Exposure to heat-inactivated Trichophyton rubrum resulting in a limited immune response of human keratinocytes

Background Trichophyton rubrum （T. rubrum） represents the most important agent of dermatophytosis in humans. T. rubrum infection causes slight inflammation, and tends to be chronic and recurrent. It is suggested that it may result from the failure of epithelial cells to recognize T. rubrum effectively and initiate effective immune responses. The C-type lectin receptors （CLR） and toll-like receptors （TLR） are the two major pattern recognition receptors （PRRs） that recognize fungal components. Therefore, the purpose of the study was to analyze the expression of those PRRs and the cytokines in HaCaT cells stimulated with heat-inactivated T. rubrum conidia and hyphae, respectively. Methods HaCaT cells were unstimulated or stimulated with heat-inactivated T. rubrum conidia and hyphae （l×106 and 1.5×105 colony-forming unit （CFU） in 2 ml medium, respectively） for 6, 12 and 24 hours. The mRNA expression of PRRs involved in recognizing fungal pathogen-associated molecular patterns （PAMPs） and signaling molecules were measured by quantitative reverse transcription polymerase chain reaction （RT-PCR）. Meanwhile, surface toll-like receptor （TLR） 2, TLR4 and Dectin-1 were analyzed by fluorescence-activated cell sorter （FACS） 24 hours after treatment. The cytokines were detected in cell culture supernatants of HaCaT cells in 12 and 24 hours after treatment. Results HaCaT cells constitutively expressed mRNA of membrane-bound TLR1,2, 4 and 6, Dectinl and DC-SIGN, but not Dectin-2 or Mincle. Heat-killed T. rubrum did not significantly upregulate gene transcriptions of the PRRs of HaCaT cells. Heat-inactivated T. rubrum conidia significantly reduced the surface expression of TLR2 and Dectin-1, and suppressed the secretions of interferon-inducible protein-10 （IP-10） and monocyte chemotactic protein-1 （MCP-1） of HaCaT cells, while heat-killed T. rubrum hyphae significantly induced the secretions of IP-10 and MCP-I. Improving mechanical properties and high-temperature oxidation of press hardened steel by adding Cr and Si

This work investigated the effect of Cr and Si on the mechanical properties and oxidation resistance of press hardened *** indicated that the microstructure of the Cr-Si micro-alloyed press hardened steel consisted of lath martensite,M_(23)C_(6)carbides,and retained *** retained austenite and carbides are responsible for the increase in elongation of the micro-alloyed *** addition,after oxidation at 930℃for 5 min,the thickness of the oxide scales on the Cr-Si micro-alloyed press hardened steel is less than 5μm,much thinner than 45.50μm-thick oxide scales on *** oxide scales of the Cr-Si micro-alloyed steel are composed of Fe_(2)O_(3),Fe_(3)O_(4),mixed spinel oxide(FeCr_(2)O_(4)and Fe_(2)SiO_(4)),and amorphous SiO_(2).Adding Cr and Si significantly reduces the thickness of the oxide scales and prevents the generation of the FeO *** to the increase of spinel FeCr_(2)O_(4)and Fe_(2)SiO_(4)phase in the inner oxide scale and the amorphous SiO_(2)close to the substrate,the oxidation resistance of the Cr-Si micro-alloyed press hardened steel is improved.

Vanadium oxide nanospheres encapsulated in N-doped carbon nanofibers with morphology and defect dual-engineering toward advanced aqueous zinc-ion batteries

Vanadium-based electrodes are regarded as attractive cathode materials in aqueous zinc ion batteries(ZIBs)caused by their high capacity and unique layered ***,it is extremely challenging to acquire high electrochemical performance owing to the limited electronic conductivity,sluggish ion kinetics,and severe volume expansion during the insertion/extraction process of Zn^(2+).Herein,a series of V_(2)O_(3)nanospheres embedded N-doped carbon nanofiber structures with various V_(2)O_(3)spherical morphologies(solid,core-shell,hollow)have been designed for the first time by an electrospinning technique followed thermal *** N-doped carbon nanofibers not only improve the electrical conductivity and the structural stability,but also provides encapsulating shells to prevent the vanadium dissolution and aggregation of V_(2)O_(3)***,the varied morphological structures of V_(2)O_(3)with abundant oxygen vacancies can alleviate the volume change and increase the Zn^(2+)***,the phase transition between V_(2)O_(3)and Zn_XV_(2)O_(5-m)·n H_(2)O in the cycling was also *** a result,the as-obtained composite delivers excellent long-term cycle stability and enhanced rate performance for coin cells,which is also confirmed through density functional theory(DFT)*** assembled into flexible ZIBs,the sample still exhibits superior electrochemical performance,which may afford new design concept for flexible cathode materials of ZIBs.

