The application of microorganisms as probiotics is limited due to lack of safety evaluation.Here,a novel multi-stress-tolerant yeast Meyerozyma guilliermondii GXDK6 with aroma-producing properties was identified from marine mangrove microorganisms.Its safety and probiotic properties were assessed in accordance with phenotype and whole-genome sequencing analysis.Results showed that the genes and phenotypic expression of related virulence,antibiotic resistance and retroelement were rarely found.Hyphal morphogenesis genes(SIT4,HOG1,SPA2,ERK1,ICL1,CST20,HSP104,TPS1,and RHO1)and phospholipase secretion gene(VPS4)were annotated.True hyphae and phospholipase were absent.Only one retroelement(Tad1-65_BG)was found.Major biogenic amines(BAs)encoding genes were absent,except for spermidine synthase(JA9_002594),spermine synthase(JA9_004690),and tyrosine decarboxylase(inx).The production of single BAs and total BAs was far below the food-defined thresholds.GXDK6 had no resistance to common antifungal drugs.Virulence enzymes,such as gelatinase,DNase,hemolytic,lecithinase,and thrombin were absent.Acute toxicity test with mice demonstrated that GXDK6 is safe.GXDK6 has a good reproduction ability in the simulation gastrointestinal tract.GXDK6 also has a strong antioxidant ability,β-glucosidase,and inulinase activity.To sum up,GXDK6 is considered as a safe probiotic for human consumption and food fermentation.

The occurrence of benign prostate hyperplasia(BPH)was related to disrupted sex steroid hormones,and metformin(Met)had a clinical response to sex steroid hormone-related gynaecological disease.However,whether Met exerts an antiproliferative effect on BPH via sex steroid hormones remains unclear.Here,our clinical study showed that along with prostatic epithelial cell(PEC)proliferation,sex steroid hormones were dysregulated in the serum and prostate of BPH patients.As the major contributor to dysregulated sex steroid hormones,elevated dihydrotestosterone(DHT)had a significant positive relationship with the clinical characteristics of BPH patients.Activation of adenosine 5'-monophosphate(AMP)-activated protein kinase(AMPK)by Met restored dysregulated sex steroid hormone homeostasis and exerted antiproliferative effects against DHT-induced proliferation by inhibiting the formation of androgen receptor(AR)-mediated Yes-associated protein(YAP1)-TEA domain transcription factor(TEAD4)heterodimers.Met’s anti-proliferative effects were blocked by AMPK inhibitor or YAP1 overexpression in DHT-cultured BPH-1 cells.Our findings indicated that Met would be a promising clinical therapeutic approach for BPH by inhibiting dysregulated steroid hormone-induced PEC proliferation.

Evaluating the performance of genomic selection on purebred population by incorporating crossbred data in pigs

Genomic selection(GS)has been widely used in livestock,which greatly accelerated the genetic progress of complex traits.The population size was one of the significant factors affecting the prediction accuracy,while it was limited by the purebred population.Compared to directly combining two uncorrelated purebred populations to extend the reference population size,it might be more meaningful to incorporate the correlated crossbreds into reference population for genomic prediction.In this study,we simulated purebred offspring(PAS and PBS)and crossbred offspring(CAB)base on real genotype data of two base purebred populations(PA and PB),to evaluate the performance of genomic selection on purebred while incorporating crossbred information.The results showed that selecting key crossbred individuals via maximizing the expected genetic relationship(REL)was better than the other methods(individuals closet or farthest to the purebred population,CP/FP)in term of the prediction accuracy.Furthermore,the prediction accuracy of reference populations combining PA and CAB was significantly better only based on PA,which was similar to combine PA and PAS.Moreover,the rank correlation between the multiple of the increased relationship(MIR)and reliability improvement was 0.60-0.70.But for individuals with low correlation(Cor(Pi,PA or B),the reliability improvement was significantly lower than other individuals.Our findings suggested that incorporating crossbred into purebred population could improve the performance of genetic prediction compared with using the purebred population only.The genetic relationship between purebred and crossbred population is a key factor determining the increased reliability while incorporating crossbred population in the genomic prediction on pure bred individuals.

Tumor-resident microbiota in breast cancer promotes cancer initiation and malignant progression.However,targeting microbiota to improve the effects of breast cancer therapy has not been investigated in detail.Here,we evaluated the microbiota composition of breast tumors and found that enterotoxigenic Bacteroides fragilis(ETBF)was highly enriched in the tumors of patients who did not respond to taxane-based neoadjuvant chemotherapy.ETBF,albeit at low biomass,secreted the toxic protein BFT-1 to promote breast cancer cell stemness and chemoresistance.Mechanistic studies showed that BFT-1 directly bound to NOD1 and stabilized NOD1 protein.NOD1 was highly expressed on ALDH+breast cancer stem cells(BCSCs)and cooperated with GAK to phosphorylate NUMB and promote its lysosomal degradation,thereby activating the NOTCH1-HEY1 signaling pathway to increase BCSCs.NOD1 inhibition and ETBF clearance increase the chemosensitivity of breast cancer by impairing BCSCs.

