Production and characterization of monoclonal antibodies against grass carp CD4-1 and CD4-2

CD4 T helper cells are an important group of cells in the immune system of vertebrates and express CD4 receptor on the cell surface.In mammals,the CD4 receptor is encoded by a single copy gene,whilst in fish,two copies of cd4 genes,namely cd4-1 and cd4-2,are found.In this study,the ectodomains of grass carp(Ctenopharyngodon idella,Ci)CD4-1 and CD4-2 were expressed in the E.coli cells and used to generate monoclonal antibodies in mice.Western blotting,confocal microscopy and flow cytometry were performed to characterize the monoclonal antibodies.It has been shown that the CiCD4-1 and CiCD4-2 monoclonal antibodies had good specificity to react with the recombinant ectodomains of CiCD4-1 and CiCD4-2 expressed in the CHO-S cells and the native CD4 molecules of grass carp.The CiCD4-1 monoclonal antibody did not recognize CiCD4-2 and verse versa.In addition,the CD4-1 and CD4-2 monoclonal antibodies specifically recognized the CD4-1 and CD4-2 receptors expressed in the HEK293 cells and native molecules of fish cells.Further,the percentages of lymphocytes in immune tissues of healthy fish were analyzed by flow cytometry.It was found that 17.6%of lymphocytes were CD4-1^(+)cells and 22.5%were CD4-2^(+)in the head kidney.In the spleen,13.1%of lymphocytes were CD4-1^(+)and 18.6%were CD4-2^(+)while 7.3%of blood lymphocytes were CD4-1^(+)cells and 8.8%were CD4-2^(+)cells.The availability of the CD4-1 and CD4-2 monoclonal antibodies provides antibody-based tools for further elucidation of the functions of CD4 T cells in grass carp.

Stacking MoS_(2) flower-like microspheres on pomelo peels-derived porous carbon nanosheets for high-efficient X-band electromagnetic wave absorption

The key to solve increasingly severe electromagnetic(EM)pollution is to explore sustainable,easily prepared,and cost-effective EM wave absorption materials with exceptional absorption capability.Herein,instead of anchoring on carbon materials in single layer,MoS_(2) flower-like microspheres were stacked on the surface of pomelo peels-derived porous carbon nanosheets(C)to fabricate MoS_(2)@C nanocomposites by a facile solvothermal process.EM wave absorption performances of MoS_(2)@C nanocomposites in X-band were systematically investigated,indicating the minimum reflection loss(RLmin)of-62.3 dB(thickness of 2.88 mm)and effective absorption bandwidth(EAB)almost covering the whole X-band(thickness of 2.63 mm)with the filler loading of only 20 wt.%.Superior EM wave absorption performances of MoS_(2)@C nanocomposites could be attributed to the excellent impedance matching characteristic and dielectric loss capacity(conduction loss and polarization loss).This study revealed that the as-prepared MoS_(2)@C nanocomposites would be a novel prospective candidate for the sustainable EM absorbents with superior EM wave absorption performances.

Rice gene OsUGT75A regulates seedling emergence under deep-sowing conditions

Poor seedling emergence is a challenge for direct seeding of rice under deep-sowing field conditions.Here we reveal that UDP-glucosyltransferase OsUGT75A promotes rice seedling emergence under deepsowing conditions by increasing shoot length.Expression of OsUGT75A was higher in the middle regions of the shoot and in shoots under deep-sowing conditions.Levels of free abscisic acid(ABA)and jasmonates(JA)were higher in shoots of OsUGT75A mutants than in those of wild-type plants,and OsUGT75A mutants were more sensitive to ABA and JA treatments.Reduced shoot length was attributed to higher ABA INSENSITIVE 3(OsABI3)expression and lower JASMONATE-ZIM domain protein(OsJAZ)expression in shoots.Shoot extension by OsUGT75A is achieved mainly by promotion of cell elongation.An elite haplotype of OsUGT75A associated with increased shoot length was identified among indica rice accessions.OsUGT75A acts to increase seedling emergence under deep-sowing conditions.

