Molecularly imprinted polymers (MIPs) are synthetic tailor-made polymers with high selectivity towards a particular substance (template).An MIP using vinblastine (VLB) as the template molecule was synthesized and *** presence of monomer-template complexes in a non-covalent way was confirmed by UV-vis spectrometry *** polymerization was performed using methacrylic acid (MAA) as the functional monomer,ethylene glycol dimethacrylate (EGDMA) as the cross-linking agent,and toluene as the porogenic solvent by a thermo-polymerization *** characterization of the obtained MIP was evaluated by scanning electron microscopy (SEM) and Brunauer-Emmett-Teller (BET) *** was observed that the morphology of the MIP was more porous and rough,and the surface area had a significant increase compared with that of the non-imprinted polymer (NIP).This MIP was used as the sorbents of solid-phase extraction (SPE) to assess the selectivity of the MIP after optimization of the SPE *** was specifically adsorbed on the MIP cartridge,while to vincristine (VCR),the chemical analog of VLB,almost no selective binding *** the basis of the results,Catharanthus roseus extract was applied to the MIP cartridge for investigating its capability to extract VLB from the plant extract,and the capacity of the MIP cartridge was also *** was shown that the MIP could effectively enrich VLB from *** extract and the recovery amounted to 93.8%.The solvents dissolving the samples had significant influence on the capacity of the MIP cartridge;it was 750 μg/g in toluene,625 μg/g in chloroform,and 250 μg/g in methanol.

Corona virus disease 2019(COVID-19)has exerted a profound adverse impact on human *** have demonstrated that aerosol transmission is one of the major transmission routes of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Pathogenic microorganisms such as SARS-CoV-2 can survive in the air and cause widespread infection among *** monitoring of pathogenic microorganism transmission in the atmosphere and accurate epidemic prediction are the frontier guarantee for preventing large-scale epidemic *** of pathogenic microorganisms in the air,especially in densely populated areas,may raise the possibility to detect viruses before people are widely infected and contain the epidemic at an earlier *** multi-scale coupled accurate epidemic prediction system can provide support for governments to analyze the epidemic situation,allocate health resources,and formulate epidemic response *** review first elaborates on the effects of the atmospheric environment on pathogenic microorganism transmission,which lays a theoretical foundation for the monitoring and prediction of epidemic ***,the monitoring technique development and the necessity of monitoring pathogenic microorganisms in the atmosphere are summarized and ***,this review introduces the major epidemic prediction methods and highlights the significance to realize a multi-scale coupled epidemic prediction system by strengthening the multidisciplinary cooperation of epidemiology,atmospheric sciences,environmental sciences,sociology,demography,*** summarizing the achievements and challenges in monitoring and prediction of pathogenic microorganism transmission in the atmosphere,this review proposes suggestions for epidemic response,namely,the establishment of an integrated monitoring and prediction platform for pathogenic microorganism transmission in the atmosphere.

Inhibition of Aloperine on Dextran Sulphate Sodium-induced Chronic Colitis in C57BL/6 Mice

Objective To investigate the effects of aloperine (ALO) on a model of dextran sulphate sodium (DSS)-induced chronic colitis in C57BL/6 mice. Methods Repeated colitis was induced by administration of four cycles of 4% DSS. The severity of colitis was assessed on the basis of clinical signs, ratio of colon weight and colon length, and histological grading scores. Moreover, secretory immunoglobulin A (S-IgA) and plasma haptoglobin (HP) were analyzed by enzyme-linked immunosorbent assay, and the changes of mRNA expression of ICAM-1 and MIF gene in colorectal tissue were detected by quantitative reverse transcriptase real-time polymerase chain reaction using SYBR Green I. Results ALO administration significantly attenuated the colon damage, caused substantial reductions of the rise in HP, and maintained the level of cecum S-IgA. ALO inhibited the ICAM-1 mRNA expression and had no effect on MIF mRNA expression. Conclusion The effect of ALO on DSS-induced chronic colitis in mice is investigated for the first time, which suggests that ALO could be an attractive therapeutic candidate in the treatment of inflammatory bowel disease.

Clinical observation of Qushi Xiezhuo formula in reducing monosodium urate crystal deposition in patients with axial spondyloarthritis

OBJECTIVE: To investigate effect of Qushi Xiezhuo formula (QSXZF) on axial spondyloarthritis (AxSpA) with a high incidence of monosodium serum urate (MSU) crystal ***: In this prospective cohort study, 62 Ax- SpA patients diagnosed with MSU crystal deposition from October 2012 to July 2015 were recruited for follow-up observation for 1 year after discharge from the hospital. Patients were divided into a case group with QSXZF treatment and a control group without any interventions. X-ray and dual-energy computed tomography were used to assess structural damage in the pelvis and sacroiliac joint and the volume of the MSU crystals. The Wilcoxon rank-sum test and Fisher's exact test were used to compare the proportion and distribution between the groups. RESULTS: A decrease in C-reactive protein (CRP) level, relief from back pain, and an increase in MSU crystal depositions were found in control patients. Compared with the control group, QSXZF reduced CRP levels and back pain to a greater extent, as well as reduced erythrocyte sedimentation rate levels, serum uric acid levels, Ankylosing Spondylitis Disease Activity Score, morning stiffness and MSU crystal deposition. CONCLUSION: QSXZF can lower progress of radiogrphaic grade at sacroiliac joint in AxSpA/AS patients with MSU crystal deposition by decreasing the inflammation response and reducing the serum uric acid and volume of MSU crystal deposition in sacroiliac joint. The above process may be attributed to the relieving the Qi-movement disturbance in the body, and eliminating turbidity and dampness by QSXZF.

