity when a small molecule associates with DNA. The diffusion coefficients of VB12 in the absence （D0）f and presence of DNA （D0）b was calculated as 5.04 × 10^-6 and 1.13 × 10^-6 cm^2·s^-1 respectively. The results indicated that vitamin B12 can bind to DNA and the major binding mode was intercalative binding.

A metastableβTi alloy was additively manufactured by laser powder bed fusion(LPBF).Tensile testing along the build direction of the as-LPBF material(LPBF-0°)revealed signifcant work softening immediately following yielding with no uniform *** contrast,substantial work hardening and uniform elongation well over 10%were achieved perpendicular to the build direction(LPBF-90°).Similar effects were obtained in the build direction after super transus heat treatment(LPBF-0°+HT)although the strength was slightly *** addition,the yield drop phenomenon observed in both orientations of the as-LPBF materials disappeared after *** enhanced work hardening ability,and thus ductility,can be attributed to increased interactions of slip bands/slip bands owing to additional{112}slip systems becoming operative in LPBF-0°+HT and LPBF-90°while LPBF-0°was dominated by{110}*** other variations after HT may be related to the coarsening of grain structure and removal of specifc substructures in the as-LPBF microstructure.

Regulatory Micro RNA Networks:Complex Patterns of Target Pathways for Disease-related and Housekeeping MicroRNAs

Blood-based mieroRNA （miRNA） signatures as biomarkers have been reported for various pathologies, including cancer, neurological disorders, cardiovascular diseases, and also infections. The regulatory mechanism behind respective miRNA patterns is only partially understood. Moreover, ＂preserved＂ miRNAs, i.e., miRNAs that are not dysregulated in any disease, and their biological impact have been explored to a very limited extent. We set out to systematically determine their role in regulatory networks by defining groups of highly-dysregulated miRNAs that contribute to a disease signature as opposed to preserved housekeeping miRNAs. We further determined preferential targets and pathways of both dysregulated and preserved miRNAs by computing multi-layer networks, which were compared between housekeeping and dysregulated miRNAs. Of 848 miRNAs examined across 1049 blood samples, 8 potential housekeepers showed very limited expression variations, while 20 miRNAs showed highly-dysregulated expression throughout the investigated blood samples. Our approach provides important insights into miRNAs and their role in regulatory networks. The methodology can be applied to systematically investigate the differences in target genes and pathways of arbitrary miRNA sets.

Optimization of Solvent Systems for the Extraction of Vitexin as the Major Bioactive Flavonoid in <i>Prosopis farcta</i>

Prosopis farcta, a plant belongs to the mimosoideae, is characterized by a very wide spectrum of various bioactive and medical constituents. Vitexin, the marker flavonoid found in Prosopis, has potent and broad antitumour efficacy in preclinical models. Many studies had been done for the isolation of flavonoids (vitexin) by completely different chromatographically methodology. During this study, vitexin was isolated from Prosopis farcta by 6 different extraction methods in which parameters as the type, concentration and pH of the extracting solvents considered. among different solvent systems used, methanol-water (40%, containing acetic acid 0.5%) was found to be the best solvent generating the highest yield (0.554 mg·g-1 DW) from Prosopis leaves. The present work suggests an efficient method for estimation the greatest content of vitexin analyzed by HPLC technique and introduces Prosopis farcta as a suitable source of this flavonoid with several pharmacological properties.

Management and Reduction of Chemical Nitrogen Consumption in Agriculture

Nitrogen is an effective material for improving the crop production. There is a big concern about rising chemical nitrogen usage as fertilizer in soil, because some forms of nitrogen absorbs in soil and causes an increase of environment and water pollution and decrease fertilizer efficiency. Composted cow manure is an important resource of organic nitrogen which improves the soil conditions. However, low density of composted cow manure and inconstant nutrient content are two factors that limit the application of composted cow manure. The densification technology is an effective solution for both problems. In this research first the capability of single screw extruder for production of fertilizer pellets including composted cow manure and nitrogen fertilizer was studied and then the effect of fertilizer pellet on the reduction of chemical nitrogen consumption was evaluated. The response of basil plants was determined in three treatments: T1 control (no fertilizer), T2 (nitrogen fertilizer) and T3 (pellet). The results showed that pelletization can be used as a proper method for slow-release nitrogen fertilizer;enhance nitrogen uptake and reducing the chemical nitrogen consumption. The results also showed that all basil responses (chlorophyll content, total weight, leaves weight, root weight and leaf area) were significantly affected by application of N fertilizer and pellet. While maximum recorded parameters were observed with application pellet.