循环水净化养殖过程中大口黑鲈生长及肌肉品质变化

为了探究循环水净化养殖对大口黑鲈形体指标、质构特性、营养成分及土腥味物质的影响,采用质构仪测定鱼肉硬度、弹性、咀嚼性等指标;采用常规生化方法、氨基酸自动分析仪、气相色谱检测鱼肉水分、灰分、粗蛋白、粗脂肪、脂肪酸、氨基酸等成分;采用气相色谱-质谱联用仪结合电子舌对鱼肉的土腥味物质和滋味进行分析。结果显示:鱼体肝体比随着净养时间的延长显著降低;相较于净养前,净养后20d内鱼肉硬度、内聚性、咀嚼性、弹性、胶黏性和黏附性均提高;净养过程中,鱼肉中的粗脂肪含量先上升后下降,水分含量则与之相反,并且净养后15d内鱼肉中粗蛋白、灰分、呈味氨基酸、必需氨基酸、单不饱和脂肪酸含量均提高,其中在5d和15d时脂肪酸中的二十碳五烯酸甲酯和二十二碳六烯酸甲酯之和、ω-3族脂肪酸含量显著升高;鱼肉中水溶性蛋白和非蛋白氮含量均显著升高;随着净养时间延长,肌肉滋味差异增大,土腥味物质含量降低,且从15d开始低于感官阈值。综合考虑净养后大口黑鲈形体指标、肌肉质构、营养成分和土腥味物质各项指标后,建议净养时间为15d。

库岸危岩剪切带—基岩界面宏细观剪切贯通机制及力学特性研究

以三峡库区“板壁岩”危岩为研究背景,在总结岸坡危岩内部剪切带形成过程及基本特征的基础上,通过室内剪切试验和数值模拟手段,从宏细观的角度研究该岸坡危岩内部剪切带–基岩界面的剪切贯通机制及其力学特性。研究表明:(1)剪切带为危岩体失稳的主控因素,内部充填物质具有层状无胶结特征;(2)在常法向应力作用下,界面的破坏模式可以总结为台阶根部张裂–台阶爬坡,岩板损伤–台阶剪断,岩板裂纹贯通–台阶剪碎;(3)界面剪切应力–剪切位移曲线分为压密阶段、近加载端台阶根部斜裂缝出现阶段、近似弹性变形及微裂隙扩展阶段、近加载端台阶剪断阶段、剩余台阶剪断阶段及残余阶段6个阶段;(4)界面的峰值剪切强度和残余强度随法向应力、层厚和台阶高度的增加而增加,竖向累积位移随法向应力的增加而减小、随层厚和台阶高度的增加而增加;(5)界面裂纹数量变化曲线呈“S”型,共经历裂纹缓慢增加(岩板表面磨损)–裂纹迅速扩展(台阶剪断)–裂纹数量稳定(残余摩擦)3个发展阶段,试样内部颗粒应变能和胶结应变能变化趋势与剪切应力–剪切位移曲线变化相近,总能量、摩擦能及阻尼能随剪切位移的增加而增加。研究成果对剪切带整体力学性质及含剪切带危岩稳定性的研究具有借鉴意义。

A pilot study on ^(99m)Tc-3PRGD2 scintigraphy in diagnosis of brain glioma

The 99mTc-3PRGD2 targeted SPECT/CT scanning was of significance in detecting differentiated glioma.In this work,the diagnostic value of 99mTc-3PRGD2 scintigraphy in brain glioma was evaluated by the ten clinically verified brain glioma patients after obtaining informed consent.The patients first accepted X-ray imaging to localize the detecting regions before administrating with 99mTc-3PRGD2 at a mean radioactivity of 849±115 MBq via single intravenous bolus injection 2 h prior to SPECT/CT imaging.Tumor samples for detectingαvβ3 were collected by surgical operations two weeks after the scintigraphy.The results of CT and SPECT scanning were merged and compared.The correlation between tumor occupation（T/N ratio） andαvβ3 expression level were analyzed.The T/N ratios in brain glioma were proportionally correlated to av(33 positive cell percentage（R2=0.9253,p<0.05）.This study primarily evaluated the clinical application of 99mTc-3PRGD2 SPECT scintigraphy on brain glioma.The more pathological types and detecting strategies covering a large amount of samples would aid to clarify the potentials.

多糖作为生物医药和食品加工与生产中广泛使用的高分子聚合物之一,具有抗肿瘤、免疫调节、降血糖、抗氧化、抑菌等生物活性。越来越多的学者从动物、微生物和植物中分离与纯化多糖并对其生物活性及应用进行研究。多糖的生物活性受单糖组成、分子量、糖苷键类型、分支度、空间构象等多种因素影响,如何实现活性多糖的设计、构建、筛选和高效生产一直是多糖研究的重点和难点。基于分子量与多糖生物活性的构效关系进行综述,介绍分子量对多糖抗氧化、抗肿瘤、抗炎、益生、免疫调节和抑菌等活性的影响,并对由分子量差异造成的多糖生物活性变化进行了讨论和展望,以期为从分子量角度改善多糖生物活性的研究及应用提供理论参考。

目的探讨基于学习风格的培训模式在新护士规范化培训中的应用效果。方法将2021年新护士63名设为对照组,2022年新护士61名设为观察组。对照组按照《新入职护士培训大纲(试行)》进行培训;观察组在培训大纲基础上根据学习风格分类匹配带教老师,同时根据新护士的学习方式偏好和学习特点提供相对应的学习环境及相匹配的教学方法。结果培训1年后,观察组自主学习能力总分和各维度评分及临床推理能力得分显著高于对照组(均P<0.05);观察组理论考核成绩、客观结构化临床考试成绩及培训满意度显著高于对照组(均P<0.05)。结论新护士基于学习风格的规范化培训有利于提升新护士自主学习能力、临床推理能力,提高培训效果及培训满意度。
