Oxidative stress-initiated one-carbon metabolism drives the generation of interleukin-10-producing B cells to resolve pneumonia

The metabolic reprogramming underlying the generation of regulatory B cells during infectious diseases remains unknown.Using a Pseudomonas aeruginosa-induced pneumonia model,we reported that IL-10-producing B cells(IL-10+B cells)play a key role in spontaneously resolving infection-mediated inflammation.Accumulated cytosolic reactive oxygen species(ROS)during inflammation were shown to drive IL-10+B-cell generation by remodeling one-carbon metabolism.Depletion of the enzyme serine hydroxymethyltransferase 1(Shmt1)led to inadequate one-carbon metabolism and decreased IL-10+B-cell production.Furthermore,increased one-carbon flux elevated the levels of the methyl donor S-adenosylmethionine(SAM),altering histone H3 lysine 4 methylation(H3K4me)at the Il10 gene to promote chromatin accessibility and upregulate Il10 expression in B cells.Therefore,the one-carbon metabolism-associated compound ethacrynic acid(EA)was screened and found to potentially treat infectious pneumonia by boosting IL-10+B-cell generation.Overall,these findings reveal that ROS serve as modulators to resolve inflammation by reprogramming one-carbon metabolism pathways in B cells.

The Unprecedented Extreme Anticyclonic Anomaly over Northeast Asia in July 2021 and Its Climatic Impacts

This study investigates the evolution of an extreme anomalous anticyclone(AA)event over Northeast Asia,which was one of the dominant circulation systems responsible for the catastrophic extreme precipitation event in July 2021 in Henan,and further explores the significant impact of this AA on surface temperatures beneath it.The results indicate that this AA event over Northeast Asia was unprecedented in terms of intensity and duration.The AA was very persistent and extremely strong for 10 consecutive days from 13 to 22 July 2021.This long-lived and unprecedented AA led to the persistence of warmer surface temperatures beyond the temporal span of the pronounced 500-hPa anticyclonic signature as the surface air temperatures over land in Northeast Asia remained extremely warm through 29 July 2021.Moreover,the sea surface temperatures in the Sea of Japan/East Sea were extremely high for 30 consecutive days from 13 July to 11 August 2021,persisting well after the weakening or departure of this AA.These results emphasize the extreme nature of this AA over Northeast Asia in July 2021 and its role in multiple extreme climate events,even over remote regions.Furthermore,possible reasons for this long-lasting AA are explored,and it is suggested to be a byproduct of a teleconnection pattern over extratropical Eurasia during the first half of its life cycle,and of the Pacific-Japan teleconnection pattern during the latter half.

Structural Engineering of Hierarchical Magnetic/Carbon Nanocomposites via In Situ Growth for High-Efficient Electromagnetic Wave Absorption

Materials exhibiting high-performance electromagnetic wave absorption have garnered considerable scientific and technological attention,yet encounter significant challenges.Developing new materials and innovative structural design concepts is crucial for expanding the application field of electromagnetic wave absorption.Particularly,hierarchical structure engineering has emerged as a promising approach to enhance the physical and chemical properties of materials,providing immense potential for creating versatile electromagnetic wave absorption materials.Herein,an exceptional multi-dimensional hierarchical structure was meticulously devised,unleashing the full microwave attenuation capabilities through in situ growth,selfreduction,and multi-heterogeneous interface integration.The hierarchical structure features a three-dimensional carbon framework,where magnetic nanoparticles grow in situ on the carbon skeleton,creating a necklace-like structure.Furthermore,magnetic nanosheets assemble within this framework.Enhanced impedance matching was achieved by precisely adjusting component proportions,and intelligent integration of diverse interfaces bolstered dielectric polarization.The obtain Fe_(3)O_(4)-Fe nanoparticles/carbon nanofibers/Al-Fe_(3)O_(4)-Fe nanosheets composites demonstrated outstanding performance with a minimum reflection loss(RLmin)value of−59.3 dB and an effective absorption bandwidth(RL≤−10 dB)extending up to 5.6 GHz at 2.2 mm.These notable accomplishments offer fresh insights into the precision design of high-efficient electromagnetic wave absorption materials.

