Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(P<0.05). Furthermore, our research revealed that monotropein induced cell apoptosis by inhibiting Bcl-2 and increasing Bax, induced G_1–S cycle arrest in colorectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(P<0.05), and might play an anticancer role through Akt signaling pathway. 针对基于侧信道信号的硬件木马检测普遍面临的两类问题,即需要黄金芯片和信号测量成本大,提出一种利用路径延迟故障序列的检测方法。基于时序冲突时电路路径产生延迟故障的先后顺序,间接利用路径延迟之间相对大小关系产生芯片检测ID;检测不依赖于黄金芯片,且在大范围的环境变化和工艺偏差下具有稳定性;故障序列在芯片设计阶段仿真获得,无需额外硬件开销。对ISCAS-89基准和AES-128电路实例验证了检测的有效性,其可成功检测插入的两种类型硬件木马。

Min-max disagreements are an important generalization of the correlation clustering problem(CorCP).It can be defined as *** a marked complete graph G=(V,E),each edge in the graph is marked by a positive label"+"or a negative label"_"based on the similarity of the connected *** goal is to find a clustering C of vertices V,so as to minimize the number of disagreements at the vertex with the most ***,the disagreements are the positive cut edges and the negative non-cut edges produced by clustering *** paper considers two robust min-max disagreements:min-max disagreements with outliers and min-max disagreements with *** parameter δ∈(0,1/14),we first provide a threshold-based iterative clustering algorithm based on LP-rounding technique,which is a(1/δ,7/(1-14δ))-bi-criteria approximation algorithm for both the min-max disagreements with outliers and the min-max disagreements with outliers on one-sided complete bipartite ***,we verify that the above algorithm can achieve an approximation ratio of 21 for min-max disagreements with penalties when we set parameter δ=1/21.

Electrical discharge and arc milling with automatic tracking of optimal flushing direction:A novel high-efficiency compound machining method

The arc milling method has the advantages of high machining efficiency and low cost and is independent of the strength and hardness of machined ***,frequent electrode back-offs and the risk of workpiece burning may occur if erosion products are not removed *** this study,it was found that the flushing method of the working medium had a significant impact on the machining performance of arc *** on this,a novel highefficiency compound machining method of electrical discharge and arc milling with automatic tracking of the optimal flushing direction was *** automatic tracking optimizer for external working medium injection was designed to determine the optimal external flushing direction according to the feed *** influence of flushing methods,working mediums,and machining parameters on the machining efficiency,tool electrode wear rate,machining error,and surface integrity of titanium alloys were *** results indicated that better machining performance and environmental friendliness were achieved using the compound flushing method of outer compressed air and inner deionized ***,the automatic tracking flushing method in the opposite direction of the feed direction showed superior results compared to other *** material removal rate with the opposite direction injection could be increased up to 1.62 times that of the same direction,and the relative electrode wear rate could be reduced by 14.76%.This novel method has broad application prospects for machining parts with difficult-to-cut materials in aerospace and military industries.

Retrieval of Antarctic sea ice freeboard and thickness from HY-2B satellite altimeter data

Antarctic sea ice is an important part of the Earth’s atmospheric system,and satellite remote sensing is an important technology for observing Antarctic sea *** Chinese Haiyang-2B(HY-2B)satellite altimeter data could be used to estimate sea ice freeboard and provide alternative Antarctic sea ice thickness information with a high precision and long time series,as other radar altimetry satellites can,needs further *** paper proposed an algorithm to discriminate leads and then retrieve sea ice freeboard and thickness from HY-2B radar altimeter *** first collected the Moderate-resolution Imaging Spectroradiometer ice surface temperature(IST)product from the National Aeronautics and Space Administration to extract leads from the Antarctic waters and verified their accuracy through Sentinel-1 Synthetic Aperture Radar ***,a surface classification decision tree was generated for HY-2B satellite altimeter measurements of the Antarctic waters to extract leads and calculate local sea surface *** then estimated the Antarctic sea ice freeboard and thickness based on local sea surface heights and the static equilibrium ***,the retrieved HY-2B Antarctic sea ice thickness was compared with the CryoSat-2 sea ice thickness and the Antarctic Sea Ice Processes and Climate(ASPeCt)ship-based observed sea ice *** results indicate that our classification decision tree constructed for HY-2B satellite altimeter measurements was reasonable,and the root mean square error of the obtained sea ice thickness compared to the ship measurements was 0.62 *** proposed sea ice thickness algorithm for the HY-2B radar satellite fills a gap in this application domain for the HY-series satellites and can be a complement to existing Antarctic sea ice thickness products;this algorithm could provide long-time-series and large-scale sea ice thickness data that contribute to research on global climate change.

