Decreased AKAP4/PKA signaling pathway in high DFI sperm affects sperm capacitation

The sperm DNA fragmentation index(DFI)is a metric used to assess DNA fragmentation within sperm.During in vitro fertilizationembryo transfer(IVF-ET),high sperm DFI can lead to a low fertilization rate,poor embryo development,early miscarriage,etc.A kinase anchoring protein(AKAP)is a scaffold protein that can bind protein kinase A(PKA)to subcellular sites of specific substrates and protects the biophosphorylation reaction.Sperm protein antigen 17(SPA17)can also bind to AKAP.This study intends to explore the reason for the decreased fertilization rate observed in high sperm DFI(H-DFI)patients during IVF-ET.In addition,the study investigates the expression of AKAP,protein kinase A regulatory subunit(PKARIl),and SPA17 between H-DFI and low sperm DFI(L-DFI)patients.SPA17 at the transcriptional level is abnormal,the translational level increases in H-DFI patients,and the expression of AKAP4/PKARIl protein decreases.H,O,has been used to simulate oxidative stress damage to spermatozoa during the formation of sperm DFI.It indicates that H,O,increases the expression of sperm SPA17 protein and suppresses AKAP4/PKARIl protein expression.These processes inhibit sperm capacitation and reduce acrosomal reactions.Embryo culture data and IVF outcomes have been documented.The H-DFI group has a lower fertilization rate.Therefore,the results indicate that the possible causes for the decreased fertilization rate in the H-DFI patients have included loss of sperm AKAP4/PKARIl proteins,blocked sperm capacitation,and reduced occurrence of acrosome reaction.

Nanoplastics-induced developmental and reproductive toxicity,neurotoxicity and immunotoxicity are a focus of widespread attention.However,the effects of nanoplastics(NPs)on glycolipid metabolism and the precise underlying mechanisms are unclear at present.Here,we showed that oral administration of polystyrene nanoparticles(PS-NPs)disrupts glycolipid metabolism,with reactive oxygen species(ROS)identified as a potential key signaling molecule.After PS-NPs treatment,excessive production of ROS induced the infammatory response and activated the antioxidant pathway through nuclear factor-erythroid factor 2-related factor 2.The activation of nuclear factor-κB(NFκB)signaling pathway induced the phosphorylation of the mitogen-activated protein kinases(MAPK)signaling pathway,which induced the activation of extracellular regulated kinases(ERK)and p38.Constitutive activation of the MAPK signaling proteins induced high continued phosphorylation of insulin receptor substrate-1,in turn,leading to decreased protein kinase B(Akt)activity,which weakened the sensitivity of liver cells to insulin signals and induced insulin resistance.In parallel,phosphorylation of Akt led to loss of control of Fo XO1,a key gene of gluconeogenesis,activating transcription of glucose-6-phosphatase(G6PC)and phosphoenolpyruvate carboxykinase(PEPCK)in a manner dependent on PGC1α.Moreover,the activated ERK promoted lipid accumulation through ERK-PPARγcascades.Therefore,sterol regulatory element-binding protein-1 and levels of its downstream lipogenic enzymes,ACC-1,were up-regulated.Upon treatment with the antioxidant resveratrol,PS-NPs-induced glucose and lipid metabolic disorders were improved by inhibiting ROS-induced activation of NFκB and MAPK signaling pathway in mice.Based on above,PS-NPs exposure disrupts glycolipid metabolism in mice,with ROS identified as a potential key signaling molecule.

Application and progress of artificial intelligence technology in the segmentation of hyperreflective foci in OCT images for ophthalmic disease research

With the advancement of retinal imaging,hyperreflective foci(HRF)on optical coherence tomography(OCT)images have gained significant attention as potential biological biomarkers for retinal neuroinflammation.However,these biomarkers,represented by HRF,present pose challenges in terms of localization,quantification,and require substantial time and resources.In recent years,the progress and utilization of artificial intelligence(AI)have provided powerful tools for the analysis of biological markers.AI technology enables use machine learning(ML),deep learning(DL)and other technologies to precise characterization of changes in biological biomarkers during disease progression and facilitates quantitative assessments.Based on ophthalmic images,AI has significant implications for early screening,diagnostic grading,treatment efficacy evaluation,treatment recommendations,and prognosis development in common ophthalmic diseases.Moreover,it will help reduce the reliance of the healthcare system on human labor,which has the potential to simplify and expedite clinical trials,enhance the reliability and professionalism of disease management,and improve the prediction of adverse events.This article offers a comprehensive review of the application of AI in combination with HRF on OCT images in ophthalmic diseases including age-related macular degeneration(AMD),diabetic macular edema(DME),retinal vein occlusion(RVO)and other retinal diseases and presents prospects for their utilization.

