Inhibition of tumor necrosis factor-α reduces alveolar septal cell apoptosis in passive smoking rats

Background Recent studies have revealed that lung cell apoptosis plays an important role in pathogenesis of cigarette-induced chronic obstructive pulmonary disease （COPD）. Tumor necrosis factor alpha （TNF-α） is one of the most important cytokines which are involved in COPD. This study aimed at investigating the influence of its inhibitor, recombinant human necrosis factor-alpha receptor II：lgG Fc fusion protein （rhTNFR：Fc） on alveolar septal cell apoptosis in passive smoking rats. Methods Forty-eight rats were randomly divided into a normal control group, a passive smoking group, an rhTNFR：Fc intervention group and a sham intervention group. The passive smoking rats were treated by exposure to cigarette smoking daily for 80 days. After smoking for one month the rhTNFR：Fc intervention group was treated with rhTNFR：Fc by subcutaneous injection, the sham intervention group injected subcutaneously with a neutral preparation （normal saline 0.1 ml, manicol 0.8 ml, cane sugar 0.2 mg, Tris 0.024 mg as a control. Lung function was determined and the levels of TNF-a in serum and broncho-alveolar lavage fluid （BALF） were measured with enzyme-linked immunosorbnent assay （ELISA）. Lung tissue sections stained by hematoxylin and eosin （HE） were observed for study of morphological alternations. Mean linear intercept （MLI） and mean alveolar numbers （MAN） were measured and the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling （TUNEL） method was carried out to determine the percentage of positive cells and distribution of apoptotic cells. Results Increased MLI and decreased MAN were found in the passive smoking group compared with both the normal control group and the rhTNFR：Fc intervention group （P〈0.05）. Forced expiratory volume in 0.3 second （FEVo.3）/forced vital capacity （FVC） and peak expiratory flow （PEF） were lower in the passive smoking group than that in the normal control group （P〈0.05）. 目的探讨重组人Ⅱ型肿瘤坏死因子受体-抗体融合蛋白(rhTNFR:Fc)对大鼠慢性阻塞性肺疾病(COPD)模型肺功能的影响。方法 48只大鼠随机分为正常对照组、COPD 组、rhTNFR:Fc 干预组(简称干预组)及假干预组。单纯吸烟法建立 COPD 模型,干预组皮下注射 rhTNFR:Fc 进行干预,假干预组皮下注射空白模拟制剂。酶联免疫吸附法(ELISA)测定各组血清和支气管肺泡灌洗液(BALF)中的肿瘤坏死因子α(TNF-α)浓度。肺组织切片行苏木精-伊红染色观察形态学改变,定量测定肺平均内衬间隔和平均肺泡数,用小动物肺功能仪测定系统测定肺功能。结果 COPD 组0.3秒用力呼气容积占用力肺活量比值(FEV_(0.3)/FVC)和呼气峰流速(PEF)[(65.1±8.4)%和(18.8±1.6)ml/s]显著低于正常对照组[(85.6±5.9)%和(47.2±7.3)ml/s]和干预组[(77.7±2.7)%和(38.2±3.3)ml/s],假干预组 FEV_(0.3)/FVC 和 PEF[(65.4±9.8)%和(19.0±1.9)ml/s]显著低于干预组。COPD 组血清中TNF-α浓度[(118±34)ng/L]显著高于正常组[(74±16)ng/L]和干预组[(79±14)ng/L],假干预组血清中 TNF-α浓度[(120±39)ng/L]显著高于干预组;COPD 组 BALF 中TNF-α浓度[(155±28)ng/L]显著高于正常对照组[(79±28)ng/L]。COPD 组肺平均内衬间隔[(77±29)×10^(-6)m/个]显著高于正常对照组[(44±7)×10^(-6)m/个],其平均肺泡数[(252±97)×10~6个/m^2]显著低于正常对照组[(393±24)×10~6个/m^2]和干预组[(379±33)×10~6个/m^2]。假干预组平均肺泡数[(213±99)×10~6个/m^2]显著低于干预组[(379±33)×10~6个/m^2]。COPD 组+假干预组血清和 BALF 中 TNF-α浓度与 PEF 和 FEV_(0.3)/FVC 均呈显著负相关。结论 TNF-α与吸烟大鼠的 COPD 形成有关,并影响其肺功能;rhTNFR:Fc 对延缓肺功能损害具有一定的作用。

The Advanced Space-based Solar Observatory(ASO-S)is a mission proposed for the 25 th solar maximum by the Chinese solar *** scientific objectives are to study the relationships between the solar magnetic field,solar flares and coronal mass ejections(CMEs).Three payloads are deployed:the Full-disk vector Magneto Graph(FMG),the Lyman-αSolar Telescope(LST)and the Hard X-ray Imager(HXI).ASO-S will perform the first simultaneous observations of the photospheric vector magnetic field,non-thermal imaging of solar flares,and the initiation and early propagation of CMEs on a single ***-S is scheduled to be launched into a 720 km Sun-synchronous orbit in *** paper presents an overview of the mission till the end of Phase-B and the beginning of Phase-C.