目的通过胰胆管型胆管内乳头状瘤(pbIPNB)的影像学表现与病理对照分析,提高对该病的认识及影像诊断水平。方法回顾性分析2020年1月至2023年9月河南省人民医院收治的经手术病理学证实的pbIPNB患者15例,将影像学(CT及MRCP)表现与病理对照。结果15例患者中,13例患者胆管内显示乳头状肿瘤,2例患者胆管内未见肿瘤显示;13例肿瘤均边界清楚,CT平扫为软组织密度,CT增强扫描均表现为动脉期轻中度强化,静脉期强化程度稍减低;10例患者表现为肿瘤上下游胆管扩张,3例患者表现为上游胆管扩张;7例患者CT平扫表现为胆管内密度增高;胆管扩张分型,Ⅰ型3例,Ⅱ型3例,Ⅲ型2例,Ⅳ型7例;6例患者有肝实质异常灌注,15例患者均无肝实质萎缩;3例患者MRCP表现为螺纹征,3例患者表现为肝左叶肝内胆管、肝总管及胆总管团状充盈缺损。病理学表现:镜下见细而薄的分枝和乳头被立方或低柱状上皮覆盖,胞浆呈嗜酸或嗜双色性,外观黏液较少,核圆、深染,核仁突出,包括不规则的乳头状结构,细胞核分层较多,实性或粉刺样结构,细胞和细胞核不典型。结论pbIPNB在CT及MRCP上有一定的影像学特征,且有相应的病理学基础作为支撑,基于病理基础,并结合影像学征象进行分析,有助于提高该病诊断准确率。

Detecting fake reviewers in heterogeneous networks of buyers and sellers:a collaborative training-based spammer group algorithm

It is not uncommon for malicious sellers to collude with fake reviewers(also called spammers)to write fake reviews for multiple products to either demote competitors or promote their products'reputations,forming a gray industry *** detect spammer groups in a heterogeneous network with rich semantic information from both buyers and sellers,researchers have conducted extensive research using Frequent Item Mining-based and graph-based ***,these methods cannot detect spammer groups with cross-product attacks and do not jointly consider structural and attribute features,and structure-attribute correlation,resulting in poorer detection ***-fore,we propose a collaborative training-based spammer group detection algorithm by constructing a heterogene-ous induced sub-network based on the target product set to detect cross-product attack spammer *** jointly consider all available features,we use the collaborative training method to learn the feature representations of *** addition,we use the DBSCAN clustering method to generate candidate groups,exclude innocent ones,and rank them to obtain spammer *** experimental results on real-world datasets indicate that the overall detection performance of the proposed method is better than that of the baseline methods.

Solid polymer electrolytes(SPEs)have become increasingly important in advanced lithium-ion batteries(LIBs)due to their improved safety and mechanical properties compared to organic liquid ***-linked polymers have the potential to further improve the mechanical property without trading off Li-ion *** this study,focusing on a recently developed cross-linked SPE,i.e.,the one based on poly(vinylene carbonate)-poly(ethylene oxide)cross-linked network(PVCN),we used solid-state nuclear magnetic resonance(NMR)techniques to investigate the fundamental interaction between the chain segments and Li ions,as well as the lithium-ion *** utilizing homonuclear/heteronuclear correlation,CP(cross-polarization)kinetics,and spin-lattice relaxation experiments,etc.,we revealed the structural characteristics and their relations to lithium-ion *** is found that the network formation prevents poly(ethylene oxide)chains from crystallization,which could create sufficient space for segmental tumbling and Li-ion co nductio *** such,the mechanical property is greatly improved with even higher Li-ion mobilities compared to the poly(vinylene carbonate)or poly(ethylene oxide)based SPE analogues.
