Cystamine ameliorates liver fibrosis induced by carbon tetrachloride via inhibition of tissue transglutaminase

AIM： To investigate the anti-fibrosis effect of the tissue transglutarninase （tTG） specific inhibitor cystarnine on liver fibrosis. METHODS： Sixty-eight male Sprague Dawley rats were divided into three groups： normal control, liver fibrosis control and cystamine-treated group. Liver fibrosis was induced by intraperitoneal injection of carbon tetrachloride （CCl4）, and Cystarnine was administrated by intraperitoneal injection starting 2 d before the first administration of CCl4. Animals in each group were further divided into 2 subgroups according to two time points of 4 wk and 8 wk after treatment. Hepatic function, pathological evaluation （semi-quantitative scoring system, SSS） and liver hydroxyproline （Hyp） content were examined. Real-time PCR was used to detect the expression of tTG, smooth muscle alpha actin （α-SMA）, tissue inhibitor of metalloproteinase 1 （TIMP-1） and collagen-1 mRNA. The expressions of tTG and α-SMA protein were detected by Western Blotting. RESULTS： Eight weeks after treatment, the SSS score of liver was significantly less in the cystamine group than that in the fibrosis control group （P 〈 0.01）. The levels of alanine arninotransferase （ALT） and total bile acid （TBA） at the 4 wk and 8 wk time points were decreased in the cystamine group compared with those in fibrosis controls （P 〈 0.01）. Liver hydroxyproline content at the 4 wk and 8 wk time points showed a substantial reduction in the cystamine group compared to fibrosis controls （P 〈 0.01）. The expression of tTG, α-SMA, collagen-1, TIMP-1 mRNA and tTG, as well as α-SMA protein was downregulated in the cystamine group compared to fibrosis controls. CONCLUSION： Cystamine can ameliorate CCl4 induced liver fibrosis and protect hepatic function. The possible mechanism is related to the reduced synthesis of the extracellular matrix （ECM） caused by the inhibition of hepatic stellate cell activation and decreased expression of TIMP-1.

Effectiveness and safety of splenectomy for gastric carcinoma:A meta-analysis

AIM： To evaluate the impact of splenectomy on longterm survival, postoperative morbidity and mortality of patients with gastric cancer by performing a metaanalysis METHODS： A search of electronic databases to identify randomized controlled trials in The Cochrane Library trials register, Mediine, CBMdisc （Chinese Biomedical Database） and J-STAGE, etc was performed. Data was extracted from the studies by 2 independent reviewers. Outcome measures were survival, postoperative morbidity and mortality and operation-related events. The meta-analyses were performed by RevMan 4.3. RESULTS： Three studies comprising 466 patients were available for analysis, with 231 patients treated by gastrectomy plus splenectomy. Splenectomy could not increase the 5-year overall survival rate [RR = 1.17, 95% confidence interval （CI） 0.97-1.41]. The postoperative morbidity （RR = 1.76, 95% CI 0.82-3.80） or mortality （RR = 1.58, 95% CI 0.45-5.50） did not suggest any significant differences between the 2 groups. No significant differences were noted in terms of number of harvested lymph nodes, operation time, length of hospital stay and reoperation rate. Subgroup analyses showed splenectomy did not increase the survival rate for proximal and whole gastric cancer. No obvious differences were observed between the 2 groups when stratified by stage. Sensitivity analyses indicated no significant differences regarding the survival rates （P 〉 0.05）. CONCLUSION： Splenectomy did not show a beneficial effect on survival rates compared to splenic preservation. Routinely performing splenectomy should not be recommended.

Metal surfaces play a crucial role in numerous applications,from self-cleaning and anti-icing to anti-fogging and oil-water *** regulation of their wettability is essential to enhance their performance in these *** paper proposes a multi-state regulation method for metal surface wettability,leveraging nanosecond laser *** creating non-uniform microstructures on a metal surface,the contact area between the solid and liquid phases can be increased,resulting in the attainment of superhydrophilic properties(contact angle(CA),ranging from 4.6°to 8.5°).Conversely,the construction of uniform microstructures leads to a decreased solid-liquid contact area,thereby rendering the metal surface hydrophilic(CA=12.2°–53°).Furthermore,through heat treatment on a surface with uniform microstructures,organic matter adsorption can be promoted while simultaneously reducing surface *** process results in the metal surface acquiring hydrophobic properties(CA=92.1°–133.5°),facilitated by the“air cushion effect.”Building on the hydrophobic surface,stearic acid modification can further reduce surface energy,ultimately bestowing the metal surface with superhydrophobic properties(CA=150.1°–152.7°,and sliding angle=3.8°).Performance testing has validated the durability and self-cleaning effectiveness of the fabricated superhydrophobic surface while also highlighting the excellent anti-fog performance of the superhydrophilic *** findings strongly indicate the immense potential of these surfaces in various engineering applications.

