In order to detect the damage locations of complex spatial structures, a sensor region-based damage detection approach was developed based on the damage locating vectors method. A normalized damage locating index was introduced to identify the damage regions. An experiment on damage detection of a substructure model of the National Swimming Center 'Water Cube' was carried out. Two damage patterns were involved in the experiment. The test model was excited by using hammer impacts. Acceleration responses of the undamaged and damaged structure model were measured. Modal parameters were identified from the acceleration responses by utilizing the eigensystem realization algorithm (ERA). By using the developed sensor region-based method, the damage regions of the substructure model were located. The results show that the proposed method is able to effec- tively locate the damage regions.

Cell mechanics plays an important role in cellular physiological activities. Recent studies have shown that cellular mechanical properties are novel biomarkers for indicating the cell states. In this article, temperature-controllable atomic force microscopy(AFM) was applied to quantitatively investigate the effects of temperature and cellular interactions on the mechanics and morphology of human cancer cells. First, AFM indenting experiments were performed on six types of human cells to investigate the changes of cellular Young's modulus at different temperatures and the results showed that the mechanical responses to the changes of temperature were variable for different types of cancer cells. Second, AFM imaging experiments were performed to observe the morphological changes in living cells at different temperatures and the results showed the significant changes of cell morphology caused by the alterations of temperature. Finally, by co-culturing human cancer cells with human immune cells, the mechanical and morphological changes in cancer cells were investigated. The results showed that the co-culture of cancer cells and immune cells could cause the distinct mechanical changes in cancer cells, but no significant morphological differences were observed. The experimental results improved our understanding of the effects of temperature and cellular interactions on the mechanics and morphology of cancer cells.

Atomic force microscopy (AFM) can probe single living cells and single native membrane proteins in natural fluid environments with label-free high spatial resolution. It has thus become an important tool for cellular and molecular biology that significantly complements traditional biochemical and biophysical techniques such as optical and electron microscopy and X-ray crystallog-raphy. Imaging surface topography is the primary application of AFM in the life sciences. Since the early 1990s, researchers have used AFM to investigate morphological features of living cells and native membrane proteins with impressive results. Steady improvements in AFM techniques for imaging soft biological samples have greatly expanded its applications. Based on the authors' own research in AFM imaging of living cell morphologies, a review of sample preparation procedures for single-cell and single-molecule imaging experiments is presented, along with a summary of recent progress in AFM imaging of living cells and native membrane proteins. Finally, the challenges of AFM high-resolution imaging at the single-cell and single-molecule levels are discussed.

目的探究通络煎对脑梗死(气虚血瘀型)的临床疗效.方法随机将我院收治的68例脑梗死患者分为两组,每组各34例.对照组行常规治疗,治疗组加用通络煎.对比两组的中医证候积分、同型半胱氨酸(Hcy)水平、日常生活活动能力(ADL)评分、美国国立卫生研究院卒中量表(NIHSS)评分.结果治疗2周,两组中医证候积分、NIHSS评分、血浆Hcy水平均降低,且治疗组比对照组低(P＜0.05).治疗后3个月,两组ADL评分均升高,且治疗组比对照组高(P＜0.05).结论通络煎可减轻脑梗死(气虚血瘀型)患者的中医证候表现,降低血浆Hcy水平,改善其神经功能及日常生活能力.

Elucidating the underlying mechanisms of cell physiology is currently an important research topic in life sciences. Atomic force microscopy methods can be used to investigate these molecular mechanisms. In this study, single-molecule force spectroscopy was used to explore the specific recognition between the CD20 antigen and anti-CD20 antibody Rituximab on B lymphoma cells under near-physiological conditions. The CD20-Rituximab specific binding force was measured through tip functionalization. Distribution of CD20 on the B lymphoma cells was visualized three-dimensionally. In addition, the relationship between the intramolecular force and the molecular extension of the CD20-Rituximab complex was analyzed under an external force. These results facilitate further investigation of the mechanism of Rituximab’s anti-cancer effect.

