Characterization of mulberry leaf instant tea and evaluation of its hypolipidemia effect via regulation of intestinal microbiota

The purpose of this study was to characterize mulberry leaf instant tea(MLIT)powder prepared from the'Longsang No.1'(Morus abla L.cv.Longsang 1)mulberry leaves in Heilongjiang Province(China)and assess its obesity-preventing/relieving effects.A total of 174 compounds including quercetin,chlorogenic acid,1-deoxyecomycin(1-DNJ)related to antihyperlipidemia effects were identified from the MLIT powder.MLIT treatment reversed the Lee's index,fat coefficient,and serum biochemical parameters in both the obesity relieving and obesity preventing mice fed with high-fat diet.In the obesity relieving experiment,the relative abundance of Desulfovibrio in mouse feces decreased after both 0.5%and 1%MLIT treatments.In obesity preventing experiments,mouse with different amount of MLIT treatments showed increased relative abundance of Akkermansia,Bifidobacterium and Lactobacillus,while Deferribacteres,Desulfobacterota decreased.The beneficial bacteria in the intestinal tract of mice treated with MLIT increased.This study proved that MLIT had antihyperlipidemia potential via modulating intestinal microbiota in mice.

In Vitro Antifungal Activity of Dihydrochelerythrine and Proteomic Analysis in Ustilaginoidea virens

Dihydrochelerythrine(DHCHE)is an isoquinoline compound,which has distinct antifungal activity and can induce apoptosis.The antifungal activity of DHCHE against five rice pathogenic fungi was studied in vitro.At the concentration of 7.5 mg/L,DHCHE exhibited the highest efficacy among tested compounds in inhibiting mycelium growth,with an inhibition rate of 68.8%in Ustilaginoidea virens,which was approximately 2.4 times of that of validamycin(28.7%).After exposure to DHCHE,transmission electron micrographs revealed spores showed incomplete organelles,malformed cell walls and nuclear membranes,as well as irregular lipid spheres.Reactive oxygen species accumulation in treated spores was markedly higher than that in control spores.DHCHE induced cell damage increased in a dose-dependent manner,as indicated by the decrease in mitochondrial membrane potential and initiation of apoptosis.The differences of expression levels of Fip1,ACP1,PMS2 and COX13 that are important for oxidative phosphorylation and mismatch repair pathway were significant,which may be some of the reasons for the induction of apoptosis in DHCHE-treated U.virens.The protein levels of Fip1,ACP1,PMS2 and COX13 agreed with protein fold change ratio from parallel reaction monitoring Gene Ontology terms and Kyoto Encyclopedia of Genes and Genomes pathway of differentially expressed proteins were further analyzed.These findings will help to elucidate the mechanisms associated with antifungal and pro-apoptotic effects of DHCHE on U.virens,thereby aiding the potential development of novel pesticides.

On-surface synthesis and characterization of polyynic carbon chains

Carbyne, an elusive sp-hybridized linear carbon allotrope, has fascinated chemists and physicists for decades. Due to its high chemical reactivity and extreme instability, carbyne was much less explored in contrast to the sp2-hybridized carbon allotropes such as graphene. Herein, we report the on-surface synthesis of polyynic carbon chains by demetallization of organometallic polyynes on the Au（111） surface;the longest one observed consists of ～60 alkyne units（120 carbon atoms）. The polyynic structure of carbon chains with alternating triple and single bonds was unambiguously revealed by bond-resolved atomic force microscopy. Moreover, an atomically precise polyyne, C14, was successfully produced via tip-induced dehalogenation and ring-opening of the decachloroanthracene molecule(C14Cl10) on a bilayer NaCl/Au（111） surface at 4.7 K, and a band gap of 5.8 eV was measured by scanning tunnelling spectroscopy,in a good agreement with the theoretical HOMO–LUMO gap（5.48 eV）.

