Effect of local viral transfer of interleukin 10 gene on a rabbit arthritis model induced by interleukin 1β

Backgroud Interleukin 16 （IL-1β） is the principal mediator in the pathogenesis of rheumatoid arthritis. Continuous injection of interleukin 1β （IL-1β） into the knee articular cavities of anamals can induce models that resemble rheumatoid arthritis. The objective of this study was to evaluate the feasibility of local recombinant retrovirus viral interleukin 10 （rRV-vlL-10） gene transfer treatment of a rabbit model of arthritis induced by IL-16. Methods An hlL-1β-induced rabbit rheumatoid arthritis model was established using the MFG-hlL-1β-neo-HIG-82 cell line, which is capable of continuous secretion of hlL-1β. After transfecting the rabbit synovial fibroblast cell line （MFG-hlL-1β-neo-HIG-82） with rRV-vlL-10, G418 was then added to identify the positive clone. The rRV-vlL-10 positive clone was injected into the established rabbit rheumatoid arthritis model through intra-articular injection. Successful gene transfer was determined by reverse transcripUon-polymerase chain reaction （RT-PCR） and immunohistochemistry. The levels of IL-1β before and after treatment were determined by enzyme- linked immunosorbent assay. Results Retrovirus vector was an effective vector both to synoviocytes in vitro and synovium tissue in vivo as confirmed by RT-PCR and immunohistochemistry. The rabbit arthritis model treated with rRV-vlL-10 showed a dramatic remission of arthritis and a decline in the level of cytokines such as IL-1β. Conclusions Retrovirus-mediated transfecUon of vlL-10 successfully transferred the gene into rabbit synovium ex vivo and was able to suppress intra-articular inflammation response to IL-1β.

Expression of Peptidylarginine Deiminase 4 and Protein Tyrosine Phosphatase Nonreceptor Type 22 in the Synovium of Collagen-Induced Arthritis Rats

Objective To study the expression level of peptidylarginine deiminase 4(PADI4) and protein tyrosine phosphatase nonreceptor type 22(PTPN22) in the synovium of rat model of collagen-induced arthritis, and to explore their possible therapeutic role in rheumatoid arthritis. Methods Thirty-two female Wistar rats weighing 100±20 g were randomly assigned into 3-week collagen-induced arthritis(CIA) model group(n=8), 4-week CIA model group(n=8), 6-week CIA model group(n=8), and the control group(n=8). The body weight changes of each group were recorded. The expression levels of PADI4 and PTPN22 were detected and compared by the methods of immunohistochemical staining and Western blot. Results Arthritis of rat began to form 14 days after sensitization and the joint swelling reached peak at 28 days. The weights of the rats slowly grew both in CIA model groups and the control group. Immunohistochemical staining results showed that the positive expression of PADI4 and PTPN22 was mainly located in cartilage peripheral mononuclear cells, the cytoplasm of infiltrated cells, and bone marrow cavity. There were significant differences in the optical density of PADI4 and PTPN22 among CIA model groups and the control group(PADI4, 0.2898±0.012, 0.2982±0.022, 0.2974±0.031, 0.2530±0.013 in 3-week CIA model, 4-week CIA model, 6-week CIA model and control groups; PTPN22, 0.2723±0.004, 0.2781±0.010, 0.2767±0.008, 0.2422±0.019; all P <0.05). The expression bands of PADI4 were observed in Western blot 3 weeks after initial immunization, the thickest in the 4th week, and decreased in the 6th week. The expression bands of PTPN2 were observed at all the time points, with no obvious time-dependent trend. Conclusions PADI4 and PTPN22 are obviously correlated with CIA in rat model. PADI4 is expressed at early stage of the disease, while the expression of PTPN22 sustains throughout the course.

High-dose thalidomide increases the risk of peripheral neuropathy in the treatment of ankylosing spondylitis

Thalidomide is an effective drug for the treatment of ankylosing spondylitis but might induce peripheral neuropathy. This major adverse reaction has attracted much concern. The current study aimed to observe the incidence of thalidomide-induced peripheral neuropathy among an- kylosing spondylitis patients for 1 year after treatment. In this study, 207 ankylosing spondylitis cases received thalidomide treatment, while 116 ankylosing spondylitis cases received other treat- ments. Results showed that the incidence of thalidomide-induced peripheral neuropathy in the thalidomide group was higher than that in the non-thalidomide group. There was no significant difference in the incidence of neuropathy between the 〈 6 months medication and 〉 6 months medication groups. There were no differences in the mean age, gender, or daily dose between the two groups. The incidence of peripheral neuropathy among patients receiving 25, 50, 75, or 100 mg thalidomide per day was 4.6%, 8.5%, 17.1%, 21.7%, respectively. The incidence was significantly different between the groups receiving 25 mg and 100 mg thalidomide. In conclu- sion, thalidomide can induce peripheral neuropathy within 1 year after treatment of ankylosing spondylitis; however, age and gender have no obvious impact on the incidence of peripheral neuropathy. The incidence of peripheral neuropathy is associated with increasing daily doses of thalidomide.

