Combination of fruquintinib with venetoclax for the treatment of colorectal cancer

Background:As a novel blocker of vascular endothelial growth factor receptor (VEGFR), fruquintinib has been approved for treating colorectal cancer (CRC). However, its dosage and therapeutic efficacy are limited by its widespread adverse reactions. Venetoclax, recognized as the initial inhibitor of B-cell lymphoma protein 2 (BCL2), has shown potential in boosting the effectiveness of immunotherapy against CRC. This study investigated the efficacy and mechanisms of fruquintinib combined with venetoclax in treating CRC.Methods and Materials:We developed a colon cancer mouse model with the CT26 colon cell line to demonstrate fruquintinib and venetoclax's efficacy against tumors. Then we employed various techniques to evaluate different aspects of the experimental outcomes. Immunohistochemistry was used to detect cell proliferation and angiogenesis in tumor tissues. Western blot analysis was utilized to examine the occurrence of cell apoptosis, and flow cytometry to quantitate immune cells within the tumor tissues. Moreover, immunofluorescence was employed to measure cytokine levels.Results:The strongest inhibition on tumor growth was achieved by the combination of fruquintinib with venetoclax, as opposed to individual drug use. Venetoclax was found to amplify the impact of fruquintinib, leading to decreased cancer cell proliferation, increased cancer cell apoptosis, lowered angiogenesis, better vascular structure normalization, and improved immune cell infiltration.Conclusion:Our findings indicate that the addition of venetoclax enhances the impact of fruquintinib on vascular normalization and modulation of the tumor immune microenvironment. Our study presents the justification for utilizing the fruquintinib and venetoclax combination in treating CRC. Venetoclax holds promise in being assimilated into anticancer medications for CRC.

A two-field-scan harmonic Hall voltage analysis for fast,accurate quantification of spin-orbit torques in magnetic heterostructures

We report on the development of a“two-field-scan”harmonic Hall voltage(HHV)analysis,which collects the second HHVas a function of a swept in-plane magnetic field at 45°and 0°relative to the excitation current,for the determination of the spin-orbit torques of transverse spins in magnetic heterostructures without significant perpendicular spins,longitudinal spins,and longitudinal/perpendicular Oersted *** demonstrate that this two-field-scan analysis is as accurate as the well-established but time-consuming angle-scan HHV analysis even in the presence of considerable thermoelectric effects but takes more than a factor of 7 less measurement *** also show that the fit of the HHV data from a single field scan at 0°,which is commonly employed in the literature,is not reliable because the employment of too many free parameters in the fitting of the very slowly varying HHV signal allows erroneous conclusion about the spin-orbit torque efficiencies.

Dzyaloshinskii-Moriya interaction(DMI)is the key ingredient of chiral spintronic phenomena and the emerging technologies based on such phenomena.A nonzero DMI usually occurs at magnetic interfaces or within non-centrosymmetric single ***,we report the observation of a strong unexpected DMI within a centrosymmetric polycrystalline ferromagnet that has neither a crystal inversion symmetry breaking nor a composition *** DMI is a bulk effect,increases with the thickness of the magnetic layer,and is insensitive to the symmetry of the interfaces or the neighboring *** observe a total DMI strength that is a factor of>2 greater than the highest interfacial DMI in the *** DMI most likely arises from the strong spin-orbit coupling,strong orbital hybrization,and a“hidden”long-range asymmetry in the *** discovery of the strong unconventional bulk DMI in centrosymmetric,composition-uniform magnetic single layers provides fundamental building blocks for the emerging field of spintronics and will stimulate the exploitation of unconventional spin-orbit phenomena in a wide range of materials.

Weathering-pedogenesis of Carbonate Rocks and Its Environmental Effects in Subtropical Region

We investigated the weathering-pedogenesis of carbonate rocks and its environmental effects in subtropical regions of China. The investigation demonstrated that the weathering- pedogenesis of carbonate rocks is the process of a joint action of corrosion and illuviation and metasomatism in subtropical region. It is characterized by multi-stage, multi-path and multi-style. With the persisting development of weathering-pedogenesis of carbonate rocks, metasomatic pedogenesis progressively became the main process of the weathering-pedogenesis and the dominant style of formation of minerals. And it proceeds through the whole process of evolution of the weathering-pedogenesis of carbonate rocks. The stage evolution of weathering-pedogenesis of carbonate rocks and the fractionation evolution of newly produced minerals are characterized by obvious vertically zoning structures and the rules of gradation of elements geochemical characteristics in the carbonate rocks weathering profiles. The geochemical process of weathering-pedogenesis of carbonate rocks can be divided into three geochemical evolution stages, i.e., the Ca, Mg-depletion and Si, Al-enrichment stage; the Fe, Mn enrichment stage and the Si-depletion and Al-enrichment stage in the subtropical regions. Consistent with the three geochemical evolution stages, the sequence of formation and evolution of minerals can be divided into the clay mineral stage; the Fe, Mn oxide and the gibbsite stage. The influence of weathering-pedogenesis of carbonate rocks on the chemical forms of heavy elements is mainly affected via newly produced components and minerals in the process of weathering-pedogenesis, e.g., iron oxide minerals and organic matters. The important mechanism for the mobilization, transport and pollution of F and As is affected the selective adsorption and desorption of F and As on the surface of iron oxide minerals in the subtropical karst zones, i.e., the selective adsorption and desorption on mineral surfaces of

