Meloidogyne incognita is a devastating plant-parasitic nematode.Effectors play important roles during the stages of nematodes infection and parasitism,but their molecular functions remain largely unknown.In this study,we characterized a new effector,Minc03329,which contains signal peptide for secretion and a C-type lectin domain.The yeast signal sequence trap experiments indicated that the signal peptide of Minc03329 is functional.In situ hybridization showed that Minc03329 was specifically expressed in the subventral esophageal gland.Real-time qPCR confirmed that the expression level of Minc03329 transcript was significantly increased in pre-parasitic and parasitic second-stage juveniles(pre-J2s and par-J2s).Tobacco rattle virus(TRV)-mediated gene silencing of Minc03329 in host plants largely reduced the pathogenicity of nematodes.On the contrary,ectopic expression of Minc03329 in Arabidopsis thaliana significantly increased plant susceptibility to nematodes.Transient expression of Minc03329 in Nicotiana benthamiana leaves suppressed the programmed cell death triggered by the pro-apoptotic protein BAX.Moreover,the transcriptome analysis of Minc03329-transgenic Arabidopsis and wild type revealed that many defense-related genes were significantly down-regulated.Interestingly,some different expressed genes were involved in the formation of nematode feeding sites.These results revealed that Minc03329 is an important effector for M.incognita,suppressing host defense response and promoting pathogenicity.

本研究通过建立黄芪不同炮制品HPLC指纹图谱及多成分含量测定方法,并结合化学计量法对其进行比较分析,为黄芪炮制品的综合利用提供理论支持。采用HPLC建立黄芪炮制品指纹图谱并进行6种成分的含量测定,并通过聚类分析(hierarchical clustering analysis,HCA)、主成分分析(principal component analysis,PCA)和正交偏最小二乘-判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)评价不同黄芪炮制品的差异,找寻差异性成分。结果显示,不同炮制品(各10批)共得到20个共有峰,其中指认了6个共有峰,且6个成分在各自的质量浓度范围内线性关系良好(r>0.999),平均加样回收率为100.5%~102.1%,相对标准偏差(relative standard deviation,RSD)为0.77%~2.1%;指纹图谱相似度结果显示,蜜麸黄芪、蜜炙黄芪、盐黄芪、酒黄芪各自10批的相似度均在0.900以上,但不同类型炮制品的相似度降低;聚类分析可将四种炮制品聚为3类,主成分分析与聚类分析结果一致,可相互佐证,正交偏最小二乘-判别分析能有效的将4种炮制品区分,以变量重要性投影(variable importance of projection,VIP)大于1为依据,筛选了9种主要差异成分,确认了毛蕊异黄酮苷、芒柄花苷、5-羟甲基糠醛是引起黄芪不同炮制品间成分差异的主要标志性成分。该研究所建立的黄芪炮制品HPLC指纹图谱结合化学计量法的方法准确可靠,可为黄芪炮制品的质量控制和品质评价提供参考依据。

Fuzzy ontologics are efficient tools to handle fuzzy and uncertain knowledge on the semantic web; but there are heterogeneity problems when gaining interoperability among different fuzzy ontologies. This paper uses concept approximation between fuzzy ontologies based on instances to solve the heterogeneity problems. It firstly proposes an instance selection technology based on instance clustering and weighting to unify the fuzzy interpretation of different ontologies and reduce the number of instances to increase the efficiency. Then the paper resolves the problem of computing the approximations of concepts into the problem of computing the least upper approximations of atom concepts. It optimizes the search strategies by extending atom concept sets and defining the least upper bounds of concepts to reduce the searching space of the problem. An efficient algorithm for searching the least upper bounds of concept is given.

