Is tumor cell specificity distinct from tumor selectivity in vivo?A quantitative NIR molecular imaging analysis of nanoliposome targeting

The significance and ability for receptor targeted nanoliposomes(tNLs)to bind to their molecular targets in solid tumors in vivo has been questioned,particularly as the efficiency of their tumor accumulation and selectivity is not always predictive of their efficacy or molecular specificity.This study presents,for the first time,in situ near-infrared(NIR)molecular imaging-based quantitation of the in vivo specificity of tNLs for their target receptors,as opposed to tumor selectivity,which includes influences of enhanced tumor permeability and retention.Results show that neither tumor delivery nor selectivity(tumor-to-normal ratio)of cetuximab and IRDye conjugated tNLs correlate with epidermal growth factor receptor(EGFR)expression in U251,U87,and 9L tumors,and in fact underrepresent their imaging-derived molecular specificity by up to 94.2%.Conversely,their in vivo specificity,which we quantify as the concentration of tNL-reported tumor EGFR provided by NIR molecular imaging,correlates positively with EGFR expression levels in vitro and ex vivo(Pearson’s r=0.92 and 0.96,respectively).This study provides a unique opportunity to address the problematic disconnect between tNL synthesis and in vivo specificity.The findings encourage their continued adoption as platforms for precision medicine,and facilitates intelligent synthesis and patient customization in order to improve safety profiles and therapeutic outcomes.

Human spinal cord injury (SCI) results in locomotor and sensory disabilities,which severely affect the quality of life.To restore function after SCI,it is necessary to repair and reconstruct the damaged local circuitry.Major hurdles in neural regeneration include a limited level of neurogenesis in the adult spinal cord and an inflammatory microenvironment that inhibits neurogenesis and axon regeneration.

在水合物开采过程中,管道内的液滴和气泡受环境条件影响会转化生成水合物颗粒,水合物颗粒发生聚集时会引起堵塞,导致潜在的安全问题。粘附力是探究开采管道流动安全及水合物颗粒聚集和沉积机理的关键,目前相关研究主要集中在输运过程中水合物颗粒间或颗粒-表面间粘附力的测量和描述,而对开采过程中沉积物矿物润湿性及表面粗糙度对粘附力的影响关注较少。利用粘附力测量实验系统,开展了水合物颗粒与经不同方法处理的碳钢表面及石英石、孔雀石、方解石和高岭石等矿物表面间粘附力的测量分析。结果显示,受润湿性影响,水合物颗粒与矿物表面之间粘附力是其与碳钢表面粘附力的3~6倍。此外,水合物颗粒与矿物表面粘附力受接触时间影响,与方解石和高岭石表面间的粘附力随接触时间的延长而增加。进一步探索添加离子表面活性剂对水合物晶体生长速度及粘附力的影响,并指出颗粒与矿物表面水膜接触面积变化使水合物颗粒形貌由层状结构变为针状结构。对比实验测量结果与理论计算结果的差异,分析得出矿物表面的强亲水性是造成结果差异的关键原因。

Sparse-Grid Implementation of Fixed-Point Fast Sweeping WENO Schemes for Eikonal Equations

Fixed-point fast sweeping methods are a class of explicit iterative methods developed in the literature to efficiently solve steady-state solutions of hyperbolic partial differential equations(PDEs).As other types of fast sweeping schemes,fixed-point fast sweeping methods use the Gauss-Seidel iterations and alternating sweeping strategy to cover characteristics of hyperbolic PDEs in a certain direction simultaneously in each sweeping order.The resulting iterative schemes have a fast convergence rate to steady-state solutions.Moreover,an advantage of fixed-point fast sweeping methods over other types of fast sweeping methods is that they are explicit and do not involve the inverse operation of any nonlinear local system.Hence,they are robust and flexible,and have been combined with high-order accurate weighted essentially non-oscillatory(WENO)schemes to solve various hyperbolic PDEs in the literature.For multidimensional nonlinear problems,high-order fixed-point fast sweeping WENO methods still require quite a large amount of computational costs.In this technical note,we apply sparse-grid techniques,an effective approximation tool for multidimensional problems,to fixed-point fast sweeping WENO methods for reducing their computational costs.Here,we focus on fixed-point fast sweeping WENO schemes with third-order accuracy(Zhang et al.2006[41]),for solving Eikonal equations,an important class of static Hamilton-Jacobi(H-J)equations.Numerical experiments on solving multidimensional Eikonal equations and a more general static H-J equation are performed to show that the sparse-grid computations of the fixed-point fast sweeping WENO schemes achieve large savings of CPU times on refined meshes,and at the same time maintain comparable accuracy and resolution with those on corresponding regular single grids.

