Effect of local viral transfer of interleukin 10 gene on a rabbit arthritis model induced by interleukin 1β

Backgroud Interleukin 16 （IL-1β） is the principal mediator in the pathogenesis of rheumatoid arthritis. Continuous injection of interleukin 1β （IL-1β） into the knee articular cavities of anamals can induce models that resemble rheumatoid arthritis. The objective of this study was to evaluate the feasibility of local recombinant retrovirus viral interleukin 10 （rRV-vlL-10） gene transfer treatment of a rabbit model of arthritis induced by IL-16. Methods An hlL-1β-induced rabbit rheumatoid arthritis model was established using the MFG-hlL-1β-neo-HIG-82 cell line, which is capable of continuous secretion of hlL-1β. After transfecting the rabbit synovial fibroblast cell line （MFG-hlL-1β-neo-HIG-82） with rRV-vlL-10, G418 was then added to identify the positive clone. The rRV-vlL-10 positive clone was injected into the established rabbit rheumatoid arthritis model through intra-articular injection. Successful gene transfer was determined by reverse transcripUon-polymerase chain reaction （RT-PCR） and immunohistochemistry. The levels of IL-1β before and after treatment were determined by enzyme- linked immunosorbent assay. Results Retrovirus vector was an effective vector both to synoviocytes in vitro and synovium tissue in vivo as confirmed by RT-PCR and immunohistochemistry. The rabbit arthritis model treated with rRV-vlL-10 showed a dramatic remission of arthritis and a decline in the level of cytokines such as IL-1β. Conclusions Retrovirus-mediated transfecUon of vlL-10 successfully transferred the gene into rabbit synovium ex vivo and was able to suppress intra-articular inflammation response to IL-1β.

The reaction order plays a crucial role in evaluating the response rate of ***,the conventional two-scale model typically assumes that the reaction order is constant as one,which can lead to significant deviations from *** address this issue,this study proposes a novel multi-order dynamic model for acid-rock reaction by combining rotating disk experimental data with theoretical *** numerical simulations,this model allows for the investigation of the impact of acidification conditions on different orders of reaction,thereby providing valuable insights for on-site *** analysis reveals that higher response orders require higher optimal acid liquid flow rates,and lower optimal H+diffusion coefficients,and demonstrate no significant correlation with acid ***,it is recommended to increase the displacement and use high-viscosity acid for reservoirs with high calcite content,while reducing the displacement and using low-viscosity acid for reservoirs with high dolomite content.

BACKGROUND Bipolar disorder(BD)is a severe mental *** often coexists with borderline personality disorders,making the condition more *** To explore the differences in cognitive impairment between patients with BD and those with BD comorbid with borderline personality *** Eighty patients with BD and comorbid borderline personality disorder and 80 patients with BD alone were included in groups A and B,respectively,and 80 healthy volunteers were included as *** function in each group was evaluated using the Chinese version of the repeatable battery for the assess-ment of neuropsychological status(RBANS),the Stroop color-word test,and the Wechsler intelligence scale-revised(WAIS-RC).RESULTS The indices of the RBANS,Stroop color-word test,and WAIS-RC in groups A and B were significantly lower than those of the control group(P<0.05).Group A had significantly longer Stroop color-word test times for single-character,single-color,double-character,and double-color,lower scores of immediate memory,visual breadth,verbal function dimensions and total score of the RBANS,as well as lower scores of verbal IQ,performance IQ,and overall IQ of the WAIS-RC compared with group B(P<0.05).Compared to group B,group A exhibited significantly longer single-character time,single-color time,double-character time,and double-color time in the Stroop color-word test(P<0.05).CONCLUSION The cognitive function of patients with BD complicated with borderline personality disorder is lower than that of patients with BD.

Dear Editor,Severe fever with thrombocytopenia syndrome(SFTS)is an emerging tick-borne infectious disease caused by a novel bunyavirus called SFTS virus(SFTSV).It was initially identified in China in 2009(Yu et al.,2011).Since then,the number of reported SFTS cases has rapidly increased in China,South Korea,and Japan(Li,et al.,2018;Takahashi et al.,2014;Kim et al.,2018).Sporadic SFTS cases have also been identified in several other Asian countries,such as Vietnam,Pakistan,Myanmar,and Thailand(Takahashi et al.,2014;Tran et al.,2019;Li et al.,2021).This disease is recognized as a highly lethal viral hemorrhagic fever with a mortality rate ranging from 12%to 50%(Yu et al.,2011;Li et al.,2018;Takahashi et al.,2014;Kim et al.,2013).SFTS primarily spreads to humans through bites from ticks infected with SFTSV,with the Haemaphysalis longicornis tick acting as the predominant vector for SFTSV(Zhuang et al.,2018).

