Dynamics of the Eukaryotic Microbial Community at the Breeding Sites of the Large Yellow Croaker Pseudosciaena crocea in the Southern East China Sea

Clarifying eukaryotic microbial spatial distribution patterns and their determinants is an important idea in ecological research.However,information on the distribution patterns of eukaryotic microbial community structures(EMCSs)within oceans remains unclear.In this study,surface water samples from the southern East China Sea(SECS)were collected to investigate the spatiotemporal variation in EMCSs by using 18S rRNA high-throughput sequencing technology and the impact of this variation on Pseudosciaena crocea during the breeding season.The results indicated that the distribution patterns of the eukaryotic microbial community structure were different among the Sansha Bay,Mindong and Wentai reserves and the offshore East China Sea.In addition,there were notable potential effects of EMCSs on fishery activities.The variation partitioning analysis showed the environmental and spatial factors caused 53.4%of the variation in the EMCSs,indicating that spatially structured environmental factors were the key determinants of the EMCSs spatial heterogeneity in the SECS and may have contributed to the general distribution of P.crocea.In addition,all the environmental factors were the main factors driving the distribution of eukaryotic microbes except for total phosphorus.Furthermore,it was noted some phytoplankton such as Poterioochromonas and Rhizophydium of fungi in Sansha Bay can effectively prevent Cyanobacteria blooms.Chrysophyceae are natural high-quality baits for juvenile fish distributed in Sansha Bay,Mindong and Wentai reserves.This study provides a part of the insight into potential eukaryotic community distributions in large water bodies and how they are affected by environmental factors.

In situ Assemblies of Bacteria and Nutrient Dynamics in Response to an Ecosystem Engineer,Marine Clam Scapharca subcrenata,in the Sediment of an Aquaculture Bioremediation System

The relationships between nutrient dynamics,microbial community,and macrofauna are important in bioremediation systems.In this study,we examined the effects of marine clam Scapharca subcrenata sedimentary activities on the microbial assemblages,benthic nutrient chemistry,and its subsequent remediation impacts on organic effluent in the sediment of an aquaculture wastewater bioremediation system.The results showed that microbial community composition differed significantly in the clam area(ANOSIM,R=0.707,P=0.037).Pyrosequencing of bacterial 16S rRNA gene revealed a total of 48 unique phyla,79 classes,107 orders,197 families,and 321 genera amongst all samples.The most dominant bacterial assemblages were Proteobacteria,Bacteroidetes,Acidobacteria,Firmicutes,Verrucomicrobia,and Actinobacteria,with Bacteroidetes and Firmicutes significantly higher in all treatment samples than control(P<0.001).All dominant phyla in the list were shared across all samples and accounted for 89%(control)and 97%(treatment)of the total 16S rRNA.The nutrient flux rates from the sediments into the water(treatment group)were 51%(ammonium),88%(nitrate),77%(nitrite)and 45%(phosphate)higher,relative to the control implying increased mineralization,degradability,and mobility of the benthic nutrients.Similarly,significantly increased oxygen consumption rates were evident in the clam area signifying improved oxygen distribution within the sediment.The organic effluent contents associated with total-organic matter,carbon,nitrogen,and,phosphate were lower among the clam treatments relative to the control.Our results describe the potential roles and mechanisms contributed by marine bivalve S.subcrenata on benthic-bacterial-community assembly,nutrient balance,and effluent reduction in the sediments of aquaculture wastewaters bioremediation system.