From landrace to modern hybrid broccoli:the genomic and morphological domestication syndrome within a diverse B.oleracea collection

Worldwide,broccoli(Brassica oleracea var.italica)is among the most economically important,nutritionally rich,and widely-grown vegetable crops.To explore the genomic basis of the dramatic changes in broccoli morphology in the last century,we evaluated 109 broccoli or broccoli/cauliflower intermediates for 24 horticultural traits.Genotype-bysequencing markers were used to determine four subpopulations within italica:Calabrese broccoli landraces and hybrids,sprouting broccoli,and violet cauliflower,and to evaluate between and within group relatedness and diversity.While overall horticultural quality and harvest index of improved hybrid broccoli germplasm has increased by year of cultivar release,this improvement has been accompanied by a considerable reduction in allelic diversity when compared to the larger pool of germplasm.Two landraces are the most likely founding source of modern broccoli hybrids,and within these modern hybrids,we identified 13 reduction-in-diversity genomic regions,53 selective sweeps,and 30(>1 Mbp)runs of homozygosity.Landrace accessions collected in southern Italy contained 4.8-fold greater unique alleles per accessions compared to modern hybrids and provide a valuable resource in subsequent improvement efforts.This work broadens the understanding of broccoli germplasm,informs conservation efforts,and enables breeding for complex quality traits and regionally adapted cultivars.

With the constant development of multiphoton microscopy,our ability to observe complex and dynamic biological processes deeper within living tissue,is steadily improving.Researchers use multiphoton microscopy,because experiments can be conducted with little to no invasiveness or tissue damage over a long period of time with no photodamage(Mancuso et al.,2009).This allows for the introduction of tissue into the context of a three-dimensional 3D environment in which visualization of cellular activation and interaction is viable.By circumventing a distorted reconstruction with limited z-stacks,multiphoton imaging provides enhanced spatiotemporal resolution.

Stability Analysis and Performance Evaluation of Additive Mixed-Precision Runge-Kutta Methods

Additive Runge-Kutta methods designed for preserving highly accurate solutions in mixed-precision computation were previously proposed and analyzed.These specially designed methods use reduced precision for the implicit computations and full precision for the explicit computations.In this work,we analyze the stability properties of these methods and their sensitivity to the low-precision rounding errors,and demonstrate their performance in terms of accuracy and efficiency.We develop codes in FORTRAN and Julia to solve nonlinear systems of ODEs and PDEs using the mixed-precision additive Runge-Kutta(MP-ARK)methods.The convergence,accuracy,and runtime of these methods are explored.We show that for a given level of accuracy,suitably chosen MP-ARK methods may provide significant reductions in runtime.

Developing an identities-based approach to support more robust resilience and recovery in heritage planning and management

Resilience has become an increasingly important concept in the cultural heritage feld,particularly in the aftermath of the unprecedented challenges the COVID-19 global pandemic brought.However,on a conceptual and practical level,resilience remains closely linked to the technical conservation of built heritage,and there remains a need to develop broader approaches inclusive of cultural and socioeconomic components.This article investigates the potential applicability of theoretical concepts linked to identity and identities in heritage planning to help fll these gaps and develop approaches that consider resilience and are better able to address a range of unanticipated disasters.We frst review the literature and policy documents to defne and identify the potential for identities-based approaches.We then examine the case of Matera,an extremely fragile world heritage site in southern Italy that has been continuously inhabited for more than 8000 years and provides a relevant example of resilience.We explore the trends and efects of globalised tourism development before the pandemic and the post pandemic emergence of more local/regional and slow tourism patterns,largely based on cultural solutions to local development challenges and knowledge exchange.Through this comparison,we analyse the potential and limitations of introducing identities-based concepts into heritage planning as a more robust way to enhance resilience and prepare cities for unexpected future crises.

A Multi-Point Distance-Bounding Protocol for Securing Automatic Dependent Surveillance-Broadcast in Unmanned Aerial Vehicle Applications

The Automatic Dependent Surveillance-Broadcast(ADS-B)protocol is being adopted for use in unmanned aerial vehicles(UAVs)as the primary source of information for emerging multi-UAV collision avoidance algorithms.The lack of security features in ADS-B leaves any processes dependent upon the information vulnerable to a variety of threats from compromised and dishonest UAVs.This could result in substantial losses or damage to properties.This research proposes a new distance-bounding scheme for verifying the distance and flight trajectory in the ADS-B broadcast data from surrounding UAVs.The proposed scheme enables UAVs or ground stations to identify fraudulent UAVs and avoid collisions.The scheme was implemented and tested in the ArduPilot SITL(Software In The Loop)simulator to verify its ability to detect fraudulent UAVs.The experiments showed that the scheme achieved the desired accuracy in both flight trajectory measurement and attack detection.