Plate tectonics in the Archean:Observations versus interpretations

Plate tectonics theory,established in the 1960s,has been successful in explaining many geological phenomena,processes and events that occurred in the Phanerozoic.However,the theory has often struggled to provide a coherent framework in interpreting geological records in continental interior and Precambrian period.In dealing with the relationship between plate tectonics and continental geology,continental interior tectonics was often separated from continental margin tectonics in the inheritance and development of their structure and composition.This separation led to the illusion that the plate tectonics theory is not applicable to Precambrian geology,particularly in explaining the fundamental geological characteristics of Archean cratons.Although this illusion does not mean that the Archean continental crust did not originate from a regime of plate tectonics,it led to the development of alternative tectonic models,often involving vertical movements under a regime of stagnant lid tectonics,including not only endogenous processes such as gravitational sagduction,mantle plumes and heat pipes but also exogenous processes such as bolide impacts.These vertical processes were not unique to the Archean but persisted into the Phanerozoic.They result from mantle poloidal convection at different depths,not specific to any particular period.Upgrading the plate tectonics theory from the traditional kinematic model in the 20th century to a holistic kinematic-dynamic model in the 21st century and systematically examining the vertical transport of matter and energy at plate margins,it is evident that plate tectonics can explain the common geological characteristics of Archean cratons,such as lithological associations,structural patterns and metamorphic evolution.By deciphering the structure and composition of convergent plate margins as well as their dynamics,the formation and evolution of continental crust since the Archean can be divided into ancient plate tectonics in the Precambrian and modern plate tectonics in the Phanero

Orthogonal Combinatorial Raman Codes Enable Rapid High-Throughput-Out Library Screening of Cell-Targeting Ligands

High-throughput assays play an important role in the fields of drug discovery,genetic analysis,and clinical diagnostics.Although super-capacity coding strategies may facilitate labeling and detecting large numbers of targets in a single assay,practically,the constructed large-capacity codes have to be decoded with complicated procedures or are lack of survivability under the required reaction conditions.This challenge results in either inaccurate or insufficient decoding outputs.Here,we identified chemical-resistant Raman compounds to build a combinatorial coding system for the high-throughput screening of cell-targeting ligands from a focused 8-mer cyclic peptide library.The accurate in situ decoding results proved the signal,synthetic,and functional orthogonality for this Raman coding strategy.The orthogonal Raman codes allowed for a rapid identification of 63 positive hits at one time,evidencing a high-throughput-out capability in the screening process.We anticipate this orthogonal Raman coding strategy being generalized to enable efficient high-throughput-out screening of more useful ligands for cell targeting and drug discovery.

Anti-cancer therapies usually focus on tumor cells,but non-tumor stromal components in the tumor microenvironment also play vital roles in tumor initiation and progression,which may be the prognostic factors and potential therapeutic targets.Cancer-associated fibroblasts(CAFs)are the essential component in the tumor environment,exhibiting high heterogeneity in their cell origin and phenotype with diverse functions that influence tumor angiogenesis,immune systems,and metabolism.Single-cell RNA sequencing and genetically engineered mouse models have increased our understanding of CAF diversity,and many subtypes have been defined.However,the precise functions of these subtypes need to be studied and validated.Studies of signaling pathways and epigenetic changes in CAFs facilitate understanding of the phenotypes of CAFs and the crosstalk between tumor cells and CAFs to provide potential therapeutic targets.Some clinical trials,including phase III trials targeting CAFs,have been performed recently.However,few of these trials have generated promising results,which indicates that the complexity of CAFs in the tumor microenvironment remains largely unknown,and in-depth investigations of CAFs should be performed.This review summarizes the research on CAFs,focusing on the heterogeneity of their phenotypes and functions,specific signaling pathways,and the therapeutic strategies involving CAFs.Additionally,we briefly discuss the current technologies commonly used in CAF studies and describe the challenges and future perspectives of CAF research.

A positive trend in the stability of global offshore wind energy

The recognition on the trend of wind energy stability is still extremely rare,although it is closely related to acquisition efficiency,grid connection,equipment lifetime,and costs of wind energy utilization.Using the 40-year(1979–2018)ERA-Interim data from the European Center for Medium-Range Weather Forecasts,this study presented the spatial-temporal distribution and climatic trend of the stability of global offshore wind energy as well as the abrupt phenomenon of wind energy stability in key regions over the past 40 years with the climatic analysis method and Mann-Kendall(M-K)test.The results show the following 5 points.(1)According to the coefficient of variation(C_(v))of the wind power density,there are six permanent stable zones of global offshore wind energy:the southeast and northeast trade wind zones in the Indian,Pacific and Atlantic oceans,the Southern Hemisphere westerly,and a semi-permanent stable zone(North Indian Ocean).(2)There are six lowvalue zones for both seasonal variability index(S_(v))and monthly variability index(M_(v))globally,with a similar spatial distribution as that of the six permanent stable zones.M_(v) and S_(v) in the Arabian Sea are the highest in the world.(3)After C_(v),M_(v) and S_(v) are comprehensively considered,the six permanent stable zones have an obvious advantage in the stability of wind energy over other sea areas,with C_(v) below 0.8,M_(v) within 1.0,and S_(v) within 0.7 all the year round.(4)The global stability of offshore wind energy shows a positive climatic trend for the past four decades.C_(v),M_(v) and S_(v) have not changed significantly or decreased in most of the global ocean during 1979 to2018.That is,wind energy is flat or more stable,while the monthly and seasonal variabilities tend to shrink/smooth,which is beneficial for wind energy utilization.(5)C_(v) in the low-latitude Pacific and M_(v) and S_(v) in both the North Indian Ocean and the low-latitude Pacific have an obvious abrupt phenomenon at the end of the20th century.

