A pH/ROS dual-responsive system for effective chemoimmunotherapy against melanoma via remodeling tumor immune microenvironment

Chemotherapeutics can induce immunogenic cell death(ICD)in tumor cells,offering new possibilities for cancer ***,the efficiency of the immune response generated is insufficient due to the inhibitory nature of the tumor microenvironment(TME).Here,we developed a pH/reactive oxygen species(ROS)dual-response system to enhance chemoimmunotherapy for *** system productively accumulated in tumors by specific binding of phenylboronic acid(PBA)to sialic acids(SA).The nanoparticles(NPs)rapidly swelled and released quercetin(QUE)and doxorubicin(DOX)upon the stimulation of tumor microenvironment(TME).The in vitro and in vivo results consistently demonstrated that the NPs improved anti-tumor efficacy and prolonged survival of mice,significantly enhancing the effects of the *** study revealed DOX was an ICD inducer,stimulating immune responses and promoting maturation of dendritic cells(DCs).Additionally,QUE served as a TME regulator by inhibiting the cyclooxygenase-2(COX2)-prostaglandin E2(PGE2)axis,which influenced various immune cells,including increasing cytotoxic T cells(CLTs)infiltration,promoting M1 macrophage polarization,and reducing regulatory T cells(Tregs)*** combination synergistically facilitated chemoimmunotherapy efficacy by remodeling the immunosuppressive *** work presents a promising strategy to increase anti-tumor efficiency of chemotherapeutic agents.

Recent clinical studies have shown that mutation of phosphatase and tensin homolog deleted on chromosome 10(PTEN)gene in cancer cells may be associated with immunosuppressive tumor microenvironment(TME)and poor response to immune checkpoint blockade(ICB)***,efficiently restoring PTEN gene expression in cancer cells is critical to improving the responding rate to ICB ***,we screened an adeno-associated virus(AAV)capsid for efficient PTEN gene delivery into B16F10 tumor *** demonstrated that intratumorally injected AAV6-PTEN successfully restored the tumor cell PTEN gene expression and effectively inhibited tumor progression by inducing tumor cell immunogenic cell death(ICD)and increasing immune cell ***,we developed an anti-PD-1 loaded phospholipid-based phase separation gel(PPSG),which formed an in situ depot and sustainably release anti-PD-1 drugs within 42 days in *** order to effectively inhibit the recurrence of melanoma,we further applied a triple therapy based on AAV6-PTEN,PPSG^(@anti-PD-1)and CpG,and showed that this triple therapy strategy enhanced the synergistic antitumor immune effect and also induced robust immune memory,which completely rejected tumor *** anticipate that this triple therapy could be used as a new tumor combination therapy with stronger immune activation capacity and tumor inhibition efficacy.

A Polarization Control Operator for Polarized Electromagnetic Wave Designing

To describe and control the polarization state of electromagnetic waves, a polarization control operator of the complex vector form is proposed. Distinct from traditional descriptors, the proposed operator employs an angle parameter to configure the polarization state of the polarized wave. By setting the parameter in the proposed operator,the amplitude of the field components can be modified, resulting in changes in the magnitude and direction of the field vector, and thus realizing control of the polarization state of the electromagnetic wave. The physical meaning,orthogonal decomposition, and discrete property of the proposed operator are demonstrated through mathematical derivation. In the simulation examples, the polarization control operator with fixed and time-varying parameters is applied to the circularly polarized wave. The propagation waveform, the trajectory projection and the waveform cross section in different reception directions of the new electromagnetic waves are observed. The simulation results indicate that complex electromagnetic waves with more flexible polarization states can be obtained with the aid of the polarization operator.

Effects of the fertilizer and water management on amino acids and volatile components in Cabernet Sauvignon grapes and wines

The amino acids and the volatile substances in grapes and wines play important roles in their quality,and the concentrations of these substances can be changed by how a vineyard is managed,e.g.,irrigation and fertigation *** study aimed to evaluate the effect of fertilizer and water management on the distribution of amino acids,the volatile component profiles,and the sensory characteristics of Cabernet Sauvignon grapes and *** results showed that the amino acid concentration in grape berries was the highest under the 100%local fertilizer rate(HF)and 100%water irrigation quota(HW)treatment,and the volatile component concentration in wine was the highest under HF and 80%water irrigation quota(MW)*** effect of irrigation on the amino acid content in grapes was greater than that of *** synergistic effect of fertilizer and water on arginine,serine,and glutamine in grape berries was *** interactive effect of fertigation and irrigation on the volatile substance in grapes was greater than that of fertigation and irrigation *** influence of irrigation on volatile substances in wines was greater than that of *** addition,there was also a correlation between the concentrations of multiple amino acids in grapes and volatile components in *** component analysis showed that the wine from the HFMW treatment had the best quality among all treatments.

Owing to the distinctive structural characteristics,vanadium nitride(VN)is highly regarded as a catalyst for oxygen reduction reaction(ORR)in zinc-air batteries(ZABs).However,VN exhibits limited intrinsic ORR activity due to the weak adsorption ability to O-containing ***,the S-doped VN anchored on N,S-doped multi-dimensional carbon(S-VN/Co/NS-MC)was constructed using the solvothermal and in-situ doping *** sulfur atoms into VN species alters the electron spin state of vanadium in the S-VN/Co/NS-MC for regulating the adsorption energy of vanadium sites to oxygen *** introduced sulfur atoms polarize the V 3d_(z)^(2) electrons,shifting spin-down electrons closer to the Fermi level in the S-VN/Co/***,the introduction of sulfur atoms into VN species enhances the adsorption energy of vanadium sites for oxygen ****OOH dissociation transitions from being unspontaneous on the VN surface to a spontaneous state on the S-doped VN ***,the ORR barrier on the S-VN/Co/NS-MC surface is *** S-VN/Co/NS-MC demonstrates a higher half-wave potential and limiting current density compared to the VN/Co/*** S-VN/Co/NS-MC-based liquid ZABs display a power density of 195.7 m W cm^(-2),a specific capacity of 815.7 m A h g^(-1),and a cycling stability exceeding 250 *** S-VN/Co/NS-MC-based flexible ZABs are successfully employed to charge both a smart watch and a mobile *** approach holds promise for advancing the commercial utilization of VN-based catalysts in ZABs.