Thermally Conductive,Healable Glass Fiber Cloth Reinforced Polymer Composite based onβ-Hydroxyester Bonds Crosslinked Epoxy with Improved Heat Resistance

To simultaneously endow thermal conductivity,high glass transition temperature(Tg)and healing capability to glass fiber/epoxy(GFREP)composite,dynamic crosslinked epoxy resin bearing reversibleβ-hydroxyl ester bonds was reinforced with boron nitride nanosheets modified glass fiber cloth(GFC@BNNSs).The in-plane heat conduction paths were constructed by electrostatic self-assembly of polyacrylic acid treated GFC and polyethyleneimine decorated ***,the GFC@BNNSs were impregnated with the mixture of lower concentration(3-glycidyloxypropyl)trimethoxysilane grafted BN micron sheets,3,4-epoxycyclohexylmethyl 3,4-epoxycyclohexanecarboxylate and hexahydro-4-methylphthalic anhydride,which accounted for establishing the through-plane heat transport pathways and avoiding serious deterioration of mechanical *** resultant GFREP composite containing less boron nitride particles(17.6 wt%)exhibited superior in-plane(3.29 W·m^(-1)·K^(-1))and through-plane(1.16 W·m^(-1)·K^(-1))thermal conductivities,as well as high Tg of 204℃(Tg of the unfilled epoxy=177℃).The reversible transesterification reaction enabled closure of interlaminar cracks within the composite,achieving decent healing efficiencies estimated by means of tensile strength(71.2%),electrical breakdown strength(83.6%)and thermal conductivity(69.1%).The present work overcame the disadvantages of conventional thermally conductive composites,and provided an efficient approach to prolong the life span of thermally conductive GFREP laminate for high-temperature resistant integrated circuit application.

Self-Powered Flexible Multicolor Electrochromic Devices for Information Displays

The development of self-powered flexible multicolor electrochromic(EC)systems that could switch different color without an external power supply has remained extremely ***,a new trilayer film structure for achieving self-powered flexible multicolor EC displays based on self-charging/discharging mechanism is proposed,which is simply assembled by sandwiching an ionic gel film between 2 cathodic nickel hexacyanoferrate(NiHCF)and Prussian blue(PB)nanoparticle films on indium tin oxide *** display exhibits independent self-powered color switching of NiHCF and PB films with fast responsive time and high reversibility by selectively connecting the Al wire as anodes with the 2 EC *** switching is thus achieved through a color overlay effect by superimposing the 2 EC films,including green,blue,yellow,and *** bleaching/coloration process of the displays is driven by the discharging/self-charging mechanism for NiHCF and PB films,respectively,ensuring the self-powered color switching of the displays reversibly without an external power *** is further demonstrated that patterns can be easily created in the self-powered EC displays by the spray-coating method,allowing multicolor changing to convey specific ***,a self-powered ionic writing board is demonstrated based on the self-powered EC displays that can be repeatedly written freehand without the need of an external power *** believe that the design concept may provide new insights into the development of self-powered flexible multicolor EC displays with self-recovered energy for widespread applications.

Dynamic analysis of a latent HIV infection model with CTL immune and antibody responses

This paper develops a mathematical model to investigate the Human Immunodeficiency Virus(HIV)infection *** model includes two transmission modes(cell-to-cell and cell-free),two adaptive immune responses(cytotoxic T-lymphocyte(CTL)and antibody),a saturated CTL immune response,and latent HIV *** existence and local stability of equilibria are fully characterized by four reproduction *** sensitivity analyses,we assess the partial rank correlation coefficients of these reproduction numbers and identify that the infection rate via cell-to-cell transmission,the number of new viruses produced by each infected cell during its life cycle,the clearance rate of free virions,and immune parameters have the greatest impact on the reproduction ***,we compare the effects of immune stimulation and cell-to-cell spread on the model's *** findings highlight the significance of adaptive immune responses in increasing the population of uninfected cells and reducing the numbers of latent cells,infected cells,and ***,cell-to-cell transmission is identified as a facilitator of HIV *** analytical and numerical results presented in this study contribute to a better understanding of HIV dynamics and can potentially aid in improving HIV management strategies.