Preparation of chitosan/N-doped graphene natively grown on hierarchical porous carbon nanocomposite as a sensor platform for determination of tartrazine

In this work,the chitosan and N-doped graphene natively grown on hierarchical porous carbon（N-PC-G/CS） nanocomposite was obtained by ultrasonic method,as a novel sensor platform for determination of tartrazine（TT）.The nanocomposite as prepared had well dispersivity in water and excellent *** N-PC-G/CS nanocomposite was characterized by scanning electron microscopy（SEM）,transmission electron microscopy（TEM）,nitrogen adsorption-desorption,fourier transform infrared（FTIR） and electrochemical impedance spectroscopy（EIS）.The application of N-PC-G/CS for determination of tartrazine（TT） was investigated by chronocoulometry（CC）,cyclic voltammetry（CV） and differential pulse voltammetry（DPV）.Under optimized conditions,the sensor displayed a sensitive response to TT within a wide concentration range of 0.05-15.0 μmol/L,the detection limits is 0.036 μmol/L（S/N = 3）.Furthermore,this nanocomposite could be efficiently applied for determination of TT in soft drink samples.

Effects of nitrogen form on growth,CO_2 assimilation,chlorophyll fluorescence,and photosynthetic electron allocation in cucumber and rice plants

Cucumber and rice plants with varying ammonium (NH4+) sensitivities were used to examine the effects of different nitrogen (N) sources on gas exchange, chlorophyll (Chl) fluorescence quenching, and photosynthetic electron allocation. Compared to nitrate (NO3-)-grown plants, cucumber plants grown under NH4+-nutrition showed decreased plant growth, net photosynthetic rate, stomatal conductance, intercellular carbon dioxide (CO2) level, transpiration rate, maximum photochemical efficiency of photosystem II, and O2-independent alternative electron flux, and increased O2-dependent alternative electron flux. However, the N source had little effect on gas exchange, Chl a fluorescence parameters, and photosynthetic electron allocation in rice plants, except that NH4+-grown plants had a higher O2-independent alternative electron flux than NO3--grown plants. NO3- reduction activity was rarely detected in leaves of NH4+-grown cucumber plants, but was high in NH4+-grown rice plants. These results demonstrate that significant amounts of photosynthetic electron transport were coupled to NO3- assimilation, an effect more significant in NO3-- grown plants than in NH4+-grown plants. Meanwhile, NH4+-tolerant plants exhibited a higher demand for the reduced form of nicotinamide adenine dinucleotide phosphate (NADPH) for NO3- reduction, regardless of the N form supplied, while NH4+-sensitive plants had a high water-water cycle activity when NH4+ was supplied as the sole N source.

Lattice damage in InGaN induced by swift heavy ion irradiation

The microstructural responses of In_(0.32)Ga_(0.68)N and In_(0.9)Ga_(0.1)N films to 2.25 GeV Xe ion irradiation have been investigated using x-ray diffraction,Raman scattering,ion channeling and transmission electron *** was found that the In-rich In_(0.9)Ga_(0.1)N is more susceptible to irradiation than the Ga-rich In_(0.32)Ga_(0.68)*** ion irradiation with a fluence of 7×10^(11)ions·cm^(-2)leads to little damage in In_(0.32)Ga_(0.68)N but an obvious lattice expansion in In_(0.9)Ga_(0.1)*** level of lattice disorder in In_(0.9)Ga_(0.1)N increases after irradiation,due to the huge electronic energy deposition of the incident Xe ***,no Xe ion tracks were observed to be formed,which is attributed to the very high velocity of 2.25 Ge V Xe *** defects and/or small defect clusters are probably the dominant defect type in Xe-irradiated In_(0.9)Ga_(0.1)N.