One-year results for myopia control of orthokeratology with different back optic zone diameters: a randomized trial using a novel multispectral-based topographer

AIM:To present the 1-year results of a prospective cohort study investigating the efficacy,potential mechanism,and safety of orthokeratology(ortho-k)with different back optic zone diameters(BOZD)for myopia control in children.METHODS:This randomized clinical study was performed between Dec.2020 and Dec.2021.Participants were randomly assigned to three groups wearing ortho-k:5 mm BOZD(5-MM group),5.5 mm BOZD(5.5-MM group),and 6 mm BOZD(6-MM group).The 1-year data were recorded,including axial length,relative peripheral refraction(RPR,measured by multispectral refractive topography,MRT),and visual quality.The contrast sensitivity(CS)was evaluated by CSV-1000 instrument with spatial frequencies of 3,6,12,and 18 cycles/degree(c/d);the corneal higher-order aberrations(HOAs)were measured by iTrace aberration analyzer.The one-way ANOVA was performed to assess the differences between the three groups.The correlation between the change in AL and RPR was calculated by Pearson’s correlation coefficient.RESULTS:The 1-year results of 20,21,and 21 subjects in the 5-MM,5.5-MM,and 6-MM groups,respectively,were presented.There were no statistical differences in baseline age,sex,or ocular parameters between the three groups(all P>0.05).At the 1-year visit,the 5-MM group had lower axial elongation than the 6-MM group(0.07±0.09 vs 0.18±0.11 mm,P=0.001).The 5-MM group had more myopic total RPR(TRPR,P=0.014),with RPR in the 15°–30°(RPR 15–30,P=0.015),30°–45°(RPR 30–45,P=0.011),temporal(RPR-T,P=0.008),and nasal area(RPR-N,P<0.001)than the 6-MM group.RPR 15–30 in the 5.5-MM group was more myopic than that in the 6-MM group(P=0.002),and RPR-N in the 5-MM group was more myopic than that in the 5.5-MM group(P<0.001).There were positive correlations between the axial elongation and the change in TRPR(r=0.756,P<0.001),RPR 15–30(r=0.364,P=0.004),RPR 30–45(r=0.306,P=0.016),and RPR-N(r=0.253,P=0.047).The CS decreased at 3 c/d(P<0.001),and the corneal HOAs increased in the 5-MM group(P=0.030).CONCLUSION:Ortho-k with 5 mm BOZD can control myopia

Learning to represent 2D human face with mathematical model

How to represent a human face pattern?While it is presented in a continuous way in human visual system,computers often store and process it in a discrete manner with 2D arrays of pixels.The authors attempt to learn a continuous surface representation for face image with explicit function.First,an explicit model(EmFace)for human face representation is pro-posed in the form of a finite sum of mathematical terms,where each term is an analytic function element.Further,to estimate the unknown parameters of EmFace,a novel neural network,EmNet,is designed with an encoder-decoder structure and trained from massive face images,where the encoder is defined by a deep convolutional neural network and the decoder is an explicit mathematical expression of EmFace.The authors demonstrate that our EmFace represents face image more accurate than the comparison method,with an average mean square error of 0.000888,0.000936,0.000953 on LFW,IARPA Janus Benchmark-B,and IJB-C datasets.Visualisation results show that,EmFace has a higher representation performance on faces with various expressions,postures,and other factors.Furthermore,EmFace achieves reasonable performance on several face image processing tasks,including face image restoration,denoising,and transformation.

End-Triassic storm deposits in the lacustrine Sichuan Basin and their driving mechanisms

Storm deposits or tempestites are event sequences formed by storms,requiring at least a water temperature of 26.5℃.While inland lakes are unlikely to form storm deposits because of their limited width and water temperature.The Upper Triassic Xujiahe Formation in the Sichuan Basin is a set of coal-bearing,clastic sequences with dominant sedimentary facies varying from braided river delta to lacustrine settings,with storm deposits widely reported.In the Zilanba of Guanyuan area,in situ tree trunks on a palaeosol surface in Member Vof the Xujiahe Formation provide new evidence of a storm event.Six fallen-down directions of nine in situ tree trunks were predominant in the NW direction,contrary to the palaeocurrent direction of the underlying strata,suggesting that the southeasterlies prevailed during the end-Triassic in the northern Sichuan Basin.Massive mud clasts were frequently recorded in sandstones of the Xujiahe Formation,as well as in the Xindianzi section.These mud clasts showed a rip-up or a plastic deformation with upside-down V-shapes,were capped on an erosional surface,showed no transport traces and were therefore interpreted as a storm lag deposit.The megamonsoonal climate prevailed during the Late Triassic,although the megamonsoons themselves could not generate a storm deposition in the Xujiahe Formation due to its low maximum surface wind speed.The driving mechanism for generating storm deposits in the Xujiahe Formation is suggested to be tropical cyclones over the Tethys Ocean moving eastward,further landfalling on the western margin of the Sichuan Basin.Statistics of storm events in the circum-Tethys region show a widespread storm surge in low latitudes during the end-Triassic.The storm deposits at the top of the Xujiahe Formation represent a sedimentary response to the end-Triassic hyperthermal event.