全身麻醉下闭合复位加髋人位石膏固定是治疗24月龄以下发育性髋关节脱位(developmental dislocation of the hip,DDH)的通用方法,但闭合复位后仍有约1/3的患儿存在残余髋臼发育不良。尽管骨盆截骨术正越来越多地应用于残余髋臼发育不良的治疗,但其手术时机和手术指征仍不明晰。本研究拟通过一项多中心前瞻性非随机对照试验探讨DDH闭合复位后残余髋臼发育不良的手术干预时机和指征。研究设计拟纳入283例DDH闭合复位后残余髋臼发育不良患儿,包括观察组183例,手术组(采用骨盆截骨术)100例。手术组根据患儿年龄采用Salter、Pemberton、骨盆三联或髋臼周围截骨术。所有患儿随访10年,拟在DDH闭合复位后8个时间点(闭合复位术后1、2、3、4、5~6、7~8、9~10年以及10年以上)拍摄髋关节正位X线片,测量髋臼指数(acetabular index,AI)、中心边缘角(center-edge angle of wiberg,CEA)和Reimer指数(Reimer's index,RI),根据末次随访时正位X线片上髋关节Severin分级分为满意组(Severin分级为Ⅰ、Ⅱ级)和不满意组(Severin分级为Ⅲ、Ⅳ级),利用Logistic回归和Cox回归分析残余髋臼发育不良的风险因素,比较两组患儿AI、CEA、RI及Severin分级情况。这一多中心前瞻性研究可以为DDH闭合复位后残余髋臼发育不良的手术干预时机和指征提供循证医学证据。

The volume-sensitive outwardly rectifying chloride channel(VSOR) involves in essential physiological and pathophysiological processes,and represents a promising and developing target on the treatment of cancer. Novel strategies based on new interesting targets are needed to improve the conventional *** (DS),also named antabuse or antabus,a traditional drug to treat alcoholism,is currently being announced to be capable of binding copper and a novel drug to cure *** this study,we aimed to investigate the role of the VSOR in cell apoptosis induced by disulfiram/copper mixture(DS/Cu) in poorly differentiated nasopharyngeal carcinoma cells(CNE-2Z).We found that:(1) DS/Cu induced substantial cell *** to 1μmol/L DS plus 10μmol/L CuCl for 24 h and 48 h,the apoptotic rates were increased greatly to(20.7±2.3)%and(53.2±4.8)% (n=3,P<0.01) *** with those in the blank control,the change was *** with CuCl(10μmol/L) or DS(1μmol/L) alone did not affect apoptotic rate significantly.(2) Apoptosis induced by DS/CuCl was effectively prevented by the administration of chloride channel blocker,NPPB(100μmol/L).The apoptotic rate reduced to(13.7±3.8)%and(38.5±6.5)%at 24 h and 48 h,respectively(n=3,P<0.01 vs DS/Cu group).(3) Chloride currents were activated by DS/Cu mixture in a dose-dependent manner with EC values of 7.7 umol/L with(36.7±4.5) pA/pF and(-29.4±3.9) pA/pF(n=6,P<0.01 vs control) in response to±80 mV,respectively. While EC values of DS only was 48.1μmol/L,and the currents were(28.8±8.5) pA/pF and(-28.4±7.6) pA/pF(n=7, P<0.01 vs control) at±80 mV clamp *** properties of the currents induced by DS/Cu showed a weak outward-rectified and sensitive to cell volume *** currents were inhibited reversibly by extracellular hypertonic solution.(4) Chloride channel blockers inhibited the DS/Cu-activated chloride currents *** (100μmol/L) inhibited(93.3±7.9)%of the outward current at +80 mV a

Exploring Mechanism of Key chinese Herbal medicine on Breast Cancer by Data Mining and Network Pharmacology Methods

Objective To screen the key chinese Herbal medicines(KCHMs)against breast cancer by data mining,and analyze the potential mechanism of KCHMs using network pharmacology *** Clinical prescriptions consisted of CHMs for treating breast cancer were screened,and then traditional chinese medicine Inheritance Support System(TCMISS)was applied to obtain the ***,active ingredients and corresponding target genes of KCHMs were searched by traditional chinese medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database,and target genes of breast cancer were collected using OMIM and *** that,the overlapping target genes of KCHMs and breast cancer were screened,and the protein-protein interaction(PPI)network was *** addition,a network of“KCHMs-active ingredients-breast cancer-targets”was constructed by Cytoscape ***,Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene Ontology(GO)analysis were performed with Database for Annotation,Visualization and Integrated Discovery(DAVID)database to reveal the action mechanism of *** A total of 7 KCHMs were identified,whose active ingredients include quercetin,luteolin,nobiletin,kaempferol,isorhamnetin,naringenin,and be-ta-sitosterol,*** on protein-protein interaction analysis,core targets were ESR1,MYC,CCND1,EGFR,CASP3,ERBB2,*** KEGG pathways(e.g,PI3K-Akt,p53,ErbB,and HIF-1 signaling pathways)were *** Based on the combination of the data mining method and network pharmacology approach,the therapeutic effect of KCHMs on breast cancer may be realized by acting on target genes and signaling pathways related to the formation and progression of breast cancer.
