Unexpected BrdU inhibition on astrocyte-to-neuron conversion

5-Bromo-2′-deoxyuridine(BrdU)is a halogenated pyrimidine that can be incorporated into newly synthesized DNA during the S phase of the cell *** is widely used in fate-mapping studies of embryonic and adult neurogenesis to identify newborn neurons,however side effects on neural stem cells and their progeny have been *** vivo astrocyte-to-neuron(AtN)conversion is a new approach for generating newborn neurons by directly converting endogenous astrocytes into *** BrdU-labeling strategy has been used to trace astrocyte-converted neurons,but whether BrdU has any effect on the AtN conversion is ***,while conducting a NeuroD1-mediated AtN conversion study using BrdU to label dividing reactive astrocytes following ischemic injury,we accidentally discovered that BrdU inhibited AtN *** initially found a gradual reduction in BrdU-labeled astrocytes during NeuroD1-mediated AtN conversion in the mouse *** most NeuroD1-infected astrocytes were converted into neurons,the number of BrdU-labeled neurons was surprisingly *** exclude the possibility that this BrdU inhibition was caused by the ischemic injury,we conducted an in vitro AtN conversion study by overexpressing NeuroD1 in cultured cortical astrocytes in the presence or absence of ***,we also found a significantly lower conversion rate and a smaller number of converted neurons in the BrdU-treated group compared with the untreated *** results revealed an unexpected inhibitory effect of BrdU on AtN conversion,suggesting more caution is needed when using BrdU in AtN conversion studies and in data interpretation.

Aero-Hydro-Elastic-Servo Modeling and Dynamic Response Analysis of A Monopile Offshore Wind Turbine Under Different Operating Scenarios

This paper constructs a coupled aero-hydro-elastic-servo simulation framework for a monopile offshore wind turbine(OWT).In this framework,a detailed multi-body dynamics model of the monopile OWT including the gearbox,blades,tower and other components(nacelle,hub,bedplate,etc.)has been explicitly *** effects of pile−soil interaction,controller and operational conditions on the turbine dynamic responses are studied systematically in time domain and frequency *** results show that(1)a comprehensive drivetrain model has the capability to provide a more precise representation of the complex dynamic characteristics exhibited by drivetrain components,which can be used as the basis for further study on the dynamic characteristics of the drivetrain.(2)The pile−soil interaction can influence the wind turbine dynamic responses,particularly under the parked condition.(3)The effect of the pile−soil interaction on tower responses is more significant than that on blade responses.(4)The use of the controller can substantially affect the rotor characteristics,which in turn influences the turbine dynamic responses.(5)The tower and blade displacements under the operational condition are much larger than those under the parked *** model and methodology presented in this study demonstrate potential for examining complex dynamic behaviors of the monopile *** ensure accuracy and precision,it is imperative to construct a detailed model of the wind turbine system,while also taking into account simulation efficiency.

EuCd_(2)As_(2)was theoretically predicted to be a minimal model of Weyl semimetals with a single pair of Weyl points in the ferromagnet ***,the heavily p-doped Eu Cd_(2)As_(2)crystals in previous experiments prevent direct identification of the semimetal ***,we present a comprehensive magneto-transport study of high-quality Eu Cd_(2)As_(2)crystals with ultralow bulk carrier density(10^(13)cm^(-3)).In contrast to the general expectation of a Weyl semimetal phase,Eu Cd_(2)As_(2)shows insulating behavior in both antiferromagnetic and ferromagnetic states as well as surface-dominated conduction from band ***,the application of a dc bias current can dramatically modulate the resistance by over one order of magnitude,and induce a periodic resistance oscillation due to the geometric *** nonlinear transport results from the high nonequilibrium state induced by an electrical field near the band *** results suggest an insulating phase in Eu Cd_(2)As_(2)and put a strong constraint on the underlying mechanism of anomalous transport properties in this system.

