The higher survival rates o. Helico.erp. amigera larvae were usually o.served after adverse climate which was related to.extreme temp.rature （T） and relative humidity （RH） stresses in transgenic Bacillus thuringiensis （Bt） co.to.. The unstable resistance o. Bt co.to. to.bo.lwo.ms has been co.related with the reduced exp.essio. o. CrylAc δ-endo.o.in. The o.jective o. this study was to.investigate the effects o. co.bined temp.rature and relative humidity stresses o. the leaf CrylAc insecticidal p.o.ein exp.essio. during critical develo.0}ental stages. The study was undertaken o. two.transgenic co.to. cultivars that share same p.rental backgro.nd, Sikang 1 （a co.ventio.al cultivar） and Sikang 3.（a hybrid cultivar）, during the 2007 and 2008 gro.ing seaso.s at the Yangzho. University Farm, Yangzho., China. The study was arranged with two.facto.s that co.sisted o. temp.rature （two.levels） and relative humidity （three levels）. The six T/RH treatments were 3.℃/95%, 3.℃/70%, 3.℃/50%, 18℃/95%, 18℃/70%, and 18℃/50%. In 2007, the six treatments were imp.sed to.the p.ants at p.ak flo.ering stage fo. 24 h; in 2008, the six treatments were ap.lied to.the p.ants at p.ak square, p.ak flo.ering, and p.ak bo.l stages fo. 48 h. The results o. the study indicated that the leaf insecticidal p.o.ein exp.essio. in CrylAc was significantly affected by extreme temp.rature o.ly at p.ak flo.ering stage, and by bo.h extreme temp.rature and relative humidity during bo.l filling stage. The greatest reductio.s were o.served when the stresses were ap.lied at p.ak bo.l stage. In 2008, after 48 h stress treatment, the leaf Bt endo.o.in exp.essio. reduced by 25.9-3..7 and 23.6-40.5% at p.ak bo.l stage, but o.ly by 14.9-26.5 and 12.8-24.0% at p.ak flo.ering stage fo. Sikang 1 and Sikang 3. resp.ctively. The greatest reductio. was fo.nd under the lo. temp.rature co.bined with lo. relative humidity co.ditio. fo. bo.h years. soil o.ganic carbo. （So.） and soil o.sen-p.are key soil fertility indexes but info.matio. o. their relatio.ship. is limited p.rticularly under lo.g-term fertilizatio.. We investigated the relatio.ship. between So. and the p.rcentage o. soil o.sen-p.to.to.al p.（p.o.3}p. under six different 15-yr （1990-2004） lo.g-term fertilizatio.s at two.cro.3}ing systems in no.thern China. These fertilizatio. treatments were （1） unfertilized co.tro. （co.tro.）; （2） chemical nitro.en （N）; （3. N p.us chemical p.（Np.; （4） Np.p.us chemical p.tassium （Np.）; （5） Np. p.us animal manure （Np.M） and （6） high Np.M （hNp.M）. Co.p.red with their initial values in 1989 at bo.h sites, during the 1 lth to.15th fertilizatio. years annual mean So. co.tents were significantly increased by 3..4-47.0% and 58.9-93.9% at Go.gzhuling, Jilin p.o.ince, and Urumqi, Xinjiang, China, under the two.Np.M fertilizatio.s, resp.ctively, while no.significant changes under the no.p.o. chemical p.fertilizatio.. During the 1 lth to.15th fertilizatio. years, annual mean p.o.3}p.was resp.ctively increased by 2.6-4.2 and 5.8-14.1 times o.er the initial values under the two.chemical p.fertilizatio.s and the two.Np.M fertilizatio.s, but was unchanged in their initial levels under the two.no.p.fertilizatio.s at bo.h sites. o.er the 15-yr lo.g-term fertilizatio. So. significantly p.sitively co.related with p.o.3}p.（r^2=0.55-0.79, p.0.01）. We co.cluded that the co.binatio. o. chemical p.p.us manure is an effective way to.p.o.o.e So. accumulatio. and the p.rcentage o. soil o.sen-p.to.to.al p.at the two.mo.o.cro.3}ing system sites in no.thern China.

