Host-Induced Gene Silencing of Effector AGLIP1 Enhanced Resistance of Rice to Rhizoctonia solani AG1-IA

Rice sheath blight, caused by Rhizoctonia solani AG1-IA, is a major disease in rice-growing areas worldwide. Effectors of phytopathogenic fungi play important roles during the infection process of fungal pathogens onto their host plants. However, the molecular mechanisms by which R. solani effectors regulate rice immunity are not well understood. Through prediction, 78 candidate effector molecules were identified. Using the tobacco rattle virus-host induced gene silencing(TRV-HIGS) system, 45 RNAi constructs of effector genes were infiltrated into Nicotiana benthamiana leaves. The results revealed that eight of these constructs resulted in a significant reduction in necrosis caused by infection with the AG1-IA strain GD-118. Additionally, stable rice transformants carrying the double-stranded RNA construct for one of the effector genes, AGLIP1, were generated to further verify the function of this gene. The suppression of the AGLIP1 gene increased the resistance of both N. benthamiana and rice against GD-118, and also affected the growth rate of GD-118, indicating that AGLIP1 is a key pathogenic factor. Small RNA sequencing showed that the HIGS vectors were processed into si RNAs within the plants and then translocated to the fungi, leading to the silencing of the target genes. As a result, AGLIP1 might be an excellent candidate for HIGS, thereby enhancing crop resistance against the pathogen and contributing to the control of R. solani infection.

Linear inversion of geoacoustic parameters using reflection characteristics of sediment layer with low sound speed

In order to improve the inversion efficiency and reduce the coupling of the geoacoustic parameters, a linear inversion method of the geoacoustic parameters of sediment layer with low sound speed is proposed. Based on the reflection coefficient of the plane wave of the two-layer seafloor from the quasi-linear sound speed profile, the relationship between the different reflection characteristics and the relevant geoacoustic parameters is analyzed, including the angle of intromission and the maximum reflection loss at the angle of intromission, interference fringe, and the corresponding cost function is *** with sensitivity analysis and stepwise inversion strategy, geoacoustic parameters of sedimentary layer with low sound speed are inversed step by step. This method is applicable to the case that the sound speed gradient of the low sound speed sediment layer is constant. The method is verified by part of the measured seabed reflection loss data from the new England mud patch. The inversion results are basically consistent with the relevant trans-dimensional inversion results, the inversion error is 0.1% for the sound velocity ratio, 0.3% for the density ratio, 14.6% for the absorption coefficient, and 15.8% for the sound speed gradient. The proposed method not only effectively weakens the coupling between some geoacoustic parameters, but also gives the best estimate of the power term that determines the shape of the quasi-linear sound speed profile.

Insights into the dwarfing mechanism of pear(Pyrus betulaefolia) based on anatomical and structural analysis using X-ray scanning

The lack of a suitable rootstock to control scion growth has limited the development of high-density plantations in pear production, which is partly attributed to poor understanding of the dwarfing mechanism. In the present study, the rootstock of the dwarf-type pear (Pyrus betulaefolia)PY-9’ was identified and used as the material for anatomical ***-9’ grew to half the tree height of the normal cultivar Zhengdu’, along with fewer internodes and shorter length. Significant differences in growth rate betweenPY-9’ andZhengdu’ were detected at approximately 30 days after full bloom, which corresponded with the time of the greatest difference in water potential between the dwarf and normal ***-9’ showed a higher photosynthetic rate thanZhengdu’. Anatomical analysis showed thatPY-9’ had higher area ratios of both phloem and xylem and more developed vascular tissues thanZhengdu’. The three-dimensional reconstructed skeleton of the xylem from X-ray computed tomography scanning revealed greater intervessel connectivity inZhengdu’ than inPY-9’, which could contribute to the more vigorous growth ofZhengdu’. This study thus provides the first comparison of the microstructural properties of xylem elements between a dwarfing-type and vigorous-type pear rootstock, providing new insights into the dwarfing mechanism in pear and facilitating breeding of dwarf pear rootstocks to increase crop productivity.