Cloning of 9-cis-epoxycarotenoid dioxygenase (NCED) gene encoding a key enzyme during abscisic acid (ABA) biosynthesis and ABA-regulated ethylene production in detached young persimmon calyx

Unlike the typical climacteric fruits, persimmons (Diospyros kaki Thunb.) produce higher levels of ethylene when they are detached from trees at a younger stage. In order to obtain detailed information on the role of abscisic acid (ABA) in ripening, we cloned the DKNCED1, DKACS2, and DKACO1 genes from the calyx. Water loss was first noted in the calyx lobe, and DKNCED1 was highly expressed 1 d after the fruits were detached, coinciding with an increase in the ABA content. Then, the DKACS2 and DKACO1 genes were expressed after some delay. In the calyx, the ABA peak was observed 2 d after the fruits were harvested, and this peak preceded the ethylene peak observed on day 3. The fruit firmness rapidly decreased on day 4, and the fruits softened completely 6 d after they were harvested. The increases in the expressions of ABA, ethylene, and the genes in the calyxes occurred earlier than the corresponding increases in the pulp, although the 3 increases occurred on different days. Exogenous ABA treatment increased ABA concentration, induced expression of both ACS and ACO, and promoted ethylene synthesis and young-fruit softening; by contrast, treatment with NDGA inhibited the gene expressions and ethylene synthesis and delayed young-fruit softening. These results indicate that ethylene biosynthesis in the detached young persimmon fruits is initially triggered by ABA, which is induced by water loss in the calyx, through the induction of DKACS2 and DKACO1 expressions. The ethylene produced in the calyx subsequently diffuses into the pulp tissue, where it induces autocatalytic ethylene biosynthesis, resulting in an abrupt increase in ethylene production.

NMR-based metabolomic responses of freshwater gastropod Bellamya aeruginosa to MC-producing and non MC-producing Microcystis aeruginosa

Molluscan metabolomic analysis is essential for the understanding of the regulatory mechanism of aquatic invertebrate in response to hepatotoxic microcystins(MCs)stress.To understand the system responses of the gastropod to MC exposure,metabolomic alterations caused by two strains(MC-producing and non MC-producing)of Microcystis aeruginosa were characterized indiff erent biological matrices(hepatopancreas and muscle)of Bellamya aeruginosa(Gastropoda)using 1 H nuclear magnetic resonance(NMR)spectroscopy combined with MCs detections after exposure for 1,7,and 14 d.Although ELISA analysis showed that no MCs was detected in both tissues after non MC-producing M.aeruginosa exposure,MCs concentrations were increasing in the hepatopancreas(from 1.29±0.48μg/g to 3.17±0.11μg/g)and foot muscle(from 0.07±0.02μg/g to 0.21±0.08μg/g)after 14-d exposure of MC-producing M.aeruginosa.Meanwhile,we observed that MC induced signifi cant increase in creatine,a variety of amino acids(leucine,isoleucine,valine,threonine,alanine,methionine,glutamate,aspartate,and lysine),carboxylic acids(lactate,acetate,and D-3-hydroxybutyrate),and choline and its derivatives(phosphocholine and glycerophosphocholine)but decreased the energy substance(lipids,glucose,and glycogen)in the hepatopancreas.However,no signifi cant metabolite diff erences were observed in the muscle between MC-producing and non MC-producing cyanobacteria treated groups.These results suggest that MC exposure may cause hepatic energy expenditure accompanied with various metabolic disorders that involve lipid metabolism,protein catabolism,osmoregulation,glycolysis,glycogenolysis,and tricarboxylic acid(TCA)cycle.Moreover,metabolic perturbation was aggravated as the level of accumulated MCs raised over time in the MC-producing cyanobacteria treatment.These fi ndings indicated that MCs accumulation might lead to oxidative-stress-mediated damage of mitochondria functions.