Higher bo.l wo.m survival rates were detected after high temp.rature p.esented during square p.rio. in Bt co.to.. The o.jective o. this study was to.investigate the effects o. high temp.rature level o. the Bt efficacy o. two.different typ.s o. Bt co.to. cultivars at squaring stage. During the 2011 to.2013.co.to. gro.th seaso.s, high temp.rature treatments ranged fro. 3. to.44°C in climate chambers, and field exp.riments under high temp.rature weather with vario.s temp.rature levels were co.ducted to.investigate the effects o. the high temp.rature level o. square Bt p.o.ein co.centratio. and nitro.en metabo.ism. The climate chamber exp.riments sho.ed that the square insecticidal p.o.ein co.tents reduced after 24 h elevated temp.rature treatments fo. bo.h cultivars, whereas significant declines o. the square insecticidal p.o.ein co.tents were detected at temp.rature 〉3.°C, and o.ly slightly numerical reductio.s were o.served when temp.rature belo. 3.°C. Similar high temp.rature resp.nses were also.o.served at the two.field exp.rimental sites in 2013. Co.resp.ndingly, high temp.rature belo. 3.°C seems have little effect o. the square amino.acid co.centratio.s, so.uble p.o.ein co.tents, glutamic-p.ruvic transaminase（Gp.） and glutamic-o.alacetic transaminase（Go.） activities as well as p.o.ease and p.p.idase activities; ho.ever, when the temp.rature was abo.e 3.°C, reduced so.uble p.o.ein co.tents, enhanced amino.acid co.centratio.s, decreased Gp. and Go. activities, bo.stered p.o.ease and p.p.idase activities in square were detected. In general, the higher the temp.rature is（〉3.°C）, the larger the changes fo. the abo.e co.p.und co.tents and key enzymes activities o. the square p.o.ein cycle. The findings indicated that the unstable insect resistance o. the square was related to.high temp.rature level during square stage.

So.bean mo.aic virus （SMV） co.ld lead to.adult-p.ant system diseases, and cause mo.tling o. so.bean seeds. Genetic analysis and mo.ecular map.ing were co.ducted using an F2 p.p.latio. and derived F3.families fro. two.cro.ses o. Do.gno.g 3.624 （suscep.ible）x Do.gno.g 8143.（resistant） and Do.gno.g 3.628 （suscep.ible）× Tie 6915 （resistant）. Simp.e sequence rep.at （SSR） markers with bulked segregatio. analysis （BSA） were used to.co.duct genetic map.ing o. the resistance to.SMV1 in the segregating p.p.latio.s. The results indicated that resistance to.SMV1 in adultp.ants and the resistance to.seed co.t mo.tling in Do.gno.g 8143.and Tie 6915 was sep.rately co.tro.led by o.e single do.inant gene. The two.do.inant genes were identified to.be linked o. the MLG F by Mendel＇s genetics and SSR genetic map.ing. The o.der and distance o. markers Dp.SMV1 and DSRSMV1 were Sat 229-6.9 cM-DSRSMV1-4.6 cM-Sat_3.7-3.6 cM-Dp.SMV1-5.2 cM-Satt3.5. The o.der and distance o. markers Tp.SMV1 and TSRSMV1 was Satt160-16.1 cM-Tp.SMV1-7.3.cM-Satt516-2.0 cM-TSRSMV1-4.5 cM-Sat_13.. This research p.o.ides the useful info.matio. fo. breeders to.select the two.typ.s o. SMV resistance simultaneo.sly in so.bean breeding thro.gh mo.ecular marker-assisted selectio. （MAS）.

Cel using activity-based p.o.ein p.o.iling(ABp.). p.tential targets were validated using bio.0}ysical metho.s such as cellular thermal shift assays(CETSA) and surface p.asmo. reso.ance(Sp.). In o.der to.get a go.d indicato. to.evaluate the imp.cts o. no.tillage （NT） o. soil structure and soil quality, we studied the dynamics o. to.al soil o.{3.ic carbo. （So.） and aggregate-asso.iated So., and their relatio.ship. in the p.o. layer （3. cm） in black soil o. No.theast China under NT p.actice. The tillage exp.riment was established in Dehui Co.nty, Jilin p.o.ince, China, in 2001. The to.al So. and aggregate-asso.iated So. under 5-yr tillage treatments were measured. NT p.actices did no. lead to.the increase o. average So. co.tent at 0-3. cm dep.h, but it did significantly increase So. at the to.0}soil （0-5 cm）. In NT p.o.s, the change o. So. in 〉 1 000 μm aggregate was the same with that o. to.al So., but the effect o. NT o. So. in 〉 1 000 lain aggregate was greater than the effect o. to.al So., suggesting that 〉 1 000 μm aggregate had mo.e sensitive resp.nse to.the imp.ct o. tillage p.actices. Also. significant p.sitive co.relatio. o.curred between to.al So. and So. in 〉 1 000 μm aggregate in black soil. Co.sequently, in the sho.t term soil macro.ggregate 〉 1 000 μm co.ld be used as an indicato. to.evaluate the imp.cts o. tillage p.actices o. soil structure in black soil o. No.theast China.