Impaired brain glucose metabolism is an early indicator of Alzheimer’s disease(AD);however,the fundamental mechanism is unknown.In this study,we found a substantial decline in isocitrate dehydrogenase 3β(IDH3β)levels,a critical tricarboxylic acid cycle enzyme,in AD patients and AD-transgenic mice’s brains.Further investigations demonstrated that the knockdown of IDH3βinduced oxidation-phosphorylation uncoupling,leading to reduced energy metabolism and lactate accumulation.The resulting increased lactate,a source of lactyl,was found to promote histone lactylation,thereby enhancing the expression of paired-box gene 6(PAX6).As an inhibitory transcription factor of IDH3β,the elevated PAX6 in turn inhibited the expression of IDH3β,leading to tau hyperphosphorylation,synapse impairment,and learning and memory deficits resembling those seen in AD.In AD-transgenic mice,upregulating IDH3βand downregulating PAX6 were found to improve cognitive functioning and reverse AD-like pathologies.Collectively,our data suggest that impaired oxidative phosphorylation accelerates AD progression via a positive feedback inhibition loop of IDH3β-lactate-PAX6-IDH3β.Breaking this loop by upregulating IDH3βor downregulating PAX6 attenuates AD neurodegeneration and cognitive impairments.

As a ceramic material,AlN has very good thermophysical and mechanical properties.In addition,AlN is an effective refining agent for Mg alloys because it has a lattice constant similar to that of Mg.Therefore,AlN is an ideal reinforcement for magnesium matrix composites(MMCs),and is attracting increasing attention.This review addresses the development of preparation technologies for AlN-reinforced Mg matrix composites.The mainstream preparation techniques include stir casting,melt infiltration,powder metallurgy,and in-situ methods.In addition,the advantages and disadvantages of these techniques are analyzed in depth,and it is pointed out that the next direction for the preparation of high-performance AlN-reinforced MMCs is less aluminization and multiple technologies integration.

Treatment of hemolymphangioma by robotic surgery: A case report

BACKGROUND Hemolymphangioma of the jejunum is rare and lacks clinical specificity,and can manifest as gastrointestinal bleeding,abdominal pain,and intestinal obstruction.Computed tomography,magnetic resonance imaging,and other examinations show certain characteristics of the disease,but lack accuracy.Although capsule endoscopy and enteroscopy make up for this deficiency,the diagnosis also still re-quires pathology.CASE SUMMARY A male patient was admitted to the hospital due to abdominal distension and abdominal pain,but a specific diagnosis by computed tomography examination was not obtained.Partial resection of the small intestine was performed by robotic surgery,and postoperative pathological biopsy confirmed the diagnosis of hemo-lymphangioma.No recurrence in the follow-up examination was observed.CONCLUSION Robotic surgery is an effective way to treat hemolymphangioma through minima-lly invasive techniques under the concept of rapid rehabilitation.

Mechanism of high-energy pulsed current-assisted rolling of 08AL carbon steel ultra-thin strip

To improve the plastic deformation performance of a 08AL carbon steel ultra-thin strip,a pulsed electric field was integrated into the plastic processing of the ultra-thin strip,and the effects of high-energy current on its deformation ability were investigated.Current-assisted tensile tests were employed,and the results clarified that the pulsed current could reduce the activation energy of faults and promoted dislocation slip within grains and at grain boundaries,leading to a decrease in the deformation resistance of the metal and an increase in its plastic properties.Under the current density of 2.0 A/mm2,the yield strength,tensile strength,and elongation of the rolled sample reached 425 MPa,467 MPa,and 12.5%,respectively.During the rolling process,it was found that the pulsed current promoted the dynamic recrystallization of the ultra-thin strip,reduced its dislocation density and deformation resistance,and promoted the coordinated deformation of the metal.