Response of porcine hepatocytes in primary culture to plasma from severe viral hepatitis patients

AIM： To observe the effects of plasma from patients with severe viral hepatitis （SVHP） on the growth and metabolism of porcine hepatocytes and the clinical efficiency of bioartificial liver device.METHODS： Hepatocytes were isolated from male porcines by collagenase perfusion. The synthesis of DNA and total protein, leakages of AST and LDH, changes in glutathione （GSH）, catalase and morphology of porcine hepatocytes exposed to SVHP were investigated to indicate the effect of plasma from patients with severe hepatitis on the growth, injury, detoxification, and morphology of porcine hepatocytes.RESULTS： The synthesis of DNA and protein was inhibited in the medium containing 100% SVHP compared to the controls. The leakages of LDH and AST increased in porcine hepatocytes following exposure to 100% SVHP for 5 h. The difference between 100% SVHP and 10% newborn calf serum （NCS） was significant in t-test （LDH： t = 24.552, P = 0.001; AST： t = 4.169, P = 0.014）. After exposure to SVHP for 24 h, alterations in GSH status were significant （F = 2.746, P〈0.05） between porcine hepatocytes in 100% SVHP and 10% NCS, but no alteration occurred in the culture medium after 48 h （F = 4.378, ,P〈0.05）. A similar profile was observed in catalase activity. Many round vacuoles were observed in porcine hepatocytes cultured in SVHR The membranes of these cells became indistinct and almost all the cells died on d 5.CONCLUSION： Plasma from patients with severe hepatitis inhibits the growth, injures membrane, disturbs GSH homeostasis and induces morphological changes of porcine hepatocytes, It is suggested that SVHP should be pretreated to reduce the toxin load and improve the performance of porcine hepatocytes in extracorporeal liver-support devices.

Wave Attenuation Performance and the Influencing Factors of A Lower Arc-Plate Breakwater

Comprehensive experimental and numerical studies have been undertaken to investigate wave energy dissipation performance and main influencing factors of a lower arc-plate breakwater. The numerical model, which considers nonlinear interactions between waves and the arc-plate breakwater, has been constructed by using the velocity wave- generating method, the volume of fluid (VOF) method and the finite volume method. The results show that the relative width, relative height and relative submergence of the breakwater are three main influencing factors and have significant influence on wave energy dissipation of the lower arc-plate open breakwater. The transmission coefficient is found to decrease with the increasing relative width, and the minimum transmission coefficient is 0.15 when the relative width is 0.45. The reflection coefficient is found to vary slightly with the relative width, and the maximum reflection coefficient is 0.53 when the relative width is 0.45. The transmission and reflection coefficients are shown to increase with the relative wave height for approximately 85% of the experimental tests when the relative width is 0.19 0.45. The transmission coefficients at relative submergences of 0.04, 0.02 and 0 are clearly shown to be greater than those at relative submergences of 0.02 and 0.04, while the reflection coefficient exhibits the opposite relationship. After the wave interacts with the lower arc-plate breakwater, the wave energy is mainly converted into transmission, reflection and dissipation energies. The wave attenuation performance is clearly weakened for waves with greater heights and longer periods.

Thermally Conductive,Healable Glass Fiber Cloth Reinforced Polymer Composite based onβ-Hydroxyester Bonds Crosslinked Epoxy with Improved Heat Resistance