Objective Exposure to microgravity results in postflight cardiovascular deconditioning in astronauts.Vascular oxidative stress injury and mitochondrial dysfunction have been reported during this process.To elucidate the mechanism for this condition,we investigated whether mitochondrial oxidative stress regulates calcium homeostasis and vasoconstriction in hindlimb unweighted(HU)rat cerebral arteries.Methods Three-week HU was used to simulate microgravity in rats.The contractile responses to vasoconstrictors,mitochondrial fission/fusion,Ca^(2+) distribution,inositol 1,4,5-trisphosphate receptor(IP3 R)abundance,and the activities of voltage-gated K+channels(KV)and Ca^(2+)-activated K+channels(BKCa)were examined in rat cerebral vascular smooth muscle cells(VSMCs).Results An increase of cytoplasmic Ca^(2+) and a decrease of mitochondrial/sarcoplasmic reticulum(SR)Ca^(2+) were observed in HU rat cerebral VSMCs.The abundance of fusion proteins(mitofusin 1/2[MFN1/2])and fission proteins(dynamin-related protein 1[DRP1]and fission-mitochondrial 1[FIS1])was significantly downregulated and upregulated,respectively in HU rat cerebral VSMCs.The cerebrovascular contractile responses to vasoconstrictors were enhanced in HU rats compared to control rats,and IP3 R protein/mRNA levels were significantly upregulated.The current densities and open probabilities of KV and BKCa decreased and increased,respectively.Treatment with the mitochondrial-targeted antioxidant mitoTEMPO attenuated mitochondrial fission by upregulating MFN1/2 and downregulating DRP1/FIS1.It also decreased IP3 R expression levels and restored the activities of the KV and BKCa channels.MitoTEMPO restored the Ca^(2+) distribution in VSMCs and attenuated the enhanced vasoconstriction in HU rat cerebral arteries.Conclusion The present results suggest that mitochondrial oxidative stress enhances cerebral vasoconstriction by regulating calcium homeostasis during simulated microgravity.

目的:探讨肌骨超声对骨性膝关节炎患者滑膜病变的评估价值。方法:选取95例骨性膝关节炎患者作为观察组;同期70名体检健康者为对照组。比较两组对象软骨厚度、滑膜厚度及积液深度;X线与肌骨超声检查对观察组患者膝关节各类病变的检出情况;分析软骨厚度、滑膜厚度及积液深度与膝关节HSS评分的相关性。结果:观察组患者软骨、积液与滑膜厚度均高于对照组(P<0.05)。X线对关节间隙变窄、骨质疏松与骨质增生的检出率均高于肌骨超声(P<0.05);对滑膜增厚、腘窝囊肿、髌上囊积液、关节软骨退化、软骨下骨破坏、半月板损伤退变、韧带损伤等的检出率均低于肌骨超声(P<0.05)。相关性分析显示,骨性膝关节炎患者软骨厚度、积液厚度、滑膜厚度与膝关节HSS评分均呈负相关关系(r=-0.586、-0.712、-0.684,P<0.05)。结论:肌骨超声对于骨关节炎的滑膜病变临床诊断的准确率高,早期诊断敏感性好,有临床推广价值。

The lungs were long thought to be sterile until technical advances uncovered the presence of the lung microbial community.The microbiome of healthy lungs is mainly derived from the upper respiratory tract(URT)microbiome but also has its own characteristic flora.The selection mechanisms in the lung,including clearance by coughing,pulmonary macrophages,the oscillation of respiratory cilia,and bacterial inhibition by alveolar surfactant,keep the microbiome transient and mobile,which is different from the microbiome in other organs.The pulmonary bacteriome has been intensively studied recently,but relatively little research has focused on the mycobiome and virome.This up-to-date review retrospectively summarizes the lung microbiome’s history,composition,and function.We focus on the interaction of the lung microbiome with the oropharynx and gut microbiome and emphasize the role it plays in the innate and adaptive immune responses.More importantly,we focus on multiple respiratory diseases,including asthma,chronic obstructive pulmonary disease(COPD),fibrosis,bronchiectasis,and pneumonia.The impact of the lung microbiome on coronavirus disease 2019(COVID-19)and lung cancer has also been comprehensively studied.Furthermore,by summarizing the therapeutic potential of the lung microbiome in lung diseases and examining the shortcomings of the field,we propose an outlook of the direction of lung microbiome research.

Cyber-physical systems(CPSs)have emerged as an essential area of research in the last decade,providing a new paradigm for the integration of computational and physical units in modern control systems.Remote state estimation(RSE)is an indispensable functional module of CPSs.Recently,it has been demonstrated that malicious agents can manipulate data packets transmitted through unreliable channels of RSE,leading to severe estimation performance degradation.This paper aims to present an overview of recent advances in cyber-attacks and defensive countermeasures,with a specific focus on integrity attacks against RSE.Firstly,two representative frameworks for the synthesis of optimal deception attacks with various performance metrics and stealthiness constraints are discussed,which provide a deeper insight into the vulnerabilities of RSE.Secondly,a detailed review of typical attack detection and resilient estimation algorithms is included,illustrating the latest defensive measures safeguarding RSE from adversaries.Thirdly,some prevalent attacks impairing the confidentiality and data availability of RSE are examined from both attackers'and defenders'perspectives.Finally,several challenges and open problems are presented to inspire further exploration and future research in this field.