AIM:To report a one-year clinical outcomes of low-dose laser cycloplasty(LCP)among malignant glaucoma ***:In this prospective,multicenter,noncomparative clinical study,participants with malignant glaucoma were recruited and underwent LCP at eight ophthalmic centers in *** were followed up at 1wk,1,3,6,and *** pressure(IOP),number of glaucoma medications,anterior chamber depth(ACD),and complications were *** success was defined as the reformation of the anterior chamber based on slit-lamp *** was defined by the presence of a shallow orﬂat anterior chamber after initial recovery from ***:A total of 34 eyes received *** IOP and medications decreased from 36.1±11.5 mm Hg with 3.3±1.5 glaucoma medications pre-treatment to 20.9±9.8 mm Hg(P<0.001)with 2.9±1.6 medications(P=0.046)at 1d,and 17.4±6.7 mm Hg(P<0.001)with 1.3±1.7 medications(P<0.001)at *** ACD increased from 1.1±0.8 mm at baseline to 1.7±1.0 mm and to 2.0±0.5 mm at 1d and 12mo,respectively.A total of 32(94.1%)eyes achieved initial anatomical *** follow-up,2(5.9%)eyes failed and 8(23.5%)eyes relapsed,yielding a 12-month anatomical success rate of 64.3%.Complications including anterior synechia(8.82%),choroidal/ciliary detachment(5.88%)and hypopyon(2.94%)were observed within ***:LCP is simple,safe,and effective in reforming the anterior chamber in malignant glaucoma.

Cholangiocarcinoma refers to malignant tumors that develop in epithelial lining of biliary system, and it is divided into two categories according to tumor location, intrahepatic cholangiocarcinoma （ICC） and extrahepatic cholangiocarcinoma （ECC）. ICC occurs from the epithelial cells of the intrahepatic bile duct, its branches and interlobular biliary tree; and ECC is divided into hilar cholangiocarcinoma and distal cholangiocarcinoma by the circumscription at the confluence of cystic duct and the common hepatic duct.

As one of the three payloads for the Advanced Space-based Solar Observatory(ASO-S)mission,the Lyman-alpha(Lyα)Solar Telescope(LST)is composed of three instruments:a Solar Corona Imager(SCI),a LyαSolar Disk Imager(SDI)and a full-disk White-light Solar Telescope(WST).When working in-orbit,LST will simultaneously perform high-resolution imaging observations of all regions from the solar disk to the inner corona up to 2.5 R⊙(R⊙stands for the mean solar radius)with a spatial resolution of 4.8′′and 1.2′′for coronal and disk observations,respectively,and a temporal resolution of 30–120 s and 1–120 s for coronal and disk observations,*** maximum exposure time can be up to20 s due to precise pointing and image stabilization *** the three telescopes of LST,SCI is a dual-waveband coronagraph simultaneously and independently observing the inner corona in the HI Lyα(121.6±10 nm)line and white light(WL)(700±40 nm)wavebands by using a narrowband Lyαbeam splitter and has a field of view(FOV)from 1.1 to 2.5 R⊙.The stray-light suppression level can attain<10^-6 B⊙(B⊙is the mean brightness of the solar disk)at 1.1 R⊙and≤5×10^-8 B⊙at 2.5 R⊙.SDI and WST are solar disk imagers working in the Lyαline and 360.0 nm wavebands,respectively,which adopt an off-axis two-mirror reflective structure with an FOV up to 1.2 R⊙,covering the inner coronal edge area and relating to coronal *** present the up-to-date design for the LST payload.

The second(O2)observational campaign of gravitational waves(GWs)organized by the LIGO/Virgo Collaborations has led to several breakthroughs such as the detection of GW signals from merger systems involving black holes or neutrons *** O2,14 GW alerts were sent to the astronomical community with sky regions mostly covering over hundreds of square *** them,six were finally confirmed as real astrophysical *** 2013,a new set of ground-based robotic telescopes called Ground-based Wide Angle Camera system(GWAC)project and its pathfinder mini-GWAC has been developed to contribute to the various challenges of multi-messenger and time domain *** GWAC system is built up in the framework of the ground-segment system of the SVOM mission that will be devoted to the study of the multi-wavelength transient sky in the next *** O2,only the mini-GWAC telescope network was fully *** to the wide field of view and fast automatic follow-up capabilities of the mini-GWAC telescopes,they were adept to efficiently cover the sky localization areas of GW event *** this paper,we present the mini-GWAC pipeline we have set up to respond to GW alerts and we report our optical follow-up observations of eight GW alerts detected during the O2 *** observations provided the largest coverage of the GW localization areas with a short latency made by any optical *** found tens of optical transient candidates in our images,but none of those could be securely associated with any confirmed black hole-black hole merger *** on this first experience and the near future technical improvements of our network system,we will be more competitive in detecting the optical counterparts from some GW events that will be identified during the upcoming O3 run,especially those emerging from binary neutron star mergers.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

目的探讨香烟暴露对大鼠肺组织及人血管内皮细胞基质金属蛋白酶9(MMP-9)和金属蛋白酶组织抑制因子1(TIMP-1)表达的影响。方法采用烟熏法建立慢性阻塞性肺疾病(COPD)模型,记录模型组(M组)及对照组(N组)大鼠的一般观察指标、肺功能指标、肺组织病理学指标、支气管肺泡灌洗液(BALF)中白细胞计数和分类及肺组织中MMP-9和TIMP-1的表达。用Western blot检测不同浓度香烟提取物干预后人血管内皮细胞中MMP-9和TIMP-1表达的变化趋势。结果M组大鼠出现毛发脱落、反应迟钝、呼吸频率加快等变化。M组与N组比较,大鼠BALF中炎性细胞计数、肺组织形态学指标、肺功能指标、肺组织MMP-9和TIMP-1的表达及MMP-9/TIMP-1的差异均有统计学意义(P<0.05)。随着香烟提取物浓度增高,人血管内皮细胞中MMP-9和TIMP-1的表达逐渐增强。结论吸烟是COPD的一个重要致病因素。肺内MMP-9/TIMP-1的失衡及血管内皮细胞中MMP-9和TIMP-1的表达可能在COPD的发生发展中起一定作用。
