Relationship between plasma risperidone concentrations and clinical features in chronic schizophrenic patients in China

BACKGROUND Prior studies have noted great variability in the plasma levels of risperidone(RIS).Plasma concentrations of RIS and its active moiety are highly variable and depend on absorption,metabolism,and other predictors of metabolic dysregulation;however,these factors are poorly understood and the association between metabolic change and change in psychopathology is *** To ascertain the characteristics of chronic schizophrenic patients treated with RIS,and to assess their relationship with plasma RIS *** This was a descriptive cross-sectional study of 50 patients with a diagnosis of schizophrenic psychosis treated with RIS in a psychiatric *** plasma concentrations of RIS and its metabolite 9-hydroxyrisperidone were determined by high performance liquid *** patients’demographic and clinical characteristics,and psychopathologies were assessed,and the associations between clinical variables and plasma levels of RIS were *** Male patients received higher doses of RIS than female ones,but plasma concentrations of RIS and risperidone+9-hydroxyrisperidone(active moiety)were higher in female *** and the mean scores of the general psychopathology subscale of the Positive and Negative Syndrome Scale(PANSS)were significantly positively correlated with plasma concentrations of risperidone+9-hydroxyrisperidone adjusted for weight and dose in all 50 *** male subjects,we found a statistically significant positive correlation between the concentrations of risperidone+9-hydroxyrisperidone in plasma/(dose×kg)and age,mean PANSS negative subscale scores,mean PANSS general psychopathology subscale scores,and mean PANSS total *** Long-term use of RIS should be closely monitored in older patients and females to minimize the risk of high concentrations which could induce side effects.

The Arbuzov reaction(also known as the Michaelis–Arbuzov reaction)is one of the most fundamental transformations in organic chemistry and has been extensively used for the synthesis of organophosphorus compounds for over a ***,the reaction generally requires an elevated temperature and is applicable mainly to primary alkyl halides with poor functional group *** report herein a radical alternative to the conventional nucleophilic substitution ***,new radical phosphonylation reagents,9-fluorenyl o-phenylene phosphite and benzhydryl o-phenylene phosphite,have been developed for this *** photoredox-catalyzed reaction of a wide range of alkyl bromides or iodides with 9-fluorenyl(or benzhydryl)o-phenylene phosphite followed by workup with methanol(or ethanol)provides the corresponding dimethyl(or diethyl)alkylphosphonates in satisfactory yields at room temperature under mild *** method is applicable to primary,secondary,and even tertiary alkyl iodides and bromides and exhibits an excellent functional group tolerance,thereby enabling the late-stage modification of complex molecules and rapid synthesis of phosphonate-containing natural products and drug molecules.A mechanism involving the formation and subsequentβ-scission of phosphoranyl radicals is proposed.

Ultrahigh-throughput screening of antagonistic bacteria against Erwinia carotovora Ecc15 based on droplet microfluidics

Soft rot is a destructive disease that inflicts significant losses on agricultural production and the economy post-harvest. Biocontrol strategies based on antagonistic microorganisms have a broad application prospect to fight against plant pathogens. This study utilized fluorescence-activated droplet sorting （FADS） technology as an alternative to traditional plate culture methods to isolate microorganisms with antagonistic activity against the soft rot pathogen Erwinia carotovora Ecc15. Initially， the culture performance of the FADS platform was evaluated by analyzing bacterial diversity in droplet culture samples and agar plate culture samples， our data showed that droplet culture exhibited higher species richness and diversity than plate culture， and more than 95% of the OTUs in the droplet samples belonged to the rare biosphere. Additionally， we developed a GFP-Ecc15-based FADS screening system， which achieved an enrichment ratio of up to 148. Using this system， we successfully screened 32.antagonistic bacteria from rhizosphere soil sample of healthy konjac plants， and some may be novel microbial resources， including the genera Lelliottia， Buttiauxella and Leclercia. Notably， strain D-62.exhibited the strongest antibacterial ability against Ecc15， with an inhibition zone diameter of （2..86 ± 1.56） mm. In vivo experiments conducted on the corms of Amorphophallus konjac demonstrated that strain D-62.could effectively reduce the infection ability of Ecc15 to the corms， indicating that strain D-62.has the potential to be developed as a biocontrol agent. Our findings suggested that the FADS screening system showed a screening efficiency approximately 3 × 103times higher than plate screening system， while significantly reducing costs of infrastructure， labor and consumables， it provides theoretical guidance for the screening of other plant pathogen biocontrol bacteria.

Ultrashort and powerful laser interactions with a target generate intense wideband electromagnetic pulses(EMPs).In this study,we report EMPs generated by the interactions between petawatt(30 fs,1.4×10^(2.) Association of acute glycemic parameters at admission with cardiovascular mortality in the oldest old with acute myocardial infarction

OBJECTIVES Stress-related glycemic indicators,including admission blood glucose(ABG),stress-hyperglycemia ratio(SHR),and glycemic gap(GG),have been associated with worse outcomes after acute myocardial infarction(AMI).However,data regarding their prognostic value in the oldest old with AMI are ***,this study aimed to investigate the association of stress-related glycemic indicators with short-and long-term cardiovascular mortality(CVM)in the oldest old(≥80 years)with *** In this prospective study,a total of 933 consecutive old patients with AMI admitted to FuWai hospital(Beijing,China)were *** admission,ABG,SHR,and GG were assessed and all participants were classified according to their ***-Meier,restricted cubic splines(RCS),and multivariate Cox regression analyses were performed to evaluate the association between these glycemic indicators and CVM within 30 days and long-term *** During an average of 1954 patient-years of follow-up,a total of 2.0 cardiovascular deaths were ***-Meier analyses showed the lowest CVM in quartile 1 of ABG and in quartile 2.of SHR and *** adjusting for potential covariates,patients in quartile 4 of ABG,SHR,and GG had a respective 1.67-fold(95%CI:1.03-2.69;P=0.036),1.80-fold(95%CI:1.16-2.79;P=0.009),and 1.78-fold(95%CI:1.14-2.79;P=0.011)higher risk of long-term CVM risk compared to those in the reference groups(quartile 1 of ABG and quartile 2.of SHR and GG).Furthermore,RCS suggested a J-shaped relationship of ABG and a Ushaped association of SHR and GG with long-term ***,we observed similar associations of these acute glycemic parameters with 30-day *** Our data first indicated that SHR and GG consistently had a U-shaped association with both 30-day and longterm CVM among the oldest old with AMI,suggesting that they may be useful for risk stratification in this special population.

Smartly tuning supramolecular chirality in polymer gels via photoexcitation-induced cooperative self-assembly

Although supramolecular chirality tuning is prosperous in systems with small molecular building blocks, these systems lack structural strength and have limited practical applications. Conversely, polymer systems do not exhibit these drawbacks but suffer from topological chain entanglement, which often leads to large dynamic obstacles and therefore limits the tunability of supramolecular chirality. Herein, we report that by manipulating stoichiometry and employing light to alter the H-bond crosslinking between a photoexcitation-induced aggregatable molecule and high molecular weight poly(L-lactic acids), supramolecular chirality tuning of the polymer gel systems can be achieved in different ways through a cooperative ***, the Cotton effects of the examined systems can be negatively tuned by varying their stoichiometry, while positive tuning can be realized by photoirradiation. After deexcitation, negative Cotton effect tuning occurs again spontaneously in some cases. Such supramolecular chirality tuning is accompanied by rheological property tuning, demonstrating that our strategy can overcome polymer chain entanglement limitations. The proposed strategy inspires further works on the precise control of polymer chirality and, hence, development of asymmetric materials with changeable optical activity and modulus.