Predicting Credit Bond Default with Deep Learning: Evidence from China

China’s credit bond market has rapidly expanded in recent years.However,since 2014,the number of credit bond defaults has been increasing rapidly,posing enormous potential risks to the stability of the financial market.This study proposed a deep learning approach to predict credit bond defaults in the Chinese market.A convolutional neural network(CNN)was selected as the classification model and to reduce the extreme imbalance between defaulted and non-defaulted bonds,and a generative adversarial network(GAN)was used as the oversampling model.Based on 31 financial and 20 non-financial indicators,we collected Wind data on all credit bonds issued and matured or defaulted from 2014 to 2021.The experimental results showed that our GAN+CNN approach had superior predictive performance with an area under the curve(AUC)of 0.9157 and precision of 0.8871 compared to previous research and other commonly used classification models-including the logistic regression,support vector machine,and fully connected neural network models-and oversampling techniques-including the synthetic minority oversampling technique(SMOTE)and Borderline SMOTE model.For one-year predictions,indicators of solvency,capital structure,and fundamental properties of bonds are proved to be the most important indicators.

The combined effect of cavitation and silt abrasion presents a great challenge and threat to secure the operation and the efficiency of hydraulic machineries working in sediment-laden fluid.The present paper critically reviews the current research progress on the interaction mechanisms of the bubbles and the particles.Firstly,the analytical models including boundary treatment methods for predicting the jet dynamics of the bubble collapse near particles are demonstrated.Secondly,the bubble collapsing dynamics,jet dynamics and shock wave characteristics near particles are revealed both experimentally and numerically.Finally,the bubble-particle-wall system is investigated with a focus on microjets.

Boosted Electrocatalytic Glucose Oxidation Reaction on Noble-Metal-Free MoO_(3)-Decorated Carbon Nanotubes

Electrocatalytic glucose oxidation reaction(GOR)has attracted much attention owing to its crucial role in biofuel cell fabrication.Herein,we load MoO_(3)nanoparticles on carbon nanotubes(CNTs)and use a discharge process to prepare a noblemetal-free MC-60 catalyst containing MoO_(3),Mo_(2)C,and a Mo_(2)C–MoO_(3)interface.In the GOR,MC-60 shows activity as high as 745μA/(mmol/L cm^(2)),considerably higher than those of the Pt/CNT(270μA/(mmol/L cm^(2)))and Au/CNT catalysts(110μA/(mmol/L cm^(2))).In the GOR,the response minimum on MC-60 is as low as 8μmol/L,with a steady-state response time of only 3 s.Moreover,MC-60 has superior stability and anti-interference ability to impurities in the GOR.The better performance of MC-60 in the GOR is attributed to the abundant Mo sites bonding to C and O atoms at the MoO_(3)–Mo_(2)C interface.These Mo sites create active sites for promoting glucose adsorption and oxidation,enhancing MC-60 performance in the GOR.Thus,these results help to fabricate more effi cient noble-metal-free catalysts for the fabrication of glucose-based biofuel cells.

Deformable catalytic material with excellent flexible structure is a new type of catalyst that has been applied in various chemical reactions,especially electrocatalytic hydrogen evolution reaction(HER).In recent years,deformable catalysts for HER have made great progress and would become a research hotspot.The catalytic activities of deformable catalysts could be adjustable by the strain engineering and surface reconfiguration.The surface curvature of flexible catalytic materials is closely related to the electrocatalytic HER properties.Here,firstly,we systematically summarized self-adaptive catalytic performance of deformable catalysts and various micro–nanostructures evolution in catalytic HER process.Secondly,a series of strategies to design highly active catalysts based on the mechanical flexibility of lowdimensional nanomaterials were summarized.Last but not least,we presented the challenges and prospects of the study of flexible and deformable micro–nanostructures of electrocatalysts,which would further deepen the understanding of catalytic mechanisms of deformable HER catalyst.
