GuanxinⅤRelieves Ventricular Remodeling by Inhibiting Inflammation:Implication from Virtual Screening,Systematic Pharmacology,Molecular Docking,and Experimental Validation

Objective:To reveal the anti-inflammatory mechanism of GuanxinⅤ,which is prescribed for ventricular remodeling in clinical ***:GuanxinⅤ-,ventricular remodeling-,and inflammationrelated targets were obtained through an integrated strategy of virtual screening and systematic pharmacology,and then the shared targets were visualised with a Venn ***Ⅴnetwork and the protein-protein interaction network were drawn,and enrichment analysis was ***,the main results obtained from the integrated strategy were validated by molecular docking and in vivo ***:A total of 251,11,425,and 15,246 GuanxinⅤ-,ventricular remodeling-,and inflammation-related targets were acquired,***,211 shared targets were considered to contribute to the mechanism of ventricular remodeling treated by GuanxinⅤ.Guanxin network and the protein-protein interaction network were drawn,and enrichment analysis showed some cardiovascular-related biological processes and signaling *** docking revealed that the GuanxinⅤ-derived compounds could align with key *** in vivo experiments proved that GuanxinⅤreverses ventricular remodeling by inhibiting ***:GuanxinⅤrelieves ventricular remodeling by regulating inflammation,which provides new ideas for the anti-ventricular remodeling mechanism of GuanxinⅤ.

农村脱贫家庭返贫风险评估与预警机制的构建——基于CFPS2020微观数据分析

有效识别返贫风险并及时预警是阻断脱贫人口返贫的前提。在分析生计脆弱性导致脱贫家庭返贫风险产生的作用机理基础上,结合CFPS2020数据,借助VSD模型,从暴露度、敏感度、适应能力3个维度形成脱贫家庭返贫风险评价指标体系,运用综合指标评估模型量化测度脱贫家庭返贫风险,并在此基础上构建返贫预警机制。结果表明:从整体来看,我国脱贫家庭脱贫稳定较强,返贫风险较小;从地理位置来看,西部地区属于中度返贫风险区域,面临着较高的内源风险和外源风险,“三区三州”仍是防止返贫的重点关注对象,东部地区返贫风险最低;从不同维度来看,影响四组脱贫家庭返贫风险差异主要原因是其自身适应性能力不同,其次是风险敏感性,风险暴露度影响最小;从具体指标来看,4组脱贫家庭风险敏感性和适应能力指数主要贡献因子是人力和金融指标,自然指标对其贡献率最低;基于返贫风险测算临界值,预警机制应结合警度、警源,从警度等级判断、警源识别与确认、预警帮扶介入、预警监督保障4个方面构建,以期增强预警识别的精确度,达到“精准帮扶”。因此,后扶贫时代应对五大风险靶向精准施策,构建有效的防返贫预警机制,根据不同返贫风险等级脱贫家庭出台针对性的防控措施。

环状RNA(circular RNA,circRNA)是前体信使RNA(pre-messenger RNA,pre-mRNA)经可变剪接产生的单链、共价闭合非编码RNA,具有高稳定性、序列保守性和疾病特异性表达的优势,因此吸引了广大关注。N6甲基腺苷(N6-methyladenosine,m 6A)甲基化发生在N6位置,是包括mRNA和非编码RNA(non-coding RNA,ncRNA)在内的真核生物内部转录修饰中最丰富和保守的一类。CircRNA和m 6A修饰已被证实,在调节脂质代谢在内的多种关键生物过程中发挥重要作用。随着测序技术的飞速发展,在转录物组范围内进行m 6A甲基化修饰的circRNA被广泛报道,并且发现它们参与了脂质代谢过程。即m 6A修饰参与调控circRNA的生物发生、降解、输出和翻译,circRNA也可以通过影响甲基化转移酶、去甲基化酶和甲基化阅读蛋白来调节m 6A修饰调控脂质代谢过程。因此,本文总结了与脂质代谢相关的m 6A修饰、circRNA及两者之间相互作用的最新突破,讨论了m 6A修饰的circRNA通过调控脂质合成、分解及脂质过氧化等脂代谢过程,尤其在脂质合成过程中发挥重要作用。对m 6A修饰的circRNA在脂质代谢中调控功能的综合分析揭示了其在脂质代谢途径中的重要作用,并为未来脂代谢异常相关疾病的研究提供了新的视角和理论依据。