Suppressing Wnt signaling of the blood-tumor barrier to intensify drug delivery and inhibit lipogenesis of brain metastases

Background: Idiosyncratic drug-induced liver injury(IDILI) is a serious side effect of drugs, Epimedii Folium(EF) is unequivocally implicated in idiosyncratic liver injury onset, potentially due to its ability to perturb the NOD-like receptor family pyrin domain containing 3(NLRP3) inflammasome. Fructus Ligustri Lucidi(FLL), a frequently used medicinal combination with EF, has not yet been investigated for its ability to ameliorate EF-associated hepatotoxicity. Aims and Objectives: Study on the mechanism of compatibility of FLL to alleviate liver injury caused by EF. Materials and Methods: Western blot was used to determine the expression of related proteins, ELISA was used to detect the secretion of related inflammatory factors IL-1β, IL-18, IL-6 and TNF-α, liver injury indexes were detected and liver pathological tissue staining was used to evaluate the liver injury. Results: Our results demonstrated that EF exerted a particular augmenting effect on the stimulation of the NLRP3 inflammasome mediated by nigericin or ATP, whereas FLL suppressed the NLRP3 inflammasome stimulation. Furthermore, an equal EF to FLL ratio significantly reduced the stimulatory effects of EF. Moreover, EF has the potential to induce hepatic injury and augment pro-inflammatory cytokine synthesis in rats subjected to LPS. However, when combined with FLL, the detrimental effects of EF were mitigated. Conclusions: FLL possesses the capacity to attenuate EF-associated hepatotoxicity by suppressing EF-triggered NLRP3 inflammasome activation. Thus, FLL holds promise for improving the clinical safety profile of EF, shedding light on the potential of compatibility and detoxification theories in traditional Chinese medicine.

Orbital angular momentum(OAM)holography has become a promising technique in information encryption,data storage and opto-electronic computing,owing to the infinite topological charge of one single OAM mode and the orthogonality of different OAM *** this paper,we propose a novel OAM hologram generation method based on a densely connected U-net(DCU),where the densely connected convolution blocks(DCB)replace the convolution blocks of the ***,the reconstruction process of the OAM hologram is integrated into DCU as its output layer,so as to eliminate the requirement to prepare training data for the OAM hologram,which is required by conventional neural networks through an iterative *** experimental and simulation results show that the OAM hologram can rapidly be generated with the well-trained DCU,and the reconstructed image's quality from the generated OAM hologram is significantly improved in comparison with those from the Gerchberg-Saxton generation method,the Gerchberg-Saxton based generation method and the U-net *** addition,a 10-bit OAM multiplexing hologram scheme is numerically demonstrated to have a high capacity with OAM hologram.

BACKGROUND Scrub typhus is a naturally occurring acute infectious disease that is primarily transmitted through the bites of chiggers or larval mites infected by Orientia tsutsugamushi(***).Omadacycline,a novel tetracycline,exhibits potent antibacterial efficacy against both typical bacteria and atypical ***,omadacycline application in the treatment of scrub typhus remains *** SUMMARY In the present work,we report several cases of scrub typhus,with the main clinical symptoms being fever,the formation of eschars or ulcers,local or systemic lymphadenopathy,headache,myalgia and *** samples were collected before omadacycline was administered,and *** infection was confirmed through targeted next-generation sequencing(tNGS).After two days of treatment,the patients’symptoms,including fever,were alleviated,with no adverse drug *** tNGS is an effective method for diagnosing scrub *** can be considered an alternative option for antiinfective therapy in patients with *** infections.

横向磁通永磁直线电机的电负荷与磁负荷相互解耦,且各相之间相互解耦,易于实现模块化和多相化,因此在石油开采领域有着良好的应用前景。首先,该文基于磁场调制原理,对横向磁通永磁直线电机的径向电磁力波进行了推导计算,得出对径向电磁力波影响最大的因素为轴向气隙磁通密度分量与径向气隙磁通密度分量,在此基础上对径向电磁力波的时空特性与频谱特性进行仿真计算,得出电枢磁通密度对电机径向电磁力波的频谱特性的影响;然后,考虑到电机实际运行中可能出现的次级偏心问题,揭示偏心度的大小对径向电磁力波幅值的影响,并对比不同偏心度下的电机径向磁拉力与轴向磁拉力,定量分析偏心度对不平衡磁拉力的影响;最后,为避免共振,利用有限元软件对电机的初级进行了模态分析与谐响应分析,结果表明低速状态下横向磁通永磁直线电机的额定运行频率较低,不会引起初级的共振,适合用作往复式潜油电泵的驱动电机。
