Development of RPA-Cas12a-fluorescence assay for rapid and reliable detection of human bocavirus 1

Human bocavirus(HBoV)1 is considered an important pathogen that mainly affects infants aged 6–24 months,but preventing viral transmission in resource-limited regions through rapid and affordable on-site diagnosis of individuals with early infection of HBoV1 remains somewhat challenging.Herein,we present a novel faster,lower cost,reliable method for the detection of HBoV1,which integrates a recombinase polymerase amplification(RPA)assay with the CRISPR/Cas12a system,designated the RPA-Cas12a-fluorescence assay.The RPA-Cas12a-fluorescence system can specifically detect target gene levels as low as 0.5 copies of HBoV1 plasmid DNA per microliter within 40 min at 37℃without the need for sophisticated instruments.The method also demonstrates excellent specificity without cross-reactivity to non-target pathogens.Furthermore,the method was appraised using 28 clinical samples,and displayed high accuracy with positive and negative predictive agreement of 90.9%and 100%,respectively.Therefore,our proposed rapid and sensitive HBoV1 detection method,the RPA-Cas12a-fluorescence assay,shows promising potential for early on-site diagnosis of HBoV1 infection in the fields of public health and health care.The established RPA-Cas12a-fluorescence assay is rapid and reliable method for human bocavirus 1 detection.The RPA-Cas12a-fluorescence assay can be completed within 40 min with robust specificity and sensitivity of 0.5 copies/μl.

The RFI Fast Mitigation Algorithm Based on Block LMS Filter

The radio telescope possesses high sensitivity and strong signal collection capabilities.While receiving celestial radiation signals,it also captures Radio Frequency Interferences(RFIs)introduced by human activities.RFI,as signals originating from sources other than the astronomical targets,significantly impacts the quality of astronomical data.This paper presents an RFI fast mitigation algorithm based on block Least Mean Square(LMS)algorithm.It enhances the traditional adaptive LMS filter by grouping L adjacent time-sampled points into one block and applying the same filter coefficients for filtering within each block.This transformation reduces multiplication calculations and enhances algorithm efficiency by leveraging the time-domain convolution theorem.The algorithm is tested using baseband data from the Parkes 64 m radio telescope's pulsar observations and simulated data.The results confirm the algorithm's effectiveness,as the pulsar profile after RFI mitigation closely matches the original pulsar profile.

In this study,online water-soluble inorganic ions were detected to deduce the formation mechanism of secondary inorganic aerosol in Xianyang,China during wintertime.The dominant inorganic ions of sulfate(SO_(4)^(2-)),nitrate(NO_(3)^(-)),and ammonium(NH_(4)^(+))(the sum of those is abbreviated as SNA)accounted for 17%,21%,and 12% of PM_(2.5)mass,respectively.While the air quality deteriorated from excellent to poor grades,the precursor gas sulfur dioxide(SO_(2))of SO_(4)^(2-)increased and then decreased with a fluctuation,while nitrogen dioxide(NO_(2))and ammonia(NH_(3)),precursors of NO_(3)^(-)and NH_(4)^(+),and SNA show increasing trends.Meteorological factors including boundary layer height(BLH),temperature,and wind speed also show decline trends,except relative humidity(RH).Meanwhile,the secondary conversion ratio shows a remarkable increasing trend,indicating that there was a strong secondary transformation.From the perspective of chemical mechanisms,RH is positively correlated with sulfur oxidation ratios(SOR),nitrogen oxidation ratios(NOR),and ammonia conversion ratios,representing that the increase of humidity could promote the generation of SNA.Notably,SOR and NOR were also positively related to the ammonia.On the one hand,the low wind speed and BLH led to the accumulation of pollutants.On the other hand,the increases of RH and ammonia promoted more formations of SNA and PM_(2.5).The results advance our identification of the contributors to the haze episodes and assist to establish more efficient emission controls in Xianyang,in addition to other cities with similar emission and geographical characteristics.

