Fluoropyrimidine-based combination chemotherapy plus targeted therapy is the standard initial treatment for unresectable metastatic colorectal cancer(mCRC),but the prognosis remains *** phase 3 trial(***:NCT039501.4)assessed the efficacy and adverse events(AEs)of the combination of PD-1.blockade-activated DC-CIK(PD1.T)cells with XELOX plus bevacizumab as a first-line therapy in patients with mCRC.A total of 2.2.participants were enrolled and randomly assigned in a 1.1.ratio to receive either first-line XELOX plus bevacizumab(the control group,n=1.2.or the same regimen plus autologous PD1.T cell immunotherapy(the immunotherapy group,n=1.0)every 2. days for up to 6 cycles,followed by maintenance treatment with capecitabine and *** main endpoint of the trial was progression-free survival(PFS).The median follow-up was 1..5 *** PFS was 1..8 months(95%CI,1..6-1..0)for the immunotherapy group compared with 9.9 months(8.0-1..8)for the control group(hazard ratio[HR],0.60[95%CI,0.40-0.88];p=0.009).Median overall survival(OS)was not reached for the immunotherapy group and 2..6 months(95%CI,1..3-32.8)for the control group(HR,0.57[95%CI,0.33-0.98];p=0.043).Grade 3 or higher AEs occurred in 2..0%of patients in the immunotherapy group and 2..5%in the control groups,with no toxicity-associated deaths *** addition of PD1.T cells to first-line XELOX plus bevacizumab demonstrates significant clinical improvement of PFS and OS with well tolerability in patients with previously untreated mCRC.

The interacting boson model with isospin (IBM-3) was applied to study the band structure and electromagnetic transition properties of the low-lying states in the even-even N = Z nucleus 5{1.e. The isospin excitation states with T = 0, 1.and {1.were identified, and compared with the available data. The study shows that the {1.3 state is the lowest mixed symmetry state in 5{1.e. The excitation energy of the second 0+{1.state with T = 0 in nucleus 5{1.e was identified. The model calculations with the data show a reasonably good agreement.

Since the outbreak of the coronavirus disease 2.1.(COVID-1.)epidemic in 2.1.,the public health system has faced enormous *** the individuals who test positive for severe acute respiratory syndrome coronavirus 2.SARS-CoV-2.is a key step for interrupting chains of transmission of SARS-CoV-2.and reducing COVID-1.-associated *** the increasing of asymptomatic infections,it is difficult to track asymptomatic infections through epidemiological surveys and virus whole-genome ***,due to the cross-reactivity of neutralizing antibodies produced by multiple virus subtypes,neutralizing antibody detection cannot be used to determine whether an individual has a history of infection with a specific subtype of *** recruited 4 human leukocyte antigen A2.HLA-A2.infections,1. individuals who received three doses of inactivated vaccines,and 30 breakthrough infections after vaccination and discussed a case-tracking approach to detect epitope-specific CD8^(+)T cells in the peripheral blood of close contacts,including accurate HLA typing based on ribonucleic acid(RNA)-sequencing and flow cytometry data and the comparison and characterization of SARS-CoV-2.HLA-A2.and HLA-A2. epitope-specific CD8^(+)T *** individuals who received three doses of inactivated vaccine,we observed that the CD8^(+)T cell specificity for ancestral epitopes was significantly higher than for mutated epitopes,and the fold change of CD8^(+)T cells corresponding to mutated epitopes relative to ancestral epitopes was less than *** enzyme-linked immunospot(ELISpot)results further validate this *** study forms a“method for understanding the infection history of SARS-CoV-2.subtypes based on the proportion of epitope-specific CD8^(+)T cells in the peripheral blood of subjects”,covering up to 46%of the population,including HLA-A2.and HLA-A2.+donors,providing a novel method for SARS-CoV-2.infected case tracing.

For decades micoorganisms have been engineered for the utilization of lignocellulose-based second-generation (2.) feedstocks, but with theconcerns of increased levels of atmospheric CO_(2. causing global warming there is an emergent need to transition from the utilization of 2.feedstocks to third-generation (3G) feedstocks such as CO_(2. and its derivatives. Here, we established a yeast platform that is capable ofsimultaneously converting 2. and 3G feedstocks into bulk and value-added chemicals. We demonstrated that by adopting 3G substrates such asCO_(2. and formate, the conversion of 2. feedstocks could be substantially improved. Speciﬁcally, formate could provide reducing power andenergy for xylose conversion into valuable chemicals. Simultaneously, it can form a concentrated CO_(2. pool inside the cell, providing thermodynamically and kinetically favoured amounts of precursors for CO_(2. ﬁxation pathways, e.g., the Calvin–Benson–Bassham (CBB) ***, we demonstrated that formate could directly be utilized as a carbon source by yeast to synthesize endogenous amino acids. Theengineered strain achieved a one-carbon (C1) assimilation efﬁciency of 9.2., which was the highest efﬁciency observed in the co-utilization of2. and 3G feedstocks. We applied this strategy for productions of both bulk and value-added chemicals, including ethanol, free fatty acids(FFAs), and longifolene, resulting in yield enhancements of 18.4%, 49.0%, and ~100%, respectively. The strategy demonstrated here for coutilization of 2. and 3G feedstocks sheds lights on both basic and applied research for the up-coming establishment of 3G bioreﬁneries.

对软沉积物变形构造的形成过程解析和触发因素识别一直是国内外事件沉积学研究的重点和难点。国内外学者常将地层中保存的软沉积物变形构造的形成归因于地震作用,但缺乏足够的证据来支撑地震震动触发相应的沉积过程与变形机制。由于软沉积物变形构造可以由地震、风暴和非地震参与的液化作用、重力作用及滑坡等因素触发,且可能受瑞利—泰勒不稳定性(因密度差异沿垂向变形)或开尔文—亥姆获兹不稳定性(沿水平方向变形)机制的控制,软沉积物变形构造本身并不能作为特定触发因素的判别标志。此前,通过解析事件层组合特征来揭示与软沉积物变形构造形成相关的沉积过程和变形机制,进而限定变形构造触发因素的方法已成功应用于中东地区死海盆地(死海断裂带)的事件沉积研究中。尝试应用此方法来解析灵山岛灯塔剖面底部软沉积物变形构造的变形机制与触发因素,研究发现灯塔剖面底部的软沉积物变形构造是原位形成与保存的,并被浊流沉积层上覆,且二者之间无背景沉积物。这种独特的事件层组合指示原位变形和异地搬运两种水下沉积过程准同期发生,而能够同时激发这两类物质来源与沉积过程迥异的事件沉积响应的最可能因素是区域强震震动。结合灵山岛研究案例认为,前人所做的模式化的事件沉积成因判别标志不宜直接套用,而控制事件沉积的沉积过程与物理机制具有一定的普适性,应该是事件沉积学研究的关键。