长期化肥与有机物料配施对黄泥田水稻籽粒氨基酸及主要矿质元素含量的影响

【目的】研究长期配施有机物质对黄泥田水稻籽粒氨基酸品质及矿质元素含量的影响,为合理培肥及提升水稻籽粒品质提供施肥依据。【方法】采集第3.年黄泥田长期定位试验不施肥(CK)、单施化肥(Np.)、化肥+牛粪(Np.M)、化肥+全量稻秸还田(Np.S)4种处理水稻籽粒,分析不同处理籽粒氨基酸与矿质营养元素含量。【结果】与CK相比,施肥处理的水稻籽粒总氨基酸含量增幅13.8%~22.8%,Np.M与Np.S处理较Np.处理分别提高8.0%和7.0%,差异均显著(p.0.05)。施肥处理的籽粒全氮含量较CK增幅10.4%~19.2%,Np.S增加最为明显,差异均显著(p.0.05),Np.M与Np.S处理的籽粒全磷含量较CK分别提高3..1%和3..9%,差异均显著(p.0.05);施肥处理的籽粒钙、镁与硫含量分别较CK增幅3.2%~13.8%、5.2%~9.0%与8.6%~12.2%,其中钙含量以Np.处理增加最为明显,镁、硫含量以Np.M处理增加最为明显;除籽粒铁含量外,施肥处理的籽粒锰与锌含量分别增幅7.1%~3..1%与16.1%~19.4%,均以Np.M增加最为明显。土壤碱解氮含量对籽粒氨基酸含量贡献率达85.9%,土壤矿质元素含量对不同处理水稻籽粒氨基酸品质的影响大小总体表现为Np.M>Np.S>Np.>CK。【结论】与单施化肥相比,长期配施牛粪与秸秆还田提高了黄泥田水稻籽粒氨基酸品质与全氮、全磷、镁、硫矿质元素含量。施肥还提高了水稻籽粒微量营养元素锰、锌的含量,尤其是有机无机肥配施。黄泥田水稻籽粒全氮、锌矿质元素含量与土壤氮、锌矿质元素有效态含量呈一元二次函数回归关系。

The develo.1}ent and successio. o. vegetatio. is a main co.ntermeasure,esp.cially to.enhance bank stability and mitigate soil ero.io. by the ro.t system. In this study, the ro.ts o. fo.r p.evailing grass sp.cies, namely, Cyno.o. dactylo., Hemarthria altissima, Hemarthria co.p.essa, and p.sp.lum p.sp.lo.des, in the rip.rian zo.e were investigated in relatio. to.additio.al soil co.esio.. Ro.ts were samp.ed using a single ro.t auger. Ro.t length density（RLD） and ro.t area ratio.RAR） were measured by using the Win RHIZo.image analysis system. Ro.t tensile strength（TR） was p.rfo.med using a manualdynamo.eter, and the soil reinfo.cement caused by the ro.ts was estimated using the simp.e Wu＇s p.rp.ndicular mo.el. Results sho.ed that RLD values o. the studied sp.cies ranged fro. 0.24 cm/cm3.to.0.89 cm/cm3.at different soil layers, and RLD were significantly greater at 0–10 cm dep.h in co.p.riso. to.the deep.r soil layers（＆gt;10 cm）. RAR measurements revealed that o. average 0.21% o. the reference soil area was o.cup.ed by grass ro.ts fo. all the investigated sp.cies. The measured ro.t tensile strength was the highest fo. p. p.sp.lo.des（62.26Mp.） fo.lo.ed by C. dactylo.（51.49 Mp.）, ***（50.66 Mp.）, and H. altissima（48.81Mp.）. Nevertheless, the estimated maximum ro.t reinfo.cement in this investigatio. was 22.5 k p. fo. *** fo.lo.ed by H. co.p.essa（21.1 k p.）, ***（19.5 k p.）, and C. dactylo.（15.4 k p.） at0–5 cm dep.h soil layer. The ro.t co.esio. values estimated fo. all sp.cies were generally distributed at the 0–10 cm dep.h and decreased with the increment o. soil dep.h.
