N-acetylcysteine protects against cadmium-induced germ cell apoptosis by inhibiting endoplasmic reticulum stress in testes

Cadmium （Cd） is a reproductive toxicant that induces germ cell apoptosis in the testes. Previous studies have demonstrated that endoplasmic reticulum （ER） stress is involved in Cd-induced germ cell apoptosis. The aim of the present study was to investigate the effects of N-acetylcysteine （NAC）, an antioxidant, on Cd-induced ER stress and germ cell apoptosis in the testes. Male CD-1 mice were intraperitoneally injected with CdCl2 （2.0 mg kg^-1）. As expected, acute Cd exposure induced germ cell apoptosis in the testes, as determined by terminal dUTP nick-end labelling （TUNEL）. However, the administration of NAC alleviated Cd-induced germ ceil apoptosis in the testes. Further analysis showed that NAC attenuated the Cd-induced upregulation of testicular glucose-regulated protein 78 （GRP78）, an important ER molecular chaperone. Moreover, NAC inhibited the Cd-induced phosphorylation of testicular eukaryotic translation initiation factor 2a （elF2a）, a downstream target of the double-stranded RNA-activated kinase-like ER kinase （PERK） pathway. In addition, NAC blocked the Cd-induced activation of testicular X binding protein （XBP）-1, indicating that NAC attenuates the Cd-induced ER stress and the unfolded protein response （UPR）. Interestingly, NAC almost completely prevented the Cd-induced elevation of C/EBP homologous protein （CHOP） and phosphorylation of c-Jun N-terminal kinase （.INK）, two components of the ER stress-mediated apoptotic pathway. In conclusion, NAC protects against Cd-induced germ cell apoptosis by inhibiting endoplasmic reticulum stress in the testes.

In this work,aramid nanoparticles(ANPs)were prepared in dimethyl formamide(DMF)solution containing high impact polystyrene(HIPS)via a bottom-up *** electron microscopy(TEM)images showed that the obtained ANPs were evenly distributed in the HIPS matrix without any *** composition of the ANPs,i.e.,poly(p-phenyl-p-phenylenediamine)(PPTA),was confirmed by Fourier transform infrared spectroscopy(FTIR),X-ray photoelectron spectroscopy(XPS)and X-ray diffractometer(XRD).The ANP/HIPS composites,obtained after ethanol precipitation,were added to neat HIPS as fillers to fabricate ANP/HIPS composite *** surface roughness and the glass transition temperature(T_g)of the sheets were characterized by atomic force microscope(AFM)and differential scanning calorimetry(DSC),*** with neat HIPS,the mechanical properties of the composite sheet were significantly improved,and the Young's modulus increased from 246.55 MPa to 2025.12 MPa,the tensile strength increased from 3.07 MPa to 29.76 MPa,and the toughness increased from 0.32 N/mm^2 to 3.92 N/mm^2,with an increase rate of 721%,869%and 1125%,***,the thermal stability of the composites improved with the increase in ANP content.

Objective： To determine whether additional Chinese medicine（CM） could prolong survival and improve the quality of life（QOL） in patients with advanced non-small cell lung cancer（NSCLC） compared with Western medicine（WM） alone. Methods： This was a multicenter, prospective cohort study. A total of 474 hospitalized patients with stage Ⅲ–Ⅳ NSCLC were recruited and divided into 2 groups. Patients in the WM group received radiotherapy, chemotherapy, and optimal supportive therapy according to the National Comprehensive Cancer Network（NCCN） guidelines. In the integrative medicine（IM） group, individualized CM（Chinese patent medicines and injections） and WM were administered. The primary end point was overall survival, and the secondary end points were time to disease progression, adverse events, and QOL. Follow-up clinical examinations and chest radiography were performed every 2 months. Results： The median survival was 16.60 months in the IM group and 13.13 months in the WM group（P〈0.01）. The incidences of loss of appetite, nausea, and vomiting in the IM group were significantly lower than those in the WM group（P〈0.05）. The QOL based on Functional Assessment of Cancer Therapy-Lung in the IM group was markedly higher than that in the WM group at the fourth course（P〈0.05）. Conclusions： Additional CM may prolong survival and improve the QOL patients with NSCLC. The adverse effects of radio-and chemotherapy may be attenuated as CM is used in combination with conventional treatments.

With its generality and practicality, the combination of partial charging curves and machine learning(ML) for battery capacity estimation has attracted widespread attention. However, a clear classification,fair comparison, and performance rationalization of these methods are lacking, due to the scattered existing studies. To address these issues, we develop 20 capacity estimation methods from three perspectives:charging sequence construction, input forms, and ML models. 22,582 charging curves are generated from 44 cells with different battery chemistry and operating conditions to validate the performance. Through comprehensive and unbiased comparison, the long short-term memory(LSTM) based neural network exhibits the best accuracy and robustness. Across all 6503 tested samples, the mean absolute percentage error(MAPE) for capacity estimation using LSTM is 0.61%, with a maximum error of only 3.94%. Even with the addition of 3 m V voltage noise or the extension of sampling intervals to 60 s, the average MAPE remains below 2%. Furthermore, the charging sequences are provided with physical explanations related to battery degradation to enhance confidence in their application. Recommendations for using other competitive methods are also presented. This work provides valuable insights and guidance for estimating battery capacity based on partial charging curves.