The invention of atomic force microscopy(AFM) has provided new technology for measuring specific molecular interaction *** AFM single-molecule force spectroscopy(SMFS) techniques,CD20-Rimximab rupture forces were measured on purified CD20 proteins,Raji cells,and lymphoma patient B *** molecules were linked onto AFM tips using AFM probe functionalization technology,and purified CD20 proteins were attached to mica using substrate functionalization *** cells(a lymphoma cell line) or lymphoma patient cells were immobilized on a glass substrate via electrostatic adsorption and chemical *** topography of the purified CD20 proteins,Raji cells,and patient lymphoma cells was visualized using AFM imaging and the differences in the rupture forces were analyzed and *** results showed that the rupture forces between the CD20 proteins on Raji cells and Rituximab were markedly smaller than those for purified CD20 proteins and CD20 proteins on lymphoma patient B *** findings provide an effective experimental method for investigating the mechanisms underlying the variable efficacy of Rituximab.

Biliary atresia is a rare infant disease that predisposes patients to liver transplantation and death if not treated in ***,early diagnosis is challenging because the clinical manifestations and laboratory tests of biliary atresia overlap with other cholestatic ***,it is very important to develop a simple,safe and reliable method for the early diagnosis of biliary ***,a novel NIR-Ⅱfluorescence probe,HZL2,with high quantum yield,excellent biocompatibility,low cytotoxicity and rapid excretion through the liver and gallbladder was developed based on the oil/water partition coefficient and permeability.A simple fecal sample after injection of HZL2 can be used to efficiently identify the success of the mouse model of biliary atresia for the first time,allowing for an early diagnosis of the *** study not only developed a simple and safe method for the early diagnosis of biliary atresia with great potential in clinical translation but also provides a research tool for the development of pathogenesis and therapeutic medicines for biliary atresia.

Mapping CD20 molecules on the lymphoma cell surface using atomic force microscopy

Atomic force microscopy (AFM) was used to locate CD20 molecules on the surface of lymphoma Raji cells. Rituximab (a monoclonal antibody against CD20) molecules were linked onto the AFM tip via a polyethylene glycol (PEG) linker. Raji cells were adsorbed onto glass slides coated with poly-L-lysine. First, the CD20 distribution in a local area of the cell surface was visualized using the AFM lift scan mode. Second, 16 × 16 force curves were obtained from the same cell area to construct the CD20-rituximab binding force map. Finally, free rituximab was added to block the CD20 molecules on the cell surface and the lift phase image and CD20-rituximab force map were obtained again. The experimental results indicated that when the lift height was greater than the length of the PEG linker, no recognition sites were observed in the lift phase image. However, as the lift height decreased to the length of the PEG linker, some recognition sites were observed in the lift phase image and these sites were generally consistent with the pixels in the force map. After blocking, both the recognition sites in the lift phase image and the gray pixels in the binding force map decreased markedly. These results can improve our understanding of the distribution of protein molecules on the cell surface and facilitate further investigations into cellular functions.

Enhancing convolutional neural network scheme for rheumatoid arthritis grading with limited clinical data

The gray-scale ultrasound(US) imaging method is usually used to assess synovitis in rheumatoid arthritis(RA) in clinical practice. This four-grade scoring system depends highly on the sonographer's experience and has relatively lower validity compared with quantitative indexes. However, the training of a qualified sonographer is expensive and timeconsuming while few studies focused on automatic RA grading methods. The purpose of this study is to propose an automatic RA grading method using deep convolutional neural networks(DCNN) to assist clinical assessment. Gray-scale ultrasound images of finger joints are taken as inputs while the output is the corresponding RA grading results. Firstly,we performed the auto-localization of synovium in the RA image and obtained a high precision in localization. In order to make up for the lack of a large annotated training dataset, we performed data augmentation to increase the number of training samples. Motivated by the approach of transfer learning, we pre-trained the GoogLeNet on ImageNet as a feature extractor and then fine-tuned it on our own dataset. The detection results showed an average precision exceeding 90%. In the experiment of grading RA severity, the four-grade classification accuracy exceeded 90% while the binary classification accuracies exceeded 95%. The results demonstrate that our proposed method achieves performances comparable to RA experts in multi-class classification. The promising results of our proposed DCNN-based RA grading method can have the ability to provide an objective and accurate reference to assist RA diagnosis and the training of sonographers.