Evolutionary mechanisms and practical significance of reproductive success and clonal diversity in unisexual vertebrate polyploids

Unisexual reproduction is generally relevant to polyploidy, and unisexual vertebrates are often considered an evolutionary “dead end” due to the accumulation of deleterious mutations and absence of genetic diversity. However, some unisexual polyploids have developed strategies to avoid genomic decay, and thus provide ideal models to unveil unexplored evolutionary mechanisms, from the reproductive success to clonal diversity creation. This article reviews the evolutionary mechanisms for overcoming meiotic barrier and generating genetic diversity in unisexual vertebrates, and summarizes recent research advancements in the polyploid Carassius complex. Gynogenetic gibel carp(Carassius gibelio) is a unique amphitriploid that has undergone a recurrent autotriploidy and has overcome the bottleneck of triploid sterility via gynogenesis. Recently, an efficient strategy in which ploidy changes, including from amphitriploid to amphitetraploid, then from amphitetraploid to novel amphitriploid, drive unisexual-sexual-unisexual reproduction transition and clonal diversity has been revealed.Based on this new discovery, multigenomic reconstruction biotechnology has been used to breed a novel strain with superior growth and stronger disease resistance. Moreover, a unique reproduction mode that combines both abilities of ameiotic oogenesis and sperm-egg fusion,termed as ameio-fusiongensis, has been discovered, and it provides an efficient approach to synthesize sterile allopolyploids. In order to avoid ecological risks upon escape and protect the sustainable property rights of the aquaculture seed industry, a controllable fertility biotechnology approach for precise breeding is being developed by integrating sterile allopolyploid synthesis and gene-editing techniques.This review provides novel insights into the origin and evolution of unisexual vertebrates and into the attempts being made to exploit new breeding biotechnologies in aquaculture.

Neuronal conversion from glia to replenish the lost neurons

Neuronal injury,aging,and cerebrovascular and neurodegenerative diseases such as cerebral infarction,Alzheimer’s disease,Parkinson’s disease,frontotemporal dementia,amyotrophic lateral sclerosis,and Huntington’s disease are characte rized by significant neuronal loss.Unfo rtunately,the neurons of most mammals including humans do not possess the ability to self-regenerate.Replenishment of lost neurons becomes an appealing therapeutic strategy to reve rse the disease phenotype.Transplantation of pluripotent neural stem cells can supplement the missing neurons in the brain,but it carries the risk of causing gene mutation,tumorigenesis,severe inflammation,and obstructive hydrocephalus induced by brain edema.Conversion of neural or non-neural lineage cells into functional neurons is a promising strategy for the diseases involving neuron loss,which may overcome the above-mentioned disadvantages of neural stem cell therapy.Thus far,many strategies to transfo rm astrocytes,fibroblasts,microglia,Muller glia,NG2 cells,and other glial cells to mature and functional neurons,or for the conversion between neuronal subtypes have been developed thro ugh the regulation of transcription factors,polypyrimidine tra ct binding protein 1(PTBP1),and small chemical molecules or are based on a combination of several factors and the location in the central nervous system.However,some recent papers did not obtain expected results,and discrepancies exist.Therefore,in this review,we discuss the history of neuronal transdifferentiation,summarize the strategies for neuronal replenishment and conversion from glia,especially astrocytes,and point out that biosafety,new strategies,and the accurate origin of the truly co nverted neurons in vivo should be focused upon in future studies.It also arises the attention of replenishing the lost neurons from glia by gene therapies such as up-regulation of some transc ription factors or downregulation of PTBP1 or drug interfe rence therapies.

Urinary exosomal microRNA-145-5p and microRNA-27a-3p act as noninvasive diagnostic biomarkers for diabetic kidney disease

BACKGROUND Diabetic kidney disease(DKD),characterized by increased urinary microalbumin levels and decreased renal function,is the primary cause of end-stage renal di-sease.Its pathological mechanisms are complicated and multifactorial;Therefore,sensitive and specific biomarkers are needed.Urinary exosome originate from diverse renal cells in nephron segments and partially mirror the pathological changes in the kidney.The microRNAs(miRNAs)in urinary exosome are remark-ably stable and highly tissue-specific for the kidney.METHODS Type 2 diabetic mellitus(T2DM)patients were recruited from the Second Hospital of Hebei Medical University and were divided into two groups:DM,diabetic pa-tients without albuminuria[urinary albumin to creatinine ratio(UACR)luated.The diagnostic values of exosomal miR-145-5p and miR-27a-3p in DKD were determined using receiver operating characteristic(ROC)analysis.Biological functions of miR-145-5p were investigated by performing RESULTS Urinary exosomal expression of miR-145-5p and miR-27a-3p was more upregulated in the DKD group than in the DM group(miR-145-5p:4.54±1.45 vs 1.95±0.93,Plucose,gly-cosylated hemoglobin A1c,and low-density lipoprotein cholesterol.ROC analysis revealed that miR-145-5p had a better area under the curve of 0.88[95%confidence interval(CI):0.784-0.985,P<0.0001]in diagnosing DKD than miR-27a-3p with

