Chiglitazar(Carfloglitazar)is a novel peroxisome proliferator-activated receptor(PPAR)pan-agonist that has shown promising effects on glycemic control and lipid regulation in patients with type 2 *** this randomized phase 3 trial,we compared the efficacy and safety of chiglitazar with sitagliptin in patients with type 2 diabetes who had insufficient glycemic control despite a strict diet and exercise *** patients were randomized(1:1:1)to receive chiglitazar 32 mg(n=245),chiglitazar 48 mg(n=246),or sitagliptin 100 mg(n=248)once daily for 24 *** primary endpoint was the change in glycosylated hemoglobin A_(1C)(HbA_(1c))from baseline at week 24 with the non-inferiority of chiglitazar over *** chiglitazar and sitagliptin significantly reduced HbA1c at week 24 with values of-1.40%,-1.47%,and-1.39%for chiglitazar 32 mg,chiglitazar 48 mg,and sitagliptin 100 mg,*** 32 and 48 mg were both non-inferior to sitagliptin 100 mg,with mean differences of-0.04%(95%confidential interval(Cl)-0.22 to 0.15)and-0.08%(95%Cl-0.27 to 0.10),*** with sitagliptin,greater reduction in fasting and 2-h postprandial plasma glucose and fasting insulin was observed with *** adverse event rates were similar between the groups.A small increase in mild edema in the chiglitazar 48 mg group and slight weight gain in both chiglitazar groups were *** overall results demonstrated that chiglitazar possesses good efficacy and safety profile in patients with type 2 diabetes inadequately controlled with lifestyle interventions,thereby providing adequate supporting evidence for using this PPAR pan-agonist as a treatment option for type 2 diabetes.

通过红外透过成像研究了 Cd/Zn 气氛退火过程中 Cd0．9 Zn0．1 Te∶In 晶体内 Te 夹杂的密度及尺寸分布的演变.结果发现,Cd/Zn 气氛退火前,晶体中的 Te 夹杂密度分布比较均匀；退火后,晶体高温端近表面区域的 Te 夹杂密度较退火前提高了1个数量级,而晶体内部的 Te 夹杂密度则较退火前降低了1个数量级,且其密度沿温度梯度方向逐渐增加.退火前,晶体表面和内部的 Te 夹杂的直径主要分布在1~25μm；退火后,在晶体表面,直径＜45μm 的 Te 夹杂密度显著增大；而在晶体内部,直径＜5μm 和＞25μm的 Te 夹杂密度显著增大.导致这些现象的原因是退火过程中,Te 夹杂沿着温度梯度方向不断向晶体表面迁移,在迁移过程中尺寸相近的 Te 夹杂通过合并长大,尺寸相差较大的 Te 夹杂则以 Ostwald 熟化方式长大,并使小尺寸的 Te 夹杂更小.但由于熟化不充分,在 Ostwald 熟化长大过程中留下了很多尺寸＜5μm 的 Te 夹杂颗粒.

目的:构建含小鼠GITRL基因原核表达载体,经转化***以表达GITRL融合蛋白,纯化蛋白并制备相应的抗血清。方法:将GITRL-pMD18-T重组质粒经EcoRⅠ和SalⅠ双酶切回收目的基因,与原核表达载体PET32a连接,应用电转法转化BL21(DE3)菌。阳性克隆经鉴定后用IPTG诱导GITRL蛋白表达,经Ni+-NTA层析柱纯化融合蛋白,通过SDS-PAGE、West-ernblot进行特异性鉴定,CD4+CD25+T细胞免疫抑制试验验证其生物学功能。采用弗氏完全佐剂常规免疫方案,制备抗GITRL抗血清,抗血清的效价和特异性测定采用双向琼脂免疫扩散法。结果:经酶切鉴定和测序分析证实原核表达质粒构建正确,SDS-PAGE和Westernblot结果显示,在40kD处呈现单一蛋白条带,该蛋白具有阻断CD4+CD25+T细胞免疫抑制功能的作用,双向免疫扩散结果显示兔抗小鼠GITRL抗血清效价为1∶16。结论:成功构建GITRL原核表达质粒,制备和纯化的GITRL融合蛋白具有较好的纯度和生物学功能,制备并获得特异性抗血清,可用于GITRL融合蛋白生物学活性的进一步研究。

目的:观察益气温阳养血方治疗乳腺癌术后患者(阳气亏虚证)的临床疗效。方法:将32例乳腺癌术后患者随机分为治疗组和对照组各16例,两组均接受相同方案化疗,治疗组在对照组基础上内服益气温阳养血方。比较两组治疗前后临床症状体征改善情况及临床疗效。结果:治疗组总有效率为81.3%,高于对照组的31.3%(P0.05);组内比较,治疗组治疗后KPS评分和FACT-B量表总分均高于治疗前(P0.05);治疗后KPS评分和FACT-B量表总分治疗组均高于对照组(P0.05)。结论:益气温阳养血方辅助化疗治疗乳腺癌术后阳气亏虚证患者有较好的疗效,可有效缓解患者临床症状,提高患者生存质量。