Design study of charge-stripping scheme of heavy ion beams for HIAF-BRing

Charge strippers play an essential role in heavy-ion accelerators by stripping the projectile ions to higher charge states to enhance the acceleration efficiency downstream of the stripper.In the high-energy mode of the booster ring(BRing)of the high-intensity heavy-ion accelerator facility,the pre-accelerated ions from the iLinac will be stripped by a carbon foil to higher charge states and then injected into the BRing.The key parameters of the stripper and stripped ions were calculated,and the influence of stripping on the beam quality was discussed.To get high stripping efficiencies,the foil thicknesses and resultant charge state distributions for the typical ions were determined by the code ETACHA.The equilibrium thickness was obtained for the U beam,while the stripper thicknesses for the Xe and Kr beams were determined based on a compromise between the stripped charge states and the stripping efficiency.The energy loss,energy straggling,and emittance growth due to stripping have a non-negligible impact on the transport of the stripped beams and the injection of the ring.Therefore,these parameters were simulated by GEANT4.In addition,the foil’s temperature evolution,which greatly affects the foil lifetime,was simulated by ANSYS.The maximum temperature of the foil bombarded by the U and Xe beams with the nominal parameters will exceed the safe value in terms of the impact of evaporation on the foil’s lifetime.Given the foil temperature constraint,the highest tolerable beam intensity and the injected ion number into the ring were derived for different beam sizes.The results of this paper will present important reference data for the optimization design and commissioning of the beamline and injection to the BRing for the stripped ions.

Impact pressure of waves generated by landslides on bank slopes

Impulse waves that are generated by landslides in narrow reservoir areas threaten the stability of buildings and bank slopes.To discuss the action process and evolution law of the wave pressure on bank slopes,a three-dimensional physical model test that considers impulse waves generated by landslides was performed,and factors including landslide width,thickness,slope angles of the sliding surface,and bank slope angle were considered.Based on wave forms on the bank slopes,wave pressure curve characteristics,and peak value,the action process of wave pressure could be divided into the following stages:maximum pulsating pressure stage,wave impact stage(when waves break),and stationary pulsation stage.It was found that wave breaking is dependent on the value of the surf similarity parameterξ.The distribution pattern of impact pressure decays linearly on both sides of the maximum impact pressure point,and the attenuation degree decreases when it attains 40%of the maximum value.Thus,it is proposed that the prediction formula for the maximum effective impact pressure of the bank slope be related to the reciprocal of wave steepness,relative water depth,and slope rate.The prediction formula provides strong theoretical support for early safety warning and for predicting the bank slope under impulse waves generated by landslides.

Snap-through behaviors and nonlinear vibrations of a bistable composite laminated cantilever shell:an experimental and numerical study

The snap-through behaviors and nonlinear vibrations are investigated for a bistable composite laminated cantilever shell subjected to transversal foundation excitation based on experimental and theoretical approaches.An improved experimental specimen is designed in order to satisfy the cantilever support boundary condition,which is composed of an asymmetric region and a symmetric region.The symmetric region of the experimental specimen is entirely clamped,which is rigidly connected to an electromagnetic shaker,while the asymmetric region remains free of constraint.Different motion paths are realized for the bistable cantilever shell by changing the input signal levels of the electromagnetic shaker,and the displacement responses of the shell are collected by the laser displacement sensors.The numerical simulation is conducted based on the established theoretical model of the bistable composite laminated cantilever shell,and an off-axis three-dimensional dynamic snap-through domain is obtained.The numerical solutions are in good agreement with the experimental results.The nonlinear stiffness characteristics,dynamic snap-through domain,and chaos and bifurcation behaviors of the shell are quantitatively analyzed.Due to the asymmetry of the boundary condition and the shell,the upper stable-state of the shell exhibits an obvious soft spring stiffness characteristic,and the lower stable-state shows a linear stiffness characteristic of the shell.

Roles of nanoparticles in arsenic mobility and microbial community composition in arsenic-enriched soils

Environmental effects of nano remediation engineering of arsenic(As)pollution need to be considered.In this study,the roles of Fe_(2)O_(3)and TiO_(2)nanoparticles(NPs)on the microbial mediated As mobilization from As contaminated soil were investigated.The addition of Fe_(2)O_(3)and TiO_(2)NPs restrained As(V)release,and stimulated As(Ⅲ)release.As(V)concentration decreased by 94% and 93% after Fe_(2)O_(3)addition,and decreased by 89% and 45% after Ti O_(2)addition compared to the Biotic and Biotic+Acetate(amended with sodium acetate)controls,respectively.The maximum values of As(Ⅲ)were 20.5 and 27.1μg/L at 48 d after Fe_(2)O_(3)and TiO_(2)NPs addition,respectively,and were higher than that in Biotic+Acetate control(12.9μg/L).The released As co-precipitated with Fe in soils in the presence of Fe_(2)O_(3)NPs,but adsorbed on TiO_(2)NPs in the presence of TiO_(2)NPs.Moreover,the addition of NPs amended with sodium acetate as the electron donor clearly promoted As(V)reduction induced by microbes.The NPs addition changed the relative abundance of soil bacterial community,while Proteobacteria(42.8%-70.4%),Planctomycetes(2.6%-14.3%),and Firmicutes(3.5%-25.4%)were the dominant microorganisms in soils.Several potential As/Fe reducing bacteria were related to Pseudomonas,Geobacter,Desulfuromonas,and Thiobacillus.The addition of Fe_(2)O_(3)and TiO_(2)NPs induced to the decrease of arr A gene.The results indicated that the addition of NPs had a negative impact on soil microbial population in a long term.The findings offer a relatively comprehensive assessment of Fe_(2)O_(3)and TiO_(2)NPs effects on As mobilization and soil bacterial communities.