Various infection status and molecular evidence for horizontal transmission and recombination of Wolbachia and Cardinium among rice planthoppers and related species

Wolbachia and Cardinium are widely distributed and are considered important for their ability to disturb reproduction and affect other fitness-related traits of their hosts. By using multilocus sequence typing （MLST）, RFLP （restriction fragment length poly- morphism） and 16S ribosomal DNA gene sequencing methods, we extensively surveyed Wolbachia and Cardinium infection status of four predominant rice planthoppers and one kind of leafhopper in different rice fields. The results demonstrated that Sogatellafurcifera （Horv^ith） and Laodelphax striatellus （Fallrn） were infected with the same Wolbachia strain （wStri）, while Nilaparvata lugens （St^l） and its closely related species Nilaparvata muiri China were infected with two phylogeneticlly distant strains, wLug and wMui, respectively. Three new Wolbachia strains （provisionally named wMfasl, wMfas2 and wMfas3） were detected in the leafhopper Macrostelesfascifrons （Stal）. Only S. furcifera was co-infected with Cardinium, which indicated that the distribution of Cardinium in these rice planthop- pers was narrower than that of Wolbachia. Unambiguous intragenic recombination events among these Wolbachia strains and incongruent phylogenetic relationships show that the connections between different Wolbachia strains and hosts were more complex than we expected. These results suggest that horizontal transmission and host associated special- ization are two factors affecting Wolbachia and Cardinium infections among planthoppers and their related species.

[目的]球囊霉素相关土壤蛋白(glomalin-related soil protein,GRSP)在自然界中广泛分布,其疏水性和稳定性有助于土壤团聚、增强土壤碳固存,因此准确测定GRSP含量对于探究它对土壤健康的影响至关重要.[方法]将青藏高原高寒草甸、浙东沿海森林、农田、城市绿化地土壤和东海近岸海洋沉积物筛分为粒径2 mm四个径级的团聚体,在不同离心时间和离心延迟时间下测定各级团聚体中易提取GRSP(easily extracted GRSP,EE-GRSP)的含量.[结果]高寒草甸土壤中EE-GRSP含量最高,为0.79～1.09 mg g-1,海洋沉积物中含量最少,仅为 0.33～0.49 mg g-1.除草甸土和海洋沉积物外,粒径为 0.053～0.25 mm团聚体中EE-GRSP含量较高.离心时间和离心延迟时间对各级团聚体中EE-GRSP含量测定结果的影响规律不同.粒径>2 mm团聚体中EE-GRSP含量的测定结果对离心时间变化不敏感,而粒径为 0.25～2 mm团聚体中EE-GRSP含量的测定结果受到离心时间影响最大.离心延迟时间对自然陆地生态系统森林和草甸土壤中EE-GRSP含量测定结果的影响不显著,但对人工生态系统农田和绿化地土壤都有一定程度的影响,对海洋沉积物中EE-GRSP含量的测定结果影响最大.[结论]对不同生态系统土壤中各级团聚体进行EE-GRSP含量测定时,离心时间或离心延迟时间的不同可能造成测定结果显著偏大或偏小.因此,建议根据土壤样品选择适宜的离心时间,并严格控制离心延迟时间.

