鸟嘌呤核苷酸解离抑制剂(GDP dissociation inhibitor,GDI)在植物、哺乳动物等物种中普遍存在,是鸟苷三磷酸酶(GTPases)活性的关键调控因子之一,在GTPases活性调控的过程中起着至关重要的作用。GTPases能够结合并水解GTP,GDI可参与调节小G蛋白在GTP结合的激活态和GDP结合的失活态之间转换,控制众多信号转导途径。结合已有的文献报道,总结GDI在哺乳动物和植物等不同生物中作用的研究进展。其中,在疾病发生过程中,GDI主要参与人类癌症、血管透性等相关疾病的调控;除此之外,GDI也在胰岛素分泌、男性不育和癫前期病程等过程中发挥着重要作用。在植物中,GDI参与调控花粉管或根毛的极性生长,影响植物的形态建成,同时在植物抗逆过程中也扮演着重要角色。为GDI生物学功能的研究提供理论基础。

Background:Cancer incidence and mortality have received critical attention during the long-term management of morbidities in patients with autoimmune diseases(AIDs).This study aimed to investigate and compare the risk of cancer associated with five major AIDs in a large-scale Chinese ***:A total of 8,120 AID patients consecutively admitted to a national tertiary referral center in China were included and followed-up for 38,726.55 patient-years,including those with systemic lupus erythematosus(SLE),rheumatoid arthritis(RA),Sjoren’s syndrome(SS),systemic scleroderma(SSc),and idiopathic inflammatory myositis(IIM).Demographic data,cancer incidence,predilecting sites and cancer onset time were recorded and compared among the five ***:Four hundred and thirty(5.3%)patients developed *** median agewas 57.5 years and AID durationwas 79.8 *** estimated total standardized incidence ratio(SIR)of cancer in AIDs patients was 3.37,with the highest SIR observed in IIM(4.31),followed by RA(3.99),SSc(3.77),SS(2.88)and SLE(2.58).The increased SIR of cancers in AID patients showed a female predominance(female ***:3.59 vs.2.77)and younger patient involvement(age<50 vs.≥50 years:4.88 vs.3.04).Patientswith SLE had increased SIRs for developing hematologic malignancies and solid tumors located in the urinary bladder,corpus uteri and cervix *** with SS had a significantly high SIR for developing non-Hodgkin’s *** 3 years of IIM diagnosis,74.6%of the patients developed cancer and they had a high risk of ovarian *** was associated with a wide distribution of scancers,including non-Hodgkin’s lymphoma,gynecologic,urinary tract,thyroid gland and lung *** patients had increased SIRs for developing cervical uterine,lung,and breast ***:Patients with five major AIDs in China had an increased risk of developing cancer,with a predominance in women and younger patients,although cancer incidence,predilection site

Alphaviruses are a group of important viruses that cause significant diseases in *** them,Semliki Forest vi-rus(SFV)not only causes symptoms such as joint pain but also infects neuron cells and induces encephalitis in ***,the very-low-density lipoprotein receptor(VLDLR)was identified as the cellular receptor for SFV *** this study,we present the cryo-electron microscopy structure of SFV bound to human *** targets E1-DIII region of SFV using its membrane-distal LDLR class A(LA)*** and functional analyses emphasize the synergistic role of multiple VLDLR repeats in the SFV ***,VLDLR’s binding mode to SFV closely mirrors that of minor group human rhinoviruses but differs significantly from other alphaviruses’interactions with receptors in the canyon re-gion of the E *** also assessed SFV binding to VLDLR or apolipoprotein E receptor 2(ApoER2)proteins in horses and mosquitoes and revealed their use of multiple but different LA repeats for *** findings illuminate SFV’s cross-species infectivity,offering insights into potential antiviral strategies against alphavirus infections.

Transcription factors of the APETALA2( AP2) subfamily have been reported to be involved in plant growth, development and abiotic ***, little is known about the function of AP2 in plants response to virus *** this study, TARGET OF EAT3 of Nicotiana tabacum( NtTOE3), one of the clades of AP2 subfamily, was identified as a virus response gene in *** demonstrated that NtTOE3 played a positive role in attenuating virus infection in tobacco by NtTOE3 transgenic lines, including overexpression, RNA interference and knockout mutants of ***, we found that overexpression of NtTOE3 increased the expression of genes related to salicylic acid pathway in transgenic tobacco ***, we verified that NtTOE3 could activate the expression of NtPRl by binding to the promoter of *** of AP2 subfamily are targeted by miR172, so we speculated that miR172 might act as an upstream regulatory factor of *** expected, the result of qRT-PCR analysis showed that the expression of miR172 was correlated with changes of the expression of NtTOE3 in *** plants infected by tobacco mosaic virus( TMV).Importantly,we demonstrated that miR172 was a negative regulator of viral resistance in tobacco.5’ RACE assay was then employed to confirm that NtTOE3 was a target of *** evidence indicated that the miR172/TOE3 module enhanced susceptibility of tobacco to TMV infection by regulating the expression of *** conclusion, our findings reveal a role of miR172/TOE3 module in the regulation of plant immune system and elucidate a novel antiviral molecular mechanism.

Background:With the accumulation of experience and evidence,guidelines for invasive intervention for acute pancreatitis(AP)have continuously evolved,followed in Peking Union Medical College Hospital(PUMCH).We aimed to review AP case series to help understand the impact of guideline evolution on the management and prognosis of AP in real-world ***:This was a single-center study of AP patients who had received invasive interventions from 1988 to *** demographics,baseline severity,imaging findings,and the indication,timing,and specific modalities of invasive interventions were *** composite primary endpoint was death during hospitalization or major complications after *** endpoints included pancreatic fistula,incision infection,number of interventions,length of intensive care stay,length of hospitalization,and total medical ***:A total of 195 patients were *** most common indication for invasive intervention was suspected infection,followed by persistent *** step-up and delayed strategies reduced the incidence of major complications or *** 35 years,the number of patients requiring surgery has gradually declined,and more patients need only minimally invasive procedures for *** incidence of the primary outcome decreased as well as the duration of ***:The management of AP patients at PUMCH in the last 35 years has undergone therapeutic guideline changes that support the efficacy and safety of the deferred step-up strategy in real-world practice.

以目前性能最为先进的第六代半潜式钻井海洋平台为研究背景,介绍了平台的两种供电模式;鉴于平台复杂的供电模式转换过程的可行性和设备[5]的性能约束[5],提出了一种预诊断的思想以提高供电模式转换的成功率;并应用多智能体的研究方法对供电模式的转换过程的预诊断进行了系统的描述。最后用组态软件仿真了在柴油发电机组和断路器故障状况下的供电模式转换预诊断界面,仿真结果与设定结果相同,证实了基于多智能体的预诊断策略能为供电模式的转换决策提供准确信息,提高了供电转换的成功率。