Laparoscopic hepatectomy for liver neoplasms in 15 cases

Objective： The aim of our study was to retrospectively analyze 15 patients＇ clinical materials with laparoscopic resection of liver neoplasms. Methods： From December 2007, a total of 15 patients with liver neoplasms were performed with laparoscopic hepatectomy, and their clinical materials, perioperative dates, postoperative complications, postoperative recovery and short-term curative effects were analyzed and summarized respectively. Results： Laparoscopic hepatectomy （LH） were performed in 15 patients, including 1 case underwent laparoscopic hepatic left Iobectomy, 1 case of left lateral hepatectomy, 13 cases of partial liver resection. Fourteen cases of total laparoscopic liver resections for liver neoplasms, 1 case of hand-assisted laparoscopic liver resection of the tumor, there was no conversion to open approach. Of the 15 patients with liver neoplasms, 13 cases of hepatic neoplasms with the maximum diameter was 8 cm× 8 cm × 9 cm, 9 cases of the borderline micro hepatocellular carcinoma （MHCC） with the diameter not more than 2 cm, 3 cases of hepatic benign tumor. The mean operation time was （120 ± 30） min, and the intraoperative average hemorrhage was 100 mL, beginning to eat and get out of bed following 1-2 days of operation. The average postoperative hospitalization was 8 days, WBC, ALl＇, AST, albumin, bilirubin returned to normal after one week of operation. There were no postoperative complications such as hemorrhage, bile leakage or air embolism etc. Twelve patients with HCC were confirmed by postoperative pathology, 1 case of liver smooth muscle lipoma, 2 case of hepatic hemangioma. By one year of followed-up in 12 cases of HCC, the longest survival was 38 months, and no recurrence or death, 1 year survival rate was 100%. Conclusion： Among the choice of cases, the advantages of LH for liver neoplasms compared with open surgery were less trauma, faster recovery and less blood loss. it is safe and effective for choosing a reasonable surgical indication, es

Background: To date, there is no effective medicine to treat coronavirus disease 2019 (COVID-19), and the antiviral efficacy of arbidol in the treatment for COVID-19 remained equivocal and controversial. The purpose of this study was to evaluate the efficacy and safety of arbidol tablets in the treatment of ***: This was a prospective, open-label, controlled and multicenter investigator-initiated trial involving adult patients with confirmed severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Patients were stratified 1:2 to either standard-of-care (SOC) or SOC plus arbidol tablets (oral administration of 200 mg per time, three times a day for 14 days). The primary endpoint was negative conversion of SARS-CoV-2 within the first week. The rates and 95% confidential intervals were calculated for each ***: A total of 99 patients with laboratory-confirmed SARS-CoV-2 infection were enrolled;66 were assigned to the SOC plus arbidol tablets group, and 33 to the SOC group. The negative conversion rate of SARS-CoV-2 within the first week in patients receiving arbidol tablets was significantly higher than that of the SOC group (70.3% [45/64]vs. 42.4% [14/33];difference of conversion rate 27.9%;95% confidence interval [CI], 7.7%-48.1%;P=0.008). Compared to those in the SOC group, patients receiving arbidol tablets had a shorter duration of clinical recovery (median 7.0 daysvs. 12.0 days;hazard ratio [HR]: 1.877, 95% CI: 1.151-3.060,P=0.006), symptom of fever (median 3.0 daysvs. 12.0 days;HR: 18.990, 95% CI: 5.350-67.410,P 0.05). The most common adverse event in the arbidol tablets group was the elevation of transaminase (5/200, 2.5%), and no one withdrew from the study due to adverse events or disease progression.C

目的:获得重组人活化诱导的肿瘤坏死因子超家族配体胞外区蛋白(sAITRL),分析AITRL蛋白的生物学功能。方法:从全长质粒AITRL-pMD18-T中扩增sAITRL cDNA,亚克隆至表达载体pQE30;将重组质粒转化至***15,IPTG诱导蛋白表达;表达蛋白用Ni2+-IMAC柱纯化、复性并Western blot鉴定,流式细胞仪检测sAITRL重组蛋白的结合活性,增殖试验分析sAITRL的生物学活性。结果:成功构建了sAITRL-pQE30原核表达载体;表达相对分子质量约15kD的重组蛋白;sAITRL能与活化淋巴细胞上的天然受体AITR结合,低浓度的sAITRL能促进淋巴细胞的增殖,而在高浓度下则抑制了淋巴细胞的增殖。结论:成功表达了具有生物学活性的sAITRL重组蛋白,AITR-AITRL相互作用在调节淋巴细胞增殖过程中起重要作用。

为丰富即食水产品种类,探索真空油炸技术,以单冻小龙虾虾尾为原料,通过低温真空油炸工艺制备小龙虾虾尾酥产品。在单因素试验的基础上,以水分、油脂含量、酥脆性、硬度、丙烯酰胺含量为指标,选择油炸温度、油炸时间、油料质量比和脱油时间为变量,利用正交试验设计对真空油炸工艺进行优化。结果表明,真空油炸工艺的最优条件:真空度0.09 MPa,油炸温度95℃、油炸时间120 min、油料质量比3.0∶1.0、脱油时间25 min,此条件下,小龙虾虾尾酥水分含量为(2.65±0.11)%,粗脂肪含量为(25.66±1.88)%,硬度为(443.21±42.36)g,酥脆性与常压油炸小龙虾虾尾酥无显著差别,丙烯酰胺含量为(11.2±0.17)μg/kg。相比于常压油炸,低温真空油炸工艺不仅减少营养损失,而且降低了油脂及丙烯酰胺含量,为提升小龙虾油炸制品品质提供理论依据和科学指导。