Random Terpolymer Based on Simple Siloxane-functionalized Thiophene Unit Enabling High-performance Non-fullerene Organic Solar Cells

Incorporation of siloxane-functionalized units into polymers backbone has proven to be an efficient strategy to improve photovoltaic performance. In this work, a low-cost siloxane-containing unit was developed to construct a series of terpolymers, and the effects of siloxane on the polymer performance were systematically studied. Different contents of thiophene containing siloxane-functionalized side chain were introduced into PM6 to obtain a series of polymers(PM6, PM6-SiO-10, PM6-SiO-20 and PM6-SiO-30). The siloxane-functionalized side chains in polymers have only a slight effect on the absorption behavior and frontier molecular orbitals. However, when the siloxane content increased, the terpolymers' aggregation property decreased and the temperature-dependency increased, leading to improved donor-acceptor compatibility. The power conversion efficiency(PCE) based on PM6:Y6, PM6-SiO-20:Y6 and PM6-SiO-30:Y6 devices was 15.64%, 16.03% and 15.82%, respectively. In comparison, the active layer based on PM6-SiO-10:Y6 exhibits the most appropriate phase separation morphology, resulting in effective exciton dissociation, more balanced hole-electron transport and less recombination. Consequently, the highest PCE of 16.69% with an outstanding shortcircuit current density of 26.96 mA·cm^(-2) was obtained, which are one of the highest values for siloxane-functionalized polymer-based *** work demonstrates that finely controlling the content of siloxane-functionalized thiophene is beneficial for obtaining high-performance terpolymer donors and provides a novel and low-cost method to improve photovoltaic performance.

Dietary copper requirement of broilers fed a corn-soybean meal diet during 22—42 d of age

This research was to assess the dietary copper(Cu) requirement of broiler chickens fed a practical cornsoybean meal diet during 22—42 d of age. A total of 288 numbered Arbor Acres male broilers at 22 d of age were randomly allotted 6 treatments with 8 replicate cages(6 broilers per cage) per *** were fed a Cu-unsupplemented corn-soybean meal basal diet(control, containing 7.36 mg Cu/kg) or the basal diet added with 3, 6, 9, 12, or 15 mg Cu/kg from CuSO4·5H2O for 21 d. Quadratic,asymptotic and broken-line models were fitted and the best fitted models were selected to determine dietary Cu requirements. The results revealed that the contents of Cu in serum and liver, m RNA expression levels of Cu-and zinc-containing superoxide dismutase(CuZnSOD) in liver and monoamine oxidase b(MAO B) in heart, as well as protein expression level of CuZnSOD in liver were affected(P bove indices varied linearly and quadratically(P bove indexes were 10.45—13.81 mg/kg. It was concluded that mRNA expression levels of CuZnSOD in liver and MAO B in heart, as well as liver CuZnSOD protein expression level were new specific sensitive biomarkers for estimating dietary Cu requirements, and the dietary Cu requirement was recommended to be 14 mg/kg to support Cu metabolic needs related to key Cucontaining enzymes in broilers fed the corn-soybean meal diet during 22—42 d of age, which was higher than the dietary Cu requirement(8 mg/kg) for broilers at the corresponding stage suggested by the Chinese Feeding Standard of Chicken.

Insight into the microbial community of denitrification process using different solid carbon sources: Not only bacteria

There is a lack of understanding about the bacterial,fungal and archaeal communities’composition of solid-phase denitrification(SPD)*** investigated four SPD systems with different carbon sources by analyzing microbial gene sequences based on operational taxonomic unit(OTU)and amplicon sequence variant(ASV).The results showed that the corncob-polyvinyl alcohol sodium alginate-polycaprolactone(CPSP,0.86±0.04 mg NO_(3)^(−)-N/(g·day))and corncob(0.85±0.06 mg NO_(3)^(−)-N/(g·day))had better denitrification efficiency than polycaprolactone(PCL,0.29±0.11 mg NO_(3)^(−)-N/(g·day))and polyvinyl alcoholsodium alginate(PVA-SA,0.24±0.07 mg NO_(3)^(−)-N/(g·day)).The bacterial,fungal and archaeal microbial composition was significantly different among carbon source types such as Proteobacteria in PCL(OTU:83.72%,ASV:82.49%)and Rozellomycota in PVA-SA(OTU:71.99%,ASV:81.30%).ASV methods can read more microbial units than that of OTU and exhibit higher alpha diversity and classify some species that had not been identified by OTU such as Nanoarchaeota phylum,unclassified_f_Xanthobacteraceae genus,etc.,indicating ASV may be more conducive to understand SPD microbial *** co-occurring network showed some correlation between the bacteria fungi and archaea species,indicating different species may collaborate in SPD *** KEGG function prediction results were obtained in two bioinformatic methods generally and some fungi and archaea functions should not be ignored in SPD *** results may be beneficial for understanding microbial communities in SPD systems.