Stochastic resonance is a phenomenon in which adding random noise to a system enhances the detection or transmission of a weak signal. It occurs when the noise interacts with the system's nonlinearity, thus improving signal-to-noise ratio and increasing sensitivity. Although parametric resonances in an atomic spinor Bose-Einstein condensate have been investigated, the question of whether one can observe stochastic resonance in such a system persists. In this study we propose a scheme for generating stochastic resonance in a cavity-spinor Bose-Einstein condensate coupling system. We demonstrate stochastic resonance through numerical calculations using the mean-field theory and truncated Wigner approximation methods. Furthermore, the characteristics of the system's response to noise and periodic signals are studied in detail. This study unravels a new scheme for observing stochastic resonance via linking atomic many-body physics with cavity quantum electrodynamics.

Echo State Network Based on Improved Knowledge Distillation for Edge Intelligence

Echo state network(ESN) as a novel artificial neural network has drawn much attention from time series prediction in edge intelligence. ESN is slightly insufficient in long-term memory, thereby impacting the prediction performance. It suffers from a higher computational overhead when deploying on edge devices. We firstly introduce the knowledge distillation into the reservoir structure optimization, and then propose the echo state network based on improved knowledge distillation(ESN-IKD) for edge intelligence to improve the prediction performance and reduce the computational overhead. The model of ESN-IKD is constructed with the classic ESN as a student network, the long and short-term memory network as a teacher network, and the ESN with double loop reservoir structure as an assistant network. The student network learns the long-term memory capability of the teacher network with the help of the assistant network. The training algorithm of ESN-IKD is proposed to correct the learning direction through the assistant network and eliminate the redundant knowledge through the iterative pruning. It can solve the problems of error learning and redundant learning in the traditional knowledge distillation process. Extensive experimental simulation shows that ESN-IKD has a good time series prediction performance in both long-term and short-term memory, and achieves a lower computational overhead.

Towards adaptive graph neural networks via solving prior-data conflicts

Graph neural networks(GNNs)have achieved remarkable performance in a variety of graph-related tasks.Recent evidence in the GNN community shows that such good performance can be attributed to the homophily prior;i.e.,connected nodes tend to have similar features and labels.However,in heterophilic settings where the features of connected nodes may vary significantly,GNN models exhibit notable performance deterioration.In this work,we formulate this problem as prior-data confict and propose a model called the mixture-prior graph neural network(MPGNN).First,to address the mismatch of homophily prior on heterophilic graphs,we introduce the non-informative prior,which makes no assumptions about the relationship between connected nodes and learns such relationship from the data.Second,to avoid performance degradation on homophilic graphs,we implement a soft switch to balance the effects of homophily prior and non-informative prior by learnable weights.We evaluate the performance of MPGNN on both synthetic and real-world graphs.Results show that MPGNN can effectively capture the relationship between connected nodes,while the soft switch helps select a suitable prior according to the graph characteristics.With these two designs,MPGNN outperforms state-of-the-art methods on heterophilic graphs without sacrificing performance on homophilic graphs.

Photonic neuromorphic architecture for tens-of-task lifelong learning

Scalable,high-capacity,and low-power computing architecture is the primary assurance for increasingly manifold and large-scale machine learning tasks.Traditional electronic artificial agents by conventional power-hungry processors have faced the issues of energy and scaling walls,hindering them from the sustainable performance improvement and iterative multi-task learning.Referring to another modality of light,photonic computing has been progressively applied in high-efficient neuromorphic systems.Here,we innovate a reconfigurable lifelong-learning optical neural network(L2 ONN),for highly-integrated tens-of-task machine intelligence with elaborated algorithm-hardware codesign.Benefiting from the inherent sparsity and parallelism in massive photonic connections,L2 ONN learns each single task by adaptively activating sparse photonic neuron connections in the coherent light field,while incrementally acquiring expertise on various tasks by gradually enlarging the activation.The multi-task optical features are parallelly processed by multi-spectrum representations allocated with different wavelengths.Extensive evaluations on freespace and on-chip architectures confirm that for the first time,L2 ONN avoided the catastrophic forgetting issue of photonic computing,owning versatile skills on challenging tens-of-tasks(vision classification,voice recognition,medical diagnosis,etc.)with a single model.Particularly,L2 ONN achieves more than an order of magnitude higher efficiency than the representative electronic artificial neural networks,and 14×larger capacity than existing optical neural networks while maintaining competitive performance on each individual task.The proposed photonic neuromorphic architecture points out a new form of lifelong learning scheme,permitting terminal/edge AI systems with light-speed efficiency and unprecedented scalability.
