Total Saponin Fraction of Dioscorea Nipponica Makino Improves Gouty Arthritis Symptoms in Rats via M1/M2 Polarization of Monocytes and Macrophages Mediated by Arachidonic Acid Signaling

Objective:To explore the mechanism of effects of total saponin fraction from Dioscorea Nipponica Makino(TSDN)on M1/M2 polarization of monocytes/macrophages and arachidonic acid(AA)pathway in rats with gouty arthritis(GA).Methods:Seventy-two Sprague Dawley rats were randomly divided into 4 groups(n=18 in each):normal,model,TSDN at 160 mg/kg,and celecoxib at 43.3 mg/kg.Monosodium urate crystal(MSU)was injected into the rats’ankle joints to induce an experimental GA model.Blood and tissue samples were collected on the 3rd,5th,and 8th days of drug administration.Histopathological changes in the synovium of joints were observed via hematoxylin and eosin(HE)staining.The expression levels of arachidonic acid(AA)signaling pathway were assessed via real-time polymerase chain reaction(q PCR)and Western blot.Flow cytometry was used to determine the proportion of M1 and M2 macrophages in the peripheral blood.An enzyme-linked immunosorbent assay(ELISA)was used to detect interleukine(IL)-1β,tumor necrosis factoralpha(TNF-α),IL-4,IL-10,prostaglandin E_(2)(PGE_(2)),and leukotriene B4(LTB4).Results:HE staining showed that TSDN improved the synovial tissue.q PCR and Western blot showed that on the 3rd,5th and 8th days of drug administration,TSDN reduced the m RNA and protein expressions of cyclooxygenase(COX)2,microsomal prostaglandin E synthase-1 derived eicosanoids(m PGES-1),5-lipoxygenase(5-LOX),recombinant human mothers against decapentaplegic homolog 3(Smad3),nucleotide-binding oligomerization domain-like receptor protein 3(NALP3),and inducible nitric oxide synthase(i NOS)in rats’ankle synovial tissues(P<0.01).TSDN decreased COX1 m RNA and protein expression on 3rd and 5th day of drug administration and raised it on the 8th day(both P<0.01).It lowered CD68 protein expression on days 3(P<0.01),as well as m RNA and protein expression on days 5 and 8(P<0.01).On the 3rd,5th,and 8th days of drug administration,TSDN elevated the m RNA and protein expression of Arg1 and CD163(P<0.01).Flow cytometry results showed that TSDN decreased

Integration of urchin-like MnCo_(2)O_(4)@C core–shell nanowire arrays within porous copper current collector for superior performance Li-ion battery anodes

Spinel MnCo_(2)O_(4) is a promising energy storage candidate as anode materials in lithium-ion batteries owing to synergistic effects of two intrinsic solid-state redox couples.However,low conductivity,poor rate capacity and rapid capacity fading have seriously impaired its practical applications.To overcome the inferiorities,urchin-like MnCo_(2)O_(4)@C core–shell nanowire arrays have been fabricated directly within a porous copper current collector via a facile hydrothermal method followed by a chemical vapor deposition carbonization process.In a typical nanowire,the core is composed of interconnected MnCo_(2)O_(4)nanoparticles and the shell shows as a thin amorphous carbon layer.The integrated MnCo_(2)O_(4)@C/Cu structure could act as working anodes without using additives or polymer binders.While MnCo_(2)O_(4)@C/Cu possesses slightly longer Li-ion insertion/desertion pathway than that of MnCo_(2)O_(4)/Cu,the carbon shell could effectively prevent the pulverization of MnCo_(2)O_(4) and lower down charge transfer resistance and actively participate in Li-ion cycles.The rearrangement of carbon atoms during lithiation/delithiation cycling could inhibit the formation of passive solid electrolyte interphase films.As a result,the MnCo_(2)O_(4)@C/Cu electrode presents superior rate capacity(600 mAh·g^(−1) at 1 A·g^(−1)) and better stability(797 mAh·g^(−1) after 200 cycles at 100 mA·g^(−1)).The excellent reversible Li ion storage capacity,cycling stability and rate capacity endow MnCo_(2)O_(4)@C/Cu great potential as stable and high output integrated anode materials in Li-ion batteries.

