Assessment of pancreatic carcinoma cell chemosensitivity using a three-dimensional culture system

Background Monolayer cell culture models are the traditional culture models used for in vitro research of pancreatic carcinoma chemosensitivity. However, these models neglect the interactions between tumor cells and the impact of the tumor microenvironment. Such tumor cell monolayers poorly mimic the solid tumor microenvironment. The present study aimed to investigate the chemosensitivity characteristics of pancreatic cancer cells in a three-dimensional culture system by analyzing the differences in drug sensitivity between a scattered cell culture model and a multicellular spheroid culture model. Methods Three pancreatic cancer cell lines （SW1990, ASPC-1 and PCT-3） were cultured in three-dimensional collagen gels as well as in traditional two-dimensional monolayers. The chemosensitivities of the pancreatic carcinoma cells to 5-fluorouracil （5-FU）, gemcitabine, and oxaliplatin in vitro were detected by both the Cell Counting Kit-8 test and the collagen gel droplet-embedded culture drug-sensitivity test. Results In the two-dimensional culture model, differences in the chemosensitivities of the cloned pancreatic carcinoma cells and scattered cells existed for some concentrations of 5-FU, gemcitabine and oxaliplatin. In the three-dimensional culture model, there were significant differences in the chemosensitivities of the pancreatic cancer cells between the scattered cells and multicellular spheroids （P 〈0.05）. Conclusion Pancreatic carcinoma cells exhibit multicellular resistance in three-dimensional cultures.

Gradient maturation experiment on hairs implies taphonomic changes in fossil hairs

Mammalian hair, as a key feature related to endothermy, physical protection, and camouflage, has a long geohistorical record that dates back even to the Mesozoic. However, the limited information on the taphonomy of hair hinders the understanding of its evolutionary significance in deep time. To address this, we conducted taphonomic experiments to simulate the transformation process of black hair. The changes in the morphological structure, microscopic characteristics, and chemical composition of hair during the simulated fossilization process were documented by scanning electron microscopy, Fourier transform infrared(FTIR) spectroscopy, and Raman spectroscopy. Our results revealed that damage to the morphology of hair began at a mild temperature(150–175°C), and hair transformed into a fluid-like mass at a relatively high temperature(200°C). In addition, the morphological structure of melanosomes had relatively greater preservation potential than keratins did, and the microscopic features of melanosomes were persistently stable. The Raman and FTIR spectra indicated that the characteristic melanin and keratin peaks changed as the temperature increased. Although the chemical signals were not lost completely, the variations in the characteristic amide bands in the FTIR spectra indicate that the keratins severely degraded at high temperatures(>200°C), whereas the characteristic melanin bands in the Raman spectra suggested that melanin signals were still recognizable, even at high temperatures(>200°C). These results were confirmed by principal component analysis(PCA) and principal component analysis linear discriminant analysis(PCA–LDA). In addition, the Raman signals of all the samples in the simulated experiments were distinguished from the signals of common geological carbonaceous materials(humic acid), which suggested that Raman spectroscopy can be used to detect melanin in fossils.

本文以聚氯乙烯(polyvinyl chloride,PVC)和松木屑为原料,开展共水热实验,借助SEM、FTIR和ICP-OES等手段研究了松木屑中碱/碱土金属迁移特性和水热炭燃料性能。元素分析和工业分析的结果表明,温度是影响水热炭性能的最重要因素,其次是水热时间,而木屑粒径几乎无影响。温度为260℃时,碳含量保持在73.75%左右。本文研究范围内,水热条件对碱及碱土金属脱除率无明显影响,所测元素钾(K)、钠(Na)、钙(Ca)、镁(Mg)在水热炭中含量的变化顺序依次为:钙(Ca)>钾(K)>镁(Mg)>钠(Na)。温度由220℃增加至260℃时,钙(Ca)、镁(Mg)、钾(K)、钠(Na)的脱除率分别增加了1.02%、2.77%、4.10%和9.22%。与松木屑水热空白实验对比,添加PVC增强了反应体系的酸性,促进了木屑水热脱除碱/碱土金属。水热温度为260℃时,钙(Ca)、镁(Mg)、钾(K)、钠(Na)的最高脱除率由约26.59%上升到98.59%,93.17%,86.27%和85.84%。水热温度越高,半纤维素、纤维素和木质素分解度越高,生成的气体也越多,生成的水热炭的孔隙结构越发达,为碱金属的逸出提供了良好的通道。本文研究结果可为含氯有机废物和高碱固体燃料协同提质提供参考。

以松木屑为原料,采用高压反应釜,在高压、液固质量比12：1、搅拌转速150 r/min、反应时间1 h的条件下,在220-260℃温度范围内进行水热提质实验,对产物进行分析,研究温度对燃料特性、燃料性能及动力学特性的影响.结果表明,松木屑的碳含量和热值随水热提质温度升高逐渐增大,260℃时含碳量由48.10%增加到69.05%,热值由19.12 MJ/kg增加到26.66 MJ/kg.松木屑的热值增长趋势与其它生物质相同,但增长幅度异于其它生物质.水热提质过程中,木屑各组分发生脱羧、脱水、聚合、芳香化等反应,大分子物质分解成小分子物质如气体和有机酸等.水热提质有助于改善着火性能,随水热温度提高,最大燃烧速率对应的温度降低,燃烧时间延长.原木屑的燃烧反应为二级反应,水热提质后燃烧反应为一级反应,且活化能显著降低,为33.68 k J/mol.水热提质可显著提高木屑的能量密度,改善其燃烧性能.

Inhibiting SHP2 reduces glycolysis, promotes microglial M1 polarization, and alleviates secondary inflammation following spinal cord injury in a mouse model

Reducing the secondary inflammatory response, which is partly mediated by microglia, is a key focus in the treatment of spinal cord injury. Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.

激光雷达探测大气中的CO2气体目前还处在探索阶段,在差分吸收原理基础上,利用气体的受激拉曼散射增益效应,设计出测量CO2气体的非线性拉曼激光雷达系统,并模拟实验得到了增益开关两种状态下的光子计数信号,从而反演出CO2气体的浓度分布结果,在垂直方向上,CO2气体浓度随距离变化不大,合肥地区的浓度含量大约在400ppm左右。

Distributed fiber optic sensing system for vibration monitoring of 3D printed bridges

The fiber optic sensing technology provides data support in structural health monitoring of the macro facilities, including design, construction, and maintenance of bridges, tunnels, ports and other infrastructures. In this paper, a distributed vibration sensing system is proved to be responsive to a single touch over a 1.8-m-long equivalent fiber segment, covering a vibration frequency from 5 Hz to 25 kHz. The sensing fiber was arranged as an S type layout on the bridge to recognize the standing state, windblown disturbance, and walking vibration. Moreover, the knocking and climbing events are recognized fiber laying spinning lines and hanging on the fences, respectively. The demonstration shows an accurate positioning and sensitive vibration monitoring applied on the automated three-dimensional(3D) printed bridge, which is applicable to all kinds of 3D printed facilities as intelligent sensory neuro-networks.