The adsorption behaviors of Cr(VI) on laterite from Guizhou Province were studied in this paper, and the adsorption mechanism was discussed as well. Results showed that different mineral compositions in the laterite would cause differences in the capacity of laterite to absorb Cr(VI). Gibbsite, iron oxide minerals and non-crystalloids are the main contributors to enhancing the capacity of laterite to absorb Cr(VI). The pH of the solution is an important factor affecting the adsorption of Cr(VI) on laterite. Acidic environment (pH=2-5) is favorable to the adsorption of Cr(VI). The amount of adsorbed Cr(VI) decreases with increasing pH of the solution. With increasing initial concentrations of Cr(VI), the amount of adsorbed Cr(VI) increases first, and then decreases. The optimal adsorption concentration of Cr(VI) on laterite is 250 μg/mL. The adsorption of Cr(VI) on laterite is a rapid process, about 80% Cr(VI) will be adsorbed within 2 hours. And the adsorption of Cr(VI) on kaolinite is a slow process.

Enrichment of heavy metals in coal gangue by puff balls and mechanism research

Coal gangue with/without wild puff balls growing on or covering up were analyzed by means of inductively coupled plasma atomic emission spectrometry(ICP-AES) and atomic fluorescence spectrometry(AFS) to detect the contents of heavy metal elements in the plants. Seven heavy metals were chosen to do the experiments, which showed that the puff balls have strong enrichment capacity for heavy metals(As, Pb, Mn and Ni). The highest enrichment level was Mn, 220.3 mg/kg(dry weight) for puffball fruiting bodies, and the highest As, Cr and Pb enrichment levels for puffball mycelium were 111, 265.3 and 86 mg/kg, respectively. Cd, Mn and Ni could transfer from puffball mycelium to its fruiting bodies, while As, Cr, Pb and Co mainly accumulated in the mycelium. The contents of five organic acids(tartaric acid, glycolic acid, lactic acid, citric acid and malic acid) in puffballs were determined with high performance liquid chromatography. With water, ethanol and ethyl acetate as solvents, the puffball extracts performed dissolution reactions with coal gangue, in which the water extract(containing a large amount of organic acids) was most effective in releasing heavy metals, especially As, Pb, Cr, Mn, Co and Ni from coal gangue. As a pioneer species, puff balls absorbed the hazardous trace elements in coal gangue matrix to reduce its eco-toxicity, on the other hand, it raised the p H value of coal gangue, improving the vegetation growing conditions for coal gangue matrix to a certain extent and laying the foundation for ecological restoration of coal gangue dumps.

Preparation and Investigation of Arsenic Trioxide-loaded Polylactic Acid/Magnetic Hybrid Nanoparticles

In order to improve the therapeutic index and reduce the side effects of As2O3 so as to expand its clinical utility to solid tumors,As2O3-loaded polylactic acid（PLA）/magnetic nanoparticles（PAMNs） were prepared and the preparation conditions were optimized by the improved double emulsion solvent-evaporation method to obtain as a drug delivery system with a high As2O3 encapsulation efficiency and homogeneous nanosized *** loading of magnetic nanoparticles into the nanoparticles was confirmed by Fourier transform infrared（FTIR） *** drug loading,encapsulation efficiency and releasing properties of As2O3 in PAMNs were investigated by atomic fluorescence spectrometer ***,the in vitro antitumor efficacy of free As2O3 and As2O3-PAMNs against SMMC-7721 cell line was ***,confocal fluorescence microscopy was conducted to reveal the celluar uptake of As2O3-loaded PAMNs by SMMC-7721 cell line.

The geochemical characteristics of aqueous rare-earth elements in shallow karst groundwater in Guiyang City, China

Fifty-seven shallow groundwater samples were collected from Guiyang karst basin, China, to analyze the aqueous rare-earth elements in low-water seasons and it is shown that the total amount of rare-earth elements (ΣREE) in karst groundwater is exceedingly low compared with that in carbonate rocks or weathering crusts of carbonate rocks, and ranges from 0.01 to 0.43, from 0.03 to 0.27, from 0.03 to 0.19 and from 0.05 to 1.38 μg·L-1 for dolomite, dolomitic & limestone, limestone and clastic rock aquifer, respectively. Both distributions and contents of rare-earth elements (REE) in karst groundwater reflect the lithology of host rocks or weathering crusts of carbonate rocks through which groundwater flows. The chondrite-normalized patterns show a non-flat profile with higher enrichment of slightly light rare-earth elements (LREE) than heavy rare-earth elements (HREE), prominent fractionation between LREE and HREE, negative Ce anomalies and negative or positive Eu anomalies. There is more obvious fractionation between LREE and HREE in groundwater than that in carbonate rocks and their weathering crusts due to high contents of HCO3? and PH in groundwater. In shallow karst groundwater, REE(CO3)n2n-3 (n=1 and 2) is the main inorganic species of REE. But for a clastic rock aquifer, both REESO4+ and REECO3+ are the main inorganic species of REE. Species of REE in groundwater is closely associated with the hydrochemical type of groundwater which is predominated by the lithology of host rocks, groundwater-rock interaction and weathering-pedogenesis of carbonate rocks.