PSRDP:A Parallel Processing Method for Pulsar Baseband Data

To address the problem of real-time processing of ultra-wide bandwidth pulsar baseband data,we designed and implemented a pulsar baseband data processing algorithm(PSRDP)based on GPU parallel computing technology.PSRDP can perform operations such as baseband data unpacking,channel separation,coherent dedispersion,Stokes detection,phase and folding period prediction,and folding integration in GPU clusters.We tested the algorithm using the J0437-4715 pulsar baseband data generated by the CASPSR and Medusa backends of the Parkes,and the J0332+5434 pulsar baseband data generated by the self-developed backend of the Nan Shan Radio Telescope.We obtained the pulse profiles of each baseband data.Through experimental analysis,we have found that the pulse profiles generated by the PSRDP algorithm in this paper are essentially consistent with the processing results of Digital Signal Processing Software for Pulsar Astronomy(DSPSR),which verified the effectiveness of the PSRDP algorithm.Furthermore,using the same baseband data,we compared the processing speed of PSRDP with DSPSR,and the results showed that PSRDP was not slower than DSPSR in terms of speed.The theoretical and technical experience gained from the PSRDP algorithm research in this article lays a technical foundation for the real-time processing of QTT(Qi Tai radio Telescope)ultra-wide bandwidth pulsar baseband data.

BACKGROUND This study aimed to explore the possible etiology and treatment of severe fetal tachycardia in the absence of organic disease and provide a reference for clinical management of severe fetal tachycardia.CASE SUMMARY A 29-year-old pregnant woman,with a gravidity 1 parity 0,presented with a fetal heart rate(FHR)of 243 beats per minute during a routine antenatal examination at 31+2 wk of gestation.Before termination of pregnancy at 38 wk of gestation,the FHR repeatedly showed serious abnormalities,lasting more than 30 min.However,the pregnant woman and the fetus had no clinical symptoms,and repeated examination revealed no organic lesions.The mother and the baby were regularly followed up.CONCLUSION This was a case of severe fetal tachycardia with no organic lesions and management based on clinical experience.

Changes in extreme high temperature warning indicators over China under different global warming levels

High temperature warning indicators play a pivotal role in meteorological departments,serving as crucial criteria for issuing warnings that guide both social production and daily life.Despite their importance,limited studies have explored the relationship between different global warming levels and changes in high temperature warning indicators.In this study,we analyze data from 2,419 meteorological stations over China and utilize the Coupled Model Intercomparison Project Phase 6(CMIP6)models to examine historical changes in high temperature warning indicators used by the China Meteorological Administration.We evaluate model performance and estimate future changes in these indicators using an annual cycle bias correction method.The results indicate that since 1961,the number of high temperature days(TX35d and TX40d)and length of season(TX40d and TX40l)with daily maximum temperature reaching or exceeding 35℃ and 40℃ have increased over China.The intensity of high temperatures(TXx)has strengthened and the geographical extent affected by high temperatures has expanded.In 2022,the occurrence of 40℃ high temperatures surges,with Eastern China experiencing a two-day increase in TX40d and an extended seasonal length in TX40l by over five days.While CMIP6 models have underestimated the high temperature indictors associated with 35℃ during historical periods,notable difference is not observed between the models and observations for TX40d and TX40l,given their rare occurrence.However,future projections,after bias correction,indicate that the increasing trends for 35℃ and 40℃ high temperature days and length of season become more pronounced than the raw projection,suggesting a more severe increase than that anticipated originally.As global warming intensifies,the high temperature days and length of season are projected to increase non-linearly,while the intensity of high temperatures is expected to increase linearly.For every 1℃ increase in global temperature,the intensity is projected to rise by approximately 1.4℃.The impa