A review of the neurotransmitter system associated with cognitive function of the cerebellum in Parkinson's disease

The dichotomized brain system is a concept that was generalized from the‘dual syndrome hypothesis’to explain the heterogeneity of cognitive impairment,in which anterior and posterior brain systems are independent but partially overlap.The dopaminergic system acts on the anterior brain and is responsible for executive function,working memory,and planning.In contrast,the cholinergic system acts on the posterior brain and is responsible for semantic fluency and visuospatial function.Evidence from dopaminergic/cholinergic imaging or functional neuroimaging has shed significant insight relating to the involvement of the cerebellum in the cognitive process of patients with Parkinson’s disease.Previous research has reported evidence that the cerebellum receives both dopaminergic and cholinergic projections.However,whether these two neurotransmitter systems are associated with cognitive function has yet to be fully elucidated.Furthermore,the precise role of the cerebellum in patients with Parkinson’s disease and cognitive impairment remains unclear.Therefore,in this review,we summarize the cerebellar dopaminergic and cholinergic projections and their relationships with cognition,as reported by previous studies,and investigated the role of the cerebellum in patients with Parkinson’s disease and cognitive impairment,as determined by functional neuroimaging.Our findings will help us to understand the role of the cerebellum in the mechanisms underlying cognitive impairment in Parkinson’s disease.

The organization of the brain follows a topologi-cal hierarchy that changes dynamically during development.However,it remains unknown whether and how cognitive training administered over multiple years during develop-ment can modify this hierarchical topology.By measuring the brain and behavior of school children who had carried out abacus-based mental calculation(AMC)training for five years(starting from 7 years to 12 years old)in pre-training and post-training,we revealed the reshaping effect of long-term AMC intervention during development on the brain hierarchical topology.We observed the development-induced emergence of the default network,AMC training-promoted shifting,and regional changes in cortical gradi-ents.Moreover,the training-induced gradient changes were located in visual and somatomotor areas in association with the visuospatial/motor-imagery strategy.We found that gradient-based features can predict the math ability within groups.Our findings provide novel insights into the dynamic nature of network recruitment impacted by long-term cognitive training during development.

Characterization of Fe-containing particles in Chengdu,southwest China,using single-particle aerosol mass spectrometry

Single-particle aerosol mass spectrometry was used to study the characteristics of Fecontaining particles during winter in Chengdu,southwest China.The mass concentrations of PM_(2.5)and PM_(10)during the study period were 64±38 and 89±49μg/m~3,respectively,and NO_(2)and particulate matter were high compared with most other regions of China.The Fecontaining particles were divided into seven categories with different mass spectra,sources and aging characteristics.The highest contribution was from Fe mixed with carbonaceous components(Fe-C,23.1%)particles.Fe was more mixed with sulfate than nitrate and therefore the contribution of Fe mixed with sulfate(Fe-S,20.7%)particles was higher than that of Fe mixed with nitrate(Fe-N,12.5%)particles.The contributions from Fe-containing particles related to primary combustion were high in the small particle size range,whereas aged Fecontaining particles and dust-related particles were mostly found in the coarse particle size range.The air masses mainly originated from the west and east of Chengdu,and the corresponding PM_(2.5)concentrations were 79±36 and 55±36μg/m~3,respectively.The west and east air masses showed stronger contributions of Fe-containing particles related to biomass burning(Fe-B)and fossil fuel combustion(Fe-C and Fe-S)particles,respectively.The southwest area contributed the most Fe-containing particles.Future assessments of the effects of Fe-containing particles during heavy pollution period should pay more attention to Fe-C and Fe-S particles.Emission-reduction of Fe-containing particles should consider both local emissions and short-distance transmission from the surrounding areas.