Inhibitory Effects of Cypermethrin on Interactions of the Androgen Receptor with Coactivators ARA70 and ARA55

Objective The purpose of this study was to investigate the anti-androgenic mechanism of cypermethrin involving *** Mammalian two-hybrid assays were performed to study the effects of cypermethrin on interactions of the androgen receptor(AR)with the coactivators androgen receptor-associated protein70(ARA70)and androgen receptor-associated protein 55(ARA55).Results The results showed that AR–ARA70 and AR–ARA55 interactions were remarkably enhanced by dihydrotestosterone(DHT,P≤0.05).Cypermethrin inhibited DHT-induced AR–ARA70 and AR–ARA55 interactions significantly(P≤0.05).Conclusion The study indicates that cypermethrin exhibits inhibitory effects on AR transcription associated with repression of AR–ARA70 and AR–ARA55 interactions in a ligand-dependent *** data show novel anti-androgenic mechanisms of cypermethrin that contribute to male reproductive toxicology.

Salinity effect-induced ENSO amplitude modulation in association with the interdecadal Pacific Oscillation

A 110-year ensemble simulation of an ocean general circulation model(OGCM)was analyzed to identify the modulation of salinity interdecadal variability on El Niño-Southern Oscillation(ENSO)amplitude in the tropical Pacific during *** simulating results show that sea surface salinity(SSS)variation in the region exhibits notable and coherent interdecadal variability signal,which is closely associated with the Interdecadal Pacific Oscillation(IPO).As salinity increases or reduces,the SSS modulations on ENSO amplitude during its warm/cold events vary asymmetrically with positive/negative IPO ***,salinity interdecadal variability can enhance or reduce ENSO-related conditions in upper-ocean stratification,contributing noticeably to ENSO *** anomalies associated with the mixed layer depth and barrier layer thickness can modulate ENSO amplitude during positive and negative IPO phases,resulting in the asymmetry of sea surface temperature(SST)anomaly in the tropical *** positive IPO phases,SSS interdecadal variability contributes positively to El Niño amplitude but negatively to La Niña amplitude by enhancing or reducing SSS interannual variability,and vice versa during negative IPO ***,the results indicate that the modulation of the ENSO amplitude by the SSS interdecadal variability is 15%-28%during negative IPO phases and 30%-20%during positive IPO phases,***,the SSS interdecadal variability associated with IPO and its modulation on ENSO amplitude in the tropical Pacific are among factors essentially contributing ENSO diversity.

Two-dimensional(2D)tungsten selenide(WSe_(2))is promising candidate material for future electronic applications,owing to its potential for ultimate device *** improving the electronic performance of WSe_(2)-based field-effect transistors(FETs),the modification of surface properties is *** this study,the seamless structural phase transition in WSe_(2) lattice is achieved by soft oxygen plasma,regulating the electrical conductance of WSe_(2)-based *** found that during the soft oxygen plasma treatment with optimal processing time,the generated oxygen ions can substitute some selenium atoms and thus locally modify the bond length,inducing 2H→1T phase transition in WSe_(2) with seamless *** mosaic structures have been proven to tailor the electronic structure and increase the hole carrier concentration inside WSe_(2),significantly increasing the channel conductance of WSe_(2) *** the further increase of the oxygen plasma treatment time,the creation of more selenium vacancy defects leads to the electronic doping,resulting in the reduction of *** from the hexagonal boron nitride(h-BN)encapsulation to interrupt the partial structural relaxation from 1T to 2H phase,our WSe_(2) FET exhibits high electronic stability with conductance of 6.8×10^(-4) S,which is about four orders of magnitude higher than 2H WSe_(2)(5.8×10^(-8) S).This study could further broaden the WSe_(2) FETs in applications for functionalization and integration in electronics.