Spatial-temporal dynamics of bacterioplankton communities in the breeding area of large yellow croaker Larimichthys crocea in Sansha Bay,China

As an important spawning ground for large yellow croaker Larimichthys crocea,Sansha Bay,South China Sea has been a research hotspot.However,studies on the influence of the bacterioplankton community and assessments of its seasonal succession of bacterioplankton in different sea areas in Sansha Bay are still limited.To address the issue,we use 16S rRNA gene amplicon sequencing and functional prediction to investigate the spatial-temporal dynamics of the bacterioplankton community in three distinct areas,i.e.,Breeding Area(BA),Yantian Harbor(YH),and Bay Margin(BM)of Sansha Bay.Results show that the structure of the bacterioplankton community in Sansha Bay had a significant seasonal succession.Moreover,the representative zero-radius Operation Taxon Units in different seasons were significantly different among the three selected sea areas.Specifically,during the breeding season,bacterioplankton communities in BA were characterized by compound-degrading bacteria,such as Rhodococcus and Owenweeksia,while in YH and BM,animal parasites or symbionts such as Vibrio and Arcobacter were dominant.Furthermore,the redundancy analysis and Spearman correlation analysis further explained that water temperature,dissolved oxygen,and ammonia nitrogen were the main environmental factors responsible for the difference.In addition,the bioindicator functions screened by Functional Annotation of Prokaryotic Taxa and random forest machine learning mainly relied on compound degradation,nitrite oxidation,and photoheterotrophy.The compound-degradationcorresponded bacterioplankton genera such as Rhodococcus had relatively higher abundance in BM,while Nitrospina corresponding to nitrite oxidation tended to be abundant in YH and BA.Based on the spatial and temporal variation in the composition and function of bacterioplankton,our findings provide a basis for understanding the theory of bacterioplankton community structure in the inner-bay habitat of the large yellow croaker in Sansha Bay.

Two-dimension(2D)van der Waals heterojunction holds essential promise in achieving high-performance flexible near-infrared(NIR)photodetector.Here,we report the successful fabrication of ZnSb/Ti_(3)C_(2)T_(x)MXene based flexible NIR photodetector array via a facile photolithography technology.The single ZnSb/Ti_(3)C_(2)T_(x)photodetector exhibited a high light-to-dark current ratio of 4.98,fast response/recovery time(2.5/1.3 s)and excellent stability due to the tight connection between 2D ZnSb nanoplates and 2D Ti_(3)C_(2)T_(x)MXene nanoflakes,and the formed 2D van der Waals heterojunction.Thin polyethylene terephthalate(PET)substrate enables the ZnSb/Ti_(3)C_(2)T_(x)photodetector withstand bending such that stable photoelectrical properties with non-obvious change were maintained over 5000 bending cycles.Moreover,the ZnSb/Ti_(3)C_(2)T_(x)photodetectors were integrated into a 26×5 device array,realizing a NIR image sensing application.

Eukaryotic microbial distribution pattern and its potential effects on fisheries in the fish reserves of Qiantang River in breeding season

To examine the eukaryotic biodiversity of aquatic ecosystems in the Qiantang River,China,eukaryotic microbes in the river were investigated using 18S rRNA gene sequencing during the breeding season(July to August 2018).Four distinct distribution patterns(1.Jiande;2.Tonglu and Fuyang;3.Jiubao;4.Yanguan)of the microbial community and their potential effects on fishery activities were observed.Results show lower abundances of Dinophyta and Fungi and higher abundances of Cryptophyta and Chlorophyta in Tonglu and Fuyang than those in the other three sections.In addition,the reserves(Tonglu and Fuyang)destabilized the original eukaryotic microbial co-occurrence network.Among all the environmental factors measured,nitrogen(nitrite,nitrate,ammonium),water temperature and total chlorophyll a acted as major driving factors that controlled the eukaryotic microbial distribution.Furthermore,the existence of some algae(e.g.,Chrysophyceae,Cryptophytes,and Chlorophyceae)and fungi(e.g.,Rhizophydium)in Tonglu and Fuyang was beneficial to juvenile fish growth and water quality,although some detrimental species(e.g.,Aphanomyces)needed attention.This study provides further insights into the sustainable protection and utilization of rivers.