Unexpected/contrary behavior of aerosol mass concentration in response to the individual components'concentration reduction in Kitakyushu,Japan

In the suburbs of Kitakyushu,Japan,the inorganic aerosol mass concentration(IAM)was about 32.7μg/m^(3),with the aerosol pH of 3.3.To study the thermodynamics of aerosol when its individual components'concentration is reduced,sensitive tests were performed using the ISORROPIAⅡmodel,in which the seven control species—TNaCl,TNH_(4)^(+),TSO_(4)^(2-),TNO_(3)^(-),TMg^(2+),TK^(+),and TCa^(2+)—were taken into account.IAM and inorganic aerosol pH after reducing TNaCl,TNO_(3)^(-),TMg^(2+),TK^(+),and TCa^(2+)responded linearly(0%≤concentration reduction ratio(CRR)≤100%,with the exception of 100%in TNaCl);the nonlinear variations of these two parameters could be observed by controlling TNH_(4)^(+)and TSO_(4)^(2-).Unexpected aerosol behavior occurred at 100%reduction of TNaCl,which was caused by the sudden increase of NO_(3)^(-),NH_(4)^(+),and aerosol liquid water content(ALWC);the increase of IAM was also observed after controlling TSO_(4)^(2-)(60%≤CRR≤100%)and TCa^(2+)(0%≤CRR≤100%),which was mainly related to the variation of ALWC driven by the response of CaSO_(4).Multiple regression analysis showed that ALWC was statistically and strongly related to the variations of NO_(3)^(-),Cl-,SO_(4)^(2-),HSO_(4)^(-),HNO_(3),and NH_(3)(P<0.05),with regression coefficients of 1.68,5.23,1.83,2.81,0.34,and 0.57,respectively.The highest coefficient(5.23)was found for Cl^(-),revealing that sea salts significantly influenced particle responses.Overall,this study comprehensively investigated aerosol characteristics and inner responses for the reduction of components,which is of great significance for a better understanding of atmospheric chemistry in Kitakyushu,Japan.

Improving lipid production by Rhodotorula glutinis for renewable fuel production based on machine learning

Microbial lipid fermentation encompasses intricate complex cell growth processes and heavily relies on expert experience for optimal production.Digital modeling of the fermentation process assists researchers in making intelligent decisions,employing logical reasoning and strategic planning to optimize lipid fermentation.It this study,the effects of medium components and concen-trations on lipid fermentation were investigated,first.And then,leveraging the collated data,a variety of machine learning algorithms were used to model and optimize the lipid fermentation process.The models,based on artificial neural networks and support vector machines,achieved R?values all higher than 0.93,ensuring accurate predictions of the fermentation process.Multiple linear regression was used to evaluate the respective target parameter,which were affected by the medium components of lipid fermentation.Lastly,single and multi-objective optimiza-tion were conducted for lipid fermentation using the genetic algorithm.Experimental results demonstrated the maximum biomass of 50.3 g·L^(-1) and maximum lipid concentration of 14.1 g:L^(-1) with the error between the experimental and predicted values less than 5%.The results of the multi-objective optimization reveal the synergistic and competitive relationship between biomass,lipid concentration,and conversion rate,which lay a basis for in-depth optimization and amplification.

