Elevation of serum apelin-13 associated with proliferative diabetic retinopathy in type 2 diabetic patients

AIM: To compare apelin-13, a ligand of G-proteincoupled receptor which has been shown to be involved in retinal angiogenesis, and vascular endothelial growth factor(VEGF) serum levels in type 2 diabetes mellitus(T2DM) with or without retinopathy, and to investigate the relationship between the serum concentration of apelin-13 and diabetes ***: Sixty-nine patients with T2 DM were *** the 69 patients, 16 had proliferative diabetic retinopathy(PDR group), 23 had non-PDR(NPDR group)and 30 had no retinopathy(T2DM group). Subjects’ information, including demographics, medical history,and use of medications were recorded. Their serum samples were collected for measuring the levels of C-reactive protein(CRP), serum lipid and glycosylated hemoglobin. Apelin-13 and VEGF serum levels were measured by enzyme-linked immunosorbent ***-Wallis test and one-way ANOVA were used to compare the differences among these groups. chi-square test was used to assess categorical *** between variables were investigated by Spearman rho correlation test and stepwise regression analysis. All statistical analyses were performed through SPSS 17.0 ***: Sex, age, body mass index(BMI), blood pressure, CRP, hemoglobin A1c(Hb A1c) have no significantly difference in the three groups. Serum level of apelin-13 was significantly elevated in PDR group as compared with T2 DM group(P =0.041). Differences of VEGF serum concentration in the three groups were statistically significant(P =0.007, P =0.007 and P <0.001,respectively). Spearman rho correlation test showed that serum apelin-13 was positively correlated with BMI,serum triglycerides, VEGF, but not with age, duration of diabetes, blood pressure, CRP, Hb A1 c and total-cholesterol. Stepwise regression analysis showed that BMI also significantly associated with serum apelin-13(P =0.002), while VEGF and serum triglycerides were irrelevant. Objective Obesity is recognized as a significant risk factor for diabetes and hypertension. The present study aimed to examine the associations between adults' obesity risk and childhood and parental obesity. Methods A total of 204 children aged 6-17 years were recruited in 2002 with an average follow-up period of 13.2 years. Height and body weight were measured by trained staffs. Overweight and obesity were defined based on the chinese standard for children and adults. T-test, analysis of variance, and chi-square analysis were used for single factor analysis. Multiple linear and logistic regression analyses were used to perform multifactor analysis. Results The percentage of non-obese children who grew up to be non-obese adults was 62.6%, and that of obese children who grew up to be obese adults was 80.0%. There was a significant association between childhood body mass index(BMI) and adulthood BMI with a β regression coefficient of 3.76 [95% confidence interval(CI): 1.36-6.16], and between childhood obesity and adulthood obesity with an odds ratio of 5.76(95% CI: 1.37-24.34). There was no statistical difference between parental obesity at baseline and children's adulthood obesity, after adjustment of confounders. Male participants and those aged 10.0-13.0 years had a higher risk of adulthood obesity with odds ratios of 2.50(95% CI: 1.12-5.26) and 3.62(95% CI: 1.17-11.24), respectively. Conclusion childhood obesity is an important predictor of adulthood obesity.

Objective： To investigate the proteasome inhibitor MG132-induced apoptosis pathway in HL-60 cells and the role of allogeneic mixed lymphocyte reaction. Methods： Cell apoptosis was analyzed by flow cytometry. The expressions of p21 protein, p27 protein and p53 protein in HL-60 ceils treated with MG132 were measured by Western blot. The proliferation of, peripheral blood mononuclear cells （PBMNCs） after treatment with 75 Gy irradiated HL-60 cells treated with MG132 was measured with CCK-8. Results： High-dose MG132 induced apoptosis in HL-60 cells. No significant change was observed in MG132-induced apoptosis after inhibiting caspase-8 and caspase-9 pathway. The expressions of p21 protein and p27 protein increased in MG132-induced apoptosis. HL-60 cells treated with low-dose MG132 improved the proliferation of PBMNCs from healthy volunteers. Conclusion： High-dose MG132 induced apoptosis and directly killed HL-60 ceils. MG132 induced apoptosis in a caspase-8- and caspase-9-independent pathway, p21 protein and p27 protein were involved in MG132-induced apoptosis in HL-60 cells. HL-60 cells treated with Low-dose MG132 improved the effect of promoting the proliferation of PBMNCs from healthy volunteers.

Severe fever with thrombocytopenia syndrome(SFTS)virus(SFTSV)is an emerging tick-borne virus with high fatality and an expanding ***,effective anti-SFTSV intervention remains ***(T-705)was recently reported to show in vitro and in animal model antiviral efficacy against ***,we conducted a single-blind,randomized controlled trial to assess the efficacy and safety of T-705 in treating SFTS(chinese Clinical Trial Registry website,number chiCTR1900023350).From May to August 2018,laboratory-confirmed SFTS patients were recruited from a designated hospital and randomly assigned to receive oral T-705 in combination with supportive care or supportive care *** outcome occurred in 9.5%(7/74)of T-705 treated patients and 18.3%(13/71)of controls(odds ratio,0.466,95%Cl,0.174-1.247).Cox regression showed a significant reduction in case fatality rate(CFR)with an adjusted hazard ratio of 0.366(95%Cl,0.142-0.944).Among the low-viral load subgroup(RT-PCR cycle threshold>26),T-705 treatment significantly reduced CFR from 11.5 to 1.6%(P=0.029),while no between-arm difference was observed in the high-viral load subgroup(RT-PCR cycle threshold<26).The T-705-treated group showed shorter viral clearance,lower incidence of hemorrhagic signs,and faster recovery of laboratory abnormities compared with the *** in vitro and animal experiments demonstrated that the antiviral efficacies of T-705 were proportionally induced by SFTSV mutation rates,particularly from two transition mutation *** mutation analyses on T-705-treated serum samples disclosed a partially consistent mutagenesis pattern as those of the in vitro or animal experiments in reducing the SFTSV viral loads,further supporting the anti-SFTSV effect of T-705,especially for the low-viral loads.