Properties of polydimethylsiloxane hydrophobic modified duplex microarc oxidation/diamond-like carbon coatings on AZ31B Mg alloy

A reliable,high-performance coating procedure was developed using PDMS to modify a duplex MAO/DLC coating on an AZ31B Mg ***,the duplex MAO/DLC coating was fabricated via a combined MAO and unbalanced magnetron sputter ***,a PDMS solution was used to modify the MAO/DLC coating via a conventional dip-coating *** surface characteristics,bond strength,hardness,tribological behaviour,and corrosion resistance of the coated samples were evaluated via SEM,CA,Raman spectroscopy,friction and wear behaviour,polarisation curve,and NSS *** PDMS modification reduced the HIT of MAO/DLC coating from 15.96 to 8.34GPa;this is ascribed to the penetration of PDMS,which has good rheological properties to form a viscoelastic Si-based organic polymer layer on the MAO/DLC ***,the PDMS-modified MAO/DLC coating was denser,hydrophobic,and had higher bond strength compared with MAO-and MAO/DLC-coated ***,the PDMS modification reduced the COF and wear rate of the duplex MAO/DLC *** indicates that the PDMS improved the tribological behaviour owing to the transferred Si oxide that originated from the Si-O network of the PDMS,as well as the low graphitisation of the DLC layer during ***,the corrosion current density of the MAO/DLC-coated sample modified by PDMS for 10min decreased by two order of magnitude compared with that of the MAO/DLC-coated sample but by five orders of magnitude compared with that of the bare *** NSS tests proved that the PDMS layer slowed the corrosion of the Mg alloy under long-term service,enhancing the corrosion protection *** results are attributed to the high bond strength and lubricant MAO/DLC layer,and the dual role of sealing and hydrophobicity of ***,PDMS modification is promising for the fabrication of protective materials for Mg alloys that require corrosion and wear resistance.

The reaction order plays a crucial role in evaluating the response rate of ***,the conventional two-scale model typically assumes that the reaction order is constant as one,which can lead to significant deviations from *** address this issue,this study proposes a novel multi-order dynamic model for acid-rock reaction by combining rotating disk experimental data with theoretical *** numerical simulations,this model allows for the investigation of the impact of acidification conditions on different orders of reaction,thereby providing valuable insights for on-site *** analysis reveals that higher response orders require higher optimal acid liquid flow rates,and lower optimal H+diffusion coefficients,and demonstrate no significant correlation with acid ***,it is recommended to increase the displacement and use high-viscosity acid for reservoirs with high calcite content,while reducing the displacement and using low-viscosity acid for reservoirs with high dolomite content.

Overexpression of a Cytosolic Ascorbate Peroxidase Gene, OsAPX2, Increases Salt Tolerance in Transgenic Alfalfa

Alfalfa （Medicago sativa L.） is an important forage crop in the world and it is of great signiifcance for the improvement of its salt tolerance. To improve salt tolerance in alfalfa, a rice ascorbate peroxidase gene （OsAPX2） was introduced into alfalfa using Agrobacterium tumefaciens-mediated transformation with marker gene bar. The different T-DNA insertions in T1 transgenic alfalfa were identiifed by Southern hybridization. Three independent T2 transgenic lines were selected for stress analysis and the results showed that all of them were salt tolerant compared with wild-type plants. The transgenic plants had low levels of H2O2, malondialdehyde and relative electrical conductivity under salt and drought stresses. Moreover, the contents of chlorophyll and proline, and APX activity were high in transgenic plants under salt and drought stresses. Taken together, the overexpression of OsAPX2 enhances salt tolerance in alfalfa through scavenging reactive oxygen species.

Generation and dynamic manipulation of frequency degenerate polarization entangled Bell states by a silicon quantum photonic circuit

A silicon quantum photonic circuit was proposed and realized for the generation and the dynamic manipulation of telecom-band frequency-degenerate polarization entangled Bell *** degenerate biphoton states were generated in four silicon waveguides by spontaneous four wave *** were transformed to polar-ization entangled Bell states through on-chip quantum interference and quantum superposition,and then coupled to optical *** property of polarization entanglement in generated photon pairs was demonstrated by two-photon interference under two non-orthogonal polarization *** output state could be dynamically switched between two Bell states,which was demonstrated by the simplified Bell state *** experiment results indicated that the manipulation speed supported a modulation rate of several tens kHz,showing its potential on applications of quantum communication and quantum information processing requiring Bell state encoding and dynamic control.