Biosynthesizing Au nanoparticles(AuNPs)from gold-bearing scraps provides a sustainable method to meet the urgent demand for AuNPs.However,it remains challenging to efficiently biosynthesize AuNPs of which the diameter is less than 10 nm from a trace amount of Au^(3+)concentration at the level of tens ppm.Here,we constructed an exoelectrogenic cell(eCell)-conductive reduced-graphene-oxide aero-gel(rGA)biohybrid by assembling Shewanella sp.S1(SS1)as living biocatalyst and rGA as conductive ad-sorbent,in which Au^(3+)at trace concentrations would be enriched by the adsorption of rGA and reduced to AuNPs through the extracellular electron transfer(EET)of SS1.To regulate the size of the synthe-sized AuNPs to 10 nm,the strain SS1 was engineered to enhance its EET,resulting in strain RS2(pYYD-P tac-ribADEHC&pHG13-P_(bad)-omcC in SS1).Strain RS2 was further assembled with rGA to construct the RS2-rGA biohybrid,which could synthesize AuNPs with the size of 7.62±2.82 nm from 60 ppm Au^(3+)so-lution.The eCell-rGA biohybrid integrated Au^(3+)adsorption and reduction,which enabled AuNPs biosyn-thesis from a trace amount of Au^(3+).Thus,the required Au^(3+)ions concentration was reduced by one or two orders of magnitude compared with conventional methods of AuNPs biosynthesis.Our work devel-oped an AuNPs size regulation technology via engineering eCell’s EET with synthetic biology methods,providing a feasible approach to synthesize AuNPs with controllable size from trace level of gold ions.

This paper investigates the origin of ultra-deep dolostone and the factors influencing large-scale dolostone reservoirs,focusing on the Sinian Dengying Formation and the Cambrian Longwangmiao Formation in the Sichuan Basin.The study involves petrology,microscale X-ray diffraction,trace element analysis,and C-O-Sr-Mg isotope experiments to provide a detailed analysis.The research findings indicate that the Dengying and Longwangmiao formations comprise six types of matrix dolostone and four types of cement.The Dengying Formation,which developed under a sedimentary background of a restricted platform,contains special microbial and microcrystalline dolostones.The dolomite grains are small(30μm),high order degree(Min=0.7),small unit cell parameters,high Na content(Max=432 ppm),and low Fe and Mn content.The δ^(2

Diabetes mellitus in patients with type 1 autoimmune pancreatitis at diagnosis and after corticosteroid therapy

Background:A high prevalence of diabetes mellitus(DM)coexisting with autoimmune pancreatitis(AIP)is observed.However,evidence on the circumstances under which corticosteroid therapy(CST)for AIP improves or worsens DM is scarce.This study aimed to demonstrate and identify predictors of DM control under the influence of CST.Methods:Patients diagnosed with type 1 AIP were enrolled from a prospectively maintained cohort and were classified into three groups according to the chronology in which AIP and DM were diagnosed:pre-existing DM(pDM),concurrent DM(cDM),and non-DM(nDM).The responses of DM to CST were assessed when corticosteroid was ceased or tapered to a maintenance dose and classified as‘improvement’and‘non-improvement’(including‘no change’and‘exacerbation’).Results:Among 101 patients with type 1 AIP,52(51.5%)patients were complicated with DM at the time of AIP diagnosis,with 36 patients in the cDM group and 16 patients in the pDM group.The incidences of diffuse pancreatic swelling(72.2%)and pancreatic body/tail involvement(91.7%)were significantly higher in the cDM group than in both the pDM and nDM groups.Of the 52 patients with DM,CST was administered in 48 cases.Multivariate logistic analysis identified that elevated serum gamma-glutamyl transferase(GGT)level at AIP diagnosis[odds ratio(OR)=0.032,95%confidence interval(CI):0.003-0.412,P=0.008]and pancreatic atrophy after CST(OR=0.027,95%CI:0.003-0.295,P=0.003)were negatively associated with DM control improvement.Conclusions:Patients with diffuse pancreatic swelling and pancreatic body/tail involvement in pancreatitis tended to be complicated with cDM at AIP diagnosis.CST exerted a beneficial effect on the clinical course of DM in nearly half of the AIP patients complicated with DM at diagnosis,particularly in those without elevated serum GGT levels at diagnosis and who did not experience pancreatic atrophy after CST.