The impact of tandem duplication on gene evolution in Solanaceae species

Whole genome duplication(WGD) and tandem duplication(TD) are important modes of gene amplification and functional innovation, and they are common in plant genome evolution. We analyzed the genomes of three Solanaceae species(Solanum lycopersicum, Capsicum annuum, and Petunia inflata), which share a common distant ancestor with Vitis vinifera, Theobroma cacao, and Coffea canephora but have undergone an extra whole genome triplication(WGT) event. The analysis was used to investigate the phenomenon of tandem gene evolution with(S. lycopersicum) or without WGT(V. vinifera). Among the tandem gene arrays in these genomes, we found that V. vinifera, which has not experienced the WGT event, retained relatively more and larger tandem duplicated gene(TDG) clusters than the Solanaceae species that experienced the WGT event. Larger TDG clusters tend to be derived from older TD events, so this indicates that continuous TDGs(absolute dosage) accumulated during long-term evolution. In addition, WGD and TD show a significant bias in the functional categories of the genes retained. WGD tends to retain dose-sensitive genes related to biological processes, including DNA-binding and transcription factor activity, while TD tends to retain genes involved in stress resistance. WGD and TD also provide more possibilities for gene functional innovation through gene fusion and fission. The TDG cluster containing the tomato fusarium wilt resistance gene I3 contains 15 genes, and one of these genes, Solyc07g055560, has undergone a fusion event after the duplication events. These data provide evidence that helps explain the new functionalization of TDGs in adapting to environmental changes.

Liquid Chromatography-Electrospray Quadrupole Linear Ion Trap Mass Spectrometric Method for Quantitation of Domperidone in Chinese Healthy Volunteers

A rapid, sensitive, and accurate method based on LC/MS/MS was developed and validated for the determination of domperidone in human plasma. Domperidone and internal standard, tramadol, were extracted from plasma with di- ethyl ether-dichloromethane （60： 40, volume ratio） and separated by reversed-phase HPLC with methanol-water-am- monia solution（80： 20： 0.2, volume ratio） as the mobile phase. Detection was carried out v/a multiple-reaction moni-toting（MRM） on a Q-trap^TM LC/MS/MS system（Q-trap^TM）. The assay result was linear over a concentration range of 0.1-30 ng/mL with a limit of quantitation （LOQ） of 0.1 ng/mL. The inter-and inn＇a-day precision levels were within 7.52% and 12.9%, respectively, whereas the accuracy was within a range of 87.3%-114%. This method has been successfully applied to evaluate the pharmcokinetics of domperidone in Chinese healthy volunteers given an oral dose of 10 mg.

Objective This study aimed to evaluate the clinical efficacy and safety of foldable capsular vitreous body(FCVB)implant surgery in silicone oil-dependent *** A total of 22 participants with silicone oil-dependent eyes who received treatment with FCVB implant surgery between January 2019 and June 2020 were included in this retrospective *** intraocular pressure(IOP),best-corrected visual acuity(BCVA),demographic data,and any recorded complications were *** The postoperative IOP(12.73±4.20 mmHg)was significantly improved(P=0.03)compared to the preoperative IOP(10.23±3.69 mmHg)(the main endpoint).There was no significant difference(P=0.33)in the final BCVA preoperation and 3rd month postoperation(the secondary endpoint).The most common postoperative complication was *** common postoperative complications included corneal opacity,a shallow anterior chamber,and a low *** FCVB implant surgery is a safe and effective method for treating silicone oil-dependent eyes;however,attention should be paid to the prevention and timely treatment of complications.

