Variation of photosynthesis,secondary metabolites and antioxidant activities in third generation of spaceflight-induced Salvia miltiorrhiza

Objective: Spaceflight has long been perceived as an effective way to improve the quantity and quality of plants with wide applications. In order to obtain stable and inheritable descendants of spaceflightinduced Salvia miltiorrhiza lines, we investigated and analyzed four lines m16, m50, m51, m57(three individuals of each line) and the ground control(three individuals) of the third generation of spaceflight-induced S. miltiorrhiza from primary/secondary metabolism and antioxidative ***: A portable photosynthesis system(Li-6400) with red/blue LED light source was used to perform the photosynthetic characteristics to evaluate their primary productivity. The secondary metabolites(phenolic acids, tanshinones, total phenolics and flavonoids) and antioxidant activity of roots were analyzed to assess their ***: Compared with control, line m16 presented weak photosynthetic ability, but high apparent quantum yield(AQY), higher contents of secondary metabolites, and stronger antioxidative *** m57 had a strong gas exchange ability, relatively higher secondary metabolites contents, and ascending antioxidative abilities. Lines m50 and m51 were in the middle level of lines m16 and *** principal component analysis for all the original data revealed three components including a rootrelated index, a leaf-related index, and a CO_(2) response parameter could be used to distinguish spaceflight-induced S. miltiorrhiza ***: Line m57 could be an appropriate material for the investigation of targeted breeding towards high production, and line m16 could be used to identify essential genes and unravel sophisticated pathways underlying the secondary metabolisms.

MicroRNAs (miRNAs) are a class of non-coding RNAs that play critical roles in post-transcriptional regulation. Their target genes are involved in a variety of biological processes such as development, metabolism, and stress response. Panicum miliaceum L. (Panicum) is an important grain crop, but, until now, no miRNAs have been identified in this plant. Using a homology search based on expressed sequence tag (EST) analysis and miRNA precursor secondary structure, a total of 43 new miRNAs were identified. The miRNAs were found to be unevenly distributed among 11 miRNA families. Target analysis using the plant small RNA target analysis server psRNATarget showed that the newly identified miRNAs can potentially regulate 68 target genes. Ten of the 11 miRNA families were annotated as involved in RNA regulation, suggesting they may play an essential role in post-transcriptional regulation in Panicum. Selected miRNAs representing eight of the families were verified by northern blotting, indicating that the prediction method that we used to identify the miRNAs was effective.

Andrographis paniculata(***)is a Chinese herbal medicine that clears away heat,reduces inflammation,protects the liver,and promotes *** WRKYs of *** are still not well characterized,although many WRKYs have been identified in various plant *** the present study,59 *** WRKY(ApWRKY)genes were identified and renamed on the basis of their respective chromosome *** ApWRKYs were divided into three groups via phylogenetic analysis according to their WRKY domains and combined with WRKY of *** 59 identified ApWRKY transcription factors were nonuniformity distributed on 23 chromosomes of *** the structural analysis of the conserved motifs,different ApWRKYs structures showed different biological functions,and the ApWRKY transcription factor had certain species-specificity in the evolutionary *** expression patterns of the 41 ApWRKYs were examined through quantitative real-time PCR(qRT-PCR)in various tissues and under abiotic stresses(salt).The results showed that most of the ApWRKY had different reactions to salt *** addition,the content of the four main secondary metabolites in *** leaves was determined under salt *** results show that under a low concentration of salt treatment,the synthesis of andrographolide can be improved.

ABA is one of the 5 phytohormones in higher plants, which is also the most important hormone that regulates higher plants in response to environmental stress, by ABA signal transduction. Understanding ABA signal transduction at the molecular level is crucial to biology and ecology, and rational breeding complied with corresponding eco-environmental changes. Great advancements have taken place over the past 10 years by application of the Arabidopsis experimental system. Many components involved in ABA signal transduction have been isolated and identified and a clear overall picture of gene expression and control for this transduction has become an accepted fact. On the basis of the work in our laboratory, in conjunction with the data available at the moment, the authors have attempted to integrate ABA signal transduction pathways into a common one and give some insights into the relationship between ABA signal transduction and other hormone signal transduction pathways, with an emphasis upon the ABA signal transduction during higher plant seed development. A future challenge in this field is that different experimental systems are applied and various receptors and genes need to be characterized through the utilization of microarray chips.

Interactions between HLH and bHLH Factors Modulate Light-Regulated Plant Development

Phytochromes （Phy） and phytochrome-interacting factor （PIF） transcription factors constitute a major signal- ing module that controls plant development in response to red and far-red light. A low red：far-red ratio is interpreted as shading by neighbor plants and induces cell elongation--a phenomenon called shade-avoidance syndrome （SAS）. PAR1 and its closest homolog PAR2 are negative regulators of SAS; they belong to the HLH transcription factor family that lacks a typical basic domain required for DNA binding, and they are believed to regulate gene expressions through DNA binding transcription factors that are yet to be identified. Here, we show that light signal stabilizes PAR1 protein and PAR1 interacts with PIF4 and inhibits PIF4-mediated gene activation. DNA pull-down and chromatin immunoprecipitation （CHIP） assays showed that PAR1 inhibits PIF4 DNA binding in vitro and in vivo. Transgenic plants overexpressing PAR1 （PARIOX） are insensitive to gibberellin （GA） or high temperature in hypocotyl elongation, similarly to the pifq mutant. In addition to PIF4, PAR1 also interacts with PRE1, a HLH transcription factor activated by brassinosteroid （BR） and GA. Overexpression of PRE1 largely suppressed the dwarf phenotype of PARIOX. These results indicate that PAR1-PRE1 and PAR1-PIF4 het- erodimers form a complex HLH/bHLH network regulating cell elongation and plant development in response to light and hormones.

Salvia miltiorrhiza and *** are rich in diterpenoids and have therapeutic effects on cardiovascular *** this study,the spatial distribution of diterpenoids in both species was analyzed by a combination of metabolomics and mass spectrometry imaging *** results indicated that diterpenoids in *** were mainly abietane-type norditerpenoid quinones with a furan or dihydrofuran D-ring and were mainly distributed in the periderm of the roots,*** and tanshinone *** compounds in *** were mainly phenolic abietane-type tricyclic diterpenoids with six-or seven-membered C-rings,and were widely distributed in the periderm,phloem,and xylem of the roots,e.g.11-hydroxy-sugiol,11,20-dihydroxy-sugiol,and 11,*** addition,the leaves of *** were rich in tanshinone biosynthesis precursors,such as 11-hydroxy-sugiol,while those of *** were rich in phenolic *** in the upstream pathway of tanshinone biosynthesis were highly expressed in the root of ***,and genes in the downstream pathway were highly expressed in the root of ***,we describe the specific tissue distributions and mechanisms of diterpenoids in two Salvia species,which will facilitate further investigations of the biosynthesis of diterpenoids in plant synthetic biology.
