Functional gastrointestinal disorders, mental health, genetic susceptibility, and incident chronic kidney disease

Background: Whether functional gastrointestinal disorders (FGIDs) are associated with the long-term risk of chronic kidney disease (CKD) remains unclear. We aimed to investigate the prospective association of FGIDs with CKD and examine whether mental health mediated the association.Methods: About 416,258 participants without a prior CKD diagnosis enrolled in the UK Biobank between 2006 and 2010 were included. Participants with FGIDs (including irritable bowel syndrome [IBS], dyspepsia, and other functional intestinal disorders [FIDs; mainly composed of constipation]) were the exposure group, and non-FGID participants were the non-exposure group. The primary outcome was incident CKD, ascertained from hospital admission and death registry records. A Cox proportional hazard regression model was used to investigate the association between FGIDs and CKD, and the mediation analysis was performed to investigate the mediation proportions of mental health.Results: At baseline, 33,156 (8.0%) participants were diagnosed with FGIDs, including 21,060 (5.1%), 8262 (2.0%), and 6437 (1.6%) cases of IBS, dyspepsia, and other FIDs, respectively. During a mean follow-up period of 12.1 years, 11,001 (2.6%) participants developed CKD. FGIDs were significantly associated with a higher risk of incident CKD compared to the absence of FGIDs (hazard ratio [HR], 1.36; 95% confidence interval [CI], 1.28–1.44). Similar results were observed for IBS (HR, 1.27; 95% CI, 1.17–1.38), dyspepsia (HR, 1.30; 95% CI, 1.17–1.44), and other FIDs (HR, 1.60; 95% CI, 1.43–1.79). Mediation analyses suggested that the mental health score significantly mediated 9.05% of the association of FGIDs with incident CKD and 5.63–13.97% of the associations of FGID subtypes with CKD. Specifically, the positive associations of FGIDs and FGID subtypes with CKD were more pronounced in participants with a high genetic risk of CKD.Conclusion: Participants with FGIDs had a higher risk of incident CKD, which was partly explained by mental health scores and was more pronounced

Characteristics and sources of peroxyacetyl nitrate(PAN)in the rural North China Plain:Results from1-year continuous observations

Peroxyacetyl nitrate(PAN)is an important photochemical pollutant in the troposphere,whereas long-term measurements are scarce in rural areas in North China Plain(NCP),resulting in unclear seasonal variations and sources of PAN in rural NCP.In this study,we conducted a 1-year observation of PAN during 2021-2022 at the rural NCP site.The average concentrations of PAN were 1.10,0.75,0.65,and 0.88 ppbv in spring,summer,autumn,and winter,respectively,with a 1-year average of 0.81±0.60 ppbv.Calculations indicate that the loss of PAN through thermal decomposition in summer accounts for 43.2% of the total formed PAN,which is an important reason for the low concentration of PAN in summer.We speculate that since the correlation between PAN and O_(3) in winter is significantly lower than that in other seasons,the observed regional transport of PAN cannot be ignored in winter.Through budget analysis,regional transport accounted for 12.8% and 55.9% of the observed PAN on the spring and winter pollution days,respectively,which showed that regional transport played key roles during the photochemical pollution of the rural NCP in winter.The potential source contribution function revealed that the transported PAN mainly comes from southern Hebei in spring.In winter,the transported PAN was mainly from Langfang,Hengshui,and southern Beijing.Our findings may aid in understanding PAN variations in different seasons in rural areas and highlight the impact of regional transport on the PAN budget.

GCARDTI:Drug–target interaction prediction based on a hybrid mechanism in drug SELFIES

The prediction of the interaction between a drug and a target is the most critical issue in the fields of drug development and repurposing.However,there are still two challenges in current deep learning research:(i)the structural information of drug molecules is not fully explored in most drug target studies,and the previous drug SMILES does not correspond well to effective drug molecules and(ii)exploration of the potential relationship between drugs and targets is in need of improvement.In this work,we use a new and better representation of the effective molecular graph structure,SELFIES.We propose a hybrid mechanism framework based on convolutional neural network and graph attention network to capture multi-view feature information of drug and target molecular structures,and we aim to enhance the ability to capture interaction sites between a drug and a target.In this study,our experiments using two different datasets show that the GCARDTI model outperforms a variety of different model algorithms on different metrics.We also demonstrate the accuracy of our model through two case studies.

Crimean-Congo hemorrhagic fever(CCHF)is a zoonotic disease caused by the CCHF virus(CCHFV),which is primarily transmitted by ticks(Lorenzo Juanes et al.2023).It is an emerging disease that occurs sporadically in Africa,Asia,and Europe,with a high morbidity and mortality rate,as high as 30%in humans(Ceylan et al.2013).CCHFV,belonging to genus Nairovirus,family Bunyaviridae,was first identified in the Congo in the 1960s.