Plastic deformation bonding(PDB)has emerged as a promising solid state bonding technique with limited risk of phase transformations and residual thermal stresses in the *** this study,the PDB behavior of IN718 superalloy was systematically investigated by performing a series of isothermal compression tests at various processing *** was revealed that,with increasing PDB strain rate at 1000?C,different extents of dynamic recrystallization(DRX)occur in the bonding area of IN718 *** extent of DRX,average size of DRXed grains,and a newly proposed"interfacial bonding ratio(?Bonding)"parameter(to quantify the bond quality)were initially reduced with increase in the strain rate up to 0.1 s-1 and later increased at further higher strain *** backscattered diffraction(EBSD)and transmission electron microscopy(TEM)based interfacial microstructure analyses indicated that the quality of the bonded joints is closely related with the development of fine DRXed grains at the bonding interface with the increasing strain,which promotes adiabatic temperature *** was revealed that the initial bulging and subsequent migration of the original interfacial grain boundary(IGB)were the main mechanisms promoting DRX in the well bonded IN718 superalloy ***,the mechanical properties of the bonded joints were not only controlled by the recrystallized microstructure but also depended upon the Bonding parameter of the joints.

A new substrate,aligned Ag nanowires decorated with silver nanoparticle composite structure(AgNWs@AgNPs),was fabricated to investigate metalenhanced fluorescence(MEF) and its *** new composite structure was fabricated via a three-phase interface assembly method followed by SnCl2 sensitization and AgNO3 reduction *** size and distribution of the nanoparticles on silver nanowires increased with the sensitization and reduction *** formation of AgNPs on the surfaces of AgNWs was confirmed by multiple characterization methods including scanning electron microscopy(SEM),transmission electron microscope(TEM),atomic force microscopy(AFM) and X-ray diffraction(XRD).The results show that the fluorescence intensity of the poly(3-hexylthiophene)(P3HT) on the composite structure was greatly enhanced compared with that on bare glass substrate,and the intensity increased with the increase in particle sizes and *** mechanism was basedo n the increase in excitation rate and the radiation decay *** new type of substrate could serve as a good and efficient MEF substrate for high-performance fluorescence-based devices.

Induction of P450 genes in Nilaparvata lugens and Sogatella furcifera by two neonicotinoid insecticides

Nilaparvata lugens and Sogatella furcifera are two primary planthoppers on rice throughout Asian countries and areas. Neonicotinoid insecticides, such as imidaclo- prid （IMI）, have been extensively used to control rice planthoppers and IMI resistance consequently occurred with an important mechanism from the over-expression of P450 genes. The induction of P450 genes by IMI may increase the ability to metabolize this insecticide in planthoppers and increase the resistance risk. In this study, the induction of P450 genes was compared in S. furcifera treated with IM] and nitromethyleneimidazole （NMI）, in two planthopper species by IMI lethal dose that kills 85% of the population （LD85）, and in N. lugens among three IMI doses （LD15, LD50 and LD85）. When IMI and NMI at the LD85 dose were applied to S. furcifera, the expression changes in most P450 genes were similar, including the up-regulation of nine genes and down-regulation of three genes. In terms of the expression changes in 12 homologous P450 genes between N. lugens and S. furcifera treated with IMI at the LD85 dose, 10 genes had very similar patterns, such as up-regulation in seven genes, down-regulation in one gene and no significant changes in two genes. When three different IMI doses were applied to N. lugens, the changes in P450 gene expression were much different, such as up-regulation in four genes at all doses and dose-dependent regulation of the other nine genes. For example, CYP6AY1 could be induced by all IMI doses, while CYP6ER1 was only up-regulated by the LDs0 dose, although both genes were reported important in IMI resistance. In conclusion, P450 genes in two planthopper species showed similar regulation patterns in responding to IMI, and the two neonicotinoid insecticides had similar effects on P450 gene expression, although the regulation was often dose-dependent.

（-）-Tetrahydrolipstatin 1 was synthesized in three steps and 33.8% overall yield from aldehyde 6 by tandem Mukaiyama-aldol lactonization, which offered a concise, efficient and highly diastereoselective route to this antiobesity drug. We also presented a resolution method for preparation of the crucial intermediate, β- （t-butyldimethylsilyloxy）tetradecanal.