YQWY decoction reverses cardiac hypertrophy induced by TAC through inhibiting GATA4 phosphorylation and MAPKs

To investigate the effect of Yiqi Wenyang(YQWY) decoction on reversing cardiac hypertrophy induced by the transverse aortic constriction(TAC). Wistar rats aged 7-8 weeks were subjected to TAC surgery and then randomly divided into 4 groups(n = 5/group): Sham group, TAC group, low-dose group and high dose group. After 16-week intragastric administration of YQWY decoction, the effect of YQWY decoction on alleviating cardiomyocyte hypertrophy was examined by transthoracic echocardiography(TTE), hematoxylin/eosin(HE), wheat germ agglutinin(WGA) staining, enzyme linked immunosorbent assay(ELISA), Western blot(WB), immunohistochemistry(IHC) and immunofluorescence(IF), respectively. The results showed significant differences in left ventricle volume-diastole/systole(LV Vol d/s), N-terminal pro-B-type brain natriuretic peptide(NT-proBNP)(P < 0.01), Ejection Fraction(EF), LV mass and fractional shortening(FS)(P < 0.05) between YQWY-treated group and TAC group. HE and WGA staining showed that treatment with YQWY decoction dramatically prevented TAC-induced cardiomycyte hypertrophy. Moreover, the results of WB, IHC and IF indicated that administration of YQWY could suppress the expressions of cardiac hypertrophic markers, which included the atrial natriuretic peptide(ANP), BNP and myosin heavy chain 7(MYH7)(P < 0.05) and inhibit phosphorylation of GATA binding protein 4(P-GATA4)(P < 0.05), phosphorylation of extracellular signal-regulated kinase(P-ERK)(P < 0.05), phosphorylation of P38 mitogen activated protein kinase(P-P38)(P < 0.05) and phosphorylation of c-Jun N-terminal kinase(P-JNK)(P < 0.05). Thus, we concluded that YQWY decoction suppressed cardiomyocyte hypertrophy and reversed the impaired heart function, and the curative effects of YQWY decoction were associated with the decreased phosphorylation of GATA4 and mitogen activated protein kinases(MAPKs), as well as the reduced expression of the downstream targets of GATA4, including ANP, BNP, and MYH7.

In this paper,we respectively discuss passivity of multi-weighted complex dynamical networks with fixed and switching ***,several passivity criteria for the complex dynamical network with fixed topology and multi-weights are established by employing Lyapunov functional approach and inequality ***,similar methods are used to derive some sufficient conditions for passivity of multi-weighted complex dynamical networks with switching ***,a numerical example with simulation results is given to show the correctness of the obtained passivity criteria.

Alpha-decay properties of nuclei around neutron magic numbers

By combining experimental α-decay energies and half-lives, the α-particle preformation factor for nuclei around neutron magic numbers N of 126, 152, and 162 were extracted using the two-potential approach. The nuclei around the shell closure were more tightly bound than adjacent nuclei. Additionally, based on the WS4 mass model (Wang et al., Phys. Lett.B 734, 215 (2014)), we extended the two-potential approach to predict the α-decay half-lives of nuclei around N values of178 and 184 with Z of 119 and 120. We believe that our findings will serve as guidelines for future experimental studies.

活性锰氧化物(MnO_(x))滤料在去除水中氨氮和锰污染物方面表现出高效催化氧化性能,但在去除双酚A(BPA)方面稳定性不足,且其与其他无机污染物相互作用机理尚不明确.采用过硫酸钠(NPS)增强活性滤料(FM)去除BPA,实验发现,0.1mmol/LNPS即可将去除率提升至80%以上,且随着NPS浓度增加,去除效率进一步提升,可达100%.NH_(4)^(+)和Mn^(2+)的加入分别通过促进电子转移和提供电子,增加活性物质产生,从而提高BPA去除效率.循环实验显示,虽然FM稳定性较好,但连续使用11次后去除率降至48%,而加入NPS后去除率可维持在65%以上.自由基淬灭和EPR实验以及X射线光电子能谱(XPS)分析证实,滤料/NPS体系中的主要活性物质为SO_(4)^(·-)、·OH和^(1)O_(2),Mn(Ⅲ)在催化氧化去除BPA过程中起关键作用,NPS的加入有助于Mn(Ⅲ)的形成,促进BPA去除.