To simultaneously endow thermal conductivity,high glass transition temperature(Tg)and healing capability to glass fiber/epoxy(GFREP)composite,dynamic crosslinked epoxy resin bearing reversibleβ-hydroxyl ester bonds was reinforced with boron nitride nanosheets modified glass fiber cloth(GFC@BNNSs).The in-plane heat conduction paths were constructed by electrostatic self-assembly of polyacrylic acid treated GFC and polyethyleneimine decorated ***,the GFC@BNNSs were impregnated with the mixture of lower concentration(3-glycidyloxypropyl)trimethoxysilane grafted BN micron sheets,3,4-epoxycyclohexylmethyl 3,4-epoxycyclohexanecarboxylate and hexahydro-4-methylphthalic anhydride,which accounted for establishing the through-plane heat transport pathways and avoiding serious deterioration of mechanical *** resultant GFREP composite containing less boron nitride particles(17.6 wt%)exhibited superior in-plane(3.29 W·m^(-1)·K^(-1))and through-plane(1.16 W·m^(-1)·K^(-1))thermal conductivities,as well as high Tg of 204℃(Tg of the unfilled epoxy=177℃).The reversible transesterification reaction enabled closure of interlaminar cracks within the composite,achieving decent healing efficiencies estimated by means of tensile strength(71.2%),electrical breakdown strength(83.6%)and thermal conductivity(69.1%).The present work overcame the disadvantages of conventional thermally conductive composites,and provided an efficient approach to prolong the life span of thermally conductive GFREP laminate for high-temperature resistant integrated circuit application.

The incidence of the inferior vena cava （IVC） tumor thrombus is reported to be 4%-10% in patients with renal cell carcinoma （RCC）. Tumor thrombus may extend through to the right atrium. Management of patients with level Ⅲ/Ⅳ tumor thrombus is usually difficult. We report two cases of level Ⅳ thrombus in our hospital in 2002 and 2004.

Association between Plasma Leptin Level and Systemic Inflammatory Markers in Patients with Aggressive Periodontitis

This paper simulated the optimal refrigerant charge inventory of a refrigeration system in air-conditioning operation and heat-pump operation respectively,and studied the refrigerant control strategies in this *** void fraction in two-phase fluid region was calculated by Harms *** based on distributed parameter model and Harms model,the refrigerant charge inventory in condenser and evaporator were calculated and analyzed in air-conditioning conditions and heat-pump conditions,*** calculating results of different refrigerant mass between refrigeration and heating conditions indicate that the optimal refrigerant charge inventory in heat-pump conditions is lower than that in air-conditioning *** avoid the decrease of COP due to the surplus refrigerant in heating conditions,we introduced the liquid reservoir control method and associate capillary control *** of them could increase the heating capacity of the air-source heat *** difference of optimal refrigerant charge inventory in air-conditioning and heat-pump system can be controlled by the liquid reservoir or the associate capillary.

Relationship between transforming growth factorβ1 and antifibrotic effect of interleukin-10

AIM： To study the effect of interleukin-10 （IL-10） on the expression of transforming growth factor β1 （TGF-β1） in hepatic fibrosis rats and the anti-fibrotic role of exogenous IL-10. METHODS： Hepatic fibrosis was induced by carbon tetrachloride administered （CCh） intraperitoneally. The experiment was performed in two stages. In the first stage, 60 SD rats were divided randomly into normal control group I（GNI, n = 8）, hepatic fibrosis group（GC, n = 28）and IL-10 intervened group（GI, n = 24）. At the beginning of the 7^th and 11^th wk, hepatic stellate cells （HSCs） were isolated, reverse transcription-polymerase chain reation （RT-PCR） and immunocytochemistry were performed to detect the expression of TGF-β1 in HSCs. Histological examination was used to determine the degree of hepatic fibrosis. In the second stage, 47 SD rats were divided randomly into normal control group 2 （GN2, n = 6）and CCh group（GZ, n = 41）. At the end of the 9th week, rats in GZ group were allocated randomly into model group（GM, n = 9）, IL-10 treatment group（GT, n = 9） and recovered group （GR, n = 9）. At the end of the 12^th week, all rats were sacrificed. RT-PCR and immuno- histochemistry were performed to detect the expression of TGF-β1 in liver tissue. ELISA was used to assay serum TGF-β1 levels. RESULTS： Hepatic fibrosis developed in rats with the increase of the injection frequency of CCI4. In the first stage, hepatic fibrosis developed and HSCs were isolated successfully. At the 7^th and 11^th week, TGF-β1 mRNA in GC group increased significantly compared with that in GN1（P = 0.001/0.042） and GI groups（P = 0.001/0.007）, whereas there was no significant difference between the two groups. The levels of TGF-β1 at the beginning of the 7^th wk was higher than that of the 11^th wk （P = 0.049）.Immunocytochemistry results of TGF-β1 were consistent with the above findings.