流域是重要的生态安全屏障和社会经济发展区域,为了更加客观准确地评价干旱和半干旱区流域生态环境质量,基于遥感生态指数(RSEI)引入盐度指标构建适用于干旱和半干旱区流域的遥感生态指数(AWRSEI)并以岱海流域为例分析其适用性.基于AWRSEI模型,采用4期Landsat TM/OLI合成影像,对2001~2020年岱海流域的生态环境质量进行定量评价,通过变异系数和空间自相关分析岱海流域生态环境质量时空演变规律,并通过地理探测器进行成因分析和驱动因子解释力分析.结果表明:(1)AWRSEI与各生态因子的平均相关系数为0.860,比单个指标分量更具代表性;第一主成分的荷载正负值和排序与RSEI一致,特征值贡献率比RSEI高3.69%,评价结果与真实地表情况更为接近,适合用于干旱区流域的生态环境质量评价.(2)岱海流域2001~2020年AWRSEI指数的年均值为0.427,生态环境质量基础较差,期间AWRSEI的均值呈现波动上升的趋势,并且在2020年均值达到最高0.502.整体生态环境质量明显好转,恶化区域减少了20.51%,改善区域增加了12.71%.空间分布上,岱海流域南部和西北部高海拔地区的林地生态环境质量优,北部和南部中海拔地区生态环境质量较差,湖区北侧生态环境质量优于南侧.(3)岱海流域AWRSEI变异系数平均值为0.280,生态环境质量状况稳定,整体变化波动较小,高波动主要集中在湖区南侧和居民点区域.岱海流域生态环境质量存在显著的空间自相关性,高-高集聚区主要分布在高海拔的林地区域和低海拔耕地区域;低-低集聚区零星分布在中海拔区域.(4)2001~2020年岱海流域生态环境的提升主要由于NDVI提高,NDBSI和NDSI降低. NDVI和NDBSI是交互作用最强的组合,对生态环境的解释力最强.土地利用是AWRSEI的主导因子,解释力最强.土地利用和气象因子的组合是交互作用最强,且各驱动因子之间均为增强关系.

An Analysis of the Responsibilities and Driving Factors for Embodied Carbon in Trade

By adopting the consumption-based accounting(CBA) method of carbon emissions based on national carbon intensity(NCI),using the International Trade Centre(ITC) database,the Emissions Database for Global Atmospheric Research(EDGAR),and the World Bank database,and taking 13 countries and regions including the European Union(EU) countries and the United States(US) as research samples,this paper conducts an accounting of the embodied carbon in trade between China and other major economies in the world,clarifies the responsibilities China has taken for foreign consumption-based carbon emissions,and analyzes the various driving factors of China’s embodied carbon in exports.The findings are as follows:(ⅰ) Most of the developed economies have higher consumption-based emissions than production-based emissions,which is contrary to most of the developing economies,with developing countries bearing responsibilities for 15-20% of the consumption-based carbon emissions for developed countries;(ⅱ) the carbon emissions of countries and regions are closely related to the embodied carbon in trade,with a correlation coefficient of 0.96.(ⅲ) China has taken responsibility for a large amount of carbon emissions from foreign consumer demand,most of which are from the US,the EU,ASEAN,and Japan,respectively accounting for 3.8%,3.5%,2.3%,and 1.2% of China’s production-based carbon emissions in 2019;(ⅳ) technique effect is the main driving factor for China’s embodied carbon in trade,while the specific driving factors vary by industry.Based on the above analysis,it is suggested that China should:(ⅰ) consider the trade factor in its efforts to achieve carbon peaking and carbon neutrality;(ⅱ) adopt differentiated measures to reduce traderelated emissions for specific industries;(ⅲ) fully consider China’s demand for carbon peaking and carbon neutrality in international negotiations;(ⅳ) give more support to the research on and measurement of consumption-based carbon emissions.

以2002—2022年全国286个城市为样本,考察空气质量对企业市场进入的影响。研究发现:(1)空气质量改善显著促进企业进入;(2)空气质量对企业选址的影响存在企业和城市异质性,空气质量对企业选址的影响在第三产业、外企占比高的城市、经济发达城市和工资水平较高的城市中更加显著;(3)作用机制检验表明,空气质量改善有助于促进劳动力健康水平、高技能劳动力流动和创新效率,进而促进企业进入;(4)通过断点回归设计有效分离出空气质量与企业选址之间的正向因果关系,发现空气质量的改善对企业选址的促进作用依旧显著。