Recent progress in functional modification and crosslinking of bioprosthetic heart valves

Valvular heart disease(VHD),clinically manifested as stenosis and regurgitation of native heart valve,is one of the most prevalent cardiovascular diseases with high mortality.Heart valve replacement surgery has been recognized as golden standard for the treatment of VHD.Owing to the clinical application of transcatheter heart valve replacement technic and the excellent hemodynamic performance of bioprosthetic heart valves(BHVs),implantation of BHVs has been increasing over recent years and gradually became the preferred choice for the treatment of VHD.However,BHVs might fail within 10-15 years due to structural valvular degeneration(SVD),which was greatly associated with drawbacks of glutaraldehyde crosslinked BHVs,including cytotoxicity,calcification,component degradation,mechanical failure,thrombosis and immune response.To prolong the service life of BHVs,much effort has been devoted to overcoming the drawbacks of BHVs and reducing the risk of SVD.In this review,we summarized and analyzed the research and progress on:(i)modification strategies based on glutaraldehyde crosslinked BHVs and(ii)nonglutaraldehyde crosslinking strategies for BHVs.

Damping Characteristic Analysis and Optimization of Wind-thermal-bundled Power Transmission by LCC-HVDC Systems

With the rapid development of renewable energy,wind-thermal-bundled power transmission by line-commutated converter based high-voltage direct current(LCC-HVDC)systems has been widely developed.The dynamic interaction mechanisms among permanent magnet synchronous generators(PMSGs),synchronous generators(SGs),and LCC-HVDC system become complex.To deal with this issue,a path analysis method(PAM)is proposed to study the dynamic interaction mechanism,and the damping reconstruction is used to analyze the damping characteristic of the system.First,based on the modular modeling,linearized models for the PMSG subsystem,the LCC-HVDC subsystem,and the SG subsystem are established.Second,based on the closed-loop transfer function diagram of the system,the disturbance transfer path and coupling relationship among subsystems are analyzed by the PAM,and the damping characteristic analysis of the SG-dominated oscillation mode is studied based on the damping reconstruction.Compared with the PAM,the small-signal model of the system is obtained and eigenvalue analysis results are presented.Then,the effect of the control parameters on the damping characteristic is analyzed and the conclusions are verified by time-domain simulations.Finally,the penalty functions of the oscillation modes and decay modes are taken as the objective function,and an optimization strategy based on the Monte Carlo method is proposed to solve the parameter optimization problem.Numerical simulation results are presented to validate the effectiveness of the proposed strategy.

Discovery of the Significant Impacts of Swell Propagation on Global Wave Climate Change

This study compared the differences in the wave climate in the South China Sea and North Indian Ocean under these two datasets:ERA-40 wave reanalysis and Mei’s hindcast wave data.In the numerical calculation of regional ocean waves,the wave climate characteristics exhibited significant bias if the influence of external swells(swells from afar)was not fully considered,which may provide an incorrect basis for global climate change analysis.1)The trends of the significant wave height(SWH)obtained from the two datasets showed significant differences,such as those of the Bay of Bengal and the Java Sea in June-July-August.For the past 45 years,SWH from ERA-40(SWH-ERA)exhibited a significant annual increase in low-latitude waters of the North Indian Ocean(0.2-0.6 cm yr^(-1))and South China Sea(0.2-0.8 cm yr^(-1)).2)In the Bay of Bengal,the SWH-ERA in each month was generally 0.5 m higher than the SWH from Mei’s hindcast wave data(SWH-Mei)and can reach 1.0 m higher in some months.3)In the Bay of Bengal,SWH-ERA and SWH-Mei increased significantly at annual rates of 0.13 and 0.27 cm yr^(-1),respectively.This increasing trend was mainly reflected after 1978.SWH-ERA showed a trough in 1975(1.33 m)and a crest in 1992(1.83 m),which were not reflected in SWH-Mei.