Psoriasis is a chronic,recurrent,inflammatory systemic disease that is not only characterized by skin manifestations but may also be accompanied by various comorbidities,imposing a heavy burden on patients’physical and mental health and affecting their quality of *** the continuous approval of various biological agents for clinical use has provided more effective treatment options to patients with psoriasis,no consensus on the evaluation of comprehensive treatment goals has yet been *** treat-to-target(T2T)strategy requires consideration of multiple dimensions of treatment outcomes,the development of long-term management goals,and regular assessments of treatment conditions,which are often used in the management of chronic ***,based on the latest consensuses and guidelines,research data,and clinical experience as well as the combination of survey results and expert group discussions,the present consensus focuses on 4 dimensions of short-and long-term integrated management goals for biological agents:alleviating skin lesions,improving quality of life,screening and managing psoriasis comorbidities,and ensuring drug *** implementation methods,evaluation time,treatment monitoring,and program adjustments are also herein described to achieve comprehensive management of psoriasis to the maximum *** consensus provides a reference for clinical practice.

Various SARS-CoV-2-related coronaviruses have been increasingly identified in pangolins,showing a potential threat to *** we report the infectivity and pathogenicity of the SARS-CoV-2-related virus,PCoV-GX/P2V,which was isolated from a Malayan pangolin(Manis javanica).PCoV-GX/P2V could grow in human hepatoma,colorectal adenocarcinoma cells,and human primary nasal epithelial *** replicated more efficiently in cells expressing human angiotensin-converting enzyme 2(hACE2)as SARS-CoV-2 *** intranasal inoculation to the hACE2-transgenic mice,PCoV-GX/P2V not only replicated in nasal turbinate and lungs,but also caused interstitial pneumonia,characterized by infiltration of mixed inflammatory cells and multifocal alveolar *** population immunity established by SARS-CoV-2 infection and vaccination may not protect people from PCoV-GX/P2V *** findings further verify the hACE2 utility of PCoV-GX/P2V by in vivo experiments using authentic viruses and highlight the importance for intensive surveillance to prevent possible cross-species transmission.

根施生物炭对设施连作土壤氮素转化及黄瓜幼苗根系氮代谢的影响

设施连作土壤普遍存在由于氮素过量积累导致的土壤障碍问题。本研究以不添加生物炭作为对照,以添加5%的生物炭(质量比)为处理,采用盆栽方式分析了生物炭对土壤微生物群落结构、氮循环功能基因丰度以及黄瓜幼苗根系生长和氮素代谢相关基因表达的影响,以期探明外源添加生物炭对设施连作土壤氮素转化的调控作用。结果表明:与对照相比,设施连作土壤添加生物炭处理后显著提高了黄瓜幼苗的株高、根系干重、总根长、根表面积和根体积;改善了根区环境,诱导黄瓜通过上调植物氮代谢相关基因的表达促进了黄瓜根系对氮素的吸收。施入生物炭显著提高了土壤微生物生物量氮、硝态氮和亚硝态氮含量,显著提高了土壤中变形菌门和蓝细菌门以及鞘脂单胞菌(土壤固氮菌)等氮代谢相关菌群丰度;增加了土壤硝化、氮同化还原功能基因丰度;增强了参与氮代谢的羟胺脱氢酶、硝基单加氧酶和碳酸酐酶活性。综上,根施生物炭改善了设施连作土壤理化性质和微生物生态系统,通过促进硝化作用和氮同化作用来调节土壤的氮循环过程,提高了植株对土壤氮素的吸收能力,最终促进了黄瓜植株生长。

Expression of Peptidylarginine Deiminase 4 and Protein Tyrosine Phosphatase Nonreceptor Type 22 in the Synovium of Collagen-Induced Arthritis Rats

Objective To study the expression level of peptidylarginine deiminase 4(PADI4) and protein tyrosine phosphatase nonreceptor type 22(PTPN22) in the synovium of rat model of collagen-induced arthritis, and to explore their possible therapeutic role in rheumatoid arthritis. Methods Thirty-two female Wistar rats weighing 100±20 g were randomly assigned into 3-week collagen-induced arthritis(CIA) model group(n=8), 4-week CIA model group(n=8), 6-week CIA model group(n=8), and the control group(n=8). The body weight changes of each group were recorded. The expression levels of PADI4 and PTPN22 were detected and compared by the methods of immunohistochemical staining and Western blot. Results Arthritis of rat began to form 14 days after sensitization and the joint swelling reached peak at 28 days. The weights of the rats slowly grew both in CIA model groups and the control group. Immunohistochemical staining results showed that the positive expression of PADI4 and PTPN22 was mainly located in cartilage peripheral mononuclear cells, the cytoplasm of infiltrated cells, and bone marrow cavity. There were significant differences in the optical density of PADI4 and PTPN22 among CIA model groups and the control group(PADI4, 0.2898±0.012, 0.2982±0.022, 0.2974±0.031, 0.2530±0.013 in 3-week CIA model, 4-week CIA model, 6-week CIA model and control groups; PTPN22, 0.2723±0.004, 0.2781±0.010, 0.2767±0.008, 0.2422±0.019; all P <0.05). The expression bands of PADI4 were observed in Western blot 3 weeks after initial immunization, the thickest in the 4th week, and decreased in the 6th week. The expression bands of PTPN2 were observed at all the time points, with no obvious time-dependent trend. Conclusions PADI4 and PTPN22 are obviously correlated with CIA in rat model. PADI4 is expressed at early stage of the disease, while the expression of PTPN22 sustains throughout the course.