Trichoderma species collected from Iran

In order to identify Trichoderma species isolated from Iran, Trichoderma selective media and malt extract agar (MEA) were used to isolate Trichoderma species from the soil samples. All the cultures were purified on 2% water agar by hyphal tip method prior to morphological examination. Morphological observations were carried out on the cultures grown on 2% MEA and oat meal agar at 20°C under ambient laboratory conditions. Macroscopic features of colony and microscopic features of conidiophore, phialid and conidium including position of phialids on conidiophore and shape and size of phialids and conidia were studied and recorded 3-5 days after inoculation. Out of 36 tested isolates, using morphological features and molecular data obtained from ITS1, ITS2 and 5.8S regions fourteen species were identified as follow: T. atroviride, T. ghanense, T. spirale, T. erinaceum, T. citrinoviride, T. saturnisporum, T. longibrachiatum, T. hamatum, T. harzianum, T. inhamatum, T. tomentosum, T. virens, T. asperellum, T. koningii. among the species T. harzianum and T. virens isolates were the most frequent species. In addition of the mentioned species two Tichoderma sp. were collected from walnut rhizospher that they are not fit to any described species so far. Although one of them are T. brevicumpactum introduced informally.

Synchrony between Ovarian Function &Sleep in Polycystic Ovary Syndrome Patients

Polycystic ovary syndrome (PCOS) is a complex, multifaceted, heterogeneous disorder, affecting 4% to 18% of reproductive-aged women and is associated with reproductive, metabolic and psychological dysfunction. In this study we determined the relationship between the time to sleep and serum levels of neurohormones in polycystic ovary syndrome (PCOS). Totally 77 PCO patients(case group) and 97 non-PCOS infertile women (control subjects) participated in this study between February 2012 and February 2013. A PSQI sleep questionnaire was completed by each patient in both groups. PSQI sleep questionnaire score and serum concentration of adrenaline, noradrenaline, melatonin, β-endorphin, cortisol and progesterone were compared in two groups. The results of the study indicate that serum levels of melatonin and β-endorphin were lower in women with PCOS. Serum level of stress hormones, adrenaline and noradrenaline were significantly correlated with patients’ sleep time in study group. Serum level of adrenaline in control group was significantly lower in women who wake up earlier in the morning. All hormones except for cortisol had no significant correlation with PSQI global score in both groups and also the people who sleep less than 8 hours had lower cortisol level. These data showed that changes in cortisol in PCO women were due to damage of disturbed sleep at night. Our preliminary work provided this study with new insight into the interactions between sleep-wake cycles in PCO women with specific sleep patterns.

Hyponeurotrophinemia in Serum of Women with Polycystic Ovary Syndrome as a Low Grade Chronic Inflammation

Introduction: This study aimed to discuss about neuroimmune-pathophysiology of polycystic ovary syndrome (PCOS). Nerve growth factor (NGF) as modulator of neuro-endocrine-immune (NEI) system plays a critical role in tissue survival and homeostasis maintenance. It is a strong marker for sympathetic nerve activity. Women with PCO have enhanced ovarian NGF production. Over the past few decades, PCOS has been discussed as a low grade chronic inflammatory disease. In this study, we investigated the neuromodulator role of serum NGF and inflammatory cytokines (IL-1α, β, 17A and TNF-α) in women with PCOS. Material and Methods: 171 patients were investigated in two groups: study (n = 85 PCO) and control (n = 86). Serum levels of NGF-α and interleukins IL-1α, IL-1β, IL-17A and TNFα were determined in both groups by ELISA kit. Results: Data of this study showed that the level of NGF-α and IL-17A in serum of PCO women were lower than control group (p α, IL-1β were higher than control group in PCO women (p ammatory background in PCOS is the main cause of increased serum levels of IL-1α, IL-1β. Reduction of IL-17A and NGF in serum of PCO patients might be under the direct influence of the hyperactivity of sympathetic nervous system (SNS).
