Background Pediatric antineutrophil cytoplasmic antibody-associated vasculitis(AAV)is a life-threatening systemic vasculitis featured by liability to renal ***,there are few studies on the risk factors and predictive models for renal outcomes of AAV in *** Data from 179 AAV children in multiple centers between January 2.12.and March 2.2. were collected *** risk factors and predictive model of end-stage renal disease(ESRD)in AAV were *** Renal involvement was the most typical manifestation(95.5%),and the crescent was the predominant pathological lesion(84.9%).The estimated glomerular filtration rate(eGFR)was evaluated in 114 patients,of whom 59.6%developed ESRD,and the median time to ESRD was 3.2. *** eGFR[P=0.006,odds ratio(OR)=0.955,95%confidence interval(CI)=0.92.–0.987]and the percentages of global glomerulosclerosis(pGGS;P=0.018,OR=1.060,95%CI=1.010–1.112.were independent risk factors for ESRD of renal *** on the pGGS and eGFR at renal biopsy,we developed three risk grades of ESRD and one predictive *** Kaplan‒Meier curve indicated that renal outcomes were significantly different in different risk grades(Pline,the predictive model had higher accuracy(0.86 versus 0.58,P2.in external *** Renal involvement is the most common manifestation of pediatric AAV in china.of which more than half deteriorates into *** predictive model based on eGFR at renal biopsy and the pGGS may be stable and accurate in speculating the risk of ESRD in AAV children.

Somatic cell nuclear transfer(SCNT)and parthenogenesis are alternative forms of reproduction and development,building new life cycles on differentiated somatic cell nuclei and duplicated maternal chromatin,*** thepreceding paper(Sun F,et al.,Cell Res 2.07;17:117-13..),we showed that an"erase-and-rebuild"strategy is used innormal development to transform the maternal gene expression profile to a zygotic ***,we investigate if the samestrategy also applies to SCNT and *** relationship between chromatin and chromatin factors(CFs)during SCNT and parthenogenesis was examined using immunochemical and GFP-fusion protein *** fromthese studies indicated that soon after nuclear transfer,a majority of CFs dissociated from somatic nuclei and were re-distributed to the cytoplasm of the *** erasure process in oogenesis is recaptured during the initial phase in *** CFs entered pseudo-pronuclei shortly after their *** parthenogenesis,all parthenogenotes underwentnormal oogenesis,and thus had removed most CFs from chromosomes before the initiation of *** CFswere subsequently re-associated with female pronuclei in time and sequence similar to that in fertilized *** these data,we conclude that the"erase-and-rebuild"process observed in normal development also occurs in SCNTand in parthenogenesis,albeit in altered *** process is responsible for transcription reprogramming in ***"erase"process in SCNT is compressed and the efficiency is compromised,which likely contribute tothe developmental defects often observed in nuclear transfer(nt)***,results from this study indicatedthat the cytoplasm of an egg contains most,if not all,essential components for assembling the zygotic program and canassemble them onto appropriate diploid chromatin of distinct origins.

Within the electronic design automation(EDA) domain, artificial intelligence(AI)-driven solutions have emerged as formidable tools, yet they typically augment rather than redefine existing methodologies. These solutions often repurpose deep learning models from other domains, such as vision, text, and graph analytics, applying them to circuit design without tailoring to the unique complexities of electronic circuits. Such an “AI4EDA” approach falls short of achieving a holistic design synthesis and understanding,overlooking the intricate interplay of electrical, logical, and physical facets of circuit data. This study argues for a paradigm shift from AI4EDA towards AI-rooted EDA from the ground up, integrating AI at the core of the design process. Pivotal to this vision is the development of a multimodal circuit representation learning technique, poised to provide a comprehensive understanding by harmonizing and extracting insights from varied data sources, such as functional specifications, register-transfer level(RTL) designs, circuit netlists,and physical layouts. We champion the creation of large circuit models(LCMs) that are inherently multimodal, crafted to decode and express the rich semantics and structures of circuit data, thus fostering more resilient, efficient, and inventive design methodologies. Embracing this AI-rooted philosophy, we foresee a trajectory that transcends the current innovation plateau in EDA, igniting a profound “shift-left” in electronic design methodology. The envisioned advancements herald not just an evolution of existing EDA tools but a revolution, giving rise to novel instruments of design-tools that promise to radically enhance design productivity and inaugurate a new epoch where the optimization of circuit performance, power, and area(PPA) is achieved not incrementally, but through leaps that redefine the benchmarks of electronic systems' capabilities.

Sperm DNA Fragmentation may Influence IUI Outcome but could be Treated by ICSI: Evidence from Human Sperm Bank

Objective To determine the correlation between semen parameters, sperm DNA damage, progressive motility （PR）, morphology and intrauterine insemination （IUI）/ intracyto- plasmic sperm injection （ICSI） outcomes. Methods All the donors providing the samples in this study were recruited by Shang- hai Human Sperm Bank. For IUI, 12. donors were divided into group A （n=60） and group B （n=62.. Group A had a higher pregnancy rate while group B had a lower pregnancy rate （3.86 ± 1.50% vs 0.18 ± 0.52.）. For ICSI, 45 donors were divided into group C with a higher pregnancy rate （77. 78 ± 17.2.%, n=2.）, group D with a lower pregnancy rate （40. 73.± 19.19%, n=2.） and group E with an average preg- nancy rate in the sperm bank （48.96 ± 12.08%, n=2.）. Semen analysis, morphology and DNA damage were assessed on samples retained in the sperm bank. Fresh semen samples were also collected and corresponding semen analyses data was included along with the pregnancy rates. Results No significant difference was found in the population characteristics between groups A and B, while there was a significant difference in sperm DNA fragmetation index （DFI） and morphology between the two groups （P〈0.05）, but not in other semen parameters （P〉0.05）. There was no significant difference in population characteristics between groups C, D and E while the DFI of group D was significant higher than groups C and E （P〈0.01）, but there was no significant difference in other semen parameters （P〉0.05）.Conclusion DFI might be a good predictor for IUI outcomes. Infertile couples with a high DFI should choose ICSI treatment instead of IUI. DFI should be a routine screening marker used to screen for sperm donors.