Immune evasion has made ovarian cancer notorious for its refractory features, making the development of immunotherapy highly appealing to ovarian cancer treatment. The immune-stimulating cytokine IL-12 exhibits excellent antitumor activities. However, IL-12 can induce IFN-γ release and subsequently upregulate PDL-1 expression on tumor cells. Therefore, the tumor-targeting folate-modified delivery system F-DPC is constructed for concurrent delivery of IL-12 encoding gene and small molecular PDL-1 inhibitor(i PDL-1) to reduce immune escape and boost anti-tumor immunity. The physicochemical characteristics, gene transfection efficiency of the F-DPC nanoparticles in ovarian cancer cells are analyzed. The immune-modulation effects of combination therapy on different immune cells are also studied. Results show that compared with non-folate-modified vector, folate-modified F-DPC can improve the targeting of ovarian cancer and enhance the transfection efficiency of p IL-12. The underlying anti-tumor mechanisms include the regulation of T cells proliferation and activation, NK activation,macrophage polarization and DC maturation. The F-DPC/p IL-12/i PDL-1 complexes have shown outstanding antitumor effects and low toxicity in peritoneal model of ovarian cancer in mice. Taken together, our work provides new insights into ovarian cancer immunotherapy. Novel F-DPC/p IL-12/i PDL-1 complexes are revealed to exert prominent anti-tumor effect by modulating tumor immune microenvironment and preventing immune escape and might be a promising treatment option for ovarian cancer treatment.

Particle size dependence of the microsegregation and microstructure in the atomized Ni-based superalloy powders:Theoretical and experimental study

The microstructure and microsegregation of atomized powder,which depend on their sizes,are of great importance to the mechanical properties of the consolidated bulk materials.Therefore,it is necessary to reveal the relationship between particle size and powder attributes.The effects of particle size on the so-lidification characterization of the atomized Ni-based superalloy powders were studied via finite element simulation.Based on the simulations,a model was developed to predict the microsegregation and mi-crostructure of atomized powders with different sizes and study the influence of thermal history on the powder attributes during the atomization processes.The radiation heat transfer and temperature gradi-ent within the rapid solidification alloy powders were taken into account in this model.For validating the accuracy of the model,the predictions of the present model were compared with the microsegregation and microstructure of the specific size powder close to the screen mesh size.The results showed that mi-crostructure depended primarily on the temperature gradient within the powder,while the solidification rate had a more significant effect on the microsegregation.The model predicted microstructure features in agreement with the experiment,and for microsegregation,the deviations of prediction for most ele-ments were less than 10%.This work provides a new model to precisely predict the microsegregation and microstructure of the atomized alloy powders and sets a foundation to control the powder features for various engineering applications.

A Nonlinear Grey Bernoulli Model with Conformable Fractional-Order Accumulation and Its Application to the Gross Regional Product in the Cheng-Yu Area

This study considers a nonlinear grey Bernoulli forecasting model with conformable fractionalorder accumulation,abbreviated as CFNGBM(1,1,λ),to study the gross regional product in the ChengYu area.The new model contains three nonlinear parameters,the power exponentγ,the conformable fractional-orderαand the background valueλ,which increase the adjustability and flexibility of the CFNGBM(1,1,λ)model.Nonlinear parameters are determined by the moth flame optimization algorithm,which minimizes the mean absolute prediction percentage error.The CFNGBM(1,1,λ)model is applied to the gross regional product of 16 cities in the Cheng-Yu area,which are Chongqing,Chengdu,Mianyang,Leshan,Zigong,Deyang,Meishan,Luzhou,Suining,Neijiang,Nanchong,Guang’an,Yibin,Ya’an,Dazhou and Ziyang.With data from 2013 to 2021,several grey models are established and results show that the new model has higher accuracy in most cases.

SimHOEPI:A resampling simulator for generating single nucleotide polymorphism data with a high-order epistasis model

Epistasis is a ubiquitous phenomenon in genetics,and is considered to be one of main factors in current efforts to unveil missing heritability of complex diseases.Simulation data is crucial for evaluating epistasis detection tools in genome-wide association studies(GWAS).Existing simulators normally suffer from two limitations:absence of support for high-order epistasis models containing multiple single nucleotide polymorphisms(SNPs),and inability to generate simulation SNP data independently.In this study,we proposed a simulator SimHOEPI,which is capable of calculating penetrance tables of high-order epistasis models depending on either prevalence or heritability,and uses a resampling strategy to generate simulation data independently.Highlights of SimHOEPI are the preservation of realistic minor allele frequencies in sampling data,the accurate calculation and embedding of high-order epistasis models,and acceptable simulation time.A series of experiments were carried out to verify these properties from different aspects.Experimental results show that SimHOEPI can generate simulation SNP data independently with high-order epistasis models,implying that it might be an alternative simulator for GWAS.