A nanoagent for concurrent therapy of breast cancer bone metastasis and cancer-induced bone pain through SLC7A11 interruption and photodynamic therapy

Bone metastasis,a life-threatening complication of advanced breast cancer,is often accompanied by debilitating pain(cancer-induced bone pain,CIBP)that severely impairs life quality and survival.The concurrent treatment of bone metastases and CIBP remains a clinical challenge because the therapeutic options are limited.In this study,we construct a near-infrared light-activated nano-therapeutic system to meet this conundrum.In detail,sorafenib(SRF)and photosensitizer(chlorin e6,Ce6)are encapsulated into mesoporous hydroxyapatite nanoparticles(HANPs),which are further functionalized with hyaluronic acid(HA)to obtain HA-SRF/Ce6@HANPs system.The designed nanoplatform destroys tumor cells in vitro and in vivo via the synergism of SRF(interrupting the exchange of cystine/glutamate by inhibiting SLC7A11)and photodynamic therapy(PDT,inducing reactive oxygen species generation).The decrease in tumor burden and reduction of extracellular glutamate significantly attenuate CIBP in mice model with developing bone cancer.Moreover,the combination of HA-SRF/Ce6@HANPs and PDT inhibit osteoclasts activation,promote osteoblast differentiation and accelerate bone repair.Overall,the nanoagent with good biocompatibility may provide an effective therapy method for the concurrent treatment of breast cancer bone metastasis and CIBP.

Rice production accounts for approximately half of the freshwater resources utilized in agriculture,result-ing in greenhouse gas emissions such as methane(CH4)from flooded paddy fields.To address this chal-lenge,environmentally friendly and cost-effective water-saving techniques have become widely adopted in rice cultivation.However,the implementation of water-saving treatments(WsTs)in paddy-field rice has been associated with a substantial yield loss of up to 50%as well as a reduction in nitrogen use efficiency(NUE).In this study,we discovered that the target of rapamycin(TOR)signaling pathway is compromised in rice under WsT.Polysome profiling-coupled transcriptome sequencing(polysome-seq)analysis unveiled a substantial reduction in global translation in response to WST associated with the downregulation of TOR activity.Molecular,biochemical,and genetic analyses revealed new insights into the impact of the positive TOR-S6K-RPS6 and negative TOR-MAF1 modules on translation repression under WST.Intriguingly,ammonium exhibited a greater ability to alleviate growth constraints under WsT by enhancing TOR signaling,which simultaneously promoted uptake and utilization of ammonium and nitrogen allocation.We further demonstrated that TOR modulates the ammonium transporter AMT1;1 as well as the amino acid permease APP1 and dipeptide transporter NPF7.3 at the translational level through the 5'untranslated region.Collectively,these findings reveal that enhancing TOR signaling could mitigate rice yield penalty due to WST by regulating the processes involved in protein synthesis and NUE.Our study will contribute to the breeding of new rice varieties with increased water and fertilizer utilization efficiency.

A low-profile metamaterial absorber with ultrawideband reflectionless and wide-angular stability

An ultrawideband reflectionless metamaterial absorber(MA)is proposed by replacing the metallic ground with the complementary split-ring resonator(CSRR)structure.The proposed MA exhibits-10 d B reflectivity spectrum from 1 GHz to 20 GHz,which maintains more than 90%absorption from 1.5 GHz to20 GHz.Furthermore,it achieves angle stability for TE and TM polarization at oblique incident angles up to 40°and 65°,respectively.To achieve broadband absorption spectrum,we have adopted a single-layer high-impedance surface(HIS)loaded with a double-layer magnetic material(MM)structure.To further realize the RCS reduction into a lower frequency range,we have employed the scattering cancellation technology into the traditional metallic ground.Finally,we have fabricated a sample exhibiting the 10 d B RCS reduction from 1 GHz to 20 GHz with a thickness of 10 mm.Measurement and simulation results confirm that the proposed MA exhibits excellent comprehensive performance,making it suitable for many practical applications.
