Background:Bladder cancer,characterized by a high potential of tumor recurrence,has high lifelong monitoring and treatment *** date,tumor cells with intrinsic softness have been identified to function as cancer stem cells in several cancer ***,the existence of soft tumor cells in bladder tumors remains ***,our study aimed to develop a microbarrier microfluidic chip to efficiently isolate deformable tumor cells from distinct types of bladder cancer ***:The stiffness of bladder cancer cells was determined by atomic force microscopy(AFM).The modified microfluidic chip was utilized to separate soft cells,and the 3D Matrigel culture system was to maintain the softness of tumor *** patterns of integrinβ8(ITGB8),protein kinase B(AKT),and mammalian target of rapamycin(mTOR)were determined by Western *** immunostaining was conducted to examine the interaction between F-actin and tripartite motif containing 59(TRIM59).The stem-cell-like characteristics of soft cells were explored by colony formation assay and in vivo studies upon xenografted tumor ***:Using our newly designed microfluidic approach,we identified a small fraction of soft tumor cells in bladder cancer *** importantly,the existence of soft tumor cells was confirmed in clinical human bladder cancer specimens,in which the number of soft tumor cells was associated with tumor ***,we demonstrated that the biomechanical stimuli arising from 3D Matrigel activated the F-actin/ITGB8/TRIM59/AKT/mTOR/glycolysis pathways to enhance the softness and tumorigenic capacity of tumor ***,we detected a remarkable up-regulation in ITGB8,TRIM59,and phospho-AKT in clinical bladder recurrent tumors compared with their non-recurrent ***:The ITGB8/TRIM59/AKT/mTOR/glycolysis axis plays a crucial role in modulating tumor softness and ***,the soft tumor cells become more sensitive to chemother

叶城沙蜥是一种主要分布于中国新疆南部地区的蜥蜴,通过测量和比较采自新疆塔里木盆地且末县叶城沙蜥的个体大小以及口宽等8个形态特征,研究了叶城沙蜥的两性异形。研究共采集叶城沙蜥62只,其中成年雌性个体27只,雄性个体35只。结果显示:(1)叶城沙蜥雌雄性成体的体长有显著差异,雄性大于雌性,表明叶城沙蜥具有雄性偏大的两性异形模式;(2)叶城沙蜥其他身体形态特征中尾长、腹长和头长3个特征有显著差异,其中雄性的尾长和头长大于雌性,而雌性的腹长大于雄性;(3)叶城沙蜥雌雄性个体间的口宽、眼间距等其他形态特征则没有显著差异。叶城沙蜥的两性异形表现为雄性大于雌性,并且雄性具有更大的头部和更长的尾部,雌性具有更长有腹部,这可能是雄性个体面临的性选择压力与雌性个体面临的繁殖压力共同作用的结果。

锂离子电池内部生热导致的加速老化和热安全问题一直以来都是制约电池产业的瓶颈问题。此外,单体老化路径和一致性的差异也会造成电池老化程度不一致,引发电池内阻和发热量的差异。基于上述问题,提出了一种考虑老化一致性的电池组热管理方法,搭建锂离子电池热-电耦合模型对电池进行参数辨识,并利用有限元分析的方法对Z型槽空气冷却电池组进行三维建模。分析不同外界电流应力、入口风速以及不同老化状态对电池组温度分布的影响。进行了仿真与实验验证,结果表明实验与仿真温度结果误差在2℃以内。

One of the major challenges in single-cell data analysis is the determination of cellular developmental trajectories using single-cell *** substantial studies have been conducted in recent years,more effective methods are still strongly needed to infer the developmental processes *** work devises a new method,named DTFLOW,for determining the pseudotemporal trajectories with multiple *** consists of two major steps:a new method called Bhattacharyya kernel feature decomposition(BKFD)to reduce the data dimensions,and a novel approach named Reverse Searching on k-nearest neighbor graph(RSKG)to identify the multi-branching processes of cellular *** BKFD,we first establish a stationary distribution for each cell to represent the transition of cellular developmental states based on the random walk with restart algorithm,and then propose a new distance metric for calculating pseudotime of single cells by introducing the Bhattacharyya kernel *** effectiveness of DTFLOW is rigorously examined by using four single-cell *** compare the efficiency of DTFLOW with the published state-of-the-art *** results suggest that DTFLOW has superior accuracy and strong robustness properties for constructing pseudotime *** Python source code of DTFLOW can be freely accessed at https://***/statway/DTFLOW.

Nowadays,high-performance computing(HPC)clusters are increasingly *** volumes of job logs recording many years of operation traces have been *** the same time,the HPC cloud makes it possible to access HPC services *** executing applications,both HPC end-users and cloud users need to request specific resources for different workloads by *** users are usually not familiar with the hardware details and software layers,as well as the performance behavior of the underlying HPC *** is hard for them to select optimal resource configurations in terms of performance,cost,and energy ***,how to provide on-demand services with intelligent resource allocation is a critical issue in the HPC *** of job characteristics plays a key role for intelligent resource *** paper presents a survey of the existing work and future directions for prediction of job characteristics for intelligent resource allocation in HPC *** first review the existing techniques in obtaining performance and energy consumption data of *** we survey the techniques for single-objective oriented predictions on runtime,queue time,power and energy consumption,cost and optimal resource configuration for input jobs,as well as multi-objective oriented *** conclude after discussing future trends,research challenges and possible solutions towards intelligent resource allocation in HPC systems.

Great earthquakes often occur along or near active fault belts. Thus,monitoring and research on fault deformation are quite important. Methods such as short-leveling,shortbaseline and integrated monitoring profile across fault belts have been used to monitor fault activities for many years. GNSS observations are mainly used to obtain the horizontal velocity field in large areas and to study the activities and deformation of major *** technology has been used to monitor and study the deformation of faults from a different aspects. In this paper,some applications and new explorations of GNSS are discussed. They are:( 1) Research and monitoring of strike-slip activities of faults with GNSS.( 2) Research and monitoring of vertical activities of faults with GNSS.( 3)Investigating the laws of deformation of blocks on the sides of fault zone and setting up strain models to deduce the activities and deformation of faults with respective models and compare the deduced results with the actual measurements across fault. It is concluded that a larger discrepancy between the deduced and the observed deformation indicates a stronger interaction between the blocks,which can be important for predicting the location of a strong earthquake and assessing seismic hazard,as well as the seismicity trend.
